Abstract

This study aims to determine: (1) the optimum conditions for α-amylase enzyme activity with potato starch as a substrate including incubation time, pH, temperature, substrate concentration, and enzyme concentration, (2) the effect of Co2+ metal ions in the form of CoCl2 compounds on α-amylase enzyme activity with potato starch substrate at optimum conditions. The determination of α-amylase enzyme activity with potato starch substrate was carried out using the dinitrosalicylic acid (DNS) method. The determination of α-amylase enzyme activity with and without the addition of CoCl2 was carried out at the optimum conditions that have been obtained. The variation in the added concentration of CoCl2 were 0.01; 0.02; 0.03; 0.04; and 0.05 M. The data obtained in this study were enzyme activity in units of mg/mL/minute at 37° C. The data analysis used was descriptive qualitative by comparing the activity of the α-amylase enzyme with and without the addition of CoCl2 at optimum conditions. The results of the average α-amylase enzyme activity at optimum conditions (temperature 37° C, pH 7.2, incubation time 15 minutes, substrate concentration 20 mg/mL, and enzyme concentration 40 mg/mL), namely 0.00794 mg/mL at temperature 37° C. For the mean α-amylase enzyme activity with the addition of CoCl2 at a concentration of 0.01; 0.02; 0.03; 0.04; and 0.05 M, respectively 0.0038; 0.0013; 0.0012; 0.0011; and 0.00031 mg/mL per minute at 37° C. Based on these data it can be concluded that the tendency of the Co2+ metal ion is inhibitory and greatly affects the activity of the α-amylase enzyme.

Abstract

Abstrak: Pendidikan merupakan salah satu jalan keluar untuk membina karakter generasi muda secara terarah, terprogram, dan optimal agar dapat terbentuk generasi muda yang cerdas intelektual dan berkualitas akhlaknya. Percaya diri adalah karakter yang penting ditanamkan agar mereka menjadi generasi yang tidak mudah dipengaruhi hal-hal negatif di sekitarnya, optimis, dan tegar dalam menghadapi berbagai masalah dengan kemampuannya sendiri. Karakter keingintahuan diharapkan peserta didik suka pada tantangan, berinovasi dan kreatif menciptakan sesuatu yang dapat membanggakan dirinya, keluarga, dan negara. Jiwa wirausaha penting ditanamkan dan dikembangkan dalam diri peserta didik, mengingat di era globalisasi saat ini sangat diperlukan adanya SDM yang mampu bersaing. Karakter seperti ini tidak muncul serta merta, tetapi harus dibina secara dini. Penanaman nilai karakter yang terintegrasi dalam semua mata pelajaran dapat diharapkan nilai-nilai tersebut terinternalisasi dalam diri peserta didik. Materi yang dibelajarkan tidak hanya sebagai pengetahuan sekolah, tetapi juga menjadi pengetahuan dalam diri yang akhirnya ditunjukkan dalam bentuk perilaku. Abstract

AbstractThe school-based curriculum gives teachers freedom to design and develop alllearning activities for students by themselves, so it is expected that the teacherscompetence can be improved optimally. Besides, teachers creativity andinnovation can be developed to increase their professionalism. But, the fact showsthat the teachers responsibility is so heavy that they do not have enough time todevelop their professionalism. Almost all SMPs/MTss and SMAs/MAs inIndonesia assign teachers some duties based on classes. One of the ways toimprove teachers mastery can be done by taking specialization in teachingmaterials. This method gives some advantages to teachers and students as well. Forthe teachers, specialization in teaching materials can make them: (1) concentrate onthe teaching material development; (2) plan learning strategies effectively; (3) lookfor references easily and attentively; (4) write summaries and modules; (5) selectseminars and workshops to attend; and (6) direct students based on their ability.For the students, specialization in teaching materials can make them: (1) get theunity of concepts from the same teacher; (2) avoid needless character adaptationfrom different teachers; (3) detect self-ability; (4) look for references easily; and (5)avoid from learning over-lapping concepts. It seems difficult to implementspecialization for teaching material, but by having a good intention to increaselearning quality in Indonesia, all difficulties and obstacles can be handled. Patienceand seriousness are really needed to start a new thing. Whatever the result is, thiseffort should be made.Keywords : specialization, effort, professionalism, teacher

Abstract

AbstractThe Relevance of the Standards of Competence and Basic Competence inNatural Science Subjects to the Development of Ideational Learning in Year VII ofPilot International Standard Junior High Schools in Yogyakarta Special TerritoryProvince. This study aims to investigate the relevance of the standards ofcompetence and basic competence to the development of ideational learning inPilot International Standard Junior High Schools (PISJHSs). This study wasconducted in Yogyakarta Special Territory Province and employed a surveymethod involving 36 teachers of Year VII of PISJHSs. The survey instrument was aquestionnaire that the researchers developed. The results show that all standards ofcompetence and basic competence for Year VII students in semesters 1 and 2 arecapable of developing ideational learning. Therefore, it is not necessary to add newstandards of competence and basic competence to develop ideational learning.However, it is still necessary to add indicators leading ideational learning to thebasic competence in the syllabus of the school-based curriculum in PISJHSs.Keywords: standards of competence, basic competence, ideational learning

Abstract

Air limbah industri yang dikeluarkan oleh proses-proses industriberbeda-heda dalam jumlah maupun kekuatan pencemaranoya,sesuai dengan satuan operasi atau proses yang merupakansumbernya. Dalam Ungkup sempit. air limbah buanganyang dapat dapat mengakibatkan pencemaran antara lain:buangan dari laboratorium kimia, yang di dalamnya terdapatlogam-Iogam herat, seperti air raksa, arsen. .Logam herat tersebut dapat berubah menjadi suatusenyawa metil dari logam herat yang sangat berbahaya bagikesehatan, baik dalam bentuk gas maupun cab". Proses 1nlterjadi dengan bantuan bakteri dalam kondisi anaerob karenadi dalam bakteri ada suatu koenzim metilkobalamin. Mekanismeperpindahan metil menjadi suatu senyawa metiI darilogam berat yang utama melalui: serangan elektrofilH: Fadaikatan Co-C (metilkobalamin) atau perpindahan CH3-, danserangan radikal bebas (CH3.).Agar hasil dari reaksi' di a tas tidak terjadi (da pa t dikurangi)diperlukan suatu teknik pembuangan, proses pengolahansedemikian rupa sehingga proses metilasi oleh bakteritidak dapat berlangsung. Misalnya, penghilangan ion-ionlogam berat sebelum dibuang ke lingkungan.

Abstract

The aim of this study was to know whether there was any positive and significant relationship between the ability of effectively listening and recording learning material and the chemistry study achievement of students of MAN Maguwoharjo Yogyakarta. The study was designed as of one sample and three variables. The population was designated to be all the students of class X semester 2 MAN Maguwoharjo  Yogyakarta it the academic year of 2006/2007 consisting of 96 students. Sixty-four students were taken as the subjects by cluster random sampling for two classes (class XB and XC). The free variable was effective listening and recording in chemistry classes, and the binding variable was chemistry study achievement of the students. Three instruments were utilized; namely a questionnaire of listening ability, a value sheet of the recording books, and a test of chemistry study achievement. The data were analyzed by double regression and yielded an Ry(1,2) of 0.546. The first and second grade correlation coefficients (r1-2) were 0,393 and 0.369, respectively. The significance test yielded a value of F of 12,933 against the F­ table of 3.148 on the significance level of 5% with the df of 2 versus 61. The statistics  showed  that  there was  a positive  and  significant  relationship between the ability of effectively listening and recording of chemistry matter, either separately or together, on the chemistry study achievement  of the students. The relative contributions of the ability of effective listening on the chemistry study achievement were 53,05% and 15,80, The effective contributions of the ability of effective recording on the chemistry study achievement were 46,95% and 13,98%.

Abstract

Penelitian ini bertujuan untuk mengembangkan dan memperoleh Instrumen Pendeteksi Miskonsepsi Ikatan Kimia (IPMIK) untuk peserta didik SMA. Instrumen penelitian berupa tes pilihan ganda dengan alasan setengah terbuka. Produk awal (IPMIK I) yang telah dikembangkan divalidasi melalui expert jugdment dengan menggunakan teknik FGD yang melibatkan ahli kimia, pendidikan kimia, miskonsepsi, dan psikometri serta reviewer (dosen kimia dan guru kimia SMA di DIY). Masukan dalam FGD digunakan untuk merevisi instrumen menjadi produk IPMIK II. Selanjutnya, instrumen inti soal divalidasi empiris pada peserta didik SMA di DIY. Hasil uji coba dianalisis IRT tiga parameter untuk menentukan butir-butir soal yang valid. Penelitian ini telah berhasil mengembangkan IPMIK untuk peserta didik SMA yang telah divalidasi empiris terhadap 811 peserta didik SMA kelas XI dari 13 SMA di DIY dan diperoleh 20 butir soal yang valid (produk IPMIK III) berdasarkan analisis IRT tiga parameter

Abstract

The objectivity of this research is to determine media that effective to studying in biochemistry practical work to improve student achievement.The sample of this research is all students of biochemistry practical work at odd semester in 2003/2004 in Chemistry Department in Faculty of Mathematics and Natural Science, at Yogyakarta State University. The student’s number was 28 people. The research was carried out using 3 media, i.e. Over Head Transparent presented by Over Head projector (OHP) at carbohydrate topic; Power Point presented by LCD projector at protein topic, and Audio-Visual presented by LCD projector at enzyme topic. The instruments of this research are observation sheet, pre-test and post-test, and evaluation questionnaire. The increase of post-test to pre-test was evaluated using t-test. The analyzed of observation data was carried out by qualitative descriptive using calculation percentage continued by qualitative analyzed. The mean of observation data was analyzed by ANAVA-A test followed by LSDT test. The data of evaluation questionnaire was calculated as percentage of student choice at this questionnaire.The result of observation data showed that there was the increasing in practical work skilled using OHT, Power Point, and Audio Visual media. There were 18.16 (very low), 36.39 (medium); and 49.31 (very high) respectively. The ANAVA-A test and LSDT indicated that there was significant difference between three score of the practical work skilled with three different media at three topics. The result of pre-test and post-test showed that there was the increasing score for all topics. The great increasing was enzyme topic when using Audio-visual media. The t-test indicated that there was the differentia of the increase score at three topics. The evaluation questionnaire showed that Audio-Visual media presented by LCD was effective

Abstract

The aim of the research is to study  the effect of the duration of fermentation process toward the content of soluble protein in tempe from soybean, and the activity of trypsin enzym toward that protein.The content of soluble protein was analyzed by Lowry method using casein as solution standard, and the tripsin activity was determined by Anson method. The duration offermentation process were 0,24,48,72, 96, and 120 hours. The research indicated that the content ofsoluble protein in tempe duringfermentation process for 0, 24, 48, 72,96, and  120 hours were 0.172;  0.212; 0.217; 0.212; 0.197, and0.158% (wlw) respectively, and the activity of trypsine toward the that soluble protein were 1.35; 2.33; 2.73; 2.13; 1.17 and o. 78 unit. It was indicated that the duration of fermentation process  influenced thecontent of soluble protein and tripsin activity. The highest of trypsin activity was infermentation process for 48 hours.
