Abstract

Abstract The purpose of this study was to determine the resistance of several antibiotics phenotypically and genotypically to detect the tetA gene from broiler chicken liver samples at Dukuh Kupang market, Surabaya. A total of 30 samples were taken and then prepared aseptically and sterile. Isolation on Eosin Methylene Blue Agar (EMBA) media, then microscopic examination using gram staining and biochemical tests of Triple Sugar Iron Agar (TSIA), Sulfide Indole Motility (SIM), Methyl Red (MR), Voges Prouskauers (VP) and Simons Citrate Agar (SCA). The identified Escherichia coli colonies were tested for antibiotic sensitivity using the Kirby Bauer method, then isolates that were proven to be resistant to tetracycline antibiotics were followed by genetic testing using the Polymerase Chain Reaction (PCR) method. The results showed that 20 of the 30 samples were positive for Escherichia coli. Escherichia coli isolates from chicken liver samples showed resistance to 30 µg tetracycline antibiotics by 85% (17 of 20 samples) Researchers also compared with other antibiotics, the highest resistance to ampicillin 10 µg was 90% (18 out of 20 samples), gentamicin resistance was 10 µg by 50% (10 of 20 samples) and 30 µg chloramphenicol antibiotic resistance by 30% (6 of 20 samples). The isolates that were resistant to tetracycline were confirmed by Polymerase Chain Reaction to detect the tetA gene with the final product in the form of a band with a length of 210 bp. Bacterial isolates resistant to Tetracycline antibiotics did not always show TetA gene expression in the PCR test. Keywords: Antibiotic Resistance; Escherichia coli; Market; TetA gene Abstrak Tujuan dari penelitian ini yaitu untuk mengetahui resistansi beberapa antibiotik secara fenotip dan secara genotip mendeteksi gen tetA dari sampel hati ayam broiler di pasar Dukuh Kupang Surabaya. Sebanyak 30 sampel diambil kemudian dipreparasi secara aseptis dan steril. Isolasi pada media Eosin Methilen Blue Agar (EMBA), selanjutnya dilakukan pemeriksaan mikroskopis menggunakan pewarnaan gram dan uji biokimiawi Triple Sugar Iron Agar (TSIA), Sulfide Indol Motility (SIM), Methyl Red (MR), Voges Prouskauers (VP), dan Simons Citrat Agar (SCA). Koloni Escherichia coli yang teridentifikasi dilakukan uji sensitifitas antibiotik dengan metode Kirby bauer, selanjutnya isolat yang terbukti resistan terhadap antibiotik tetrasiklin dilanjutkan pemeriksaan genetik dengan metode Polymerase Chain Reaction (PCR). Hasil penelitian menunjukkan bahwa 20 dari 30 sampel positif Escherichia coli. Isolat Escherichia coli asal sampel hati ayam menunjukkan resistansi terhadap antibiotik Tetrasiklin 30 µg sebesar 85 % (17 dari 20 sampel) Peneliti juga melakukan perbandingan dengan antibiotik lainnya, resistensi tertinggi pada antibiotik ampisilin 10 µg sebesar 90 % (18 dari 20 sampel), resistensi gentamisin 10 µg sebesar 50 % (10 dari 20 sampel) dan resistensi antibiotik kloramfenikol 30 µg sebesar 30 % (6 dari 20 sampel). Isolat yang resisten terhadap tetrasiklin dikonfirmasi dengan Polymerase Chain Reaction untuk mendeteksi gen tetA dengan produk akhir berupa band dengan panjang 210 bp. Isolat bakteri yang resistan terhadap antibiotik Tetrasiklin tidak selalu menunjukkan ekspresi gen tetA pada uji PCR. Kata kunci: Escherichia coli; Gen TetA; Pasar; Resistansi Antibiotik.

Abstract

Slaughtering of sacrificial animals is a worship activity for slaughtering ruminant livestock which is carried out on Eid al-Adha and carried out in mosques or outside Slaughterhouses (RPH). The limited facilities affect the welfare of the animals so that it affects the quality of the meat. Supervision in the production chain with antemortem and postmortem inspections is carried out to obtain good quality meat. This community service aims to help the community get a guarantee of safe, healthy, whole and halal, sacrificial animal meat with antemortem and postmortem examinations, as well as holding counseling on the handling of sacrificial animals and meat during the covid-19 pandemic through webinars. The activity was carried out in 13 mosques in the areas of Surabaya, Sidoarjo, Gresik, and Pasuruan. The antemortem examination will be held on 19-22 July 2021 followed by a post-mortem examination which will be held on 20-22 July 2021 with a total number of 112 cows, 220 goats and 1 sheep. The Webinar was held earlier, namely on July 14, 2021, by 3 competent speakers. The webinar was attended by the general public, mosque takmir, lecturers, and students. The results of the antemortem examination indicated that the animals were suitable for slaughter, although 2–6 (0.6–1.8%) animals showed diarrhea, nasal and eye discharge, and infection of the prepuce. The results of postmortem examination stated that the animals were fit for consumption with a note that the organs infected with worms had to be removed, namely 15.2% of the liver of cattle and 0.9% of the liver of goats with fasciolosis, 0.5% of goats experiencing parampistomiasis and 1.7% of cattle experiencing eurytrematosis.

Abstract

Limbah peternakan umumnya meliputi semua kotoran yang dihasilkan dari suatu ke¬giatan usaha peternakan, baik berupa limbah padat dan cairan, gas, ataupun sisa pakan. Limbah peternakan adalah semua buangan dari usaha peternakan yang bersifat padat, cair dan gas. Limbah padat merupakan semua limbah yang berbentuk padatan atau dalam fase padat (kotoran ternak, ternak yang mati atau isi perut dari pemotongan ternak). Limbah cair adalah semua limbah yang berbentuk cairan atau berada dalam fase cair (air seni atau urine, air pencucian alat-alat. Penelitian ini untuk mengetahui analisis usaha pembuatan pupuk organik cair dengan bahan baku urin sapi perah di Desa Galuhan Kecamatan Kandat Kabupaten Kediri.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian perasan kunyit terhadap jumlah monosit dan heterofil pada ayam pedaging yang diinfeksi dengan antigen Salmonella pullorum. Hewan percobaan yang digunakan adalah ayam pedaging sejumlah 40 ekor. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 5 pengulangan. Keempat perlakuan tersebut adalah P0 (kontrol), P1 (40% perasan kunyit), P2 (50% perasan kunyit), P3 (60% perasan kunyit). Pemberian perasan kunyit dilakukan mulai dari DOC sampai umur 14 hari pada saat pagi hari (maksimal 4 jam pemberian). Infeksi antigen Salmonella pullorum dilakukan pada umur 7 hari. Jumlah monosit dan heterofil dihitung dari hasil preparat ulas darah. Hasil penelitian ini menunjukkan bahwa pemberian perasan kunyit dengan konsentrasi 50% menunjukkan peningkatan jumlah monosit dan heterofil jika dibandingkan dengan kelompok perlakuan 40% dan 60%.

Abstract

The present research to determine the Extract of Pineapple (Ananas comosus) on the survival of catfish infected with A. Hydrophila. The main problem of cultivation of catfish (C. batrachus) is a disease caused by the bacterium Aeromonas hydrophila.Catfish (Clarias batrachus) is a freshwater fish that has several advantages, one of which is a high nutritional content. Test animals used were catfish measuring 15-20cm with a body weight of ± 250gr. Challenge test by injecting A. hydrophila suspension with a dose of 108sel / mm3 as much as 0.1mm intramuscularly. Observations were made for 7 days after infection which included the total number of leukocytes and neutrophils. This study was divided into 5 treatment groups with 5 replications, namely P1 (positive control), P2 (0.1%), P3 (0.3%), P4 (0.5%), P5 (0.7%) and soaking Pineapple Fruit Extract for 24 hours. Data analysis in this study used one way ANOVA statistical test. The results showed symptoms of catfish attacked by Aeromonas hydrophila including increased total number of leukocytes and neutrophils and ulceration in catfish. Soaking effect Pineapple fruit extract 0.5% effective dose can reduce the total number of leukocytes and 0.7% effective dose can reduce the total number of neutrophils.

Abstract

This study was conducted to determine the effect of antioxidant administration from green tea infusion (Camellia sinensis) on damage to the histological structure of the lung exposed to clove cigarette smoke in mice (Mus muscullus). Free radicals in cigarette smoke such as tar, nicotine, and carbon monoxide can cause various damages to the lung or This study was conducted to determine the effect of antioxidant administration from green tea infusion (Camellia sinensis) on damage to the histological structure of the lung exposed to clove cigarette smoke in mice (Mus muscullus). Free radicals in cigarette smoke such as tar, nicotine, and carbon monoxide can cause various damages to the lung organs due to oxidative stress caused by the reaction of ultrafine particles with cells. A total of 27 mice were taken from the Biochemical Laboratory of Airlangga University, Surabaya. In this study mice (Mus muscullus) were exposed to kretek cigarette smoke in the smoking chamber for 15 minutes in a period of 2 weeks, used 2 variants of antioxidant doses of green tea infusion of 0.45 gr to 0.6 gr. After the treatment was completed, the preparation of the mice's pulmonary organ preparations and damage to the structure of the lung organ were observed in 9 visual fields. The results showed that there was a relationship between the amount of cigarette smoke exposed and damage to the lung organs of mice when given antioxidant green tea. The administration of green tea antioxidants reduced inflammation of the lung organ infiltration of inflammatory cells by 44%, edema by 55%, and destruction of septa alveoli by 88%. gans due to oxidative stress caused by the reaction of ultrafine particles with cells. Abstract

Toxoplasma gondii is an intracellular parasite of the protozoan group and is obligate parasite with definitive host is a cat and other family felidae, Toxoplasma gondii can cause Toxoplasmosis and is categorized as one of zoonotic diseases. This study aims to determine the Toxoplasma gondii infection in domestic cats (Felis domesticus) in the traditional markets of Sawahan sub-district, Surabaya City. This study used a faecal sample of 46 domestic cats in the traditional markets of the Sawahan sub-district consisting of 20 female cats, 10 male cats, and 16 kittens. Then examined at the Laboratory of Parasitology, Faculty of Veterinary Medicine, Wijaya Kusuma University. The examination was carried out by the native method using eosin 2% and the floating method using saturated sugar and observed under a microscope with a magnification of 100x-400x. The results showed there were no cats infected with Toxoplasma gondii but found 3 endoparasites namely Ancylostoma sp 23.9%, Toxocara cati 41.3% and Isospora rivolta 4.3%.

Abstract

This research was conducted to determine the effect of the extract of telang flower (Clitoria ternatea) as a natural antibacterial on the growth of Stapylococcus aureus bacteria in terms of the results of the inhibition test and phytochemical screening. This study used 5 treatments with 5 replications in positive control and negative control, for the extract treatment using duplo replication. P0 as a positive control using Amoxicillin antibiotic, P0 as a negative control using a blank disk, P1 as a treatment using a telang flower extract with a concentration of 80%, P2 as a treatment using a telang flower extract with a concentration of 90%, and P3 a treatment using a telang flower extract with a concentration of 100 %. The results of the inhibition zone measurements showed the highest average in the positive control. The average diameter of the inhibition zone using the Agar Well Diffusion Method showed a significant difference (P<0.05). These results are supported by the ANOVA test with H1 accepted so that it can be said that there is an effect of telang flower extract (Clitoria ternatea) on the growth of Staphylococcus aureus bacteria. The results of phytochemical screening showed that the telang flower extract contained antibacterial active compounds, namely flavonoids, tannins, saponins, alkaloids, steroids and terpenoids.

Abstract

The aim of this study was to report cases of respiratory tract infection caused by the bacterium Ornithobacterium rhinotracheale. Observations included anatomical pathology and bacterial isolation results. Poultry was necropsed, observed changes in organs, trachea samples were taken aseptically then isolated on Blood Agar, Chocholate Agar, MCA, microscopic examination and biochemical tests, including TSIA, SIM, SCA, Catalase, Urease, MR, VP. The results of the examination showed the presence of Ornithobacterium rhinotracheale bacteria, the results showed small, grayish colored colonies. Microscopic examination, gram-negative pleomorphic rod-shaped bacteria. Bacteria did not grow on MCA, there was no change in TSIA media, non-motile bacteria, the results of the indole test, SCA, MR and urease were negative. All four types of birds showed pathological changes, including hemorrhage in the trachea and lungs, but the air sacs were normal and did not show any specific lesions, only duck samples with code I-11 had changes in the air sacs. Based on the results of isolation and bacteria, it can detect the presence of Ornithobacterium rhinotracheale from several different types of poultry, including layer chickens, broilers, local ornamental chickens and ducks.
