Abstract

Singkil (Premna cordifolia Roxb.) is a plant that is widely found un Kalimantan. The people of Kalimantan are often used singkil leaf by as traditional medicine are believed to have medicinal properties as a cure for intestinal worms, colds, and antidiabetic. In this research, it used phytochemical screening and inhibition activity amylase test on singkil’s methanol extract. The objective of this study was to determine in-vitro enzyme amilase inhibition activity against sampel by DNS method. The result of this research was phytochemical screening in singkil’s methanol extract contains alkaloids, flavonoids, saponins and phenolics. Singkil leaf methanol extract at a concentration of 10 µg/mL can inhibit the activity of the amylase enzyme with an inhibition value of 60,93 %. Acarbose used as a positive control in this study showed activity against the amylase of 44,79 %. Amylase inhibition activity of methanol extract was higher than acarbose, the singkil leaf methanol extract has the potential as an antidiabetic drug. Abstract

Pucuk Merah (Syzygium myrtifolium Walp.) is an ornamental plant that has benefits as an antioxidant, antibacterial, antiplatelet, antiulcerative, antimalarial, antiviral, antihyperglycemic and anti-inflammatory. This research aims to determine the optimum time for the production of secondary metabolites of pucuk merah endophytic bacteria for amylase and protease inhibition. Production of secondary metabolites was carried out at various times of 24 hours, 48 hours and 72 hours. The method for the amylase inhibition test used the DNS method and the protease inhibition test used the Bradford method. The results showed that the optimum time for the production of secondary metabolites was 48 hours. Keywords: Syzygium myrtifolium Walp., Endophyte Bacteria, Production Time.

Abstract

One of the causes of acne is infection by the bacteria Propionibacterium acnes. Singkil (Premna cordifolia Roxb.) is a kind of herb plant from Kutai Kartanegara which is used as a traditional herb including for acne’s treatment. This study aims to know the content of secondary metabolites compound in singkil’s extract and determine the antibacterial activity against Propionibacterium acnes KCCM 41747 bacteria. In this study, it’s phytochemical screening and antibacterial activity test using the disc diffusion method on methanol extract, n-hexane, ethyl acetate and water-methanol fractions. the result of phytochemical screening revealed that methanol extract of singkil contains alkaloids, flavonoids, steroids, and saponins. The n-hexane fraction contains steroids and the ethyl acetate fraction contains alkaloids, flavonoids, and phenolics while the water-methanol fraction contains alkaloids, flavonoids, and phenolics. The antibacterial activity test extract with a concentration of 10% showed that the diameter of clear zone of the metanol extract, n-hexane fraction, ethyl acetate fraction, and water-methanol fraction were 13 mm, 9 mm, 13,67 mm and 6 mm, respectively. The ethyl acetate fraction had the highest antibacterial activity among the four extracts. A minimum inhibitory concentration value of the ethyl acetate fraction against Propionibacterium acnes KCCM 41747 was 0,625%. The ethyl acetate fraction have the potential as an antibacterial against the bacteria Propionibacterium acnes KCCM 41747. Abstract

Kratom (Mitragyna speciosa korth) is one of the plants found in forest areas in Indonesia. Phytochemical screening and pharmacological activity of kratom plants (Mitragyna speciosa Korth) have been conducted. Some studies show kratom plants contain secondary metabolites, namely alkaloids, flavonoids, saponins, triterpenoids, tannins and terpenoid-steroids. Literature studies show kratom leaves have strong antioxidant activity with IC50 values <50 ppm. Kratom leaves have antinonceptive activity in the dichloromethane fraction at doses of 70 mg/kgBB (51%), 140 mg/kgBB (81%) and 280 mg/kgBB (86%). Antimicrobial activity against staphylococcuc pidermidis bacteria with a concentration of 5% (7.23 mm) 10% (10.50 mm), 15% (11.78 mm), 20% (12.63 mm) and 25% (13.69 mm). Stems and roots of kratom plants have antibacterial activity against propionibacterium acnes bacteria, stem concentration 5% (5.71 mm ± 0.40), 10% (6.75 mm ± 0.26) and 15% (6.78mm ± 0.29), on roots 5% (5.4 mm ± 0.12), 10% (5.8 ± 0.08) and 15% (6.21 mm ± 0.36). Anti-inflammatory activity with a dose of kratom extract 0.5 mg/grBB (46.37 mm ± 10.04), 1 mg/grBB (32.33 mm ± 11.24), 2mg/grBB (26.69 mm ± 6.57). Acute toxicity test in the reflux method LC50 value of 71.08 ppm while the maceration method LC50 value of 143, 50 the results shown are included in the toxic category. Abstract

Methylene blue is a cationic dye molecule which is quite dangerous for the body, one of which can cause irritation of the digestive tract if swallowed, cause cyanosis if inhaled, and cause skin irritation if touched. Adsorption is one of the separation methods most often used in most industries, because the process is easy, and the operating costs are also relatively cheap. Activated charcoal is widely used as an adsorbent material because it has high porosity and surface area and has good absorption capacity for organic and non-organic materials. In this research, a literature study has been carried out on the adsorption of methylene blue with activated charcoal adsorbent from various types of ingredients. There are various factors that can influence the adsorption ability of methylene blue by activated carbon, such as the type of original material, the type of activator used to include HCl, NaCl, KOH, NaOH, H2SO4, and Na2CO3, as well as the temperature range for carbonation between 300-700oC. Adsorption parameters varied, such as pH between 4-9, contact time between 20-240 minutes, and adsorption capacity between 6.90-1614.968 mg/g. Keywords: Methylene blue, Adsorption, Activated charcoal.
