Abstract

This study aims were to investigate the characteristics of the quail cement macroscopically and microscopically. The research used 30 quail with ± 6 weeks of age as a source of cement. The quail cement collection were done by using modifications of massage technique to prevent contamination of the foam produced by cloaca gland and feces. Examination of cement macroscopically include volume, color, pH and consistency (viscosity) and microscopic examination includes sperm concentration, morphology, abnormalities , the number of live sperm and sperm motility. The results showed that the quail cement (Coturnix-Coturnix Japonic) macroscopically has a ±0.02 ml of volume, beige, pH ±7.06, lumpy consistency and a distinctive odor, while the results of microscopic examination of quail has a mass wave of spermatozoa good (++), ±86% progressive motility, sperm concentration ±57.2x107, spermatozoa abnormalities ±7.4% and ±5% spermatozoa death.

Abstract

The purposes of this research were to determine the motility and viability of turkey sperm on egg yolk phosphate diluent with the addition of various concentration of vitamin C stored at 5oC. The source semen from one and half years old turkey, semen diluted with eggyolk phosphate addition with vitamin C 0 mg/ml (without vitamin C), 0.1 mg/ml, 0.2 mg/ml and 0.3 mg/ml respectively. Observation was done every 12 hours for 72 hours. The results showed thataddition of vitamin C significantly difference on motility and viability of turkey stored at 5oC theaddition of 0.3 mg/ml vitamin C gives the best results in maintaining motility and viability of turkey sperm after 72 hours. In Conclusion, the addition of 0.3 mg/ml vitamin C in egg yolk phosphate diluent able to maintain motility (69.07%) and viability (71.64%) turkey sperm stored at 5oC.

Abstract

This research was conducted to produce a formula of diluent for the manufacture of frozen semen of Gembrong goat. Yolk phosphate was used as a basic diluent with the addition of anti-cold shock, antioxidants and combination of anti-cold shock-antioxidant. The research design used a completely randomized design with three kinds of treatments; T1: the addition of lactose 0.6% (anti-cold shock), T2: the addition of astaxanthin 0.004% (antioxidant), and T3: a combination of 0.6% lactose-Astaxanthin 0.004% (combination of anti-cold shock and antioxidants). The addition of DMSO 6% was used in each treatment as intracellular cryo protectants. The freezing process was done with the gradual freezing with conventional techniques. Examination of the quality of semen by thawing prior included progressive motility, viability, and abnormalities. The results showed that anticoldshock-antioxidant combination (0.6% lactose -Astaxanthin 0.004%) produced the best semen quality with progressive motility, viability, and less abnormalities as follow 44.00 + 3.46%, 59.00 + 1.85%, and 14.00 + 0.76% respectively. It was concluded that the addition of a combination of 0.6% lactose-Astaxanthin 0.004% on the phosphate yolk diluent with 6% DMSO as intracellular produced best quality of frozen goat Gembrong semen that suitable for use in artificial insemination and in vitro fertilization.

Abstract

The aims of this research were to determine PGF2? concentration the produced by bali cattlesendometrial and seminal vesicle monolayer cell culture and in vitro luteolytic ability on luteal monolayercell culture. The endometrial and seminal vesicle epithelial cell of bali cattle were cultured in tissueculture medium (TCM) 199 growth medium supplemented with 10% fetal calf serum and 10% EstrusMare Serum. The cells were cultured at 1.9 x 106 density per ml medium. Then Followed by incubation at38.50 C in 5% CO2 atmosphere for 12 days. The level of PGF2? in the cell culture medium were assayed byRadioimmnuassay (RIA) technique. The luteal cells were cultured in 9 days incubation and divided into 2groups. Group I were added with 10% of cell culture product and group II were added with 1,25 mgdinoprost/ml. The level of progesterone produced by luteal cell culture was measured at day 9th and 11thincubation. The result showed concentration of PGF2? cell product of seminal vesicle cell culture wassignificantly higher (P < 0.05) compared to endometrial cell culture. There was no significant difference(P>0.05) in luteolytic ability between PGF2? cell culture product and dinoprost. In conclusion, the PGF2?could be produced by monolayer cell culture of bali cattle is endometrial and seminal vesicle epithelialcells more over they have similar ability with dinoprost in luteolytic ability.

Abstract

This study aims were to determine the proper time for insemination and the progesterone level ofpregnant Bali cattle. Complete randomized design method was used in this study. The study consisted ofthree treatment groups i.e. Group I, animals were inseminated at 0 hour (on estrus), group II, inseminatedat 12 h post-estrus and group III inseminated at 24 h post-estrus. Estrus was observed two times a day ie.in the morning (6:00 to 8:00 am) and afternoon (16:00 to 18:00 pm) which was characterized by transparentvaginal discharge. The results showed that the highest percentage of pregnancy occurred when the cattlewere inseminated at 24 h post-estrus (100%), however, statistically this was not significant different (P>0.05) to animals that were inseminated at 12 h post-estrus (75%). Moreover, no pregnancy (0%) wasobserved in cattle that were inseminated at estrus. The progesterone level of pregnant bali cattle increasedas the period of gestation increased, being 15.43 ± 0.50 ng/mL at 30 days of gestation, 17.16 ± 0.34 ng/mLat 60 days of gestation and 20.78 ± 0.59 ng/mL at 90 days of gestation. In conclusion, the best time forinsemination in Bali cattle is at 24 h post-estrus and progesterone level seems to increase as the older thegestation period.

Abstract

The purpose of this study was to determine the effect of postmortem time on percentage of life epididymal sperm from postmortem canine caudae epididymides. A total of 9 dog were used and divided into three group. T0 was control group, T1, 3 hours postmortem and T2, 6 hours postmortem. This way, samples were obtained at different times postmortem. Sperm were extracted from the caudae epididymis by means of cuts. The result showed that the percentage of life sperm were 67,16 ± 5.67(T0), 46.33± 5.60 (T1) and 24.00 ± 4.35 respectively. We could appreciate that percentage of life was affected by postmortem time. There was significant decrease life sperm recovered from epididymides postmortem (P<0.01). In conclusion, epididymal sperm from dog undergo decrease of percentage of life, but it could stay acceptable within many hours postmortem. We intepreted these data to indicate that it may still be possible to obtain viable spermatozoa many hours later.

Abstract

This study was conducted to investigate the reproductive performance of bali cattle in breeding instalation bali cattle of Sobangan, Badung, Bali. Totaling 288 heifers were use to represents the benchmark reproductive performance of bali cattle. Data collected were service per conception (S/C), gestation length, birth weight, and postpartum estrus. The overall means for service per conception (S/C), gestation length, birth weight, and postpartum estrus were 1,3 times; 284,63±10,73 kg; 17,89±1,04 kg; dan 101,71±32,31 days.

Abstract

Leptin is one of metabolic product that regulates hypothalamus pituitary ovaries. This study aims to determine the relationship of leptin levels during parturition with estrogen levels estrus post-partus in bali cattle. The data source of this research is bali cattle during parturition where the main blood sample is taken less than 24 hours after parturition. Continued sampling of cow's blood when showing symptoms of estrus postpartum. Determination of leptin and estrogen levels was carried out using the Direct Elisa, Double Antibody Sandwich method. Data were analyzed descriptively with the Correlation and Regression statistical tests. The results showed that the average level of leptin hormone at parturition ( ± SD) was 2.60 + 0.47 ng/ml and the average level of post-partus leptin estrus hormone ( ± SD) was 5.42 + 0.47 ng/ml. While the average levels of the hormone estrogen estrus post-partus ( ± SD) is 105.51 + 19.72 pg/ml. This shows an increase in the level of the hormone leptin from parturition to estrus post-partus which has an impact on increasing estrogen levels. By using a linear regression test it was found that leptin hormone levels had a positive effect on estrogen hormone levels. From the results of the study it can be concluded that there is a close relationship between leptin levels during parturition with estrogen levels during estrus post-partus.

Abstract

The success of artificial insemination in Badung Regency has not been maximized, this can be seen from the number of recurrent mating events. One important factor in the success of artificial insemination is the quality of the frozen semen used. This study aims to determine the quality of frozen semen of Bali cattle stored at different times in the storage depot of the Mengwi Artificial Insemination Service Unit (SPIB), Badung Regency. This study uses frozen bali cattle semen from the Baturiti Regional Artificial Insemination Center. The design used is a completely randomized design with four treatments, namely treatment shortly after arriving at the storage depot (P1), seven days storage time (P2), 14 days (P3) and 28 days (P4). And each treatment consisted of five replications. The observed variables are the percentage of viability, percentage of motility and percentage of intact plasma membrane. The results showed that frozen semen stored in the Mengwi SPIB storage depot was not significantly different (P?0.05) for storage times P1, P2 and P3 but significantly different for storage of 28 days (P4) (P?0.05) with a percentage the average viability of 54.06% up to 61.21%, the percentage of motility 41.39% up to 46.44% and the percentage of the integrity of plasma membrane obtained from 50.53% up to 57.14%. The results of this study can be concluded that the quality of frozen semen spermatozoa both in terms of viability, motility and integrity of plasma membrane decreased significantly on the 28th day.

Abstract

Maturasi oosit merupakan salah satu langkah penting dalam pelaksanaan fertilisasi in vitro untuk pelaksanaan transfer embryo. Oosit hasil koleksi baik secara aspirasi maupun slicing pada umumnya belum mencapai fase MII yang dibutuhkan untuk fertilisasi in vitro sehingga perlu dilakukan maturasi oosit. Tujuan penelitian adalah untuk mengetahui tingkat maturasi oosit sapi bali pada media TCM 199 dengan penambahan hipotaurin. Oosit berasal dari ovarium yang diperoleh dari rumah potong hewan Pesanggaran Denpasar dan dikoleksi dengan metode slicing. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari tiga kelompok yaitu kelompok I / kontrol (P0: media maturasi tanpa hipotaurin), kelompok II (P1: media maturasi dengan penambahan hipotaurin dengan konsentrasi 2 mMol dan kelompok III (P2: media maturasi dengan penambahan hipotaurin dengan konsentrasi 4 mMol dengan masing-masing kelompok terdiri dari 27 ulangan. Maturasi oosit dilakukan dengan cara inkubasi selama 24 jam dalam inkubator dengan suhu 38,5 0C dengan tekanan CO2 5 %. Hasil penelitian menunjukkan bahwa tingkat ekspansi sel kumulus hingga tahap cumulus oocyte complexes (COCs) pada kelompok P1 tidak berbeda nyata dibandingkan kelompok P0 (p>0,05), namun kelompok P2 menunjukkan perbedaan yang nyata (p<0,05). Pada tingkat kematangan inti, penambahan hipotaurin tidak menyebabkan perbedaan yang nyata (p>0,05) pada tingkat GVBD, namun pada tingkat MI terdapat perbedaan yang nyata (p<0,05) antara kelompok P0 dengan P1 dan P2, sedangkan pada tingkat MII tidak terdapat perbedaan yang nyata (p>0,05) antara P0 dengan P1 namun berbeda nyata (p<0,05) antara P0 dengan P2. Dari hasil peneltian dapat disimpulkan bahwa penambahan hipotaurin dengan konsentrasi 4 mM pada media maturasi oosit TCM 199 dapat meningkatkan tingkat ekspansi sel kumulus mencapai COCs dan meningkatkan tingkat kematangan inti mencapai fase MII.
