Abstract

Infections caused by multidrug-resistant (MDR) bacteria are on the rise globally. MDR is facilitated by overexpression of efflux pump and permeability changes of membrane. Acinetobacter baumannii is a pathogenic germ that causes a major problem in infection, also, there has been an increase in the incidence of resistance to various antibiotics. The present study highlights the synergistic of ethanolic extract of Curcuma domestica (EECD) rhizome with tetracycline against multidrug-resistant A. baumannii (MDR-Ab). Assessment of Minimum Inhibitory Concentration (MIC) was determined by microdilution using 96-well plates. The synergistic effect of EECD and tetracycline was determined by checkerboard method. The effect of EECD and tetracycline combination was investigated by bacteriolytic activity and inhibition of efflux pump by Ethidium Bromide (EtBr) accumulation assay. EECD presented the MIC value 250 µg/mL against MDR-Ab. Fractional Inhibitory Concentration Index (FICI) value of EECD and tetracycline combination was 0.4, which showed their synergistic effect. Additionally, the combination of EECD and tetracycline could inhibit the efflux pump in MDR-Ab. This combination can also compromise cell integrity by altering membrane permeability thus lysing the bacteria cells. According to these results, EECD and tetracycline combination has synergistic effects at some sites of action, and thus could be used as a breakthrough to overcome infection problems due to MDR-Ab. Abstract

Burns are a condition where there is damage to the epidermis layer down to the dermis layer. The skin structure sustains damage from the epithelium to the basal layer of the dermis. You can use the herbal Chinese pepper (Peperomia pellucida L.) for burn treatment. This study aims to evaluate the impact of administering Chinese betel leaf ointment, with variations in concentration and formula, on the duration of burn wound healing and the diameter of wound closure. Methods There were 30 Mus musculus mice in this study. Divided them into five groups: a regular group without burns, a positive control group that received gentamicin, treatment group 1 that received a low concentration 5% ethanol extract of Chinese betel leaf (EECB) ointment, a treatment group 2 that received a low concentration 10% EECB ointment, and a treatment group 3 that received a high concentration 15% EECB ointment. Results demonstrate wound closure activity by measuring the diameter of burn wounds treated with a 15% concentration of Chinese pepper ointment on the 18th day and a 15% concentration closed the burn wound. Histopathological differences were seen in the K3 group, where collagen density scored +3 (±0.06). We found a very weak positive correlation (0.000) between histopathology (collagen) and wound diameter and a strong negative correlation (-0.0527) between the histopathological parameter (collagen) and the duration of burn wound healing. Conclusion. The research concludes that the effectiveness of administering Chinese betel leaf ointment with a concentration of 15% was obtained

Abstract

Tanaman daun sembung (Blumea balsamifera L.) adalah obat tradisional yang digunakan untuk mengobati batu ginjal, diare, masalah perut, dan sebagai rempah-rempah sauna tradisional. Dikenal juga sifat antibakterinya yang alami dan dapat menahan perkembangan kanker dan tumor. Tujuan penelitian ini adalah untuk menilai karakteristik simplisia, golongan kimia tumbuhan, fraksi n-heksana daun sembung dan aktivitasnya dalam menahan perkembangan biakan bakteri S.aureus, P.acnes dan E.coli. Penelitian dilaksanakan meliputi seperti pengumpulan bahan baku daun sembung, pengujian karakteristik, maserasi serbuk simplisia daun sembung menggunakan pelarut etanol 96%, uji skrining fitokimia, fraksinasi ekstrak etanol B. balsamifera menggunakan pelarut n-heksana yang kemudian diuji aktivitas antibakterinya pada fraksi n-heksana daun sembung (FHDS) terhadap bakteri Propionibacterium acnes, Staphylococcus aureus, dan Escherichia coli dengan konsentrasi 100, 200, 300, 400 dan 500 mg/ml menggunakan metode difusi agar sumuran dengan mengamati zona hambat maksimum. Berdasarkan hasil pemeriksaan golongan senyawa pada FHDS, terdapat golongan senyawa kimia tumbuhan, yaitu alkaloid, flavonoid, tanin, saponin, dan steroid. Investigasi aktivitas antibakteri FHDS menunjukkan aktivitasnya pada FHDS (500 mg/ml, 400 mg/ml, 300 mg/ml, 200 mg/ml, dan 100 mg/ml) dalam menahan perkembang biakan bakteri E. coli, S. aureus, dan P. acnes. Aktivitas antibakteri terkuat ditunjukkan pada FHDS dengan konsentrasi 500 mg/ml (16.5 mm). Berdasarkan hasil penelitian, dapat disimpulkan bahwa FHDS mempunyai efikasi sebagai antibakteri yang dikategorikan kuat dan mampu menghambat perkembang biakan bakteri E. coli, S. aureus, dan P. acnes
