Abstract

Highlights:1. This study compared the serological markers of pristane-induced mice to humanized mouse models of lupus achieved by transplanting stem cells from lupus patients, which is a novel method in Indonesia.2. This study will allow for more accurate research into the pathophysiology of the disease and the development of new lupus treatment strategies.   Abstract More studies related to systemic lupus erythematosus (SLE) therapy are urgently needed because of the current insufficiency in treatment effectiveness. However, due to ethical limitations, researchers use experimental animals as a substitute for conducting studies on humans. Models commonly used to study lupus include the pristane-induced mouse model and the recently developed humanized mouse model. The second model involves implanting human immune cells into immunodeficient mice. This study compared the serologic profiles of lupus antibodies, the antinuclear antibodies (ANA) and anti-double stranded DNA (anti-dsDNA), in both mouse models. The aim was to determine which one is more promising for use as a lupus animal model. Thirty BALB/c mice (Mus musculus) were used as subjects and divided into three groups: K1, K2, and K3. K1 served as the control group, consisting of healthy mice that received a placebo. The K2 mice were intraperitoneally injected with 0.5 cc of pristane. The K3 mice were transplanted with stem cell cultures from SLE patients, resulting in humanized mice with immune deficiencies. The mice were observed for 16 weeks, during which the ANA and anti-dsDNA levels in their serum were obtained for analysis using the Kruskal-Wallis test (p<0.05). The comparison revealed differences in the average ANA and anti-dsDNA levels among the three groups. K3 had the highest ANA and anti-dsDNA levels, followed by K1 and K2. The Kruskal-Wallis test indicated that the differences were not significant in the mean levels of ANA (p=0.156) and anti-dsDNA (p=0.061). In conclusion, the humanized mouse model has higher ANA and anti-dsDNA antibody levels compared to the pristane-induced mouse model, albeit without a significant difference. This suggests a positive picture of the humanized mouse model of lupus, making it an invaluable tool for studying the disease and testing potential therapeutic interventions.

Abstract

Latar Belakang: Sindroma lisis tumor adalah sekumpulan gejala metabolik akibat terlepasnya subtansi intraselular kedalam sirkulasi darah. Kriteria Cairo dan Bishop digunakan untuk menegakkan sindroma lisis tumor. Pasien dengan keganasan hematologis seperti leukemia akut sering mengalami sindroma lisis tumor, begitu juga dengan komplikasi infeksi. Evaluasi sindroma lisis tumor, khususnya mengetahui parameter laboratorium yang dapat digunakan serta komplikasi yang dapat muncul seperti infeksi merupakan hal yang penting untuk dipahami. Laporan Kasus: Anak laki-laki berusia 2 tahun menjalani pengobatan kemoterapi rumatan untuk LLA L2 yang di diagnosa sejak kurang lebih 1 tahun lalu. Pasien datang tanpa keluhan, namun mengalami perburukan kondisi seiring berjalannya kemoterapi. Diapatkan imbalans elektrolit serta gejala klinis yang sesuai dengan gambaran sindroma lisis tumor. Pada saat perawatan pasien juga menderita pneumonia, infeksi saluran cerna serta sepsis (skor PELOD 15). Kesimpulan dan Saran: Pasien mengalami sindroma lisis tumor saat sedang menjalani proses kemoterapi rumatan. Meskipun pasien datang tanpa keluhan, namun terdapat hiperurisemia dan peningkatan kreatinin yang mungkin dapat dipertimbangkan sebelum inisiasi kemoterapi. Sepsis pada pasien ini mungkin disebabkan oleh bakteri pada saluran cerna mengingat neutropenia berat yang dialami.

Abstract

Kurangnya pengetahuan masyarakat tentang COVID-19, lupus, reumatik, dan alergi mempengaruhi keberhasilan terapi, penurunan angka kejadian, dan pencegahan kekambuhan. Tujuan pengabdian masyarakat adalah membangun desa-desa binaan tanggap COVID-19, lupus, reumatik, dan alergi di Malang oleh Tim Kelompok Kajian Lupus, Autoimun, Reumatik, dan Alergi (LAURA) Universitas Brawijaya. Warga desa binaan diberikan penyuluhan berupa seminar awam dan pelatihan tentang pertolongan awal pada penyakit COVID-19, lupus, reumatik, dan alergi, kemudian diminta mengisi kuesioner posttest untuk mengukur pemahaman. Desa-desa binaan diberikan thermo-gun dan wastafel untuk menerapkan protokol kesehatan. Satu bulan kemudian dievaluasi adanya kejadian COVID-19, lupus, reumatik, dan alergi di desa binaan. Hasil evaluasi menunjukkan rata-rata warga desa binaan memahami 78,3% materi yang diberikan dan menerapkan protokol kesehatan sesuai yang diajarkan saat penyuluhan. Dilaporkan tidak ada kejadian COVID-19 serta kekambuhan lupus, reumatik, dan alergi dalam satu bulan terakhir kegiatan. Kesimpulan: pembangunan desa-desa binaan di Malang meningkatkan tanggap warga terhadap COVID-19, lupus, reumatik, dan alergi.

Abstract

Background: Tumor lysis syndrome is metabolic abnormalities due to accumulation of intracellular contents in systemic circulation. Cairo and Bishop criteria is used to diagnosing it. Patient with hematological malignancies such as acute leukemia is more prone to this syndrome, and also infection that comes with it. Evaluating tumor lysis syndrome, especially knowing which laboratory parameters used and probable complication are crucial. Case Report: Two years old boy undergone maintenance chemotherapy for his ALL L2 that has been diagnosed one year ago. Patient has no complaint at admission, but went to worsened condition as the chemotherapy was given. Electrolyte imbalances and clinical manifestations depicting tumor lysis syndrome was found. Patient also experiencing pneumonia, gastrointestinal tract infection and sepsis (PELOD score 15). Conclusion and Suggestion: Patient experiencing tumor lysis syndrome when he was going through maintenance chemotherapy. Even though he was admitted without any major complaints, but hyperuricemia and elevated creatinine at the time of admission should be considered before initiating chemotherapy. Sepsis in this patient might be cause by bacterial infection of the gastrointestinal tract due to his severe neutropenia condition.

Abstract

Diabetes mellitus has raised concerns about structural and functional changes in blood cells, as well as changes in blood impedance. The purpose of this study is to investigate the relationship between electrical impedance values measured with the Bioelectrical Impedance Analyzer (BIA) in diabetes patients. Blood samples were collected from 5 healthy people and 5 diabetes patients for the study. Electrical impedance testing with a BIA. The impedance method determines the electrical properties of blood by measuring its resistance and reactance at various frequencies. The results of the BIA are analyzed and compared to body health parameters such as the blood glucose level index. The average electrical impedance value in diabetes mellitus patients measured at a frequency of 100 Hz to 100 kHz with a current injection of 10 μA was found to be lower than the average electrical impedance value in healthy people. This study shows that the electrical impedance value of diabetes mellitus patients is lower than the impedance value of healthy people. This is consistent with diabetes mellitus patients' blood glucose levels, which are higher than healthy people's blood glucose levels.
