Abstract

Aging is a natural process in human life and is triggered by the presence of free radicals (ROS). The use of antioxidants from natural ingredients is one of the breakthroughs to overcome aging and counteract the harmful effects caused by the free radicals. This study aimed to determine and compare the antioxidant activity of H2O2 scavenging and hyaluronidase inhibition of red dragon fruit peel extract (DFPE) and kaempferol-3-o-rutinoside (KOR) compounds. Dragon fruit peel extract (DFPE) is obtained through extraction by maceration method using 70% ethanol solvent. The design of this study included antioxidant and anti-aging activity assay of EKBN and KOR at the series concentration of 15.63; 31.25; 62.50; 125; 250; 500 µg/mL through H2O2 scavenging, as well as the DFPE and KOR hyaluronidase inhibition assay at the series concentration of 5.21; 10.42; 20.83; 41.7; 83.33; 166.67 µg/mL. EKBN shows that the average activity of H2O2 scavenging is lower than KOR. In addition, the IC50 values of KOR for H2O2 scavenging is lower (351.46±2.30ug/mL) than DFPE (409.64±23.17ug/mL). While, KOR also has higher values of inhibitory activity than of the DFPE. However, the IC50 value of KOR for hyaluronidase inhibition activity was 84.07±10.46µg/mL, equivalent to the IC50 value of DFPE (85.32±10.24µg/mL). The presence of antioxidant and anti-aging activity in the EKBN is probably caused by betalain and the KOR compound itself contained in red dragon fruit. The results of the paired-samples T-test on antioxidant activity and anti-aging of DFPE and KOR showed non-significant difference. Thus, DFPE has an equivalent antioxidant and anti-aging through H2O2 scavenging and hyaluronidase activity as possessed by the KOR compound.

Abstract

Diabetes merupakan penyakit long-term yang dapat terjadi ketika kadar gula dalam darah tinggi sehingga tidak dapat memproduksi zat insulin secara cukup atau insulin tidak dapat bekerja secara efektif. Diperkirakan akan meningkat di tahun 2030 menjadi 552 juta jiwa jika tidak mendapatkan penanganan yang baik. Aloksan merupakan penyebab sel-β pankreas mengalami nekrosis juga sering dipakai sebagai bahan eksperimental dalam topik diabetes dengan kasus yang bervariasi pada hewan seperti tikus, mencit, dan anjing. Studi ini bertujuan untuk menganalisis dan merangkum studi-studi terdahulu mengenai pengaruh variasi pemberian dosis aloksan terhadap angka kadar gula darah hewan coba.

Abstract

The population is unaware of lead poisoning risks. Lead can cause tissue function and metabolism issues. Celery (Apium graveolens L), an antioxidant, is widely used in medicine and as a multivitamin. This study examines how celery extract affects SOD levels and testicular Caspase-3 expression in lead-exposed Wistar white rats (Rattus norvegicus). The accurate experiment-based quantitative research. 27 Wistar white rats (Rattus norvegicus) were sampled. This study examined celery extract as an independent variable and SOD and caspase-3 expression as dependent variables. Three treatment groups were presented: Control Group, Treatment Group 1 with 200 mg/kg BW celery extract, and Treatment Group 2 with 400 mg/kg BW. The Paired T-test and normality test were performed. SPSS 26.0 processed statistical data. The study indicated that celery extract increased SOD levels, but not as much as without lead introduction. Also, the caspase-3 decrease was not significant. In normality tests, SOD and caspase-3 were p > 0.05. Similarly, the Paired T-test showed 0.000 < 0.05 for SOD caspase-3 levels. Celery leaf extract (Apium graveolens Linn) at 200 and 400 mg/kg BW provided active components like alkaloids, flavonoids, saponins, tannins, and steroids, which were beneficial. The body benefits from its antioxidant properties.

Researchers were interested in studying the impact of Community-based total sanitation on stunting in the service area of the Sei Bejangkar Health Center since the first survey indicated that 90 toddlers were stunted owing to the effects of inadequate sanitation. The study used a quantitative, cross-sectional methodology. This research was carried out near the Sei Bejangkar Health Center. All of the infants and toddlers in the puskesmas's operational region served as the study's population. Total sampling was used during the sampling process. Univariate, Bivariate, and Multivariate tests were utilized to analyze both main and secondary data in this study. Based on the data, it appears that there is no connection between the halt defecation movement and stunting in toddlers in the sei anchor health center's service area (P value of 0.529 or more than 0.05). Toddlers at the Sei Bejangkar Health Center are less likely to be stunted if open defecation is reduced. The p-value is 0.000, which means it's significantly larger than 0.05 (0.000 > 0.05).
