Abstract

Tea is well known as favourite healthy drink for almost all people over the world. Tea propagation using conventional and modern ways are now developing rapidly. However, information regarding the protein profile of tissue culture of tea plant has not been revealed yet. This study aimed to determine the difference of protein profile of tea’s tissue culture using SDS-PAGE. This study was conducted using embryonic axes of TRI2025 tea clone cultured on MS media supplemented with 2,4-D for inducing somatic embryogenesis and globular-like structure (GLS) regeneration, and MS media supplemented with BAP for inducing shoot via organogenesis. The results revealed that proteins in the size of 37.69; 54.89; 60.77; 71.35; 87.34; and 92.99 KDa might be involved at somatic embryogenesis, and about 38.69 KDa, 69.27 KDa, and 55.76 KDa respectively for GLS, initiation of shoot, and initiation of GLS derived leaf. Predicted key protein for leaf initiation both directly or through GLS was about 31-33 KDa, while for callusing were about 27.56 and 52.73 KDa, and constitutive protein was about 22.75 KDa. This study provides the first report of protein profile of tea’s tissue culture. The information obtained can be beneficial as a marker for explant for somatic embryogenesis, GLS, or organogenesis pathway and as a scientific information for further biotechnology development.

Abstract

Camellia sinensis L. is an important crop in Indonesia as healthy beverage that contains several secondary metabolism compounds, such as polyphenols and catechins. Tissue culture including somatic embryogenesis and organogenesis has been used for propagating plant for various needs. In this present short-communication, scanning electron microscopic (SEM) analysis of tea was conducted and discussed. This study aimed to investigate surface ultrastructure of TRI2025 embryo axis tea clone cultured on Murashige and Skoog (MS) medium containing 2,4-Dichlorophenoxyacetic acid (2,4-D). The results revealed two different forms of explant’s development, i.e. somatic embryo and transitional form between somatic embryogenesis and organogenesis; or called by “Globular-like Structure” (GLS). Surface ultrastructure analysis of somatic embryo and GLS revealed respectively many stages of somatic embryo development i.e. globular, torpedo, and cotyledon stage, and leaf development form GLS regeneration.

Abstract

Cayenne pepper is one of vegetables that is consumed almost every day by every household. However, cayenne pepper is characterized by not being long-lasting, so efforts are needed to extend the shelf life of cayenne peppers. Ecoenzyme is known as a multipurpose solution that has antimicrobial properties, so it is thought that ecoenzyme can be used as a natural preservative, for example to extend the shelf life cayenne pepper. This research aims to determine the effectiveness of ecoenzyme solutions in extending the shelf life of cayenne pepper. The research was carried out using fruit-derived ecoenzyme solution and vegetable-derived ecoenzyme solution, with treatment without dilution, ecoenzyme solutions with a concentration of 75%, and ecoenzyme solutions with a concentration of 50%. The results of the research showed that at full concentration without dilution, fruit-derived or vegetables-derived ecoenzyme solution can extend the shelf life of cayenne pepper to 10 days. It is hoped that these results will be useful for post-harvest purposes of cayenne pepper, both on a household and larger scale.

Abstract

Program pengabdian ini bertujuan untuk memberdayakan ibu-ibu PKK melalui peningkatan keterampilan dan pengetahuan dalam pembuatan produk ecoprint. Fokus utama adalah meningkatkan kemandirian ekonomi, kesadaran lingkungan, serta keterampilan dan pengetahuan peserta. Metode yang digunakan mencakup pelatihan langsung dan pendampingan dalam pembuatan ecoprint dari bahan alami, serta strategi pemasaran produk melalui media sosial dan jaringan lokal. Hasil menunjukkan peningkatan signifikan dalam kemandirian ekonomi, dengan ibu-ibu mampu menghasilkan produk ecoprint bernilai jual tinggi. Selain itu, terdapat peningkatan kesadaran lingkungan dan keterampilan baru yang dapat diaplikasikan dalam kegiatan ekonomi lainnya. Kegiatan ini juga berhasil memperkuat jejaring sosial antar ibu-ibu PKK, mendukung potensi pengembangan ekonomi di masa depan. Program ini berkontribusi pada pemberdayaan ekonomi keluarga dan pelestarian lingkungan melalui produk ecoprint yang inovatif dan berkelanjutan.

Abstract

The aloe vera is known as an ornamental plant that can survive almost all elevations and has a myriad of benefits for humans. This plant can plant at various altitudes, including in the districts of Sukoharjo and Wonogiri. This study aimed to identify the physiological and morphological aspects of aloe vera (Aloe vera) planted in Sukoharjo and Wonogiri districts. Parameters measured were ambient temperature, leaf length, number of stomata, and transpiration rate. Stomata number parameters were calculated using the leaf extension count method under a microscope, while for transpiration rate analysis, cobalt chloride paper was used. The results of this study showed no significant differences in the parameters of average temperature, leaf length, number of stomata, and transpiration rate. The results of this study can be used as a suggestion, especially for Sukoharjo district policy makers, that the aloe vera plant has the potential to become one of the leading agricultural commodities.

Abstract

Identifikasi jenis tumbuhan Anggrek adalah suatu tahapan untuk mengidentifikasi spesies tumbuhan Anggrek yang ditemukan di lokasi penelitian secara runtut sehingga dapat ditemukan taksonominya. Anggrek yang terdapat di kawasan UPTD Taman Anggrek Magelang sangat beragam, mulai dari yang habitat aslinya tumbuh secara  secara epifit maupun terestrial. Penelitian ini bertujuan mengetahui keragaman jenis  spesies Anggrek di Kawasan UPTD Taman Anggrek Magelang. Metode dalam peneli-tian ini yaitu jelajah (survey eksploratif). Data penelitian ini diambil di wilayah kebun Anggrek UPTD Taman Anggrek Magelang. Instrumen dalam penelitian ini mengguna-kan tabel pengamatan. Berdasar hasil penelitian terdapat 19 spesies  Anggrek yang tergolong ke dalam 10 genus dari 1 famili, yang terdapat di UPTD Taman Ang-grek Magelang, diantaranya  adalah Ang-grek Arachnis flos-aeris, Vanda dou-glas, Vanda (nilareta agrihorti), Vanda (Orange),  Grammatophy-llum  speciosum, Dendrobi-um bigibbum, Bulbophyllum lobbii, Den-drobium bicolor, Dendrobum superbiens, Dendrobium Sp., Oncydium spachelatum, Cymbidium bico-lor, Cymbi-dium golden shower, Cymbidium leucochi-llum, Doritis pullcherima pink, Phalaenopsis amabilis (L.), dan  Doritaenopsis.

Abstract

A booklet is a combination of a leaflet and a small book so it is practical and can be used as a guide. The aim of the research is to develop a booklet on pests and diseases of crystal guava as well as online-based management methods. Pest and disease booklets need to be developed considering that pests and diseases are the main obstacles to agricultural productivity, disrupting plant physiology and growth so that they can reduce crop yields. Data collection was carried out through observing samples of crystal guava plants, as well as interviews with owners and managers to obtain information about pests and diseases of crystal guava. Data was analyzed descriptively qualitatively. The results of the research found 4 crystal guava pests, namely: codot (Cynopterus spp.), ladybugs (Epilachna admirabilis), fruit flies (Bactrocera dorsalis), and mealybugs (Paracoccus marginatus). The diseases found were the fungus Capnodium citri (sooty dew), algae (Chephaleuros virescens), fungus (Festlotiopsis psidii), and nutrient deficiencies (potassium/magnesium). Crystal guava is cared for by keeping the garden clean, spacing the plants, applying fertilizer, spraying insecticide, installing fruit fly traps, and wrapping the young fruit. This research still needs to be carried out in two seasons, namely dry and rainy, so that it can produce more types of pests and diseases, so that the online booklet results are more optimal.

Abstract

Wild edible plants" is a term used to refer to wild plants that can be consumed or used as a food source by humans. Village communities often use wild edible plants to obtain additional food sources, especially in situations where access to other food ingredients is limited. The aim of this research is to find out the types of wild plants that are often used as alternative food ingredients, including the parts of the plants used and the processing efforts. The data analysis method used in this research is the qualitative descriptive method. Based on the results of interviews with informants, namely the people of Pakis Baru Village, 37 types of wild edible plants from 22 families were obtained which were used as alternative food, including tubers, vegetables and fruit. The most widely used wild edible plants, namely vegetables (24 species), followed by tubers (6 species), fruits (6 species) and other preparations in the form of grass jelly (1 species). The parts or organs of wild plants consumed are young leaves or leaf shoots, stems, flowers, fruit, bamboo shoots and tubers. The common processing methods are steaming, boiling, sautéing, roasting, botok, extracting the starch for certain food preparations, or consuming it directly as fresh vegetables. The high diversity of wild edible plant species shows that Pakis Baru Village has great potential as an alternative food source. From utilizing this diversity, society can develop varied and sustainable food strategies.

Abstract

Limbah organik rumah tangga perlu ditangani dengan baik supaya tidak mengganggu lingkungan. Kulit sayur dan atau kulit buah serta minyak jelantah merupakan contoh limbah organik rumah tangga yang perlu ditangani dengan benar. Pengabdian kepada masyarakat ini dilakukan di desa Kalitekuk yang bertujuan memberikan pelatihan kepada masyarakat mitra terkait pengelolaan limbah organik rumah tangga berupa kulit sayur dan atau kulit buah serta minyak jelantah. Tim pengabdi memberikan solusi berupa pembuatan larutan ekoenzim dan pembuatan lilin aromaterapi sebagai usaha dalam mengolah limbah organik rumah tangga. Masyarakat mitra menyambut baik pelatihan ini yang ditandai dengan meningkatnya pemahaman terkait pembuatan larutan ekoenzim dan lilin aromaterapi.

Abstract

Pengabdian masyarakat ini bertujuan untuk memberdayakan ibu rumah tangga di Desa Badran Laban melalui produksi batik ecoprint menggunakan daun-daunan sekitar rumah. Metode pengabdian meliputi pelatihan teknik pukulan dan proses pembuatan batik ecoprint. Hasil kegiatan ini menunjukkan bahwa ibu rumah tangga berhasil memperoleh pengetahuan dan keterampilan dalam memproduksi batik ecoprint yang memuaskan sehingga dapat sebagai peluang usaha baru. Kegiatan ini juga berhasil memberdayakan ibu rumah tangga dan mendorong semangat meningkatkan ekonomi keluarga. Kesimpulannya, program pengabdian ini berhasil memberikan manfaat nyata bagi masyarakat lokal dalam hal peningkatan keterampilan dan pemberdayaan ekonomi.
