Abstract

Obesity has become increasingly prevalent worldwide each year. Several studies have proven that herbs are effective in preventing obesity. The research delved into the active compounds of Boesenbergia pandurata to reveal their mechanisms of action by utilizing bioinformatics approaches. The research methods included active compounds selection, QSAR analysis, networking analysis, molecular docking, and ADME prediction. The QSAR analysis predicted that the active compounds were correlated with some theoretical activities with more than 0.5 probability, namely vasoprotective, anti-hypercholesterolemic, anti-inflammatory, free radical scavenging, and as a lipid metabolism regulator and TNF expression inhibitor. Furthermore, the results of the networking analysis showed that five compounds (pinocembrin, cardamonin, flavokawain B, flavokawain C, and tectochrysin) had direct interactions with RPS6KB1. Pinocembrin exhibited the highest binding affinityof -7.26 kcal/mol, although not as strong as that of the control ligand (FS9). The ADME prediction indicated that the five compounds were non-toxic and had excellent absorption. It can be concluded that the active compounds of B. pandurata have the ability to improve metabolic syndrome, especially obesity, in silico through several mechanisms, such as suppression of pro-inflammatory cytokine, regulation of lipid metabolism, and those associated with antioxidants.

Abstract

Diabetes mellitus is a group of metabolic disorders characterized by increased blood glucose concentration, known as hyperglycemia. Several studies report that diabetes mellitus is associated with the presence of tumor necrosis factor-α (TNF-α). One of the medicinal plants that has the potential to be used to treat diabetes mellitus is pomegranate (Punica granatum L.). Therefore, this study aimed to observe the effect of bioactive compounds of pomegranate peel as antidiabetic against TNF-α using a molecular docking approach. The stages of molecular docking include geometry optimization, validation of the docking program, docking ligand test, and visualization of the docking results. The target compounds analyzed from pomegranate peel were ellagitannin and punicalagin. The macromolecular target wasTNF-α. Redocking between the original ligand and the target TNF-α showed an RMSD value of ≤ 2.0 Å with a value of 0.4902 Å. The hydrophobic interaction was formed between ellagitannin and punicalagin on the TNF-α target shows excellent ligand-receptor interactions, this can be seen from the binding free energy value which is smaller when compared to the co-crystal ligand. These results indicate that the complex formed from tethering ellagitannin and punicalagin to the TNF-α target is more stable than the original ligand complex with the TNF-α target. Based on the research results, it can be concluded that pomegranate peel has excellent potential as an antidiabetic. In silico research shows that pomegranate peel content such as ellagitannin can bind stably with TNF-α (ΔG = -132.44kcal/mol), so it is predicted to be able to suppress TNF-α which is responsible for the emergence of diabetes mellitus. Abstract

Mefenamic acid is classified as a Biopharmaceutical Classification System (BCS) Class II drug. Crystallisation techniques can significantly influence the critical properties of mefenamic acid crystals, an essential consideration in the pharmaceutical industry. This study investigates the effect of recrystallisation on the morphology and particle size of mefenamic acid crystals using ethyl acetate as the solvent.  Recrystallisation was conducted using both conventional and ultrasonic-assisted methods. The yield was calculated, and characterization was performed to determine the crystal properties, shape, and particle size using polarized microscopy, X-ray Diffraction (XRD), and Fourier Transform Infrared Spectroscopy (FTIR). The results showed that conventional recrystallisation yielded 25.078%, while ultrasonic-assisted recrystallisation at 50 W, 60 W, 70 W, and 80 W yielded 43.47%, 47.71%, 51.90%, and 50.20%, respectively. Skewness and kurtosis values for the average crystal length and width indicated a normal distribution. XRD analysis revealed that the diffraction peak intensity of conventionally recrystallised crystals became narrow. In contrast, the diffraction peak intensity of ultrasonically assisted crystals broadened. FTIR results demonstrated that the recrystallisation process did not alter the fundamental chemical structure of mefenamic acid. Recrystallisation methods influence crystal shape and size, with ultrasonic-assisted techniques producing crystals of smaller average size than those obtained through conventional methods.

Abstract

Free radicals contribute to various diseases by inducing oxidative stress. Single black garlic (Allium sativum L.) is recognized for its phenolic compounds, which possess radical-scavenging properties. This study aimed to develop and evaluate instant granules containing black A. sativum extract for their antioxidant activity and physicochemical characteristics. An instant granule formulation was developed using a 70% ethanol extract of a single black A. sativum, with three distinct formulas containing 5%, 7.5%, and 10% extract (Formulas 1, 2, and 3, respectively). The granules were subjected to comprehensive physical quality assessments, including organoleptic properties, moisture content, angle of repose, flow rate, dissolution time, and tapped density. Antioxidant activity was quantified using the DPPH (2,2-diphenyl-1-picrylhydrazyl) assay, reporting IC50 values. All instant granule formulations successfully met the established physical quality requirements across all evaluated parameters. Furthermore, the antioxidant activity demonstrated a concentration-dependent effect, with IC50 values of 119.576 ppm for Formula 1, 82.000 ppm for Formula 2, and 59.962 ppm for Formula 3. This study confirms that instant granules formulated with a single black A. sativum extract exhibit significant antioxidant potential and possess desirable physical characteristics, indicating their promise as a natural antioxidant supplement.
