Abstract

This study had three fold objective which were finding the effectiveness of ethanol extract of permot leaf (Passiflora foetida Linn.) towards mortality of Rhpicephalus sanguineus (R. sanguineus) larvae in vitro and also identifying lethal concentration and lethal time. Completely randomized design was used with six treatments and four replications. Five R. sanguineus were used in the treatment for all replication. The observation of mortality was done every ten minutes in five hours. Rhpicephalus sanguineus larvae were dead if there was no movement. The collected data was analyzed using ANOVA factorial and followed by Duncan’s test. The obtained data showed that ethanol extract of permot’s leaf ethanol extract was effective toward mortality of R. sanguineus larvae in vitro. The suspension of 10% extract concentration of permot leaf ethanol extract has a similar effectiveness as Noticks®. Probit analysis stated that LC50 can be reached by using the 4% concentration in two hours and 2.4% concentration in five hours. LC95 can be reached by using the 6.5% concentration in two hours and 3.93% concentration in five hours. The quickest LT50 can be reached in 0.91 hours and LT95 2.37 hours after treatment with 10% concentration.

Abstract

This study aims to determine the effect of porang (Amorphophallus oncophyllus) tuber extract on the histopathological features of the testicle of alloxan induced rats (Rattus norvegicus). Thirty male rats were randomly divided into six groups. Rats in group C- were injected with distilled water, while rats in groups C+, T0, T1, T2, and T3 were injected with 120 mg/kg bw alloxan. Blood glucose was measured three days after alloxan injection. Rats in groups C- and C+ were then administered with 1% Na-CMC, whereas rats in groups T0, T1, T2, and T3 were then administered with 45mg/kg bw metformin, and 100, 200, and 400 mg/kg bw of porang tuber extract, respectively. All solution were given orally once every day for 14 days. Spermatogenic activity was assessed using the Johnsen scoring system and analyzed by Kruskal-Wallis, followed by the Mann-Whitney test. The diameter and epithelial thickness of the seminiferous tubule were measured using image raster software and analyzed using Anova followed by Duncan's test. The results showed that spermatogenesis score, diameter, and epithelial thickness of seminiferous tubule of group C+ were smaller (p <0.05) than group C-. Spermatogenesis scores of groups T0, T1, T2, and T3 were higher (p <0.05) than group C+. Seminiferous tubule diameter and epithelial thickness in groups T0, T1, and T2 were greater (p <0.05) than group C+. It could be concluded that porang tuber extract at a dose of 200 mg/kg bw improved the spermatogenesis score, diameter seminiferous tubule, and thickness of the epithelium of diabetic rats.

Abstract

This study aimed to evaluate the association of factors such as nutrition, thermal humidity index (THI), age, and lactation period with the occurrence of repeat breeding of dairy cows. The sample consisted of Holstein Friesian cows aged 1-11 years, with lactation periods ranging from 1 to 7 months, currently in lactation, and experiencing repeat breeding (inseminated three or more times without conception, despite exhibiting normal estrous cycles and intervals). The results showed that feeding practices by farmers did not significantly differ (p >0.05) in relation to the incidence of repeat breeding. THI values recorded in several barns also showed no significant differences (p >0.05), with the cows experiencing light to moderate heat stress. Similarly, the frequency of artificial insemination and the estrous cycle intervals remained within the normal range. The highest incidence of repeat breeding was observed in cows aged 2.5 to 5.5 years, accounting for 78.13%, and during lactation periods of 1-3 months, accounting for 81.26%. It can be concluded that there were no significant differences in the provision of forage and leguminous feed between normal cows and those with repeat breeding, and the crude protein content of forage did not influence its occurrence. Additionally, there was no indirect relationship between THI and repeat breeding. However, the incidence of repeat breeding tended to increase in cows aged 3-4 years, and during the second and third lactation periods.

Abstract

Predicting the turtle's reproductive status and sexual maturity is necessary for ex-situ breeding and conservation program. The histology of the seminiferous tubules of the Indonesian box turtle (Cuora amboinensis couro) at various ages has not been studied much. This study aims to develop equations to predict the sexual maturity of Indonesian box turtles based on straight carapace length (SCL). Six turtles with SCL sizes 10-21 cm were sacrificed and their testes were taken for morphometric measurements and histological preparations for Hematoxylin Eosin staining. Pearson's correlation of SCL with testicular maturity parameters was analyzed, followed by Anova Regression. The results showed that there were differences in the morphometry and topographic anatomy of the Indonesian box turtle testes between SCL below 15 cm and above 15 cm. Abstract

Lead is a harmful pollutant from engine exhaust that causes free radicals and has detrimental effects on the testicular tissue. This study aimed to determine the effects of tomato juice on the number of Leydig cells and the diameter of the seminiferous tubules of mice exposed to lead acetate. Twenty-five male mice were divided into five groups. Mice in the control (C-) group were given placebos. Meanwhile, mice in C+, T1, T2, and T3 groups were exposed to lead acetate at a dose of 100 mg/kg BW/day for 14 days and given tomato juice respectively at 0, 0.16, 0.32, and 0.64 mL/day from day 8 to day 35. On day 36, all mice were sacrificed, and the testes were collected for histological preparation. The result showed that lead exposure in the C+ group decreased (p <0.05) in the number of Leydig cells and the diameter of the seminiferous tubules compared to the mice in group C-. Administration of tomato juice in groups T1, T2, and T3 increased (p <0.05) the number of Leydig cells and the diameter of seminiferous tubule compared to the mice in the C+ and C- groups. However, tomato juice administration to the T3 group decreased the number of Leydig cells and the diameter of the seminiferous tubules (p <0.05) compared to the T2 group. In conclusion, an effective dose of 0.32 mL/day of tomato juice restored Leydig cell number and seminiferous tubules diameter in mice exposed to lead acetate.

Abstract

This study aimed to determine the effect of the administration of ethanolic extract of Moringa oleifera leaves on the number of spermatogenic and Leydig cells of gentamicin-induced rats (Rattus norvegicus). This study used 25 white male rats divided randomly into five groups. The rats were injected with Gentamicin 5 mg/kg bw/day subcutaneously for 14 days, except for the negative control group (NCG). During the next 14 days, the rats were given oral moringa leaf extract of 200, 316, and 500 mg/kg bw/day respectively for E200, E316, and E500 groups. The data were analyzed with one-way ANOVA followed by Duncan's test. The results showed that the number of spermatogonia, spermatocytes, and spermatids in NCG was higher (p <0.05) than in the other groups. The number of spermatogonia, spermatocytes, and spermatids in the E316 group was higher (p <0.05) than in PCG. The number of spermatogonia in the E500 group was higher (p <0.05) than in the E316 group, but the number of spermatocytes and spermatids in the E316 group was similar (p >0.05) to the E500 group. The number of PCG Leydig cells was the smallest (p <0.05) compared to the other groups. The administration of Moringa leaves extract in the E200 and E316 groups increased (p <0.05) the number of Leydig cells compared to PCG. The number of Leydig PCG cells in the E500 group was not significantly different (p >0.05) from that of the NCG. It could be concluded that administration of Moringa oleifera leaves extract restored the number of spermatogenic and Leydig cells of gentamicin-induced rats.
