Abstract

Button mushroom (Agaricus bisporus) is an edible mushroom that has nutritional value and bioactive compounds like β -glukan, unsaturated fatty acids, tocopherol, ergosterol, lectins, polyphenol vitamin D, and selenium  that are beneficial for health. The limiting factor of this commodity is its short shelf life, which is 3-4 days after harvest. This research aims to determine the optimum concentration of anti-browning agents (citric acid and EDTA) in the pretreatment process of fresh button mushrooms to obtain the best physical properties (water content, browning index/BI, texture, and weight reduction).  The research design uses  Response Surface Methodology with Central Composite Design through Design Expert 12 software with a combination of treatment factors are the concentration of citric acid (%) and EDTA (ppm). The results showed that the optimum concentration of citric acid and EDTA was 0.5 % and 100 ppm with the results are 93.63 % for water content, 28.89 % for BI, 23.061 N for texture, and 4.34 % for weight change.  This condition is classified as still suitable for consumption.

Abstract

Water kefir is a fermented drink made by adding water kefir grains to a water solution containing dried fruit or sugar. This research aims to determine the effect of sucrose, starter, and interactions on the chemical characteristics (antioxidant activity, total acid, medium pH) and organoleptics of fig leaf tea water kefir and to determine the interaction of antioxidant activity with total acid and medium pH. The experiment used variations in starter concentration (5% and 8%) and sucrose concentration (3%, 6%, 9% and 12%). Water kefir was analyzed through chemical analysis (antioxidants, total acid, medium pH, and sugar content) and organoleptic tests (flavor, aroma, color, and overall preference). The results showed that variations in the addition of sucrose, starter, and interactions influenced the metabolic results (primary and secondary metabolites) of microorganisms in water kefir grains and statically had a significant effect on changes in chemical and organoleptic properties (flavor, aroma, and overall preference). The best chemical characteristic parameters were obtained in the B2S3 treatment (8% starter + 9% sucrose) with antioxidant activity of 51.27%, total acid of 0.70%, and medium pH of 3.53. Meanwhile, the best organoleptic parameters were obtained in the B2S4 treatment (8% starter + 12% sucrose) with a taste value of 5.56 (liked), an aroma value of 4.80 (slightly liked), a color value of 4.97 (slightly liked), and the overall liking level was 5.45 (liked), and the antioxidant activity of fig leaf tea water kefir had a strong relationship with total acid and medium pH. Furthermore, the addition of sucrose and different starters during the water kefir fermentation process affects the chemical and organoleptic properties of fig leaf tea water kefir. It is hoped that the results of this research will make fig leaf tea water kefir an alternative probiotic drink for sufferers of lactose intolerance and allergies to milk-based drinks.

Abstract

The yidRv gene, isolated from the hypervirulent Klebsiella pneumoniae (hvKP), is a novel gene with an unknown function; however, it has exhibited high homology to the yidR, a gene recognized as potential vaccine candidate. The aim of this study was to clone the yidRv gene from the Indonesian hvKP and to investigate its overexpression in Escherichia coli. In the experiment, yidRv was cloned into pET21 to construct pYik23. Recombinant protein YidRv was produced by growing E. coli BL21 (DE3)/pYik23 in LB medium with ampicillin at 29 °C, inducing protein synthesis with 0.5 mM IPTG for 20 hours. Purification was performed using Ni‐NTA resin, and the purified protein (50 µg) was administered to BALB/c mice to test for the production of IgG, IgM and IgA on 2 days before and day 19th and 37th after the first vaccination. The results show a significant induction of IgG and IgM, but not of IgA antibodies. In conclusion, the yidRv gene was overexpressed in E. coli BL21 (DE3) at high levels in soluble form, with the recombinant protein able to be purified to 90% homogeneity. The recombinant YidRv demonstrated the ability to stimulate the generation of both IgM and IgG antibodies.

Abstract

Cocoa (Theobroma cacao L.) and chili pepper (Capsicum annuum L.) are commonly cultivated by farmers, and in some cases, both crops are grown together in intercropping systems. The main pathogen infected cocoa is the Phytophthora sp. fungus, which causes fruit rot, stem cancer, and leaf blight, while Colletotrichum sp. causes anthracnose in chili pepper plants. The ability of endophytic bacteria to inhibit the growth of fungi and bacterial pathogens is well known. In this study, the biocontrol ability of endophytic bacteria against fungal pathogens was tested, and a biocontrol preliminary test was also observed by examining their potential inhibition on Chromobacterium violaceum called antiquorum sensing test. Endophytic bacteria were isolated from cocoa plants, while Phytophthora and Colletotrichum were isolated from cocoa and chili pepper plants that showed symptoms of fruit rot and anthracnose. A total of 34 endophytic bacterial isolates were successfully obtained, with 10 isolates from leaves (DK), 12 isolates from branches (RK), and 12 isolates from roots (KA) of cocoa plant ICCRI 4. Sixteen isolates showed quorum sensing ability, which AHL degradation index ranging from 0.44 – 1.56. Antagonistic tests showed that 11 out of 16 isolates had strong antibiosis against Phytophthora sp., with inhibition zones ranging from 0.6-1.35 cm. Meanwhile, 10 out of 16 isolates had strong antibiotics against Colletotrichum sp., with clear zones ranging from 0.6 – 1.1 cm. The three best endophytic bacterial isolates that had a combination of antiquorum sensing ability on C. violaceum and biocontrol against Phytophthora sp. and Colletotrichum sp. were RK 11, KA 1, and KA 8.
