Abstract

Abstract: The development and induction of cervical cancer carcinogenesis are linked to sexually transmitted Human Papillomavirus (HPV) infection, and can affect human genes, particularly those involved in the apoptotic process, one of which is the Bcl-2 gene. Several synthetic compounds, including pyrimidineylpiperazine, phenylpyrazole, kendomycin, and navitoclax, have been shown to be effective in inhibiting the antiapoptotic activity of Bcl-2, but it is the labdane-type diterpene content of the natural legundi fruit plant (Vitex trifolia L.) that has anticancer bioactivity, particularly in the process of genetic mutations, Apoptotic inhibition and therapeutic failure have yet to be discovered, particularly in cervical cancer. The study's goal was to investigate the mechanism of interaction (molecular docking) of labdane type-diterpene compounds with the antiapoptotic protein Bcl-2. In silico (docking molecular) research was used to determine the interaction between legundi fruit ligands and the Bcl-2 target protein in cervical cancer. The docking analysis results will then be visualized with the Discovery Studio 4.1, LigPlot+, and Ligand Scout 3.1 software. The interaction of proteins and ligands was studied to determine the number and type of bonds formed, such as hydrogen and hydrophobic bonds. The results were as follows: Gibbs energy -7.8, pKi 1.91 m, 10 hydrophobic bonds, and hydrophobic residues Leu80, Leu82, Thr83, Thr86, Ala87, Cys90, Met118, Glu121, Ser122, and Leu130. The findings presented here suggest that labdane-type diterpenes are powerful anticancer agents capable of inducing apoptosis and inhibiting the antiapoptotic action of the Bcl-2 gene, which should be investigated further in (pre)clinical studies.

Abstract

Dengue is a disease caused by dengue virus (DENV) that is transmitted mainly by the female Aedes aegypti mosquito. There are four serotypes of DEN, leading to a possibility that a person may be infected four times by this virus, albeit with different serotypes. Recovery from infection with one viral serotype provides lifelong immunity to the same serotype but not to the other serotypes. Secondary infection by other serotypes increases the risk of developing severe dengue. The pathogenesis of severe dengue involves apoptosis of microvascular endothelial cells that leads to plasma leakage. In addition, there is usually a decrease in platelets and leukocytes and an increase in hematocrit. This study aimed to compare the results of the CD95L examination involved in the apoptotic process and the results of blood tests in primary and secondary dengue patients. This was a cross-sectional study performed in a four months period  (September–December 2019) involving several clinics and doctor's private practices in Medan, Indonesia. Subjects were eighty-four dengue patients, consisting of 18 (21%) patients with primary infection and 66 (79%) with secondary infection. Data collected were tested with the Mann Whitney test with p-value of <0.05 considered significant. A significant difference (p value=0.007) was observed in the lymphocyte counts between primary and secondary dengue patients, but no differences were seen in CDL95 level, platelet count, leukocyte count, and hematocrit. In conclusion, except for the lymphocyte count, there is no difference in CD95L level and blood test results between primary and secondary dengue patients.

Abstract

High-density lipoprotein (HDL) functions to transport cholesterol from the blood vessel endothelium to the liver and excreted through the digestive tract so that it does not cause dyslipidemia, which is a risk factor for cardiovascular disease. Current treatment for dyslipidemia uses long-term statin drugs, which can cause various side effects, so herbal alternatives such as ketapang leaves (Terminalia catappa L.), which contain flavonoids and have the effect of increasing HDL levels, needed to be evaluated to achieve that reason. This study aims to evaluate whether ketapang leaf extract has an effect on increasing blood HDL levels. This research used experimental animals with a pretest and a posttest with a control group design. It was conducted at the Biochemistry Laboratory and Laboratory Animal Management Unit, Faculty of Medicine, Universitas Muhammadiyah Sumatera Utara, Medan, from September to December 2023. This study used Wistar strain male white rats (Rattus norvegicus) that were induced on a high-fat diet, divided into five groups: negative control, positive control (given simvastatin), treatment 1 (given 21 mg/kgBW ketapang leaf extract), treatment 2 (given 42 mg/kgBW ketapang leaf extract) and treatment 3 (given 84 mg/kgBW ketapang leaf extract). The study results showed a significant difference (p<0.05) in the average HDL levels before and after treatment in the positive control, treatment 2, and therapy three groups. Thus, the study concluded that ketapang leaf extract affected increasing HDL levels of male white rats (Rattus norvegicus).
