Abstract

Polyethylene terephthalate (PET) is utilized in the containment of potable water, nonetheless, it is susceptible to impairment due to temperature and duration of storage. Migration of formaldehyde from PET arises as a consequence of thermal deterioration. This study aims to ascertain the extent of formaldehyde transference in PET-contained drinking water of varying condition, subjected to diverse temperature setting and storage durations. An examination into the levels of formaldehyde migration in polyethylene terephthalate (PET) bottled drinking water was effectively carried out employing a validated high-performance liquid chromatography (HPLC) technique. A total of 162 water samples from three different PET bottled water brands sourced from supermarkets in Palu City, Central Sulawesi, Indonesia, underwent analysis. The formaldehyde assessment was executed utilizing a mobile phase composition of methanol:acetonitrile:water (48:12:40 v/v) for 12.97 minutes at a flow rate of 0.8 mL/minute on the 1st day, the 14th day, and the 42nd day while stored at room temperature and exposed to sunlight. Formaldehyde, lacking effective chromophoric entities and not readily ionizable, necessitated derivatization with 2,4-dinitrophenylhydrazine before HPLC analysis. The findings of the study demonstrated exceptional method linearity within the concentration span of 0.5 to 1321 µg/mL, exhibiting a commendable correlation coefficient of 0.999. The limit of detection (LOD) and quantification (LOQ) were determined to be 17.33 and 57.76 µg/mL, correspondingly. Precision evaluations displayed average intraday and interday values of 128.46±3.48% and 103.19±0.12%, respectively (%RSD < 2/3 CV Horwitz ratio). The intraday and interday %recovery values were computed at 97.22±3.48% and 96.83±0.14% (n = 6). The scrutiny of formaldehyde concentrations in the water samples unveiled the absence of formaldehyde migration.

Abstract

Damage to vital organs such as the liver, kidneys, and pancreas is a common complication of diabetes mellitus, characterized by chronic hyperglycemia due to pancreatic cell dysfunction. Graptophyllum pictum (purple leaf) is a potential herbal therapy containing bioactive compounds with antioxidant and organ-protective effects. This study aimed to evaluate the effect of purple leaf emulsion on blood glucose levels and the histopathological features of diabetic rats induced by streptozotocin. Rats were divided into six groups: normal control, negative control, positive control (glibenclamide), and three doses of purple leaf emulsion (100, 200, and 300 mg/kg BW). After 28 days of treatment, the emulsion significantly reduced blood glucose levels and improved the histological structures of the liver, kidneys, and pancreas. The 200 mg/kg BW dose was most effective for pancreatic protection, while 300 mg/kg BW provided optimal hepatoprotective and renoprotective effects. These findings highlight the potential of purple leaf as a natural antidiabetic and organ-protective agent.

Abstract

Introduction: The use of traditional medicines derived from the active ingredients of several types of plants is preferred by the community in treating several types of diseases. One of the plants that has been used by the community as medicine is purple leaves with the Latin name Grapthophylum pictum (L.) Griff. These leaves are traditionally used to treat rheumatism, menstruation, hemorrhoids, urinary tract infections, scabies, swelling, wounds, dermatitis, ear diseases, laxatives, and cancer. The aim of this study was to determine the total levels of secondary metabolites from purple leaf extract ethanol (PLEE) and evaluate the effect of administering PLEE on acute toxicity (LD50) in Wistar rats. Methods: This research is a laboratory experiment with a post test only controlled group design. The research subjects were 20 male Wistar rats, divided into 1 control group and 3 treatment groups, each consisting of 5 rats. The control group only received distilled water, the Treatment I group (P1) was given a suspension of the purple leaf extract test preparation at a dose of 500 mg/kg. Treatment group II (P2), received a suspension of the test preparation at a dose of 2,000 mg/kg, while Treatment group III (P3) received a suspension of the test preparation at a high dose of 5,000 mg/kg of mice. The test preparation was given orally with only one administration at the beginning of the research period and observation was carried out for 14 days. Data was obtained if the rats died after being given treatment. Results: The results showed that the total secondary metabolite content of purple leaves was alkaloids (24,725.99 mg/g), saponins (89.191 mg/g), flavonoids (6.332 mg/g) and tannins (0.884 mg/g), while based on toxicity the PLEE classification was mildly toxic with an LD50 value of 3,890 mg/kg in accordance with BPOM RI's acute toxicity potential at toxicity level 4 (oral LD50 500 – 5,000 mg/kg). Conclusion: PLEE showed the highest total content of secondary metabolites in alkaloids, and consumption of PLEE in high doses caused the death of rats with a mild toxic classification.

Abstract

Good nutrition is crucial during the first 1,000 days of life, influencing a child’s physical and cognitive development. In Indonesia, malnutrition—particularly stunting—poses a significant public health problem, with 21.6% of children under five affected in 2022. Stunting leads to long-term consequences such as learning difficulties and reduced productivity, requiring a cross-sectoral approach that involves improving maternal nutrition and healthcare services. This community service program aims to enhance the awareness and skills of parents and healthcare workers in utilizing digital technology for the early detection of nutritional status and stunting risk. The activity employed a quantitative–descriptive design through training sessions, direct observation, and nutritional status assessments conducted before and after the intervention. A total of 31 children under five were evaluated using anthropometric indicators such as weight-for-age (W/A), height-for-age (H/A), and weight-for-height (W/H). The intervention involved training and the implementation of a digital application, which significantly improved the efficiency of data collection and analysis. Findings indicated a 90% improvement in cadres’ digital recording competencies and a reduction in data entry errors from 15% to 2%. Most children demonstrated good nutritional status (67.75%), while 35.48% were classified as mildly stunted and 3.22% as severely stunted. The program’s impact included enhanced data accuracy and increased parental awareness of child growth monitoring. Challenges encountered during implementation included limited internet connectivity and varying participants’ digital literacy. It is recommended that similar programs be conducted regularly with continued mentorship and technical support to ensure sustainability.

Abstract

The unsupervised use of antibiotics continues to be a pressing public health issue because it may contribute to antimicrobial resistance and increase the likelihood of unwanted health effects. A community’s understanding of when and how antibiotics should be used is one factor that can influence self-medication practices. For this reason, research sought for eva;uatng level knowledge related to independent antibiotic use among residents of East Mamungaa Village. A quantitative descriptive observational approach was selected as it enables a clear depiction of knowledge patterns within the population. A total of 172 individuals 18–65 years asked by purposive sampling, and information was obtained using a structured questionnaire assessing knowledge of indications, safety considerations, and appropriate antibiotic use. The results indicate that 60.5% of participants possessed a moderate level of knowledge, 28.5% demonstrated a good level of knowledge, and 11.0% showed limited knowledge. These outcomes reveal that although most respondents have a basic awareness of antibiotics, important gaps remain—especially in recognizing the dangers of using these medications without clinical guidance. Accordingly, continuous community-based education efforts are needed to strengthen proper antibiotic practices and minimize the risks associated with self-medication.
