Abstract

Antimicrobial peptide (AMP), as a new antibiotic agent, has promising prospects in overcoming the problem of resistance. AMP production can be carried out by proteolytic enzymes. Protein from castor bean (R. communis) is toxic, so it can potentially be a source of AMP. This study aims to obtain protein hydrolysate from the castor bean (R. communis) using several extraction methods, including SDS dialysis, SDS-gel filtration, trifluoroacetic acid (TFA), and acetone precipitation. Chymotrypsin enzyme was used to hydrolyze the protein, and the peptide hydrolysate was fractionated using RP-HPLC. The peptide fraction was tested for its antibacterial activity by agar diffusion and microdilution methods, and the most active fraction was identified for its amino acid sequence by LC-HRMS. The results showed that the acetone precipitation extraction method was the best method, with a degree of hydrolysis of 83.9%. The active fractions 6 and 10 of RP-HPLC had IC50 values of 14.1 and 14.5 µg/mL for E. coli and 13.3 and 14.4 µg/mL for S. aureus, respectively. NVLRGKGMASL peptides were found in fraction 10, and GIILLSSK, NMIAKR, and LLDILTKK peptides were found in fraction 6 with an alpha helix secondary structure that can cause membrane damage. The peptides NVLRGKGMASL, GIILLSSK, NMIAKR, and LLDILTKK are thought to have potential as antibacterial compounds.

Abstract

The regulation of proliferation and apoptosis in cancer cells, particularly breast cancer, hinges on the control exerted by the epidermal growth factor receptor (EGFR). Naturally derived EGFR inhibitors from natural compounds show significant potential for future advancements. This study aims to assess the efficacy of active compounds found in Ageratum Conyzoides L as EGFR inhibitors. Evaluation involved molecular docking studies of flavonoid, chromene, terpene, sterol, and acid compounds against EGFR using PDB ID 1m17. Additionally, the ADMET properties and drug-like characteristics of the most promising compounds were characterized. Findings reveal that compound C2 (fisetin) exhibits the most favorable binding energy of -8.9 kcal/mol. Fisetin establishes crucial interactions, particularly with the catalytic site at Met765 of the hinge protein. Fisetin also engages in hydrophobic interactions in regions I and II, involving Val702, Leu694, and Leu820. ADMET profiling of fisetin demonstrates favorable attributes, suggesting its potential as a promising anticancer agent based on drug-likeness assessments.

Abstract

ABSTRACT. Compound of N-phenyl-3-(4-chlorophenyl)-5-(3,4-dimethoxy-phenyl) pyrazoline was synthesized by cycloaddition reaction. The structure of the product was chracterized by FTIR, 1H-NMR and 13C-NMR spectrometers. The pyrazoline was screened for antibacterial activity using agar diffusion method against Staphylococcus aureus, Bacillus cereus, Bacillus subtillis, Shigella flexnerri, and Escherichia coli. The pyrazoline had been synthesized from chalcone with phenylhydrazine to give 52.90% yield. Antibacterial screening showed that pyrazoline was active against selected gram positive bacteria. The pyrazoline was found to exhibits an antibacterial activity and its zone of inhibition/concentration against S. aureus (8.75/300), B. cereus (7.75/1000), B. subtillis (6.50/1000), and E. coli (3.50/300). The result showed that pyrazoline which was subtituted with chloro and methoxy, was able to posse broad spectrum of the tested pathogenic bacteria. Keywords: chalcone, pyrazoline, antibacterial

Abstract

Reaksi metilasi adalah reaksi penggantian atom atau molekul dengan gugus metil. Beberapa parameter yang mempengaruhi proses metilasi antara lain rasio bahan dan waktu metilasi, namun hasil metilasi tidak optimal. Pada penelitian ini dilakukan penambahan katalis pada proses metilasi. Penambahan katalis berpengaruh terhadap peningkatan stabilitas warna ekstrak secang terhadap perubahan pH. Variabel yang digunakan dalam penelitian ini adalah konsentrasi katalis 0; 0,002; 0,01; 0,02; dan 0,04 mol/mL. Parameter yang diuji meliputi stabilitas terhadap pH (2, 3, 5, 7, dan 10) dan degradasi warna terhadap oksidator. Konsentrasi katalis penambahan terbaik adalah pada kisaran 0,002 hingga 0,01 mol/mL. Selain itu, adanya katalis NaOH dalam proses metilasi ekstrak secang juga dapat mengurangi degradasi warna yang disebabkan oleh adanya oksidator.
