Abstract

Sambiloto (Andrographis paniculata Nees) contains andrographolid compound which has potentialas an anticancer. The difference of growth location has affected the distinction of metabolite content. Thisstudy aims to determine the profile of anticancer activities of A. paniculata extract from several locations inEast Java. The extracts from several locations were tested for activity on T47D cancer cell using MTT assaymethod. Thus, the result of this study describes that A. paniculata extract from several different locations doesnot provide significantly different anticancer activities with the amount of (p>0.05). The extracts which havemoderate anticancer activity were A. paniculata extracts from Selopuro Subdistrict Blitar Regency, SingosariSubdistrict Malang Regency, Lowokwaru Subdistrict Malang City, Klojen Subdistrict Malang City,Purwodadi Subdistrict Pasuruan Regency, Purwosari Subdistrict Pasuruan Regency, and PandaanSubdistrict Pasuruan Regency

Abstract

Ibu-ibu di Kelurahan Merjosari RW 05 Lowokwaru Malang merupakan ibu rumah tangga dan rata-rata memiliki penghasilan rendah. Masyarakat tersebut mengeluhkan tentang sulitnya mendapatkan penghasilan dan mencari pekerjaan di masa pandemi seperti ini karena sebagian besar suami (kepala keluarga) terkena PHK. Untuk itu, masyarakat dituntut melakukan usaha secara mandiri salah satunya dengan pembuatan sabun antiseptik dengan memanfaatkan tanaman herbal yang tumbuh di depan rumah sebagai upaya menjaga kesetabilan ekonomi keluarga dan kesehatan selama pandemi. Metode yang digunakan adalah persiapan kegiatan, sosialisasi, pelatihan dan pendampingan. Evaluasi dilakukan dengan cara mengamati kinerja setiap peserta pada akhir kegiatan. Indikator pencapaian yang digunakan untuk menyatakan keberhasilan pengabdian adalah apabila setiap peserta sudah dapat membuat atau menghasilkan minimal satu sabun antiseptik dari tanaman herbal. Selain itu, juga dinilai dari hasil pre test dan post testnya. Masyarakat RW 05 Kelurahan Merjosari, Kecamatan Lowokwaru, Kota Malang dapat memanfaatkan tanaman-tanaman herbal berkhasiat secara etnofarmasi. Peserta telah berhasil memproduksi sabun antiseptik dari taman herbal yang ditanam di sekitar rumah sebagai produk lokal. Pelaksanaan program dengan tema atau topik yang dilakukan oleh Tim dapat direkomendasikan ke tempat lain atau sekolah maupun dilakukan di pondok pesantren.

Abstract

CCL4 exposure on rat resulted hepatocyte cell necrosis and activated macrofages to produce TGF-β which induced liver fibrosis. Kesambi (Schleichera oleosa (Lour.) Oken) plants have active compounds that affect the inflammatory process. The purpose of this study was to determine the effects of methanol leaf extracts from Kesambi on TGF-β levels and histologycal feature of liver in rat exposed to CCL4. Thirty male Wistar rat were divided into 5 groups, K- (injected with corn oil i.p), K+ (injected with CCL4 0,5µl/grb.w. i.p, 2x a week), and the treatment group injected with CCL4 0,5µl/grb.w. i.p, 2x a week and administrated methanol leaf extracts from kesambi dose 200mg/kgb.w. (P1), 400mg/kgb.w. (P2), and 600mg/kgb.w. (P3), everyday. After 6 weeks, the rats were dissected. Liver was taken for TGF-β levels measurement by flowcytometri, necrotizing hepatocyte cells calculation and fibrosis score by METAVIR method. TGF-β level of P3 (4,21%) was significantly different (p0,05) compared to K- (6,34%), K+ (7,21%), P1 (7,80%), and P2 (8,45%). The number of cell necrosis of K+ (54,39%) was significantly different from K- (9,62%), P2 (29,37%), and P3 (18,75%). METAVIR score calculation showed fibrosis in K+ (score 2), P1 (score 2), and P2 (score 1). The administration of methanol leaf extracts from kesambi reduced TGF-β levels and the number of necrotic hepatocyte cells and prevented the formation of liver fibrosis.
