Abstract

Karst adalah bentang alam yang unik yang terbentuk akibat dari pelarutan batuan karbonat atau batuan gamping oleh air hujan. Di dalam kawasan karst terbentuk beberapa gua yang menjadi ci khas dari kawasan karst yang terbentuk. Penelitian ini bertujuan untuk mengetahui keanekaragaman Collembola gua yang berada di Kawasan Karst Malang Selatan. Sampel yang diambil adalah berasal dari tanah yang berada di lantai gua dengan menggunakan teknik Purposiv Sampling. Penelitian dilakukan pada empat gua yaitu Gua Harta, Gua Krompyang, Gua Prapatan JLS, dan Gua Lowo. Setiap pengambilan sampel dilakukan tiga kali pengulangan pada setiap zona, yaitu zona terang, zona remang, dan zona gelap. Hasil analisis data menghasilkan nilai Indeks Keanekaragaman H’=0,46121 dan Indeks Kemerataan E=0,28656.

Abstract

Halal food means food that permitted under Islamic law and fulfills about requirements. The absence of information about halal food contained in UIN Sunan Ampel Surabaya (UINSA) campus area causes related research to be carried out. This study aims to determine the porcine DNA contamination on food around UINSA area using molecular technology. Twenty two samples used were foods that contain meat and may contain pork obtained from canteens around UINSA area, analyzed using Polymerase Chain Reaction (PCR) method. The analysis was started with DNA isolation of 22 food samples, electrophoresis, PCR, then visualization gel electrophoresis. Primer gene coding for cytochrome b (cyt b) which produces 149 bp of DNA fragments. The results showed that no porcine contamination in 22 food samples, while the positive control showed a band of 149 bp. End point PCR method potentially to detect porcine DNA contaminants in food products around UINSA. Therefore the food is halal and safe for consumption.

Abstract

Micro-entrepreneurs with basic ingredients of processed meat such as meatball who do not have a meat grinder, generally using meat grinder at the public market. The problem that occurs is that there is no clear regulation from the Government regarding the guarantee of the halal meat grinding in the Regional Company. This needs to be enhanced as a study, considering that the grinding material does not only come from Halal substances. The purpose of this study was to test pig DNA in meat grinding samples at PD Pasar Surya Surabaya City by using the conventional Polymerase Chain Reaction (PCR) method. DNA was isolated from 11 PD Pasar Surya meat grinding samples, then spectrophotometry was performed. Spectrophotometry results showed that all samples have high DNA concentrations. The primer used is the cyt b pig gene encoder. Predenaturation is performed at a temperature of 95°C-5 minutes, denaturation of 95°C-45 seconds, annealing 60°C-30 seconds, extension 72°C-40 seconds, and post extension 72°C-5 minutes. The results of PCR analysis were determined by the emergence of DNA bands of ± 149 bp as markers of pig DNA. The results showed negative on sample or no pig contamination in 11 samples tested. While the pig sample as positive control showed a band of ± 149 bp. These results prove that at 11 points of the location of meat grinding there is no contamination of pig DNA.

Abstract

Abstrak Softgellcandy adalah permen bertekstur lunak yang diproses dengan penambahan komponen hidrokoloid seperti agar, gum, pektin, pati, kaegenan, gelatin dan lain-lain yang digunakan untuk modifikasi tekstur sehingga menghasilkan produk yang kenyal. Sayangnya distribusi Softgellcandy di pasaran seringkali terlepas dari pengawasan lembaga berwenang. Banyak ditemukan bermacam merk Softgellcandy yang tidak berBPOM maupun tidak berlabel Halal. Gelatin menjadi titik kritis kehalalan Softgellcandy. Penelitian ini menguji 15 sampel Softgellcandy tak berlabel halal yang dijual bebas di Surabaya dengan primer pengkode fragmen DNA cytochrome b Babi. Metode yang digunakan adalah konvensional PCR pada suhu 98oC-2 menit; 95oC-30 detik; 61oC-30 detik; 72oC-40 detik; dan 72oC-3 menit, selama 30 siklus. Visualisasi hasil PCR menggunakan elektroforesis 2% gel agarosa menunjukkan dari 15 sampel, 8 sampel terindikasi kontaminan DNA babi ditandai dengan pita DNA sebesar ±149bp. Pemerintah perlu melakukan monitoring lebih ketat terkait peredaran produk makanan tak berlabel halal yang dijual bebas di pasaran, mengingat Halal menjadi issue yang sangat sensitif di negara dengan mayoritas penduduk muslim terbesar di dunia ini. Abstract

As one of the particulate chemicals, dust could occur in most of the production process and can create interference for workers health and safety. As one of the air pollution sources, dust could became a potential hazard which exist in room maintenances. Protection to workers is a must in order to reduce the risk of respiratory tract syndrome that often could be found in this cases. The aim of this study is to conduct a risk assessment to dust exposure in room maintenance, which held by contractors in PT. X (Persero) building in Surabaya. This is an cross sectional study with obsevation approach. The object of this research is the repairing works. The results indicate that the activities which could produce dust, such as: walls sanding using sandpaper, the tiles dismantle, sawmilling, the wood fiber refining, grinding, mixing and stirring cast materials, and room cleaning. Dust produced from a variety of works including sanddust, cement, lime, wood and dust mixed with paint. The results show that three types of works considere as high-risk activity (value > 12-25), 3 types of work consider as midle risk activities (value > 5-12), and one activity considered as a low-risk work (grades 1-5). The dusk factors controlling should be held regularly, in order to minimize the risk leveln againts the workers.

Abstract

Pangan yang berasal dari ternak sangat kita butuhkan karena mengandung protein penting dan mudah dicerna, daging merupakan salah satu pangan yang berasal dari ternak dan sangat bermanfaat bagi tubuh karena mengandung gizi yang lengkap sehingga perlu untuk dilakukan pencegahan kontaminasi terutama oleh bakteri Eschericia coli. Tujuan dari penelitian ini adalah untuk mengetahui kontaminasi bakteri Escherichia coli pada beberapa sampel daging. Penelitian ini termasuk dalam penelitian eksperimental untuk mengidentifikasi bakteri Escherichia coli pada beberapa daging segar menggunakan metode pengenceran dan lama waktu inkubasi yang berbeda dari setiap sampel. Berdasarkan penelitian yang telah dilakukan diperoleh jumlah kontaminasi bakteri Escherichia coli tertinggi pada sampel daging ayam daripada daging sapi dan daging kambing. Hasil tertinggi kontaminasi bakteri Escherichia coli daging ayam yang melebihi BMCM adalah sebesar 1108 : 1 x 109 dengan kode DA 049-DA 058. Kesimpulan berdasarkan hasil yang diperoleh bahwa 18 sampel daging ayam sebanyak (50%) sampel terkontaminasi Escherichia coli, 13 sampel daging sapi sebanyak (38%) terkontaminasi Escherichia coli. Sedangkan 1 daging kambing (0%) tidak terkontaminasi bakteri Escherichia coli.

Abstract

Spodoptera litura is a pest in vegetable plants that attacks the leaves and stems. Damage arising from the attack of S. litura can decrease the productivity of plants. Synthetic pesticides are often used in controlling the population of S. litura, but synthetic pesticides have a high negative impact. Potential entomopathogenic nematode Heterorhabditis spp. believed to be effective for controlling pest populations. This study aims to find out the pathogenic value of entomopathogenic nematode Heterorhabditis spp. to the larvae of S. litura. Based on the results obtained Heterorhabditis spp. has a positive influence in causing mortality in the larvae of S. litura. Heterorhabditis spp. mortality of up to 42%. Symptoms indicated by larvae of S. litura affected by Heterorhabditis spp. among them the behavior becomes passive, the body becomes flaccid, the cuticle turns red and the tissues inside the body are destroyed. Obtained pathogenicity value Lc 50 Heterorhabditis spp. 7,690 IJ/ml, as for the factors affecting Heterorhabditis spp. such as humidity, temperature, pH, and light intensity.Keywords – Entomopathogenic Nematodes, Pathogenicity, Spodoptera litura

Abstract

Softgel candy is soft-textured confectionery processed by the addition of several components such as gum, pectin, starch and gelatin, to obtain a supple product and packed after aging treatment first. Gelatin is one of the main components in the manufacture of soft candy derived from the hydrolysis of collagen connective tissue and animal bone that serves as the nature of gelling agents, stabilizers or emulsifiers. However, the gelatin used in products not yet labeled halal Indonesian Council of Ulama (MUI) is particularly vulnerable to pork gelatin, since pork gelatin is cheaper than cattle. The purpose of this study was to test the contaminants of pig DNA on 17 samples of soft candles not labeled halal MUI. This research used Polymerase Chain Reaction (PCR) method. Seventeen samples were isolated by DNA, then spectrophotometry was performed, followed by PCR. The PCR product is run electrophoresis. Visualize the DNA with a UV gel documentation. Primer used is primer gene encoding cyt b DNA pork. Results showed that 17 samples were negative contaminants, while the positive control of pork showed a DNA band of 149 bp. This shows that Softgel Candy 17 samples do not contain pork gelatin.

Abstract

Rice field ecosystem is an artificial ecosystem that is used in agriculture. Rice fields in Seketi Village have vegetation types of corn, sugar cane, and rice, so that it will affect of grasshopper diversity in each vegetation. The purpose of this study was to determine the diversity of grasshoppers (Caelifera) and to determine the effect of vegetation on the diversity of grasshoppers in the rice fields of Seketi Village. The method used is purposive sampling and direct capture, namely research by making 5 plots measuring 5m x 5m with different types of plant vegetation (paddy, corn, sugar cane) catching with Sweep Net (insect nets). The results showed that there were species of Phlaeoba antennata, Phlaeoba fomusa, Oxya chinensis, Valanga nigricornis, Caryanda spuria, Atractomorpha crenulata, and Tettigidea sp. with a total of 152 individuals from 3 families. The diversity index value shows that plot 4 (maize plants) has the highest H' = 1.58 with 6 species of grasshoppers found and plot 1 (cane plants) has the lowest diversity index value of H = 1.09. Based on the differences in the value of grasshopper diversity in each plot caused by different types of vegetation, habitat conditions, and abiotic factors.

Abstract

Collembola is one of the soil fauna that acts as the smallest constituent component for the balance of the ecosystem. This study aims to compare the structure of the Collembola community in the habitat of Lowo Cave and Citizen's Garden in Melirang Village, Gresik Regency. This research uses descriptive exploratory research with soil drill method for soil sampling and pitfall trap for ground surface insect traps. Based on Collembola data obtained in Lowo Cave, there are 3 species with a total of 87 individuals, while in Citizen's Garden there are 6 Collembola species with a total of 145 individuals The results of the analysis of this study indicate that the Diversity Index value in the Lowo Cave habitat is H'= 0.859735 and in the Citizen's Garden a habitat with the value H' = 1.475675, the Evenness Index value in the Lowo Cave is E = 0.782565 and in the Citizen's Garden with a value of E = 0.82359, the value of the Dominance Index in Gua Lowo is 0.48104 and in the Citizen's Garden is 0.282949, the Relative Frequency of Ascocyrtus sp. and Hypogastrura consanguinea have a relative frequency of 100%.
