Abstract

Tuberkulosis adalah penyakit   infeksi   menular   yang   disebabkan   oleh bakteri Mycobacterium  tuberculosis. Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak akar parang romang terhadap Mycobacterium tuberculosis. Sampel di ekstraksi menggunakan pelarut etanol 96%, kemudian dibuat deret konsentrasi yaitu 250 ppm, 500 ppm, 750 ppm, dan 1000 ppm. Uji penghambatan pertumbuhan Mycobacterium tuberculosis menggunakan metode MODS pada bakteri Mycobacterium tuberculosis strain H37RV. Hasil uji penghambatan  pertumbuhan Mycobacterium tuberculosis menunjukkan bahwa konsentrasi 250 ppm masih mampu menghambat pertumbuhan Mycobacterium tuberculosis strain H37RV.

Abstract

Penelitian ini bertujuan untuk menentukan aktivitas anti tuberkulosis ekstrak etanol biji beligo (Benincasa hispida (Thunb) Cogn) terhadap Mycobacterium tuberculosis strain H37RV dengan metode Microscopically Observed Drug Susceptibility. Metode penelitian meliputi ekstraksi biji beligo dengan cara maserasi menggunakan cairan penyari etanol 96%, kemudian dibuat menjadi larutan uji dengan konsentrasi 250 ppm, 500 ppm dan 750 ppm. Aktivitas antituberkulosis menggunakan metode Microscopically Observed Drug Susceptibility dan diidentifikasi dengan presentase pertumbuhan Mycobacterium tuberculosis strain H37RV pada microskop flurosence. Hasil penelitian menunjukkan ekstrak biji beligo (Benincasa hispida (Thunb) Cogn) dapat menghambat pertumbuhan Mycobacterium tuberculosis paling baik pada konsentrasi 750 ppm.

Abstract

Boehmeria virgata (Forst) Guill berasal dari keluarga Urticaceae, spesies ini mengandung metabolit sekunder dalam bentuk alkaloid, terpenoid, fenolik dan flavonoid. Penelitian ini bertujuan untuk mengetahui toksisitas fraksi fraksi ekstrak etanol Akar Parang Romang (Boehmeria virgata (Forst) Guill.) dengan metode Brine Shrimp Lethality Test terhadap Artemia salina Leach. dan mengidentifikasi golongan senyawa dari fraksi aktif. Sampel akar diekstraksi dengan metode maserasi menggunakan pelarut Metanol. Ekstrak kemudian dipekatkan dengan Rotary evaporator. Ekstrak yang diperoleh selanjutnya dipartisi terlebih dahulu dengan menggunakan pelarut n-Heksan sehingga diperoleh ekstrak larut n-Heksan dan ekstrak tidak larut n-Heksan, selanjutnya ekstrak yang tidak larut n-Heksan dipartisi kembali menggunakan pelarut Etil Asetat sehingga diperoleh ekstrak larut Etil Asetat dan tidak larut Etil Asetat, kemudian ekstrak tidak larut Etil Asetat dipartisi dengan pelarut Etanol 70% sehingga diperoleh ekstrak larut Etanol 70%. Ekstrak Etanol 70% kemudian difraksinasi dengan kromatografi kolom cair vakum sehingga diperoleh 4 fraksi gabungan yaitu fraksi A, B, C, D. Masing-masing fraksi diuji toksisitasnya dan diperoleh hasil fraksi B yang memiliki tingkat toksik yang lebih besar di bandingkan fraksi lainnya dengan nilai LC50 sebesar 16,21 μg/ml, selanjutnya fraksi B difraksinasi kembali menggunakan kolom kromatografi cair vakum dan diperoleh 3 fraksi gabungan yakni fraksi B1, B2 dan B3, setelah masing-masing fraksi diuji toksisitasnya maka diperoleh hasil fraksi B1 yang memiliki toksisitas yang lebih besar dengan nilai LC50 sebesar 2,19 μg/ml.  Hasil identifikasi fraksi B1 menunjukkan adanya golongan senyawa Alkaloid dan terpen.

Abstract

This research aims to determine the faction activity of n-hexane, ethyl acetat, and 70% beligo seeds (Benincasa hispida Thunb. Cogn) on the decrease in blood glucose levels in mice. The research method includes extraction of samples by macerasion using 96% liquid of ethanol filter. This research used 25 male mice as he test animals which were divided into 5 treatment groups. Mice were fasted for 8 hours before treatment, then measurement of initial glucose levels, after that they were induced with 10% glucose and glucose levels were measured after induction. Group I was given 0,5% Na-CMC suspense as negative control, group II was given the n-hexane fraksion, group III was given ethyl acetate fraction, group IV was given 70% ethanol fraction and group V was given 0,013 mg/20 g BB glibenclamide suspension as positive control. Giving is done perorally with a volume of 1 ml. The results showed that the faction of n-hexane, ethyl acetate, and 70% ethanol extracts of beligo seeds (Benincasa hispida Thunb. Cogn) with a dose 28 mg/20 g BB had activity in decreasing blood glucose levels in mice (Mus musculus).

Abstract

Penelitian ini bertujuan untuk membandingkan metode ekstraksi terhadap kandungan flavonoid total dan aktivitas antioksidan ekstrak batang Boehmeria virgata. Batang Boehmeria virgata diekstraksi melalui dua metode ekstraksi yaitu refluks dan maserasi. Kandungan flavonoid total ditentukan dengan menggunakan metode aluminium klorida dan dihitung sebagai ekivalen kuersetin (QE). Aktivitas antioksidan diukur dengan menggunakan spektrofotometer UV-VIS. Hasil penelitian menunjukkan aktivitas antioksidan (Uji DPPH) ekstrak Boehmeria virgata (Forst) Guill yang diperoleh melalui metode refluks lebih baik daripada dengan metode maserasi (IC50 pada 30,58 μg /mL) karena menunjukkan kandungan flavonoid lebih tinggi (sampel 2,554 mg QE / g).

Abstract

Telah dilakukan penelitian mengenai analisis kadar fenolik dan flavonoid total ekstrak daun anggur (Vitis vinifera L) dengan metode Spektrofotometri UV-Vis. Penelitian ini bertujuan untuk menentukan kadar fenolik dan flavonoid total dalam ekstrak etanol daun anggur. Sampel diekstraksi dengan metode maserasi menggunakan pelarut etanol. Penentuan kadar fenolik total ekstrak menggunakan pembanding asam galat dengan beberapa variasi konsentrasi. Penentuan kadar flavonoid total menggunakan pembanding kuersetin. Nilai absorbansi diukur dengan spektrofotometer. Nilai absorbansi kemudian dimasukkan ke dalam persamaan regresi linier. Hasil penelitian menunjukkan kadar total fenolik dan kadar total flavonoid dalam ekstrak berturut-turut adalah sebesar 95,28 mg GAE/g dan 4,07 mg QE/g.

Abstract

Study about physicochemical properties of olein and stearin fraction of palm oil has been conducted. Research aim was to determine the chemical physical properties of the olein and stearin fractions of palm oil (Elaeis guineensi Jacq) that was originated from North Mamuju, West Sulawesi. Palm oil was fractionated by dry fractionation method to collect the olein and stearin fractions. The physical chemical characteristics of fractions were determined by acid value, saponification value, peroxide value, and iodine value parameters that measured by titrimetry method. The result of analysis showed that the acid values for the olein and stearin fractions acid were 7.04 and 5.83 mg NaOH/g, saponification values were 198.53 and 194.71 mg KOH/g, peroxide values were 0.77 mek O2/kg and 0.79 mek O2/kg, and iodine values were 34.12 and 32.87 g iod/100 g.

Abstract

Botto-botto (Chromolaena odorata L.) was one of plants that its leaves used empirically in South Sulawesi society as wound and anti-inflammatory drugs. This study aimed to determine the potential of Botto-botto toward cancer cells (HeLa Cells) and normal cells (Vero cells) using the MTT assay method (Microculture Tetrazolium Salt Method). Ethyl acetate extract from methanol extract being fractionated by vacuum liquid chromatography method by a different polarity level of solvent. Cytotoxic effect test was using MTT method to cancer cells (HeLa), and normal cells (Vero) with the percent parameter of cell growth inhibition. Selectivity index was determined from percentage of cancer cell growth inhibition ratio of cancer cells to normal cells. The results showed that A, B, C, D, E, F, and G fractions had a selectivity index on HeLa cells at 0.845, 3.387, 2.004, 1.508, 1.625, 1.284, 1.299. This result proved that B fraction of methanol extract of Botto-botto leaves had a potential to be developed as chemotherapy agents in increasing the effectiveness treatment of cervix cancer.

Abstract

Parang Romang (Boehmeria virgata (Forst.) Guill) is one of the plants empirically used by the people of South Sulawesi. This study aimed to test the activity of Parang Romang Extract against Mycobacterium tuberculosis. The 96% ethanol extract obtained was fractionated by the liquid-solid extraction method: obtained n-Hexan soluble extract and n-Hexan insoluble extract. Samples of 96% ethanol extract, soluble extract of n-Hexan and insoluble extract of n-Hexan were made in series of concentrations of about 25, 50, 75, and 100 µg/ml. The inhibition test of Mycobacterium tuberculosis used the MODS method on the Mycobacterium tuberculosis strain H37RV. Color reagents then identify extracts that provide inhibition. The results of the minimum inhibition concentration of Mycobacterium tuberculosis showed that a 100 µg/ml of n-Hexan soluble extract was able to inhibit the growth of Mycobacterium tuberculosis strain H37RV.
