Abstract

Chronic exposure to ultraviolet rays from the sun might result in oxidative stress, changes in the structure and composition of the skin. Sunscreen preparations are recommended to be used to prevent or minimize the harmful effects of UV rays on the skin. Sunscreen can be obtained from natural ingredients, one of which is the flower of cherry (Muntingia calabura L.). This study aims to identify the content of flavonoids and test the sunscreen activity of the ethanol extract of cherry flower (Muntingia calabura L.) in vitro using the UV-Vis spectrophotometric method based on the SPF value. Flavonoids are claimed to be capable to blocking ultraviolet (UV) light-induced radicals that have a protective effect against UV radiation, where the SPF value is a universal measure that describes the potency of a UV defense material. The findings of this study showed that the ethanol extract of cherry flowers positively contained flavonoid compounds including the flavonol, flavanone, and anthocyanidin groups. The results of the sunscreen activity test showed that the ethanol extract of cherry flowers had sunscreen activity at a concentration of 0.01% with an SPF value of 10.9 (moderate SPF), 0.05% with an SPF value of 41.6 (high SPF), 0,15% with an SPF value of 45.5 (high SPF). and 0.25% with an SPF value of 48.7 (high SPF). The higher the extract concentration, the higher the SPF value indicating the greater the activity as sunscreen.

Abstract

Coronavirus disease is an infectious disease caused by the SARS-CoV-2 coronavirus, which is an RNA virus consisting of four structural proteins and sixteen non-structural proteins responsible for viral multiplication. One of the proteins is the main protease (Mpro). Inhibition of Mpro is potentially effective as a COVID-19 drug because it will cause the loss of the virus's replication and transcription abilities, so that the virus is unable to survive. The purpose of this study is to determine what chemical compounds in ginger (Zingiber officinale) and kersen (Muntigia calabura) plants have the potential as COVID-19 drugs with the Mpro inhibition mechanism. This research method is in silico using molecular docking method with PyRX software. The results showed that of the 70 chemical compounds from ginger (Zingiber officinale), there are 3 compounds that have potential because they have better activity than the N3 inhibitor comparison ΔG -8.0 kcal/mol, namely Zingiberoside B (-8.6 kcal/mol), Zingiberoside C (-8.5 kcal/mol) and Shogasulfonic acid D (-8.2 kcal/mol). Meanwhile, of the 10 chemical compounds from Jamaica cherry (Muntigia calabura), there are 4 chemical compounds that have potential as Mpro SARS-CoV-2 inhibitors, namely Kempferol 7-(6''-p-coumarylglucoside) or Biondnoid A (-9. 1 kcal/mol), Hiravanone (-8.4 kcal/mol), 5,7,3',4' - Tetrahydroxy - 6, 8 - di - C -prenylflavanone (-8.4 kcal/mol), and Myrtillin (-8.3 kcal/mol).

Abstract

Kersen (Cherry) (Muntingia calabura L) leaves are empirically used to treat various diseases because they contain flavonoid and tannin compounds. Based on previous research, it was reported that phenolic compounds, tannins, and flavonoids can potentially counteract free radicals so that they can improve the body's immune system. The purpose of this study was to determine the antioxidant activity of extracts of methanol, ethyl acetate, and n-hexane from cherry leaves (Muntingia calabura L.). The DPPH free radical scavenging method is based on the reduction of the colored DPPH free radical methanol solution by free radical inhibition. The extraction process of a compound is carried out to separate the material from the mixture using a suitable solvent. The existence of a level of polarity of a solvent used to attract the components of active compounds in plants is a guide for the search for new compounds in a plant. The potential of a compound to counteract free radicals can be determined by calculating the IC50. The results showed that the extract of methanol, ethyl acetate, and n-hexane of the cherry leaf (Muntingia calabura L.) had inhibitory activity against DPPH free radicals with IC50 values of 8.67; 9,097, and 18.17 g/mL. Meanwhile, gallic acid has inhibitory activity with an IC50 value of 1.77 g/mL. Methanol and ethyl acetate extracts of cherry leaves were able to inhibit DPPH activity in a very strong category, which was almost comparable to gallic acid, while n-hexane extract had antioxidant activity in a strong category.

Abstract

Free radicals are molecules that do not have an electron pair in the outer shell, making them highly reactive. Oxidation due to free radicals can be prevented and inhibited by substances, namely antioxidants. Natural antioxidants are obtained from plants or fruits, one of which is Qust al hindi (Saussurea lappa).  Qust al hindi is a plant native to India that is used as a traditional medicine. Qust al hindi contains various phytochemical compounds, one of which is flavonoid compounds that function as antioxidants. The purpose of this study was to analyze the antioxidant activity of water and ethyl acetate fractions of qust al hindi plant roots by 1,1-diphenyl-2picrylhydrazyl (DPPH) silencing method using UV-Vis spectrophotometry based on the calculation of IC50 value using GraphPad Prism Application. The results of this study indicate that the water and ethyl acetate fractions of qust al hindi plant roots have antioxidant activity against DPPH free radicals with IC50 values of 203.5 µg/mL; 193.3 µg/mL, respectively, which are included in the very weak and weak antioxidant groups.
