Abstract

Tabat Barito (Ficus deltoidea Jack.) plant has secondary metabolite compounds and several bioactivities. This study was conducted to test the allelopathic activity and phytotoxicity of methanol extract of Tabat Barito leaves against Pletekan weed (Ruellia tuberosa L.) The study used a completely randomized design (CRD) in two experimental stages, namely the seed germination stage and the plant growth stage, using the concentration of methanol extract of Tabat Barito leaves: 0%, 25%, 50%, and 75%. The results showed that the methanol extract of F. deltoidea leaves was able to inhibit the germination process of R. tuberosa weed seeds. The highest allelopathic power and phytotoxicity were produced at a concentration of 75%, which is significantly different from the other treatments. At the growth stage, the methanol extract of F. deltoidea reduced biomass (wet weight 76%; dry weight 67%), chlorophyll a 50%, chlorophyll b 50%, and total chlorophyll by 71%. The higher the F. deltoidea leaf methanol extract concentration, the higher the allelopathic power and phytotoxicity against weed growth. The study showed that the methanol extract of F. deltoidea leaves was able to inhibit weed germination and growth and had strong phytotoxicity power against Pletekan weed, so F. deltoidea leaf extract has the potential to be used as a source of allelopathy or natural herbicide ingredients (bio-herbicides) that are environmentally friendly substitutes for chemical herbicides against weed plants.

Abstract

Green synthesis of Silver nanoparticles (AgNPs) using plant-based materials is an environmentally friendly path for the synthesis of silver nanoparticles instead of traditional chemical methods. This study presents the first biosynthesis of AgNPs using an ethanolic extract obtained from in vitro callus cultures of Myrmecodia tuberosa Jack, an endangered medicinal epiphyte and conservation‑friendly biotechnological platform. Cotyledon explants were successfully induced high-frequency friable, green callus and the ethanolic extract acted as reducing and stabilizing agent. UV‑Vis spectroscopy revealed surface plasmon resonance peaks at 420–450 nm with a visible color change from light brown to dark brown, confirming the AgNP formation. The most intense and well-defined SPR peak indicating optimal nanoparticle synthesis was observed after 6–8 days static incubation at 35°C using 2 mM AgNO₃. In the DPPH radical scavenging assay, it could moderate antioxidant activities, with IC₅₀ = 51.80±0.035 μg mL⁻¹) which was more than that of parent callus extract (IC₅₀ = 58.91±0.097μgmL⁻¹). This improvement is probably a result of synergistic effects between silver core and phytochemical capping layer. These results indicate that callus cultures of M. tuberosa is an environmentally friendly, sustainable and conservation-friendly source of reducing metabolites for the preparation of green nanomaterials applicable in biomedical practices.

Abstract

Rendahnya kompetensi guru dalam menggunakan mikroskop, pembuatan preparat dan pengamatan sel akibat keterbatasan fasilitas dalam pelaksanaan praktikum IPA, menjadi kendala di sebagian sekolah di Samarinda, termasuk SMP Negeri 5 Samarinda. Tujuan pengabdian masyarakat ini dilaksanakan adalah untuk meningkatkan kompetensi guru IPA dan siswa dalam penggunaan mikroskop, pembuatan preparat, dan pengamatan sel. Kegiatan dilaksanakan dengan metode pelatihan (training) diikuti oleh 45 peserta yang terdiri dari 8 guru IPA dan 37 siswa, dilaksanakan melalui empat tahapan: tahap awal persiapan dan pretest, pembekalan teori, praktikum, dan tahap akhir posttest serta evaluasi kegiatan. Evaluasi hasil posttest menunjukkan terjadi peningkatan signifikan dalam pengetahuan dan pemahaman peserta, dari kategori "sangat kurang" dan "kurang" (19,56% dan 13,04%) pada pretest, menjadi "sangat baik" dan "baik" (34,78% dan 36,95%) pada posttest. Evauasi kegiatan, penilaian terhadap pemaparan materi dan kesesuaian antara teori dan praktik mencapai kategori sangat baik dengan persentase 80% dan 94%. Program pengabdian masyarakat ini terbukti efektif dalam meningkatkan kompetensi penggunaan mikroskop, pembuatan preparat, dan pengamatan sel, yang berkontribusi positif terhadap pembelajaran biologi di sekolah.
