Abstract

Hepatitis B virus (HBV) is a virus cause liver disease. This virus could be transmitted through blood product,and it becomes the major problems in developing countries. Genotype distribution of HBV depends ongeographical characteristics. Here, we reported HBV genotyping analysis of blood donors with positiveHepatitis Surface Antigen (HbsAg) at Indonesian Red Cross Foundation in Tuban, East Java. Total 150sera were collected from blood donors at Indonesian Red Cross Foundation in Tuban, East Java. Enzyme-Linked Immunosorbent Assay (ELISA) was conducted by PRISM®HBsAg kit (ABBOTT) according tothe manufacture’s instruction, to determine sample that has positive HbsAg. The genotyping analysis wasdone using polymerase chain reaction and sequencing method from 12 samples. Genetyx ver.10 softwarewas used for identification of HBV type. Analysis of HBV genotype revealed three different types of HBV.HBV type B was the major HBV (50%). Interestingly, there was one unidentified HBV type suggested as anovel subtype of HBV. Further genotyping analysis is needed to confirm novelty of the unidentified HBVtype. Analysis of HBV genotype revealed three different types of HBV. HBV type B was the major HBV(50%). This study was the firstevidence of blood donor’s genotyping analysis in Tuban.

Abstract

Objective: This study was conducted to detect hepatitis C virus (HCV) RNA among blood donors in Tuban, East Java, Indonesia,  using nested Polymerase Chain Reaction (PCR) technique.Material and methods: Seven blood donors  with positives anti-HCV were obtained from 500 blood donors from Red Cross Blood Donor Center (Palang Merah Indonesia = PMI) Tuban, East Java, Indonesia, consisting of 375 men (75%) and 125 women (25%). Detection of anti-HCV antibody prevalence in the blood donors was done by using Enzyme-Linked Immunosorbent Assay (ELISA).  The detection of HCV RNA in blood donors  sera  with positives  anti-HCV were performed using nested-PCR technique with three set of primer pairs targeting the NS5B and 5’UTR regions of HCV.  Further amplification products were visualized on a 2% agarose gel containing ethidium bromide under  UV illumination.Result: Seven positives anti-HCV blood donors sera (1.4%) were obtained from 500 blood donors sera, in PMI Tuban, East Java, Indonesia. Out of seven blood donors sera with positives anti-HCV, 6 sera (85,7%)  were positives HCV RNA by using nested PCR technique. Five HCV RNAs were detected with two set of primers based on NS5B HCV region and 1 positive HCV RNA was detected with another set of primer based on 5’UTR HCV region.Conclusion: HCV RNAs were detected in 85,7% positives anti-HCV blood donors using nested PCR technique, in PMI Tuban, East Java. Further research is needed to determine the  genotype  and  subtype  of  HCV.

Abstract

Objective: Salmonella typhi as a causative agent of typhoid abdominalis many militias, which was resistant to the drug, one of which is chloramphenicol. Detection of Salmonella typhi serologically generally done so if there is a problem in the diagnosis difficult to overcome so it needs a faster diagnosis techniques with accurate results. PCR can be used as an alternative solution to these problems. The purpose of this study was to detect the DNA of Salmonella typhi resistant to chloramphenicol by PCR.Material and methods: The method in this population experimental laboratory conducted in the laboratory biology and laboratory Unirow Tuban ITD Airlangga University Surabaya. Experiment done using PCR from blood samples. Using primer from CAT gene to detect this gene.Result: The results of this study with tank PCR can detect DNA of chloramphenicol resistant Salmonella typhi with a length of 293 bp.Conclusion: The conclusion PCR technique can detect DNA of Salmonella typhi resistant to chloramphenicol.

Abstract

Science is a science that learns about life in the world from all aspects. To be able to study these subjects requires a deeper understanding of the material. Application of learning innovations needs to be done to achieve the objectives of science subjects especially by teachers so as to provide hands-on experience for students to acquire innovative learning. Based on the observations at SMP Negeri 1 Bancar known that the results obtained by students less satisfactory. The process of teaching and learning is done by lecture method, when the learning process takes place many students are sleepy and chatting. To improve the learning outcomes in science subjects especially biology required the existence of learning innovation, namely by using Natural Picture learning media. The purpose of this study is to determine whether there is an influence of Natural Picture learning media on student learning outcomes in science biology subjects in SMP Negeri 1 bancar. The type of research used is Quasi Eksperimen. This research was conducted at SMP Negeri 1 Bancar with research sample that is student of class VII A which amounted to 32 students as experiment class and student of class VII E which amounted to 32 students as control class. The research design used was Pre-Test research design - Post-Test Control Group Design. Based on the results of data analysis of learning results, the average value in the experimental class given treatment using Natural Picture learning media is higher than the control class using the lecture method. The experiment class is 87.12 and the control class is 74.37. In the hypothesis test the Sig value. (2-tailed) of 0.000, when compared with α 0.05 then Sig <α then H0 is rejected and H1 accepted. Thus, it can be concluded that there is a difference in mean values of experimental class learning outcomes and control classes. That is, there is the effect of learning media Natural Picture on student learning outcomes.

Abstract

Teaching materials tailored to the student's condition will make it easier for students to understand the material delivered and provide a memorable experience. It is therefore necessary to develop a Biology handout in the form of a catalog with color images on the Respiratory Material Material of quality in terms of validity, effectiveness and practicality. The development of this handout is displayed in the form of a catalog and is complemented by color images with the aim of motivating students to learn and make it easier to understand the material with the help of color images. The development model used is the ADDIE model. Data collection techniques use validation sheet instrument, observation sheet, test sheet and questionnaire. The results showed that Biology handouts in the form of catalogs with color images on the respiratory system material were declared valid, practical and effective and suitable for use as a learning medium for students.

Abstract

Learning media is an integral part of teaching and learning process to achieve learning objectives. Teachers are required to be able to choose and use the learning media. However, teachers often choose less appropriate learning media. Learning can take place effectively, efficiently and attractively, if a teacher can make changes in delivering the subject matter. Learning media that has been applied by teachers is less fun that makes students become bored and less able to absorb the material taught, so that activity decreases and student learning outcomes become low. This study aims to analyze the level of validity of media experts and material experts to media based puzzle puzzlemake a match on sub cell subject that has been developed through the validity test. Development of this media use ADDIE learning model (Analyze, Design, Development, Implementation, and Evaluation). Data collection techniques use a sheet of validation of media experts and materials. Test of media validity of make-match puzzle based media by media expert and material expert of validity result shows that the level of validity of make-match based puzzle that has been developed reaches 73,82% from facet of valid category media and 87,38% in terms of material expert Category is very valid with the average validity of media experts and material experts reached 80.6% it shows that the validity level of make-match media based puzzle as a learning medium is valid and ready to be utilized in actual learning activities. Based on the results of the research, the learning media based on puzzlemake a match puzzle based on the sub-subject of the developed cell is declared valid to be applied as a learning medium.

Abstract

In the background exposure obtained at the observation stage shows the need for the development of other learning media that can improve students' motivation and interest to the need for development of discovery learning biology handdout on the material interaction of living creatures with the environment, this development aims to develop a learning media in the form of handout based on discovery learning that is interesting and can motivate students in learning and know the level of feasibility (valid, effective and practical) development as a biology learning media on the material interaction of living creatures with the environment in SMP Muhammadiah 2 Palang.Penelitian this is the type of research development or Research and Development (R & D) with development method using ADDIE model. This model has 5 stages: analysis, design, development of trial and evaluation. To test the feasibility of validation. Product validation is done by lecturers of material experts and instructional media to get input and suggestions on the feasibility of handouts in terms of material or media. And the results showed that the feasibility level of discovery learning biology based handout is declared valid, practical and effective

Abstract

Implementation of the learning process at SMAN 1 KEREK still uses conventional approach based learning. Learning methods such as lectures and question and answer still apply to the school. If this conventional approach-based learning is still applied in the modern era, it is feared that it will affect student's learning outcomes with 65% of the learning completeness score and KKM (70) which means that KKM has not reached the value of KKM. The currently used Student Worksheet is still simple in terms of materials, drawings and language used, so the need for the development of Student Worksheet (LKS) based on the Scientific Approach.In this environmental change and waste recycling materials LKS development uses Borg and Gall development methods. The purpose of this research is to find out the result of validity test of Student Worksheet based on scientific approach on environmental change and waste recycling materials and to improve student learning result SMAN 1 KEREK. The instrument used for this research is the questionnaire. Validity test results obtained from media experts with an average value of 83.1%, and from material experts obtained an average of 84% with criteria very valid and fit to be used as teaching aids, thus from the validity test results can be concluded that product development of Student Worksheet (LKS) based on biological approaches scientifically feasible to use and able to improve student learning outcomes SMAN 1 KEREK.

Abstract

Parasitic nematodes disease is one of impediment to the increasing rate of population and livestock production. Infection prevalence study of egg-worm parasitic nematodes established using 91 samples of cattle feces on February 11th, 2014 at Napis village, Bojonegoro. Feces examination was performed at Parasitology Laboratory , Animal Health Laboratory Unit-Animal Husbandry Department of East Java Province in Tuban by Whitlock method. Observed parameters include the type and infection prevalence of egg-worm parasitic nematodes that was infect, the data obtained were analyzed descriptively. The result showed 42 (46,15 %) of the total sample was single infection by one type of nematodes egg-worm. Worms that infect were Strongyle sp. 35 (38,46 %), Trichuris sp. 2 (2,19%) and Nematodirus sp and Strongyle sp  1 (1.09 %), Strongyle sp and Trichuris sp 4 (4,39%)The conclusion of this study is the prevalence of the highest infection of egg-worm parasitic nematodes as much as 38,46 % by Strongyle sp.and lowest prevalence as much as 1.09 % by Nematodirus sp and Strongyle sp. Key words : Infection prevalence, cattle, nematodes egg-worm, Whitlock

Abstract

Diseases affecting banana plants are very detrimental to farmers as these can lower production and economic income. The purpose of this study was to determine the type and intensity of the disease affecting banana plants. This research was an observational analytic study that observe and analyze condition or symptoms of diseases affecting banana plants in Tegalagung village, Semanding subdistrict, Tuban as many as 38 samples. Parameters observed were type of disease and measure intensity of the disease, data obtained were analyzed descriptively. Based on the symptoms that occurred on the leaves, the study found four disease types affecting banana plant that were fusarium wilt, bacterial wilt (Blood), Sigatoka leaf spot and stunting disease. The diseases intensity were 50% of Fusarium wilt; 26,66% of bacterial wilt (Blood); 26.32% of Sigatoka leaf spot and 15.38% of stunting disease. Conclusion of the study, the highest intensity of the disease that attacks banana plants is Fusarium wilt as high as 50%.
