Abstract

This study was to analyze the optimum dose of IL-6 induction in pregnant rats that could trigger an increase in mean arterial pressure and protein as two symptoms of preeclampsia. A total of 25 pregnant rats was divided into 5 groups, including pregnant rats the control group (without induction of IL-6), a group of pregnant rats were given an induction in IL-6 doses of 1.25 ng/day, a group of pregnant rats given doses of IL-6 induction 2.5 ng/day, a group of pregnant rats were given an induction of IL-6 doses of 5 ng/day, and groups of pregnant rats were given an induction in IL-6 doses of 10 ng/day. Induction of IL-6 was performed on the tenth day of gestation for 5 days. The expression of caspase-3, IL-17, and STAT3 were analyzed by confocal laser scanning microscopy. The expression of caspase-3, IL-17 and STAT3 was significantly higher in preeclampsia group than the control group (p<0.05). The expression of caspase-3 decreased significantly in all groups were treated by phycocyanin compared to the preeclamptic group (p<0.05), but has not been able to reach expression comparable to the control group (p<0.05). Expression of IL-17 decreased significantly in the group given the two highest doses of phycocyanin compared to the preeclampsia group (p<0.05). STAT3 expression decreased significantly in all groups were treatedby phycocyanin compared to the preeclamptic group (p<0.05), reached expression comparable to the control group in the group received phycocyanin at doses of 10 and 20 ng (p> 0.05). In conclusion, IL-6 on pregnant rats were able to increase apoptosis through IL-17 and STAT pathway. Inhibition of apoptosis due to phycoyanin treatment not only involve the formation of IL-17, this data is found in phycocyanin doses of 10 and 20 ng.

Abstract

Anxiety is a mental disorder common in elderly people. In the process of becoming elderly, feelings of anxiety arise as a normal response to situations that accompany development, change, new experiences and find meaning in life. Anxious looks from physiological symptoms such as shaking, sweating, heart work increases and psychological symptoms such as panic, tension, confusion, unable to concentrate. This condition will inhibit the elderlys bio-psycho-social condition in living the rest of his life. One effect is seen from their sleep problems. This is observational research with cross-sectional approach. Samples were taken by consecutive sampling technique. 43 elderly who met the exclusion criteria. Research location at Posyandu Lansia Mojo, Surabaya. Data collection using Hamilton Rating Scale for Anxiety and Insomnia Severity Index questionnaires. the results of the study described 41.9% of subjects with anxiety disorders and 67.4% of subjects had insomnia disorders. The cross-tabulation results showed 48% of subjects without anxiety can occur insomnia disorders and only mild anxiety alone can cause insomnia in 100% of subjects. Rank Spearman Test shows a very strong relationship between anxiety with the incidence of insomnia in the elderly (Sig .000; coefficient correlation .535).

Abstract

Background: Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis, can affect almost all organs of the body with the most locations in the lungs. Immunity and high nutrients such as vitamin C, zinc and other antioxidants have shown good effects for infections, including tuberculosis. One effort to help accelerate sputum conversion of people with pulmonary tuberculosis is by supplementation of Zinc Sulphate and Omega 3.Objective: Analyze the role of Zinc Sulphate and Omega 3 on sputum conversion acceleration in patients with pulmonary tuberculosis at Surabaya Paru Hospital 2015.Method: This research use Randomized Pre Post Test Post Control Group Design design. In patients with pulmonary tuberculosis with Zinc Sulphate and Omega 3 in the treatment and placebo group in the control group. A sample of 20 patients consisting of 10 treatment patients and 10 control patients. Statistical test using paired T-Test, Friedman test and Mann whitney.Results: Friedman test showed that in the treatment group showed significant difference in the number of BTA between before and after supplementation (p=0,001), while in the control group significance of 0.141. Mann-Whitney test  showed no significant difference in the amount of reduction of BTA between the treatment group and the control group (p = 0.080). This means that statistically does not show any significant difference.Conclusions: Zinc Sulphate and Omega 3 play a role in accelerating sputum smear conversion of respondents. The results of smear test in pulmonary TB patients with Zinc Sulphate and Omega 3 supplementation showed positive results

Abstract

Atherosclerosis causes significant morbidity and mortality. Carotid intima-media thickness (CIMT) can predict the onset of cardiovascular and ischemic stroke in the future. CIMT, the measurement of atherosclerosis, is determined in vivo using carotid ultrasound. CIMT is associated with traditional cardiovascular risk factors such as age, sex, race, smoking habits, alcohol consumption, exercise habits, blood pressure, dyslipidemia, dietary patterns, medications used, glycemia, hyperuricemia, anthropometric parameters associated with obesity. In addition, CIMT is also associated with novel risk factors, ie genotypes, cardiovascular anthropometry parameters, rheumatoid arthritis, immunologic diseases, inflammatory cytokines, lipid peroxidation, chemocyte anthropometric parameters, infectious diseases, vitamin D, matrix metalloproteinases, and other diseases

Abstract

The purpose of this study was to evaluate the potencial influence local product Abpo PMSG treatment on mice foetus number. Methode used for this research was by injecting 0,1 ml Abpo PMSG on superovulated mice with PMSG and hCG. There were six treatment in this research. Control group was group  of mice that only superovulated. P1 was given PMSG and Abpo PMSG together. P2 was group of mice that wich Abpo given 1 hour after PMSG, P3 given Abpo PMSG when they were estrous. P4 given Abpo PMSG 1 hour before hCG, and P5 given Abpo PMSG 1 hour after hCG. The result showed that the most foetus number resulted from P3. The least foetus number was from control group. Foetus number of P1, P2, P4 and P5 was showed no significant difference with P3 group ( P > 0,05).Key words : PMSG, Abpo PMSG, foetus number

Abstract

Wound healing is a complex dynamic process characterized by a series of events that occur in almost all type of tissue damage. In the early phase of the inflammatory response, neutrophils and macrophages enters into the injured tissue and the cells produce reactive oxygen species that can give a beneficial or detrimental effects. Oxidative stress is a condition occurs that shows imbalance between prooxidant or free radical and antioxidant that have a function to maintain the condition of the tissue damage that occurs. So Oxidative stress occurs when the production of Reactive Oxygen Species occurring is higher than the antioxidants existing as an intrinsic defense. Reactive Oxygen Species and Reactive Nitrogen Species are important components in the healing process of wounds and is necessary to be in the state of homeostasis to prevent oksidatif stress. The major components of ROS are superoxide (O2•), hydroxyl radical (OH•) and hydrogen peroxide (H2O2), which includes RNS are nitric oxide (NO•), nitrous oxide (NO2•), nitroxyl anion (HNO) and peroxynitrite (ONOO-) which could be form by the reaction between superoxide and nitric oxide. The existence of excessive O2 amount in the wound and the presence of excess NO can increase the incidence of oxidative stress that interfere with wound healing process. Oxidative stress plays a role in the inflammatory phase, proliferation and remodeling phase by increasing angiogenesis and affect other cells including endothelial cells in secreting NO. So the strategy in controlling oxidative stress is by increasing antioxidant level which is a scavenger to free radical excessive superoxide formation so preventing interference with the wound healing process.

Abstract

Obesity is a health problem whose prevalence tends to increase in Indonesia. Obesity becomes a concern, because it is one of the parameters that often followed by impaired of glucose tolerance in the metabolic syndrome. In Indonesia, Tamarindus indica is believed as a body slimming substance so it has possibility to become an alternative therapy that is easily available and cheap for obesity. The purpose of this study was to determine the ability of Tamarindus indica in inhibiting obesity. This experiment used 28 male Wistar strains in 2-3 months age that were divided into groups: group 1 (negative control, normal diet without Tamarindus indica), group 2 (positive control, obese diet without Tamarindus indica), group 3 (obese diet + Tamarindus indica 2700mg/kg BW), and group 4 (obese diet + Tamarindus indica 3600mg/kg BW). Measurement were performed in the first day after acclimization before treatment (day 8), and repeated weekly afterwards, ie at day 15, 22, 29, 36, and 43. Anova and Tukey Simultaneous statistics test were performed to see the differences in wight gain per week compared with the first week of study, then compared between groups.  Furthermore, a virtual screening that called insilico bioinformatics analyzes was performed to test the binding activity between quercetin and rutin (compounds in Tamarindus indica) with fatty acid synthase, an enzym that plays a role in de novo synthesis of fatty acids. Tamarindus indica was shown to significantly inhibit weight gain in rats with obese diet food (P=0.000), but differences dose of Tamarindus indica did not appear to have significant effect to inhibit obesity (P=0.321). The ability of Tamarindus indica is due to the compounds quercetin and rutin, where these two ingredients estimated to have the ability to inhibit fatty acid synthase with high strength.

Abstract

Alloxan causes the formation of reactive oxygen through the reduction process of alloxan (dialuric acid) in the β cells of pancreas. Free radicals cause damage  and decrease catalase enzyme activity. Black cumin seeds extract which contain carotenoids, thymoquinone, carvacrol, 4-terpineol, t-anethole, and tert-butylhydroquinone (TBHQ) has antioxidant activity that is expected to inhibit reactive oxygen species (ROS) that is expected to increase pancreatic catalase activity. This study used 24 male Wistar rats, divided into 3 groups:1)group of rats fed with standard food, 2)group of rats induced by alloxan 125mg/kg /BB intraperitoneally on day 1, 3)groups of rats induced by alloxan 125mg/kg/BB intraperitoneally on day 1 and were given by black cumin seed extract 2500mg/kg/BB orally on day 4 until 17. On the 18th day of all rats were sacrificed and pancreatic tissue catalase activity were measured by the spectrophotometric method. The results of One-Way ANOVA showed significant difference (p=0.003) pancreatic tissue catalase activity of groups of rats fed with standard food (1696,96U/g±414,10U/g) compared with group of rats induced by alloxan (1287,58U/g±88,45U/g). There was significant difference (p=0.001) of pancreatic tissue catalase activity of groups of rats induced by alloxan (1287,58U/g±88,45U/g) with a group of rats induced by alloxan and given with black cumin seed extract (1764,83U/g±199,69U/g). The conclusion of this study showed that the administration of alloxan significantly decreased pancreatic tissue catalase activity of rat and black cumin seed extract significantly increased catalase activity because cumin extract has antioxidant effects.

Abstract

Immature Granulocytes are considered as an inflammation marker. Recently Delta He is suspected to be a novel marker of inflammation. The aim of this study was to know the correlation of Immature Granulocytes and Delta He as inflammation marker in patients with lecocytosis. This research used whole blood-EDTA samples from 50 lecocytosis patients in the Dr. Soetomo Hospital Surabaya from 10 until 17 December, 2011 were studied Immature granulocytes and Delta He were examined by Flowcytometry (Sysmex XE 2000i). Analysis of the results was performed by Pearson product moment. Mean result of Immature Granulocytes and Delta He from 50 patients with lecocytosis was 0.37 x 103/µL and 3.03 pg. According to Pearson analysis test there was a correlation between Immature Granulocytes and Delta He (r = 0.26), but this was not significant (p > 0.05). Inflammation induced cytokine release and rise hepcidin, the result was decrement of ferroportin from duodenum and macrophage so the iron supply to erythropoetic precursor decrease, and the hemoglobin synthesis (Ret He) decrease. The rise of Immature Granulocytes were not happened in all lecocytosis patients, it were not certain suffered sepsis. There was no significant correlation between Immature Granulocytes and Delta He in patients with lecocytosis. A further research with more samples to determine that Delta He can be used as an inflammation marker is needed
