Abstract

Endophytic fungal isolates coded IFBZ5, IFBZ6, and IFBZ7 derived from the stem of the bidara plant (Ziziphus mauritiana Lam.) have shown potential antioxidant properties. This study aims to test the antioxidant activity of endophytic fungal isolates on the bidara plant stem (Ziziphus mauritiana Lam.) by TLC-Autography method and DPPH (1,1-diphenyl-2-picrylhydrazil) free radical silencing. This study used endophytic fungi isolates code IFBZ-5, IFBZ-6 and IFBZ-7 which were fermented for 21 days using MYB medium which was then liquid-liquid extracted with ethyl acetate (1:1) and evaporated. The fermentate extract obtained was then tested for antioxidant activity qualitatively and quantitatively. Qualitative analysis using TLC plates indicated antioxidant activity in all isolates. Quantitative analysis showed that isolate IFBZ7 exhibited strong antioxidant activity with an IC50 value of 50.120 ppm, while IFBZ5 and IFBZ6 showed very weak activity with IC50 values of 2.482.363 µg/mL and 467.847 µg/mL, respectively. The BHT comparator had an IC50 value of 6.778 µg/mL. This shows that the antioxidant power of endophytic fungal isolates coded IFBZ5, IFBZ6 and IFBZ7 is lower than the antioxidant activity of BHT as a comparison.

Abstract

Bajakah tampala root (Spatholobus littoralis Hassk.) contains phenolic compounds, flavonoids, tannins, terpenoids and saponins. Flavonoid compounds have pharmacological effects, namely anti-inflammatory. The purpose of this study was to determine the anti-inflammatory activity of ethanol extract of bajakah tampala root by inhibiting protein denaturation in vitro using UV-Vis spectrophotometer and diclofenac sodium as positive control. The extract was prepared by maceration method using 96% ethanol solvent. The results showed that the ethanol extract of bajakah tampala root had a percentage of inhibition of 31.27% at the lowest concentration (10 ppm) and 43.28% at the highest concentration (50 ppm) with an IC50 value of 77.92 µg/mL and the ic50 value of diclofenac sodium is 12.26 µg/mL. The percentage of inhibition that is more than 20% is considered to have inhibitory power in the protein denaturation process. So, it can be concluded that the ethanol extract of bajakah root has potential as an anti-inflammatory.

Abstract

The root of Bajakah Tampala (Spatholobus littoralis Hassk.) is known to contain several bioactive secondary metabolites, including saponins, flavonoids, phenolics, and tannins, which contribute to its broad pharmacological potential. Among these compounds, tannins play an important role in various therapeutic applications such as hemostasis, burn treatment, and the management of diarrhea, dysentery, and tumors. In addition, tannins possess anticholesterol properties by precipitating mucosal proteins on the surface of the small intestine, thereby reducing the absorption of cholesterol and lipids into the bloodstream. This study aimed to determine the tannin content in the ethanol extract of Bajakah Tampala root as part of an investigation into its potential as a natural anticholesterol agent. The extraction process was carried out using the maceration method with 96% ethanol as the solvent to ensure efficient recovery of phenolic compounds. Quantitative analysis of tannin content was performed using the UV-Visible spectrophotometric method at a wavelength of 750 nm, with gallic acid employed as the standard reference compound. The results showed that the tannin concentration in the ethanol extract of Bajakah Tampala root was 91.699 mg GAE/g extract (milligrams of gallic acid equivalent per gram of extract). These findings suggest that Bajakah Tampala root possesses a high tannin content, supporting its potential development as a natural anticholesterol agent and contributing to the growing evidence of its pharmacological benefits in traditional medicine.
