Abstract

This study aimed to identify the presence of formaldehyde in white tofu circulating in Pidie Regency as an indicator of food safety. The research employed a descriptive design with an exploratory approach, using qualitative testing through two techniques: the use of red dragon fruit peel filtrate as a natural indicator in the initial stage and a rapid test kit as a confirmatory method. The samples consisted of 48 white tofu samples collected from 12 sub-districts, with each sub-district represented by four samples obtained from different producers. Sampling was conducted purposively by considering market activity level, number of vendors, and ease of site access. The samples were prepared through a homogenization process prior to testing. The presence of formaldehyde was determined based on color change, reaction time, and color intensity observed during the testing process. The results showed that 12 samples (25%) were positively detected as containing formaldehyde, while the remaining 36 samples (75%) tested negative. Positive samples were generally found in areas with relatively high distribution and trading activity. Based on these findings, it can be concluded that formaldehyde is still present in a proportion of white tofu sold in Pidie Regency. Therefore, enhanced monitoring and education for business actors are needed. In addition, red dragon fruit peel filtrate shows potential as a simple, economical, and environmentally friendly preliminary screening method.

Abstract

Quickly recognize drug resistance in Mycobacterium tuberculosis is very important for the efficiency of the treatment and control of the disease. More than 95% of RIF resistance occurs due to point mutations in the 81-bp region of the rpoB gene M. tuberculosis. Generally, the mutation occurs at codon 531, 526 and 516. One of method to detect the mutation is using ARMS-PCR. In order to apply the ARMS-PCR technique to detection of mutations needed a special and specific primer. Primer design process using computer software "Primer Designer" and the specificity was confirmed with "Bioedit". Accuracy and ability of all these primers to detecting mutation in rpoB gene M. tuberculosis tested using ARMS-PCR method. PCR amplification results were then analyzed using the techniques of electrophoresis on agarose 1.5%. Electrophoresis results showed two bands produced from the amplification with the template H37RV (wild type). Both bands are expected to be in a position 238 bp and 484 bp, in the other hand the result amplification with the template from mutant strains produce one band with sized 484 bp. From these data it is known that ARMS-PCR reactions are performed can be used to detect mutations in the rpoB gene of M. Tuberculosis.

Abstract

Malaria remains a public health problem in pre-elimination areas, primarily due to the increase in cases with low parasitemia, which often go undetected by microscopic examination and potentially maintain hidden transmission. This study aims to identify the presence of malaria pathogens in clinical samples from the West Aceh Regional Health Laboratory (Labkesda) using real-time Polymerase Chain Reaction (qPCR) as a molecular confirmation approach. This study employed a quantitative descriptive design based on diagnostic surveillance. Dried Blood Spot (DBS) samples from patients exhibiting malaria symptoms were analyzed at the Banda Aceh Public Health Laboratory (Labkesmas). The results showed that four of the five samples tested positive for Plasmodium, and all were identified as Plasmodium knowlesi with Ct values <40. This finding indicates the presence of malaria infections that could potentially go undetected by conventional microscopy. Thus, qPCR has the potential to be a sensitive confirmation method to support malaria case detection in surveillance activities.
