Abstract

Barriers to cultivation activities are disease attacks, one of which is caused by infection with the bacterium Edwardsiella tarda. Treatment efforts can be made using natural ingredients such as jackfruit (A. heterophyllus) leaves. This study aims to determine the content of antibacterial compounds in the jackfruit (A. heterophyllus) leaves crude extract and their effect on Edwardsiella tarda bacteria in vitro. The inhibition test used disc test with five different concentrations of jackfruit (A. heterophyllus) leaves crude extract with three replications: 75 mg/L, 150 mg/L, 225 mg/L, 300 mg/L and 375 mg/L with a comparison using two types of controls (positive with 5 mg/L chloramphenicol antibiotics and negative with no treatment) using Tryptone Soy Agar (TSA) media and incubated for 2x24 hours. The results of phytochemical screening proved that the jackfruit (A. heterophyllus) leaves crude extract contains flavonoids, alkaloids, tannins and saponins. Treatment E with a dose of 375 mg/L gave the highest inhibition of 7.80 mm and treatment A with a dose of 75 mg/L gave the lowest inhibition of 4.38 mm.  The relationship between the treatment given to the test parameter in the form of the resulting inhibitory power obtained the equation y = 0.0111x + 3.6297 with an R2 value of 0.974 which means 97% of the treatment given in the form of crude extract of jackfruit (A. heterophyllus) leaves affects inhibition of bacteria E. tarda.

Abstract

This study aimed to analyze the antibacterial activity of red galangal extract (Alpinia purpurata) against the growth of E. tarda. This aims of this study was to determine the antibacterial compounds and the antibacterial effectiveness of red galangal extract (A. purpurata) against the growth of E. tarda bacteria. Antibacterial activity test was carried out by MIC test and disc test. The MIC test used doses of 1 ppm, 10 ppm, 100 ppm, 500 ppm, and 1000 ppm with incubation for 24 hours. The disc test used doses of 100 ppm, 200 ppm, 300 ppm, 400 ppm, 500 ppm and a positive control of 5 ppm chloramphenicol and a negative control without treatment. Photochemical test of red galangal extract (A. purpurata) contains flavonoid compounds, alkaloids, tannins, triterpenoids, and saponins. The results of the disc test showed that the highest inhibitory diameter was 7.55 mm at a dose of 500 ppm and was bacteriostatic because it decreased its inhibitory zone after 48 hours of incubation. The highest inhibitory effectiveness was 56.56% at a dose of 500 ppm after 24 hours of incubation.

Abstract

This study aimed to determine the antibacterial activity of bitter leaf extract (A. paniculata) against the growth of E. tarda bacteria. This test was carried out using Kirby Bauer method (disc test) using five different concentrations, namely 50 mg/L, 100 mg/L, 150 mg/L, 200 mg/L, and 250 mg/L in TSA (Tryptone Soya Agar) and incubated for 2x24 hours using a temperature of 30 oC. The positive control used was chloramphenicol (5 mg/L), while the negative control used PBS. Bitter leaf extract contains active compounds that have bacteriostatic antibacterial properties. After incubation for 24 hours the concentration of 250 mg/L showed the highest inhibition zone with a diameter of 7.82 mm and decreased at 48 hours to 7.46 mm. After being incubated for 24 hours, the concentration of 250 mg/L obtained an effectiveness value of 57.08% which is the best effectiveness, and there was a decrease in the 48th hour to 54.45%.

Abstract

Abstrak - Serangan penyakit merupakan pemicu kegagalan budidaya. Penyakit yang menyerang organisme budidaya air tawar yaitu penyakit bacterial salah satu agen bakteri yang menginfeksi adalah bakteri P. fluorescens. Penggunaan antibiotik seringkali digunakan untuk mengatasi penyakit yang disebabkan oleh bakteri, sehingga perlu adanya alternatif dengan penggunaan bahan alami. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak lengkuas merah (A. purpurata) terhadap daya hambat bakteri P. fluorescens secara In Vitro. Penelitian ini menggunakan metode eksperimen dengan Rancangan Acak Lengkap (RAL) yang terdiri dari 5 perlakuan, 2 kontrol dan 4 kali ulangan. Perlakuan A (100 ppm), perlakuan B (105 ppm), perlakuan C (110 ppm), perlakuan D (115 ppm), perlakuan E (120 ppm), kontrol positif menggunakan tetracyline 30 ppm, dan kontrol negatif tanpa pemberian ekstrak. Hasil penelitian didapat zona hambat tertinggi pada perlakuan E (120 ppm) dengan rerata zona hambat sebesar 10,53 mm dan terendah pada perlakuan A (100 ppm) dengan rerata zona hambat sebesar 8,15 mm. Zona hambat antar perlakuan menunjukkan grafik linier dengan persamaan y = -3,25 + 0,11x dan koefisien determinasi (R2) yaitu sebesar 0,8552. Disease tat attack freshwater aquaculture organisms, namely bacterial disease, one of the bacterial agents that infect are P. fluorescens bacteria. The use of antibiotics is still often used to treat diseases caused by bacteria, so there is a need for alternatives to the use of natural ingredients. This study aims to determine effect of red galangal extract (A. pupurata) on inhibitory of P. fluorescens bacteria in vitro. This study used an experimental method with a completely randomized design (CRD) consisting of 5 treatments, 2 controls and 4 replications. Treatment A (100 ppm), treatment B (105 ppm), treatment C (110 ppm), treatment D (115 ppm), treatment E (120 ppm), positive control with 30 ppm tetracyline, and negative control without extract. The results showed that the highest inhibition zone was in treatment E (120 ppm) with an average inhibition zone of 10,53 mm and the lowest was in treatment A (100 ppm) with an average inhibition zone of 8,15 mm. The inhibition zone between treatments shows a linear graph with the equation y = -3,25 + 0,11x and the coefficient of determination (R2) is 0,8552. Abstract

Ohoi Hako merupakan desa pesisir di Pulau Kei Besar yang ditempati oleh nelayan yang menjalankan aktivitas perikanan skala kecil, namun kondisi tersebut tidak didukung dengan informasi ilmiah terutama mengenai pola penangkapan ikan yang dilakukan nelayan. Penelitian ini dilakukan dengan tujuan untuk menganalisis pola penangkapan ikan ditinjau dari aspek temporal dan aspek spasial. Penelitian dilakukan pada bulan September 2023 di Ohoi Hako, Kecamatan Kei Besar Selatan Barat, Kabupaten Maluku Tenggara, Maluku. Pengambilan data dilakukan menggunakan teknik wawancara dan pemetaan partisipatif untuk memetakan waktu dan lokasi penangkapan melalui kalender musim dan peta daerah penangkapan. Tool yang digunakan untuk melakukan digitalisasi peta partisipatif adalah ArcGIS. Hasil penelitian menunjukkan aktivitas penangkapan ikan berlangsung sepanjang tahun, dengan konsentrasi tinggi pada bulan Maret – April dan September – Desember, dan rendah pada bulan Januari – Februari dan Mei – Agustus. Gillnet hanya dapat dioperasikan selama enam bulan, sementara hand line dan troll line dapat dioperasikan sepanjang tahun. Lokasi penangkapan ikan meliputi perairan pantai sepanjang pesisir Ohoi Nerong ke Selatan hingga pesisir Ohoi Weduar Fer, meluas ke Saaru Rumaat hingga ke perairan sekitar Tanimbar Kei. Komposisi jenis tangkapan nelayan terdiri dari berbagai jenis ikan, dimana kerapu menjadi target utama tangkapan harian dan tongkol merupakan target utama musiman (Agustus – September).

Abstract

Cultivation activities often experience obstacles due to bacterial disease attacks, one of which is caused by P. fluorescens bacteria. Commercial treatment efforts include the use of antibiotics, but this can cause bacterial resistance. Therefore, an alternative treatment that is more environmentally friendly but can still be used as medicine is needed, namely the use of natural ingredients such as jackfruit (A. heterophyllus) leaves. This study aims to determine the effect of crude jackfruit (A. heterophyllus) leaf extract on P. fluorescens bacteria in vitro. This study used an experimental method with a Completely Randomized Design (CRD) consisting of 5 treatments, 2 controls, and 3 replications. The control treatment consisted of a positive control (Tetracycline 30 ppm), and a negative control (only using disc paper), as well as 5 different extract dose treatments, namely treatment A (60 ppm), treatment B (120 ppm), treatment C (180 ppm), treatment D (240 ppm) and treatment E (300 ppm). The results of the study showed that the use of crude jackfruit (A. heterophyllus) leaf extract affected P. fluorescens bacteria. The relationship between the use of crude jackfruit (A. heterophyllus) leaf extract with different doses and the resulting inhibition zone showed a quadratic regression pattern with the equation y = -0.0001x2 + 0.05x + 6.327.
