Abstract

The prevention of fish infections caused by Aeromonas sp. bacteria can be achieved using Lactobacillus sp. bacteria. This study aims to identify and analyse the effect of Lactobacillus sp. using the microencapsulation method to treat catfish infected with Aeromonas sp. The identification of Lactobacillus sp. bacteria was conducted through microbiological and molecular tests. After obtaining the test results, the research continued with the application of microencapsulation using the emulsion technique. The impact of this microencapsulation was evaluated on catfish through an experimental approach employing a completely randomised design (CRD), with 10 catfish tested in each experimental aquarium. The study included five treatments, designated as A, B, C, D, and E. Treatment A served as the control, without the administration of Lactobacillus. Treatment B involved the application of Lactobacillus without microencapsulation. Treatments C, D, and E utilised varying doses of Lactobacillus at 0.003, 0.005, and 0.007 g/L, respectively. Each treatment was conducted in triplicate. Microbiological and molecular testing identified encapsulated Lactobacillus sp., specifically Lactobacillus helveticus and Lactobacillus johnsonii. The results indicated that treatment D, comprising a dose of 0.005 g/L of Lactobacillus helveticus and Lactobacillus johnsonii, was the most effective in promoting the healing of catfish infected with Aeromonas hydrophila bacteria. Keywords: Completely randomized design, bacterial infection mitigation, probiotic bacteria, emulsion technique.

Abstract

Insecticides can be used to overcome pest attacks in agricultural and plantation sectors. The botanical insecticide is more widely used because more friendly to the environment than chemical insecticides. Neem is one of the plants that can be used as basic ingredient for botanical insecticides because it contains the compound azadirachtin. Insecticide enter the cultivation area because carried by flowing water. Tilapia is a fish that can be used as a bioindicator of pollution levels, water quality, and changes in the aquatic environment. This study aims to determine the effect of exposure insecticide with azadirachtin active ingredient on growth and survival rate of tilapia. This research was conducted in March and August for the preliminary test and September-October 2022 for the main test at Ciparanje Inland Aquaculture Hatchery, Padjadjaran University. The fish used in this study averaged 6,70 ± 0,014 cm. This study used experimental method, Completely Randomized Design with five treatments and three replications. The treatments used were exposure to insecticides with different doses, namely A(0 ppm), B(26.25 ppm), C(52.5 ppm), D(78.75 ppm), E(105 ppm). Parameters observed included absolute length growth and weight growth, survival rate, and water quality until 28th day. Growth and survival data analyzed using the F test and continued with Duncan's test at the 95% level of confidence. The results showed that exposure insecticide can inhibited growth and affected the survival of tilapia. Treatment C(52.5 ppm) is a safe concentration for the survival of tilapia and the highest growth is the treatment without addition of insecticide.

Abstract

Catfish (Clarias gariepinus) is one of the leading aquaculture commodities in Indonesia, with a production volume reaching 1.01 million tons, accounting for approximately 15% of the national aquaculture output in 2020. The availability of high-quality fry is crucial for the sustainability of catfish farming. This study aims to evaluate the effect of high stocking densities on survival rate, condition factor, and size variation of catfish fry reared in UFBs-RAS. This research was conducted at PUI-PT Nano Powder Functional Universitas Padjadjaran (Jatinangor) from October to November 2023. The study tested catfish fry with lengths of 3–4 cm and weights of 1.2–1.4 g with four densities (10 fish/L, 15 fish/L, 20 fish/L, 25 fish/L). The results indicated that stocking density did not significantly affect the survival rate (P>0.05), but it did have a significant effect on length and weight variation (P<0.05). The condition factor did not show significant differences (P>0.05) between stocking density treatments. Overall, a density of 25 fish/L yielded the best performance within the UFBs-RAS. Therefore, this density is recommended as the optimal stocking rate for catfish fry rearing in this system. These findings suggest that the application of the UFBs-RAS system provides a strategic solution for the aquaculture industry to enhance the efficiency of catfish fry rearing at high stocking densities while maintaining optimal water quality and growth performance, thereby supporting productivity and sustainability in intensive aquaculture systems.

Abstract

This study was aimed to observe the survival rate (SR) and clinical signs, and the acute immune response of Pacific white shrimp (Litopenaeus vannamei), including total hemocyte count (THC), bacterial clearance, and prophenoloxidase (ProPO) activity due to infection with the Vibrio parahaemolyticus bacteria and its metabolites. Shrimp were infected by bacterial isolates with culture media (A), bacterial isolates without culture media (B), and metabolites suspensions from bacterial culture media (C) for 8 hours with observation every 2 hours. The results showed that experimental shrimps in groups A and B showed clinical signs and reduced SR, 24.44% and 62.22%, respectively. Experimental group C still showed the clinical signs but did not cause the mortality (SR 100.00%). The THC did not show significant changes, either based on experimental groups or duration of infection, but all groups tended to show decreasing THC at 8 hours post-infection. Bacterial clearance and ProPO activity also did not show significant differences between experimental groups or duration of infection at 8 hours post-infection. However, there was an increase in bacterial clearance activity at 6 hours post-infection in the experimental groups A and B. Based on the study results, the acute immune response in shrimp from all of the experimental groups did not show significant changes at 8 hours post-infection. However, group A significantly decreased SR compared to groups B and C.
