Abstract

Food borne disease can be transmitted through Escherichia coli and Salmonella spp contamination. The contamination of microorganisms with high pathogenic potentials on bovine carcasses results in food borne illness. The aim of this study was to identify multidrug-resistance of Escherichia coli and Salmonella spp on carcass samples that isolated from slaughter house in Banyuwangi. Samples were collected from district of Banyuwangi, Rogojampi, Genteng and Kalibaru. This study used cross sectional study with assumption of prevalence at 50% in each contaminant, confidence level 95% and standart of error at 10%. By the number of samples should reach 96 samples. The result showed that seven samples (7,3%) were positive Escherichia coli and none samples (0%) were positive Salmonella spp. The multidrug-resistance of Escherichia coli showed that Cephalotin (42,9%), Trimethoprim (14,3%) and Erythromycin (42,9%) whereas Ampicillin, Enrofloxacin, Chloramphenicol and Tetracycline were sensitive against Escherichia coli.

Abstract

The purpose of this study is to determine effect of antiinflammation of Thitonia diversifolia (TD) extracts in histopathology liver and pancretic tissue on alloxan-induced in Wistar rat. Thirty two male Wistar rats (Rattus norvegicus) with a body weight (bw)150-200 g in 3 month of age, were allocated into four groups, with eight animals per group. The control group received normal saline (P0), positive control group  received CMC Na 0.01% (P1),  the treatment group were received 100 mg/kg bw of the TD extracts (P2) and received 100 mg/kg bw of the metformin (P3)  respectively for 7 days. Bloods serum collected for measuring of  blood glucose (BG). cholesterol and insulin (INS) concentrations. The levels of insulin (INS) concentrations were analysed by Avidin-Horseradish Peroxidase (HRP) Sandwich-ELISA. All animal were sacrified for tissue staining with haematoxylin eosin. Thitonia diversifolia extract significantly decreased the level of blood glucoseand choelsterol concentrations compared to the postive control group (p<0.05). The level of INS could be detected by its level significantly increased in TD treatment group (p<0.05). Histopathological showed that induction of alloxan could effect severe multifocal degeneration, vacuolisation, cell inflammation, acute langerhan cells injury with destruction cells. Immunohistochemical staining labeled with IL-1β monoclonal antibodies (Mab) showed lowering expression of IL-1β reveal as brownish color aggregates on the langerhans cells in the TD treatment group. This study was concluded that TD extract is inhibited hepatic tissue injuries and decreased destruction in langerhan cells by decreasing IL-1β expression on alloxan-induced rats.

Abstract

Sea urchin (Echinometra mathaei) is one of the marine biota that contains high flavonoid and phenolic compounds that are potent as anti-inflammatory and antioxidant. This study aimed to measure the anti-inflammatory activity of the 70% ethanol extract of sea urchins in male white rats induced by carrageenan. Anti-inflammatory activity test using 5 groups of rats selected randomly, each group consists of 5 rats. Negative control group, positive control group, and 3 groups of extracts (doses of 100, 200, and 400mg/kgBW). Each group was induced using 1% carrageenan subplantarly and the volume of edema was measured using a mercury plethysmometer every 30 minutes for 3 hours. The data were statistically analyzed using one-way Anova. The results showed that the percentage of inflammatory inhibition in the positive control group, and the Echinometra mathaei ethanol extract group (dose of 100, 200, and 400 mg/KgBW) showed a significant difference (P0.05) with the negative control group with a percentage value of Inflammation inhibition was 61.11%, 85.19%, 85.19%, and 74.07%, respectively. The administration of sea urchin ethanol extract at all doses had a higher percentage of anti-inflammatory than diclofenac sodium, but increasing the extract dose did not significant increase inflammatory inhibition. This research concluded that 70% ethanol extract of Echinometra mathaei at doses of 100, 200, 400 mg/KgBW has strong anti-inflammatory activity potential.

Abstract

The cytochrome P-450 (CYP) and glutathione S-transferase (GST) enzyme systems may influence the biological effects of carcinogens. As such, these enzymes may predict the developmental risk of breast cancer, as well as be potential targets for chemoprevention. The purpose of this study was to compare the expression of CYP1A1 and GSTµ between treatment groups. Expression of CYP1A1 and GSTµ was quantified in liver tissue from 18 female Sprague Dawley rats aged 40 days, which randomly divided into six treatment groups. Those are base line group (without DMBA and ethanolic extract treatment), DMBA induced cancer group, the other two groups was administrated by DMBA after treated by ethanolic extract in two different doses, 300 mg /kg BW and 750 mg/kg BW.  The last two groups were given two doses of extract ethanolic only, without initiated of DMBA. The ingestion of the extract was carried for three weeks and the ingestion of DMBA was performed twice in the third week. The expression CYP1A1 and GST was quantified by immunohistochemistry. CYP1A1 expression was significantly higher (P < 0.05) in cancer group (DMBA) as compared with other groups. On the other hand, GST expression was lower in cancer group (P < 0.05). Result of this study demonstrated that ethanolic extract leaves of  G. procumbens in 300 mg/kg BW dose could  inhibit CYP1A1 stronger than are other and induced GSTµ level. Has effect on ethanolic extract leaves of G. procumbens has ability in played role of as blocking agent in preventing initiation stage of carsinogenesis, therefore it should be taken into account for chemopreventing agent in mammary carsinogenesis.Key words : Gynura procumbens, breast cancer,  hemopreventive, CYP1A1, GSTμ

Abstract

Antiangiogenesis (inhibition of new blood vessels formation) has become a strategy to inhibit cancer development. The aim of this experiment was to investigate antiangiogenic effect of Gynura procumbens (Lour) Merr focusing on the decreasing of the number of macrophages and heterophils on chick embryo chorioallantoic membrane. Nine-days-aged-eggs were divided into six groups (eight eggs each group). Group I (positive control) eggs were induced with bFGF+Tris HCl. Group II (negative control) eggs were treated with DMSO+Tris HCl. Group III (treatment I) eggs were induced with 60 ng bFGF and treated with ethanolic extract of G. procumbens leaves with the dose of 60 µg. The following treatment groups, i.e. group IV (treatment II), group V (treatment III), and group VI (treatment IV) were treated with increasing dose of extract, starting from 75 µg, 90 µg, and the last was 110 µg. Eggs were incubated until they reach the age of twelve days to observe macrophages, while to observe heterophils, eggs were incubated until the age of seventeen days. Based on haematoxylin-eosin staining, macrophages in the treatment groups were less than the control positive group (bFGF+Tris HCl), but based on giemsa staining, the effect of Gynura procumbens in decreasing the number of heterophils could not be observed because some blood smears. These analysis suggest that the ethanolic extract of Gynura procumbens leaves can perform as antiangiogenic agent decreasing the number of macrophages.Keywords: antiangiogenic, macrophages, heterophils, Gynura procumbens

Abstract

Angiogenesis is a physiological process that involves the formation of new blood vessels. In the cancer growth process, angiogenesis acts by supplying nutrition and oxygen for the cancer cells, carries metabolic waste and biological end product of cancer cells. Therefore the spread of cancer cells to the surrounding cells and tissues is accelerated. The occurrence of angiogenesis shows a change in cancer cell status, from dormant to malignant. Cancer cells induce the formation of angiogenesis through the secretion of various growth factors such as Vascular Endothelial Growth Factor (VEGF) and basic Fibroblast Growth Factor (bFGF).Currently many people turn into treatment using natural materials. One of the natural materials used for cancer therapy is pegagan (Centella asiatica). In this research 25 embryonated chicken eggs (ECE) were divided into five treatment groups. Treatment group I as the control received 60 ng bFGF, treatment group II received neither bFGF nor pegagan leaves (Centella asiatica) water extract, and  treatment groups III-V received 60 ng bFGF and  C. asiatica leaves water extract of 75, 90 and 110 µg.  Nine days old ESE were inoculated with bFGF and extract at the same time and then were incubated at 380C for three days to allow the formation of blood vessels in embryonated chicken egg chorioallantoic membrane.  Observations were performed microscopically and macroscopically by counting the new blood vessels formed around paper disc. Analysis on VEGF expression were conducted to define the role of angiogenesis growth factor.Research results showed a significant difference of treatment with Centella asiatica water extract on the number of new blood vessels formed. Centella asiatica water extract at the dose of 110 µg was the highest in inhibiting the formation of new blood vessels. Abstract

P2Y12 is a platelet receptor that is involved in ADP signal transduction and is an attractive target for antithrombotic drugs. The side effects of antithrombotic drugs are not pleasant for the patient, so research into the development of new antithrombotic agents is still necessary. Evaluation of absorption, distribution, metabolism, elimination, and the toxicity profile of candidate drugs is an important step in drug development. The aim of this study was to predict the potency of ferulic acid (FA) and its derivatives (FA1-24) as antiplatelet drugs by a docking study on the P2Y12 receptor (PDB ID: 4PXZ) and their ADMET performance. The docking study was performed using Molegro Virtual Docker, version 5.5. ADMET prediction of FA was conducted using the pkCSM online tool. The results of the in silico study showed that FA-19 had the lowest MolDock score (MDS), which means that this compound is predicted to have the greatest activity. FA-19 is also predicted to be practically non-toxic. It is expected that FA-19 will have good intestinal absorption and is similarly distributed in the intestine and in the blood plasma. Its penetration in the blood-brain barrier is moderate but does not inhibit the CYP2D6 and CYP3A4 enzymes.

Abstract

BACKGROUNDMany tumors express on their receptor tyrosine kinases vascular endothelialgrowth factor activity associated with angiogenesis. Inhibition ofangiogenesis through reduction of tyrosine kinase activity is a promisingstrategy for cancer therapy. The present study aimed to determine themechanism and potency of ethyl p-methoxycinnamate (EPMC) isolatedfrom Kaempferia galanga as angiogenesis inhibitor.METHODSA laboratory experimental study was conducted using chorio-allantoicmembranes (CAMs) of nine-day old chicken eggs induced by 60ng basicfibroblast growth factor (bFGF). Ethyl p-methoxycinnamate (EPMC) potencywas determined at dosages of 30, 60, 90 and 120 μg and compared withcelecoxib 60 μg as reference drug and one negative bFGF-induced controlgroup. Neovascularization and endothelial cell count in CAM blood vesselswere evaluated. To predict the antiangiogenic mechanism of EPMC, adocking study was performed with the Molegro Virtual Docker program ontyrosine kinase as receptor (PDB 1XKK).RESULTSAngiogenesis stimulation by bFGF was prevented significantly (p<0.05)by EPMC at dosages of 30, 60, 90 and 120 μg and this activity was dosedependent. Molecular docking showed interaction between EPMC functionalgroups and tyrosine kinase amino acids at Met766, Met793, Thr854, Thr790,Gln791 and Ala743. There was an association between EPMCantiangiogenic activity and docking study results.CONCLUSIONSEthyl p-methoxycinnamate is a potential new angiogenesis inhibitor throughinteraction with tyrosine kinase. EPMC could be a promising therapeuticagent for treatment of angiogenesis-related diseases.

Abstract

Penyebaran pandemi virus corona dari penyakit COVID-19 kian masif. Hingga kini, kasus terkonfirmasi di Indonesia sudah melebihi angka 100.000, dengan tingkat kematian (case fatality rate) sebesar 8.9%. Selain itu, terdapat 50 dari 100 kabupaten/kota yang memiliki risiko penyebaran tertinggi dengan 49 persen. Beberapa di antara 50 kabupaten/kota itu berada di Pulau Jawa. Empon-empon bukan hanya berfungsi untuk mengobati penyakit. Namun empon-empon juga berfungsi untuk menjaga kesehatan dan meningkatkan daya tahan tubuh. Di tengah maraknya wabah corona, kita perlu untuk menjaga daya tahan tubuh. Empon-empon biasa terdapat kunyit, jahe, lengkuas, temulawak, kencur, dan lainnya. Kegiatan pengabdian masyarakat dengan sasaran civitas akademika Fakultas Kedokteran Hewan Universitas Airlangga. Kegiatan dilaksanakan melalui tahapan penyuluhan, praktik mandiri, dan pembagian probiotik kombinasi empon-empon. Hasil yang didapatkan dari evaluasi pre dan post test terdapat peningkatan afektif dari 20% menjadi 60%, kognitif dari 28% menjadi 75%, dan psikomotor dari 8% menjadi 80%.
