Abstract

Diabetes Melitus is a group of metabolic diseases characterized by hyperglycemia due to lack of insulin secretion or insulin activity resistance. Chronic hyperglycemia in diabetes is associated with long-term damage, dysfunction, and failure of various organs, especially the kidneys, kidneys, nerves, heart and blood vessels. The purpose of this study was to analyze the activity of Adiponektin, superoxide dismutase, malondialdehyde, and glucose level in Streptozotocin-induced diabetic wistar rats. All wistar rats were divided into 4 groups, control (D0), induction group (D1), thitonia extract dose 100 mg / kg bw (D2) and catechin dose 10 mg/kg bw (D3) for 7 days. Model diabetes were used by induction of streptozotocin (Stz) at dosage 60 mg / kg bw intraperitoneally (ip) for 7 days. On the 8th day after the treatment, all groups of rats were sacrificed for blood samples for measured levels of superoxide dismutase and malondialdehyde and Adiponektine protein using Elisa technique, and measure of glucose level with calorimetry technique. Statistical analysis of data to examine differences in treatment and control was conducted by one way Anova test with a 95% confidence level (α=0.05). The results showed that there were differences in glucose levels (p = 0.020) and Adiponektin (p = 0.001) between the control and treatment groups, but there were no differences in level of MDA (p = 0.103) and SOD (p = 0.207 ) between groups. Base on the results of the study concluded that the administration of Tithonia Diversifolia leaf extract could reduce blood sugar and Adiponectin concentrations significantly (p≤0.05) in streptozotocin-induced diabetic on rats.

Abstract

The aims of this study is to evaluate molecular docking of flavonoid Coleus amboinicus (CA) extracts in transforming growth factor-1b and lowering MDA concentration on cisplatin-induced in Wistar rat. Eighteen male Wistar rats (Rattus norvegicus), 3 months of age with a body weight (BW) of 150-200 g, were allocated into three groups, with six animals per group. The control group received aquadest (P0), the treatment group were treated with single doses of cisplatin (5 mg/kg bw., ip) (P1) and  received 100 mg/kg bw of the CA extracts (P2) respectively for 7 days. Bloods collected for analysis of serum alkaline phospatase (AP), Blood Nitrogen Urea (BUN) and Malondialdehid (MDA) concentrations. The levels of Malondialdehid (MDA) concentrations were analysed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. All groups were sacrified for histopathology. Coleus amboinicus extract significantly decreased the level of AP, BUN and MDA concentrations compared to the control group (p<0.05). The level of MDA could be detected by its level significantly decreased in CA treatment group (p<0.05). Coleus amboinicus (CA) extract has a flavonoid as a marker compound of CA extract has stronger bind to the TGF-β1receptor than its of 3WA_601 ligand in silico analyzed. In histopathological examination showed that cisplatin-induced could alter severe multifocal hemorrhage, interstitial congestion, cell inflammatory, acute glomerular and tubular injury with necrotic cells. Immunohistochemical staining labeled with TGF-1β monoclonal antibodies (Mab) showed marked expression of brownish color aggregates on the surface of tubular epithelial cells and around glomerular mesangial cells in the CA treatment group. This study was concluded that CA extract is inhibited renal tissue injuries by lowering MDA and increasing TGF-1b expression on cisplatin-induced rats. Flavonoid as marker of CA extract has stronger bind to TGF-1b receptor by in silico.

Abstract

The purpose of this study is to determine effect of antiinflammation of Thitonia diversifolia (TD) extracts in histopathology liver and pancretic tissue on alloxan-induced in Wistar rat. Thirty two male Wistar rats (Rattus norvegicus) with a body weight (bw)150-200 g in 3 month of age, were allocated into four groups, with eight animals per group. The control group received normal saline (P0), positive control group  received CMC Na 0.01% (P1),  the treatment group were received 100 mg/kg bw of the TD extracts (P2) and received 100 mg/kg bw of the metformin (P3)  respectively for 7 days. Bloods serum collected for measuring of  blood glucose (BG). cholesterol and insulin (INS) concentrations. The levels of insulin (INS) concentrations were analysed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. All animal were sacrified for tissue staining with haematoxylin eosin. Thitonia diversifolia extract significantly decreased the level of blood glucoseand choelsterol concentrations compared to the postive control group (p<0.05). The level of INS could be detected by its level significantly increased in TD treatment group (p<0.05). Histopathological showed that induction of alloxan could effect severe multifocal degeneration, vacuolisation, cell inflammation, acute langerhan cells injury with destruction cells. Immunohistochemical staining labeled with IL-1β monoclonal antibodies (Mab) showed lowering expression of IL-1β reveal as brownish color aggregates on the langerhans cells in the TD treatment group. This study was concluded that TD extract is inhibited hepatic tissue injuries and decreased destruction in langerhan cells by decreasing IL-1β expression on alloxan-induced rats.

Abstract

Penelitian ini bertujuan untuk menganalisis gambaran maksroskopis ginjal mencit (Mus musculus) model urolithiasis dengan pemberian infusum seledri (Apium graveolens). Penelitian terbagi dalam empat perlakuan yaitu K 1 (akuades); K 2 (induser urolithiasis);  S 1 (infusum seledri 20%, setelah satu jam kemudian diberi induser urolithiasis); dan S 2 (infusum seledri 40%, setelah satu jam kemudian diberi induser urolithiasis). Induser urolithiasis adalah 0,75% etilen glikol dan 2% amonium klorida. Perlakuan dilakukan selama 10 hari, nekropsi dan analisis makroskopis ginjal dilakukan pada hari ke-11. Analisis makroskopis meliputi skoring gambaran makroskopis ginjal, panjang dan tebal ginjal, serta bobot ginjal mencit. Data hasil skoring gambaran makroskopis ginjal di analisis menggunakan uji Chi Kuadrat, sedangkan panjang dan tebal ginjal serta bobot ginjal dianalisis menggunakan uji Anova. Hasil skoring gambaran makroskopis ginjal kelompok S 1 dan S 2 terhadap kelompok K 2 menunjukan adanya perbedaan yang nyata (p>0,05). Hasil rata-rata panjang dan tebal ginjal kanan setiap kelompok adalah 0,80; 0,38(K 1); 1,08; 0,50(K 2); 1,05; 0,48 (S 1); dan 0,975; 0,47(S 2) dan rata-rata panjang dan tebal ginjal kiri setiap kelompok adalah 0,83; 0,36(K 1); 0,94; 0,44(K 2); 0,94; 0,44(S 1); 0,89; 0,39(S 2). Hasil rata-rata bobot ginjal setiap kelompok adalah 0,259 ± 0,50(K 1); 0,372 ± 0,44(K 2); 0,370 ± 0,41(S 1); dan 0,325 ± 0,079(S 2). Uji statistik ANOVA terhadap ukuran dan bobot ginjal mencit kelompok S 1 dan S 2 terhadap kelompok K 2 tidak berbeda nyata (p>0,05).

Abstract

Cisplatin adalah obat kemotarapi yang menyebabkan hepatotoksisitas.Cisplatin menyebabkan hepatotoksisitas dengan meningkatkan stres oksidatif yang memicu kematian sel. Tujuan penelitian ini adalah untuk menentukan gambaran histopatologi hepar tikus yang diinduksi cisplatin setelah pemberian ekstrak daun bangun-bangun (Coleus amboinicus).Sebanyak 18 ekor tikus Wistar dibagi menjadi 3 kelompok.Kelompok I (P0) sebagai kontrol negatif, kelompok II diinduksi cisplatin (5 mg/kg BB), kelompok III diberi ekstrak daun bangun-bangun (500 mg/kg BB) dan diinduksi cisplatin (5 mg/kg BB).Terapi dilakukan selama 7 hari dan induksi cisplatin dilakukan pada hari ke 4.Tikus dieutanasi dan hepar dikumpulkan untuk pemeriksaan histopatologi. Data analisis menggunakan uji Kruskal Wallis dan Mann Whitney Test. Hasil uji Kruskal Wllis menunjukkan bahwa skor histopatologi untuk kongesti (P<0,05), hemoragi (P<0,05), dan sel Kupffer (P<0,05). Uji Mann Whitney menunjukkan bahwa kongesti P0 dibandingkan dengan P1 (P<0,05), P0 dengan P2 (P<0,05), dan P1 dengan P2 (P<0,05). Selain itu hemoragi P0 dibandingkan dengan P1 (P<0,05), P0 dengan P2 (P<0,05), dan P1 dangan P2 (P>0,05). Namun, skor perbandingan sel Kupffer antara P0 dan P1 (P>0,05), P0 dengan P2 (P>0,05), dan P1 dengan P2 (P>0,05). Disimpulkan bahwa ekstrak daun bangun-bangun (Coleus amboinicus) mampu mencegah kongesti dan hemoragi pada hepar setelah diinduksi cisplatin.

Abstract

Probiotik adalah segala bentuk sediaan sel mikroba atau komponen sel mikroba yang mempunyai pengaruh menguntungkan bagi kesehatan dan kehidupan inangnya. Beberapa aditif pakan seperti hormon Antibiotik Growth Promoter (AGP) telah dilarang. Oleh karena itu, saat ini probiotik sering digunakan sebagai bahan tambahan pengganti antibiotik. Probiotik yang banyak digunakan adalah bakteri asam laktat karena dapat meningkatkan efisiensi pencernaan dan penyerapan nutrisi. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian probiotik kombinasi Bacillus coagulans dan Bacillus subtilis dengan dosis 2 ml, 4 ml, dan 6 ml/ekor/hari secara oral terhadap pertambahan bobot badan, konsumsi pakan, dan penurunan konversi pakan. ayam petelur. Penelitian ini dilakukan selama 2 minggu dengan menggunakan ayam pre layer umur 21 minggu sampai 23 minggu sebanyak 24 ekor, diacak menjadi 4 perlakuan dengan 6 ulangan dengan P0 sebagai kontrol, P1 = 2ml, P2 = 4 ml, dan P3 = 6 ml . Probiotik yang digunakan adalah probiotik kombinasi Bacillus coagulans dan Bacillus subtilis dengan konsentrasi 1x107 CFU/ml dalam bentuk cair. Kesimpulannya adalah pemberian oral kombinasi probiotik dengan Bacillus subtilis dan Bacillus coagulans dapat meningkatkan bobot badan, konsumsi pakan, dan menurunkan nilai konversi pakan ayam petelur dengan rendemen tertinggi bila diberikan dosis 4 ml.

Abstract

Tujuan penelitian ini adalah menganalisis daya anti radang ekstrak daun jintan (coleus amboinicus) yang diinduksi nephropathy akut. Ekstrak daun jintan diperoleh dari daun jintan (Coleus amboinicus) segar yang diekstraksi dengan metode maserasi etanol 96 % dan dianalisis secara kualitatif dengan metode kromatografi lapis tipis (KLT) untuk mengidentifikasi senyawa aktif yang terkandung di dalamnya. Aktivitas ekstrak daun jintan sebagai anti radang dilakukan dengan pemberian secara intraperitoneal ekstrak daun jintan (Coleus amboinicus) pada tikus putih galur Wistar terhadap induksi granuloma fibrosis pada tubulus ginjal yang diinduksi dengan senyawa Oxonic Acid dan Uric Acid (OA & UA). Pembentukan granuloma fibrosis diidentifikasi sebagai nephropahty akut dengan teknik sediaan histopatologi pengecatan Haematoxilin & Eosin (HE). Hasil penelitian menunjukkan bahwa ekstrak daun Jintan (coleus ambonicus) mempunyai kandungan kualitatif kumarin, saponin, minyak atsiri dan polifenol dengan teknik Kromatografi lapis Tipis (KLT). Induksi intra peritoneal Oxonic Acid dan Uric Acid menyebabkan perubahan makroskopis berupa deposisi kristal urea intraperitoneal dan keradangan pada sendi metatarsal. Secara histopatologis induksi OA & UA menimbulkan nephropathy akut yang ditandai dengan adanya lesi degenerasi, keradangan dan nekrosis pada tubulus dan glomerulus ginjal pada kelompok perlakuan. Pemberian ekstrak daun Jintan pada kelompok perlakuan menghambat terjadinya pembentukan nephropathy akut secara nyata (P<0.05) dibandingkan kontrol dan kelompok induksi.

Abstract

Hipokalsemia merupakan suatu gangguan metabolisme pada sapi perah dapat terjadi sebelum, sewaktu, atau beberapa jam sampai dengan 72 jam setelah melahirkan. Kejadian ini ditandai dengan penurunan yang tiba-tiba kadar calcium darah dari jumlah normal 9 - 10 mg/dl menjadi 3 - 7 mg/dl. Penelitian ini bertujuan untuk mengetahui tentang jumlah kasus, gejala klinis, dan penanganan penyakit hipokalsemia pada sapi perah di Koperasi Unit Desa Tani Wilis Sendang Tulungagung pada tahun 2012.

Abstract

This study was conducted to determine the histopathological picture of the pancreas by induction of alloxan and extracts of the moon flower (tithonia diversifolia) in wistar rats (rattus norvegicus). A total of 24 male Wistar rats with 4 treatments were control P0, alloxan P1 with 1% CMC Na, P2 alloxan with 100mg / kgBB extract, alloxan P3 with 200mg / kgBB extract. Mice after said diabetes mellitus with a number> 135 mg / dL were then given treatment with 1% CMC Na on P1 and lunar leaf extract at P2 and P3, necropsy was carried out on the 15th day and the pancreatic organs were taken to be made in the form of preparations with coloring. HE, lesions observed Degeneration, Inflammatory Cells and Necrosis using a microscope, the results were analyzed using Kruskal wallis followed by Mann withney. The data obtained by degeneration in the kruskal wallis test were very significantly different because there were significant differences (P = 0.01), inflammation cells in the Kruskal wallis test were very significantly different because there were significant differences (P = 0.00), and for necrosis in the Kruskal wallis test significantly different because there are significant differences (0.02).

Abstract

This study was conducted to determine the effect of antioxidant administration from green tea infusion (Camellia sinensis) on damage to the histological structure of the lung exposed to clove cigarette smoke in mice (Mus muscullus). Free radicals in cigarette smoke such as tar, nicotine, and carbon monoxide can cause various damages to the lung or This study was conducted to determine the effect of antioxidant administration from green tea infusion (Camellia sinensis) on damage to the histological structure of the lung exposed to clove cigarette smoke in mice (Mus muscullus). A total of 27 mice were taken from the Biochemical Laboratory of Airlangga University, Surabaya. In this study mice (Mus muscullus) were exposed to kretek cigarette smoke in the smoking chamber for 15 minutes in a period of 2 weeks, used 2 variants of antioxidant doses of green tea infusion of 0.45 gr to 0.6 gr. After the treatment was completed, the preparation of the mice's pulmonary organ preparations and damage to the structure of the lung organ were observed in 9 visual fields. The results showed that there was a relationship between the amount of cigarette smoke exposed and damage to the lung organs of mice when given antioxidant green tea.
