Abstract

The process of spontaneous labor is sensitive to the oxytocin hormone that causes uterine smooth muscle contraction as a receptor for the start of labor. Dates contains source of carbohydrates are simple sugars, iron, potassium, selenium, calcium, and flavonoids. The purpose of this study was to determine the effect of date’s extracts (Phoenix dactilifera L) to the oxytocin hormone day 19 of gestation age of mice (Mus musculus L). This experiment devide dose of date extract into four groups, group I : control (no treatment), group II: 3 dates (3.12 mg/kg), group III 5 dates (5.2 mg/kg), and group IV 7 dates (7.28 mg/kg) to the oxytocin hormone. The research method is done by extracting palm ajwa with methanol. Treat made orally at the age of 14-19 days gestation. Measurement of oxytocin levels using ELISA at the age of pregnancy 19 days, showed the average number of levels of the oxytocin hormone in group I in the amount of 126.494 ng / L, the second group is equal to 124.169 ng / L, group III amounting to 129.041 ng / L, and group IV is equal to 132.252 ng / L. Overall extract dates with various doses of the treatment led to increased levels of the oxytocin hormone in mice. However, these increases did not show a significant effect, although the hormones oxytocin on uterine tissue of mice kept rising. Keywords: Dates extract, Oxitocin levels, Pregnant mice

Abstract

Antioxidants can ward off free radicals in the body to resist oxidative damage caused by free radicals. The aim of this research is to know the influence of water solvent, methanol and methanol-water mixture to antioxidant activity. The extraction is done by maceration method. The extraction results in the phytochemical test and functional group analysis to determine the compounds contained in Pig leaf extract. Further testing of antioxidant activity with DPPH method. Pig leaf extract with methanol solvent has antioxidant activity with IC50 3,3005 μg / ml value, Pig leaf extract with water solvent has antioxidant activity 3,6976 μg / ml and Pig leaf extract with methanol solvent: water has antioxidant activity 13,6140 μg / ml. The IC50 <50 μg / ml value indicates that the Pig leaf extract with some solvents has potent antioxidant potential. Pig leaf extract with methanol solvent has the best antioxidant activity with IC50 3,3005 μg / ml value because according to phytochemical test and functional group analysis that methanol solvent is able to extract more active compound such as flavonoid, Triterpenoid and Sterol, Alkaloid and Saponin followed by Pig leaf extract with water solvent and methanol mixture: water.

Abstract

Salah satu tanaman yang berfungsi sebagai  antijamur  adalah bawang  putih yang diinovasikan menjadi produk bawang yang berwarna hitam atau black garlic. Penelitian ini bertujuan untuk mengetahui aktivitas antijamur ekstrak black barlic terhadap pertumbuhan jamur Candida albicans. Metoda dalam penelitian ini adalah bawang putih dipanaskan selama 35 hari pada suhu 65°C untuk mendapatkan black garlic. Black garlic diekstraksi dengan metode maserasi menggunakan pelarut methanol. Uji aktivitas antibakteri berdasarkan uji zona hambat, konsentrasi hambat minimal (KHM) dan konsentrasi bunuh minimal (KBM). Uji zona hambat dengan variasi konsentrasi ekstrak 100%, 90% dan 80%. Konsentrasi dengan nilai zona hambat yang baik akan dilanjutkan ke uji KHM dan KBM dengan pengenceran ke 1 sampai 5. Hasil penelitian menunjukkan bahwa zona hambat ekstrak Black Garlic dengan konsentrasi 100% memiliki zona hambat paling bagus yaitu 3,15 mm. Uji KHM optimum diperoleh pada pengenceran ke-5 dengan nilai OD terendah 0,460. Uji KBM dari ekstrak black garlic pada pengenceran ke-5 menunjukkan koloni jamur C. albicans masih tumbuh, sehingga dapat dikatakan bahwa ekstrak black garlic hanya mampu menghambat pertumbuhan jamur namun tidak bisa membunuh jamur C. albicans. Kata kunci: antijamur; black garlic; maserasi  ABSTRACTActivity test of black garlic extract against the growth of the fungi Candida albicans. One plant that functions as an antifungal is garlic which is innovated into black garlic. The aim of this study is to indentify black garlic extract antifungal activity against of the Candida albicans. The method in this research was heated garlic for 35 days at temperature of 65°C. Black garlic was extracted by maceration method using methanol solvent. Antibacterial activity test based on inhibition zone test, minimal inhibitory concentration (MIC) and minimal kill concentration (MBC). Inhibition zone test with various extract concentrations of 100%, 90% and 80%. Concentrations with good inhibition zone values will be continued to the MIC and MBC tests with dilutions 1 to 5. The results showed that the inhibition zone of black garlic extract with a concentration of 100%, 3.15 mm. The optimum MIC test was obtained at the 5 dilution with the lowest OD value of absorbance 0.460. The MBC test of the black garlic extract at the 5 dilution showed C. albicans fungal colonies were still growing, so it can be said that the black garlic extract was only able to inhibit fungal growth but could not kill C. albicans fungi. Keywords: antifungal; black garlic; maceration

Abstract

Abstrak Softgellcandy adalah permen bertekstur lunak yang diproses dengan penambahan komponen hidrokoloid seperti agar, gum, pektin, pati, kaegenan, gelatin dan lain-lain yang digunakan untuk modifikasi tekstur sehingga menghasilkan produk yang kenyal. Sayangnya distribusi Softgellcandy di pasaran seringkali terlepas dari pengawasan lembaga berwenang. Banyak ditemukan bermacam merk Softgellcandy yang tidak berBPOM maupun tidak berlabel Halal. Gelatin menjadi titik kritis kehalalan Softgellcandy. Penelitian ini menguji 15 sampel Softgellcandy tak berlabel halal yang dijual bebas di Surabaya dengan primer pengkode fragmen DNA cytochrome b Babi. Metode yang digunakan adalah konvensional PCR pada suhu 98oC-2 menit; 95oC-30 detik; 61oC-30 detik; 72oC-40 detik; dan 72oC-3 menit, selama 30 siklus. Visualisasi hasil PCR menggunakan elektroforesis 2% gel agarosa menunjukkan dari 15 sampel, 8 sampel terindikasi kontaminan DNA babi ditandai dengan pita DNA sebesar ±149bp. Pemerintah perlu melakukan monitoring lebih ketat terkait peredaran produk makanan tak berlabel halal yang dijual bebas di pasaran, mengingat Halal menjadi issue yang sangat sensitif di negara dengan mayoritas penduduk muslim terbesar di dunia ini. Abstract

One type of heavy metal that pollutes waters is heavy metal copper (Cu). Cu metal is basically an essential metal for living things, but with high levels of Cu metal, it can interfere with the body's metabolic processes. Therefore, there needs to be a solution to overcome Cu metal pollution in the waters. One of them is phytoremediation, or the use of plants to absorb pollutants. Spear leaf plant (Sagittaria lancifolia) is proven to be able to absorb heavy metal copper (Cu). However, the growth response is not yet known in more detail. Therefore, this study aimed to determine the growth response of pike leaf (Sagitaria lancifolia) under stress of heavy metal copper (Cu). This research is an experimental study with a completely randomized design (CRD). The experiment was conducted with 8 treatments (4 treatments with 2 tests) and 3 replications. The 4 treatments were distinguished based on differences in the concentration of Cu metal used, namely 0 mg/L, 1mg/L, 2mg/L, and 3 mg/L. The results showed that exposure to heavy metal copper (Cu) in each concentration variation and detention time variation did not have a significant effect on the growth and morphological conditions of spear leaf (Sagittaria lancifolia) plant, so this plant deserves to be used as an option in recovery efforts of water environment contaminated with heavy metal copper (Cu).Keywords – Response, Lanceleaf arrowhead, Sagittaria lancifolia, Heavy metals, Copper (Cu)

Abstract

One of the fermented traditional drinks that can have a health effect is kombucha. The materials that can be used in making kombucha is wuluh starfruit leaves. The research was conducted to determine the effect of sugar concentration of the physical, chemical characteristic, and antioxidant activities of wuluh starfruit leaf kombucha tea. This research is an experimental study with a complete randomized design method (CRD) consisting of 3 treatments. Kombucha wuluh starfruit leaves are made with a sugar concentration of 10%, 15% and 20%. The physical characteristics tested include organoleptic tests and the chemical characteristics include a test of the pH value, the total levels of titrated acids (tat), phenolics statistically  analyzed  using  the Kruskal Wallis test and the Mann Whitney test. The alcohol data and antioxidant activity (IC50) were analyzed descriptively. The results of the Kruskal Wallis test  have a significant difference in the pH. The best result based on the physical, chemical characteristic and antioxidant activity is were kombucha with a sugar concentration of 10% with a pH value of 3, a tat content of 0.11%, a phenolic content of 87.33 mg/ml GAE, an alcohol content of 0.41% and an IC50 value of 3.65 ppm.

Abstract

One of pollutant that commonly found in aquatic environment is detergent. Detergent pollution can cause skin irritation and other health problems in humans and animals that use contaminated water sources. To address this issue, it is critical to implement effective policies and regulations to control detergent pollution and promote sustainable wastewater management practices in Indonesia. One of the approaches to overcome detergent pollution is to use plants as pollutant absorbing agents, or commonly called phytoremediation techniques. Therefore, this study aims to determine the growth response of spear plant (Sagittaria lancifolia) in response to the stress of Linear Alkylbenzene Sulfonate (LAS) detergent. This research is an experimental research using a completely randomized design (CRD). There were four treatments tested with three replicates for each treatment. Treatment variations were based on the concentrations used in this study, namely 0 mg/L, 10 mg/L, 30 mg/L, and 50 mg/L. The research was conducted for four months, starting from July to October 2022, and was conducted in the greenhouse of Purwodadi Botanical Garden – BRIN. The result showed that although it experienced some morphological changes such as leaf drying, discoloration, and structural changes, Sagittaria lancifolia were able to survive and conduct metabolic activities properly during 28 days of Linear Alkylbenzene Sulfonates (LAS) pollutant stress.  Thus, Sagittaria lancifolia are able to be used as one of the strategies in restoring the aquatic environment polluted by domestic waste, including LAS detergent.

Abstract

Kurma (Phoenix dactylifera L.) merupakan buah yang banyak tumbuh di negara-negara Arab, terutama kota Madinah. Kurma memiliki lebih dari dua puluh jenis dan yang banyak beredar di Indonesia antara lain kurma ajwa, saudi arabia, tunisia, mesir madu, agal madinah, madinah, dan lulu. Identifikasi senyawa dan kandungan dari kurma jenis ajwa sendiri masih belum pernah dilakukan. Oleh karena itu tujuan dilakukannya penelitian ini adalah untuk mengidentifikasi senyawa-senyawa aktif yang terkandung dalam buah kurma ajwa. Daging buah kurma ajwa diekstraksi menggunakan metode maserasi dengan pelarut metanol. Ekstrak daging buah kurma diidentifikasi dengan melakukan uji fitokimia, analisis GC-MS, spektroskopi inframerah (FTIR) dan spektroskopi UV-Vis. Berdasarkan hasil uji fitokimia diketahui bahwa terdapat kandungan triterpenoid, flavonoid dan karbohidrat. Hasil analisis FTIR membuktikan bahwa adanya stretching dan bending gugus fungsi molekul seperti gugus –OH, -CH alifatik stretching, alkil, -C=C alifatik stretching / aromatik, aldehid, alkohol, eter, dan -CH aromatik. Pada analisis UV-Vis terdapat serapan di panjang gelombang 286 nm yang membuktikan bahwa flavonoid yang terkandung dalam ekstrak metanol daging buah kurma ajwa termasuk pada golongan flavanon.

Abstract

Stroberi (Vragaria ananassa Dutch) Var. Dorit merupakan buah yang populer di masyarakat dunia dengan ciri khas rasa buahnya yang asam manis dan segar. Penyediaan benih stroberi selama ini dilakukan secara konvensional dengan menggunakan stolon, sehingga volume perbanyakan relatif lebih sedikit dan tidak bebas penyakit. Oleh karena itu diperlukan metode kultur jaringan untuk budidaya stroberi. Kultur jaringan tanaman merupakan suatu teknik pengisolasian dan pemeliharaan sel atau potongan jaringan tanaman yang ditumbuhkan pada media buatan yang sesuai dan dalam keadaan aseptik. Medium yang dipergunakan pada kultur in vitro tumbuhan ada bermacam-macam. Medium yang digunakan dalam percobaan ini yaitu media MP yang merupakan kombinasi dari larutan MS (Murashige and skoog) dan pupuk cair. Dalam hal ini pupuk yang digunakan merupakan produk dari Balitjestro yang tentunya mengandung beberapa nutrisi dengan konsentrasi yang tepat untuk petumbuhan tanaman. Percobaan ini dilakukan untuk mengidentifikasi pengaruh berbagai modifikasi media MP serta mengidentifikasi konsentrasi kombinasi media MP yang sesuai terhadap pertumbuhan stroberi varietas dorit. Percobaan ini menggunakan Rancangan Acak Kelompok (RAK) dengan perlakuan MP1: MS 25% + pupuk 75%, MP2: MS 50% + pupuk 50%, MP3: MS 75% + pupuk 25%, MP4: pupuk 100%, Percobaan di ulang 5x, sehingga diperlukan 20 unit percobaan. Setiap unit percobaan ditanam 5 planlet. Pertumbuhan tanaman stroberi varietas dorit yang ditanam pada media MP1, MP2, MP3 dan MP4 menunjukkan pengaruh perbedaan yang signifikan terhadap pertumbuhan akar, pertumbuhan tunas dan pertambahan jumlah daun. Tanaman tersebut menunjukkan respon pertumbuhan yang paling cepat pada media MP3 karena mengandung konsentrasi larutan MS lebih banyak sehingga jumlah nitrogen yang dibutuhkan untuk pertumbuhan lebih tercukupi
