Abstract

α-glucosidase memiliki peran penting dalam metabolisme karbohidrat pada tubuh manusia dengan bekerja memutus rantai oligosakarida menjadi gula sederhana seperti monosakarida.Penghambatan aktivitas α-glucosidase menjadi salah satu mekanisme untuk mengendalikan kadar glukosa dalam darah penderita diabetes. Obat herbal sebagai inhibitor α-glucosidase bersumber dari bahan alami laut khususnya mangrove menarik perhatian. Buah pedada Sonneratia alba telah diketahui mengandung senyawa fitokimia yang berpotensi sebagai antidiabetes, akan tetapi belum pernah dilaporkan kandungan senyawa bioaktif yang berperan sebagai inhibitor α-glucosidase. Kelimpahan daun pada tanaman mangrove lebih tinggi daripada buah sehingga pada penelitian ini menggunakan daun sebagai penghambat aktivitas α-glucosidase. Tujuan dari penelitian ini adalah untuk menganalisa profil senyawa fitokimia flavonoid dari ekstrak etil asetat daun pedada S. alba, dan untuk menentukan senyawa fitokimia tersebut yang berperan sebagai inhibitor α-glucosidase secara in silico melalui penambatan molekuler. Hasil penelitian menunjukkan tiga senyawa fitokimia flavonoid dari ekstrak etil asetat daun pedada S. alba seperti luteolin, apigenin, dan diosmetin telah diidentifikasi. Menurut kaidah Lipinski, analisa druglikeness dan toksisitas, maka semua senyawa tersebut aman untuk dikonsumsi, Hasil penambatan molekuler menunjukkan energi ikatan yang dihasilkan dari interaksi reseptor dan ligan uji lebih rendah daripada energi ikatan dari reseptor dan ligan kontrol maupun alami. Sehingga dapat disimpulkan bahwa luteolin, apigenin dan diosmetin dari ekstrak etil asetat daun pedada berpotensi sebagai inhibitor α-glucosidase. α-glucosidase plays an important role in carbohydrate metabolism in the human body by breakdown oligosaccharides into monosaccharides. The inhibition of α-glucosidase activity is one of the mechanisms for controlling blood glucose levels in diabetes patients. Herbal drug as an α-glucosidase inhibitor from marine resources, especially mangroves is attracting interest. The fruit of Sonneratia alba contains phytochemical compounds potent as antidiabetic but no report related to their bioactive compound as inhibitor α-glucosidase. The abundance of leaves is higher than the fruit; therefore, in this study, the leaves will be used as α-glucosidase inhibitors. This study aimed to profile bioactive compounds of ethyl acetate extract of pedada S. alba leaves and to evaluate phytochemical compounds of S. alba leaves as α-glucosidase inhibitors using molecular docking in silico. The results showed that three phytochemical compounds of ethyl acetate extract of S. alba leaves, such as luteolin, apigenin, and diosmetin have been identified. According to the Lipinski’s rule, drug likeness, and toxicity test, all compounds are safe to consume. Docking studies showed that binding energy between receptor and phytochemical compounds is lower than binding energy between receptor and ligand control. In conclusion, luteolin, apigenin and diosmetin of ethyl acetate extract of pedada S. alba leaves have potent as α-glucosidase inhibitors.

Abstract

Sesbania grandiflora is a medicinal plant that has been used by many Indonesian people. They have the ability as antibacterial because they contain bioactive compounds that inhibit bacterial growth. Sesbania grandiflora also contains other active compounds such as tannins, saponins, steroids, flavonoids, and terpenoids. This study aims to determine the antibacterial activity of extracts, n-hexane fraction, ethyl acetate fraction and water fraction of Sesbania grandiflora against Edwardsiella tarda using the well diffusion method in different concentration (75 ppm, 150 ppm dan 225 ppm), and knowing the active compounds of FTIR absorption bands. MIC (minimum inhibitory concentration) value of extract, n-hexane, ethyl acetate, and water fractions respectively was 0.51 %, 1.11 %, 0.97 %, dan 0.79 %. InIibitory zone diameter of the n-hexane fraction is most active against Edwardsiella tarda bacteria. The results of FTIR identification showed that n-hexane fraction contained flavonoids and terpenoids, as seen from absorption bands that function as antibacterial compounds.Keywords: antibacterial, Edwardsiella tarda, FTIR, Phytochemical Sesbania grandiflora.

Abstract

α-amylase has a pivotal role in catalyzing the cleavage of α-1,4-glycosidic bonds of polysaccharides to produce oligosaccharides. The inhibition of α-amylase delays the breakdown of carbohydrates, causing a reduction of blood glucose levels absorption in diabetes patients. The exploration of α-amylase inhibitors has attracted because society assumed that utilizing herbal medicine reduced the side effect of prescribed drugs. Mangrove from genus Ceriops have been used as antidiabetic, but the mechanism as α-amylase inhibitors has not been reported. Consumption of leaves extract of C.decandra reduced blood glucose level in diabetic rats, and triterpenoids have been identified from the leaves. With this in mind, this study aims to predict the molecular interactions between α-amylase (PDB ID: 4GQR) and the inhibitors, triterpenoid identified in C.decandra leaves, and to evaluate the potency of triterpenoid as α-amylase inhibitor. There are five triterpenoids identified in C.decandra leaves used as ligand tests, including lupenone, betulin, betulonic acid, betulinic acid, and lupeol. The descriptive method was applied in this investigation. This study was carried out from June to September 2022. Based on the molecular interactions, the binding affinity of triterpenoids was lower than the native ligand and control ligand. Lupenone, lupeol, betalonic acid, and betulinic acid inhibited α-amylase activity by non-competitive inhibition. It was predicted that betulin inhibited α-amylase activity through competitive inhibition.

Some previous studies reported phytochemical screening and bioactivity test of Sonneratia sp. as anti-diabetic (in vitro and in vivo analysis). However, phytochemical identification of S. alba leaves and inhibitory activity of α-glucosidase as anti-diabetic have not been reported. The purpose of this study are to identify phytochemical compounds of methanolic extracts S. alba leaves, and to predict inhibitory mechanisms of S. alba leaves against α-glucosidase through in silico analysis. In this study, the research method consists of two steps namely identification phytochemical compounds of methanolic extracts S. alba leaves, and prediction inhibitory activities of phytochemical compounds of methanolic extracts S. alba leaves against α-glucosidase by molecular docking (in silico) analysis. Methanolic extracts S. alba leaves contained orientin, vitexin, luteolin, oleanolic acid and reserpine. Reserpine was identified for the first time in S. alba leaves. Based on in silico analysis, binding energy of orientin, vitexin, luteolin, oleanolic acid, reserpine and acarbose (positive control) against α-glucosidase were -9,7; -9,7; -9,2; -8,6; -10,0 dan -8,3 kcal/mol. This result indicated that compounds inhibited α-glucosidase activities and can be considered as antidiabetic agent.

Abstract

Inflammatory Bowel Disease (IBD) is a chronic gastrointestinal disorder driven by oxidative stress and immune dysregulation, often exacerbated by prolonged non-steroidal anti-inflammatory drug (NSAID) use. Conventional therapies provide symptomatic relief but are limited by severe adverse effects, necessitating safer alternatives. Channa striata (snakehead fish) is a rich source of albumin, essential amino acids, and bioactive compounds with potential immunomodulatory and anti-inflammatory effects. This study investigates the therapeutic efficacy of purified Channa striata albumin extract in an indomethacin-induced IBD rat model. Albumin was isolated via ammonium sulfate precipitation and characterized by SDS-PAGE, antioxidant activity (DPPH assay), and amino acid profiling. Rats were treated with Channa striata albumin (100-300 mg/kg BW) for 14 days post-indomethacin induction. Compared to controls and diclofenac-treated groups, the extract significantly reduced malondialdehyde (MDA) levels (up to 90.69%), improved jejunal histoarchitecture, and enhanced occludin expression. Immunohistochemistry showed a marked reduction in CD4+T cell infiltration, indicating immunomodulatory activity. These findings establish Channa striata albumin as a potent natural antioxidant and immunomodulator, offering a safer alternative to NSAIDs for IBD management.

Abstract

Ikan tuna merupakan komoditas laut unggulan dengan kandungan protein tinggi. Salah satu bentuk diversifikasi olahan yang potensial adalah sate lilit. Saat ini, proses pematangan sate umumnya menggunakan metode tradisional seperti pembakaran arang, yang kurang higienis dan efisien. Oleh karena itu, diperlukan metode alternatif seperti pemanasan menggunakan gelombang elektromagnetik (microwave). Penelitian ini bertujuan untuk mengevaluasi pengaruh metode pemanasan (tradisional dan microwave) serta variasi waktu pemanasan (8, 10, dan 12 menit) terhadap karakteristik organoleptik sate lilit ikan tuna. Penelitian dilakukan secara eksperimental menggunakan Rancangan Acak Lengkap (RAL) dengan pola faktorial 2 × 3, mencakup dua faktor: metode pemanasan dan lama waktu pemanasan, masing-masing diulang tiga kali. Parameter yang diamati meliputi warna, aroma, rasa, dan tekstur, dinilai secara organoleptik oleh 30 panelis tidak terlatih menggunakan uji hedonik skala 5. Hasil analisis menunjukkan bahwa metode pemanasan dan lama pemanasan berpengaruh signifikan terhadap seluruh atribut organoleptik. Perlakuan terbaik diperoleh pada pemanasan microwave selama 10 menit, menghasilkan warna kuning kecokelatan menarik, aroma tuna yang kuat, rasa gurih seimbang, dan tekstur padat namun tidak kering. Hasil ini menunjukkan bahwa penggunaan microwave selama 10 menit merupakan metode yang efektif untuk menghasilkan sate lilit tuna berkualitas, serta dapat dijadikan acuan dalam pengembangan produk olahan ikan dengan teknologi pemanasan yang higienis dan efisien.   Tuna is a leading marine commodity known for its high protein content. One potential form of product diversification is sate lilit (minced fish satay). Currently, the cooking process for this product commonly uses traditional methods such as charcoal grilling, which tend to be less hygienic and inefficient. Therefore, an alternative method is needed, such as electromagnetic heating using a microwave. This study aims to evaluate the effects of heating methods (traditional and microwave) and different heating durations (8, 10, and 12 minutes) on the organoleptic characteristics of sate lilit made from tuna.cThe research was conducted experimentally using a Completely Randomized Design (CRD) with a 2 × 3 factorial pattern, consisting of two factors: heating method and heating duration, each replicated three times. The parameters observed included color, aroma, taste, and texture, which were evaluated organoleptically by 30 untrained panelists using a 5-point hedonic scale. Statistical analysis showed that both heating method and duration had a significant effect on all organoleptic parameters. The best treatment was microwave heating for 10 minutes, which resulted in an appealing golden-brown color, a strong tuna aroma, a balanced savory taste, and a firm yet moist texture. These findings indicate that microwave heating for 10 minutes is an effective method for producing high-quality tuna sate lilit and can serve as a reference for further development in fish-based product diversification using appropriate and hygienic heating technology.

Abstract

Desa Krenceng merupakan daerah yang banyak dialiri sungai yang digunakan untuk pengairan pertanian, tetapi juga berpotensi untuk budidaya perikanan. Hasil budidaya perikanan sebagian besar dijual segar dan masih sedikikit yang diolah menjadi produk turunan seperti nugget ikan, bakso ikan, abon ikan, dan lain-lain. Pada pengabdian kepada masyarakat (PKM) ini dipilih produk  nugget ikan karena prosesnya mudah, dapat dikonsumsi semua golongan umur, dan berpotensi untuk mengatasi stunting. Tujuan pelaksanaan PKM ini untuk mengajarkan kepada mitra (kelompok Pemberdayaan Kesejahteraan Keluarga (PKK) Desa Krenceng, Kabupaten Blitar) membuat nugget ikan dan melakukan assessment mutu nugget ikan menggunakan uji organoleptik. Metode yang digunakan dalam PKM adalah Participatory Action Learning System (PALS). Mitra berpartisipasi dalam pembuatan nugget ikan dan penilaian mutu secara organoleptik melalui uji hedonik, skoring atribut, dan perbandingan jamak. Uji hedonik menilai tingkat kesukaan, skoring atribut mengukur intensitas sensori, sedangkan perbandingan jamak membandingkan karakteristik produk dengan produk sejenis di pasar. Mitra berhasil membuat produk nugget ikan nila. Hasilnya penilaian sensori hedonik diperoleh bahwa tingkat kesukaan atribut rasa, aroma, tekstur, dan kerenyahan nugget komersil masih lebih disukai daripada nugget ikan hasil praktek.  Dari perbandingan jamak diperoleh bahwa atribut nugget ikan hasil praktek yang sama dengan nugget komersil hanya pada atribut warna, sedangkan atribut rasa, aroma, tekstur, dan kerenyahan berbeda dengan nugget komersil.  Dengan demikian mitra mampu membuat produk nugget ikan dan mampu menilai mutu nugget ikan secara sensori, meskipun nugget hasil praktek masih lebih rendah mutunya daripada nugget komersil.

Abstract

This study aims to determine: (1). The effect of electronic word of mouth (eWOM) on buying interest in Nu Skin Skincare products, (2). The effect of product quality on buying interest in Nu Skin Skincare products, (3). The effect of price on buying interest in Nu Skin Skincare products, (4). The effect of brand image on buying interest in Nu Skin Skincare products, (5). The effect of promotion on buying interest in Nu Skin Skincare products, (6) The effect of electronic word of mouth (eWOM), product quality, price, brand image and promotion on buying interest in Nu Skin Skincare products.This study used a purposive sampling method with a sample size of 100 people. The measurement scale uses a Likert scale. For data processing using SPSS version 13. The results of the study obtained the regression equation as follows: Y = -1.703 + 0.020X1 + 0.369X2 + 0.208X3 + 0.559X4 + e. Hypothesis testing using the t test shows that partially the electronic word of mouth (eWOM) variable (X1), price (X3), brand image (X4)) have no significant effect on purchase intention, while product quality (X2) and promotion (X5) have a positive and significant effect on purchase intention. Then through the F test it is known that the variables electronic word of mouth (eWOM) (X1), product quality (X2), price (X3), and promotion (X4) simultaneously have a positive and significant effect on purchase intention (Y)

Abstract

As a by-product of the pre-cooking phase, canned yellow fin tuna (Thunnus sp.) is processed into body juice. A bioactive peptide derived from an organism#39;s body juice was developed to create a functional food. Hence, the purpose of the research was to evaluate the biological activity of antioxidant bioactive peptides prepared from body juice. Three stages were conducted to determine the substrate-to-enzyme and enzyme-to-substrate ratios for the production of antioxidative bioactive peptides in a batch reactor. These stages include the production of antioxidative bioactive peptides in an automated EMR with a constant flux and residence time, as well as the production of bioactive peptides using ultrafiltration with membranes of 10, 5, 4, and 2 kDa. The process was conducted at a temperature of 50°C, pH level of 7, constant flux of 8.08 L/m 2 /h, enzyme concentration of 15%, and substrate concentration of 2%. Furthermore, the optimal treatment implemented an automated EMR and filtered through a membrane of size 4 kDa, generating an IC 50 value of 0.604 mg/mL for antioxidant activity. The LC–MS analysis results indicated that the bioactive peptide, which passed through a 4 kDa membrane, contained the amino acid phenylalanine. Therefore, it can be inferred that an automated EMR could potentially be applied to create bioactive peptides from the body juice of by-products of canned yellow fin tuna (Thunnus sp.) that exhibit antioxidant activity.
