Abstract

Data collection in this study used a descriptive method, namely collecting, describing, and describing events systematically and accurately in accordance with the objects observed. This method aims to provide a systematic, factual, and accurate picture of the facts, nature, and relationships between phenomena, test hypotheses, make predictions and get the meaning and implications of the problems investigated. It can be seen that the attachment of squid eggs in the second location has not been observed during observations from August 2022 to September 2022. Observations at the East Malalayang One location did not find any laying of eggs, due to cloudy water conditions, while the two West One Malalayang locations found sticking of eggs with a total of 241 eggs attached, the number of attachments to the iron type attractor was 202 squid eggs and to the drum type namely 39 squid eggs. Squid attractors in the form of iron are mostly attached to eggs where the effectiveness rate is between 83.8% while for the type of used drum attractor, it is only around 16.2%. The form of the iron attractor is more effective because the iron attractor's cover is covered with parent nets. It can also be seen that the effectiveness of attaching squid eggs according to the level of water depth, for the effectiveness of squid sticking at a depth of 3 meters (53.53%) compared to only 5 meters (46.47%). Keywords: Attractor, Squid eggs, Manado Bay Abstrak Pengambilan data dalam penelitian ini menggunakan metode deskriptif yaitu mengumpulkan, menguraikan, dan menggambarkan peristiwa secara sistematis dan akurat sesuai dengan objek yang diamati. Tujuan penggunaan metode ini yaitu untuk memberikan gambaran secara sistematis, faktual dan akurat tentang fakta, sifat, serta hubungan antara fenomena, menguji hipotesa, membuat prediksi dan mendapatkan makna serta implikasi dari masalah yang diselidiki. Pengamatan di lokasi Malalayang Satu Timur tidak ditemukan penempelan telur, dikarenakan kondisi perairan yang keruh, sedangkan lokasi dua Malalayang Satu Barat ditemukan penempelan telur dengan jumlah penempelan 241 telur, jumlah penempelan pada atraktor tipe besi yakni 202 telur cumi dan pada tipe drum yakni 39 telur cumi. Atraktor cumi berbentuk besi lebih banyak di tempeli dengan telur dimana tingkat efektifitasnya antara 83,8% sedangkan untuk tipe atraktor drum bekas hanya sekitar 16,2%.  Bentuk atraktor besi lebih efektif karena bagian penutup atraktor besi semuanya ditutupi jaring paranet. Terlihat juga efektivitas penempelan telur cumi menurut tingkat kedalaman perairan, untuk efektivitas cumi yang menempel pada kedalaman 3 meter yakni (53,53%) dibandingkan 5 meter hanya (46,47%).             Atraktor; Telur cumi-cumi; Teluk Manado

Abstract

Observations of temperature, salinity, and dissolved oxygen have been carried out in Amurang Bay waters, North Sulawesi. This study aims to determine the temperature, salinity, and dissolved oxygen during the west and east seasonal monsoons. The statistical analysis results using the t-test showed that the temperature, salinity, and dissolved oxygen in the West monsoon with the east monsoon were significantly different. Temperature, salinity, and dissolved oxygen in the west monsoon are lower than in the east. Variations in temperature, salinity, and dissolved oxygen are influenced by atmospheric conditions, weather, rainfall, and current patterns. Implicitly, temperature and salinity can affect the solubility of oxygen in the East monsoon. The value of AOU (Apparent Oxygen Utilization) in the west monsoon is less than 0.00 mg/l (negative), which indicates that oxygen is very much needed for the process of respiration and decomposition of organic substances during the west monsoon. Abstract

Vannamei shrimp (Litopenaeus vannamei) is a brackish water commodity that is currently in great demand and is a leading producer of the aquaculture sector in Indonesia. This is because some of the advantages possessed by the vannamei shrimp, among others, can be maintained with a high stocking density, rapid growth, has a high resistance to disease and environmental changes. The mating process occurs through four stages, namely approach, pursuit, pride, and mating. This process can be known by looking at the behavior of the male parent who swims to follow the female parent. They both looked like a chase. Then the male parent swims parallel to the female and turns her body towards the female shrimp's belly. After that, the male grabs the female and releases sperm that attach to the thellycum. In the maintenance of larvae that must be considered is the management of water quality and feed management. This is because water is a living medium for aquatic organisms, so it determines the survival of larvae. Abstract

The existence and sustainability of mangrove forests have a significant role in the environment and communities around the coast. Mangrove ecosystems also have complex systems including plant and animal organisms that interact with one another and play an essential role in maintaining and protecting the coastline. This research was conducted to analyze the potential for the regeneration of mangrove seedlings in the coastal area of Dafi Village, Biak Numfor Regency. Data collection was carried out using the belt transect method. Analysis of species composition and structure of mangrove vegetation used analysis of species density, relative density, species frequency, relative frequency, and important value index. A total of seven mangrove species were found in the coastal area of Dafi Village, Biak Numfor Regency, consisting of Bruguiera gymnorrhiza, Bruguiera cylindrica, Ceriops tagal, Rhizophora apiculata, Rhizophora mucronata, Rhizophora stylosa, and Lumnitzera racemoza. Bruguiera gymnorrizha species had the highest density, frequency, and dominance values, while the lowest relative dominance and dominance were found in Lumnitzera racemoza. The highest IVI tree level values ​​were Ceriops tagal and Bruguiera gymnorrhiza. Mangrove forests in this area are still in natural condition, and the existence of mangrove ecosystems supports the lives of people around the island. Keywords: Coastal waters; Mangrove Ecosystem; Type Dominance; Important Value Value Index Abstrak Keberadaan dan kelestarian hutan mangrove mempunyai peran yang signifikan bagi lingkungan dan masyarakat yang tinggal di sekitar pesisir. Ekosistem mangrove juga memiliki sistem yang kompleks meliputi organisme tumbuhan dan hewan yang saling berinteraksi satu dengan yang lain, dan memegang peranan penting dalam menjaga dan melindungi garis pantai. Penelitian ini dilakukan dengan tujuan menganalisis struktur komunitas mangrove di Perairan Kampung Dafi  Kabupaten Biak  Numfor.Pengumpulan data ekosistem mangrove dilakukan dengan menggunakan metode belt transek. Analisis komposisi jenis dan struktur vegetasi mangrove menggunakan analisis kerapatan jenis, kerapatan relatif, frekuensi jenis, frekuensi relatif, dan  indeks nilai penting. Total tujuh jenis mangrove ditemukan di Pesisir Kampung Dafi Kabupaten Biak Numfor yang terdiri atas jenis Bruguiera gymnorrhiza, Bruguiera cylindrica, Ceriops Tagal, Rhizophora apiculata, Rhizophora mucronata, Rhizophora Stylosa, dan Lumnitzera racemoza. Jenis Bruguiera gymnorrizha memiliki nilai kerapatan, Frekuensi dan dominansi tertinggi, sedangkan dominansi jenis dan dominansi relatif terendah pada mangrove jenis Lumnitzera racemoza. Nilai INP tingkat pohon yang paling tinggi adalah Ceriops tagal dan Bruguiera gymnorrhiza. Hutan mangrove pada kawasan ini masih berada dalam kondisi alami, dan keberadaan ekosistem mangrove menjadi penopang bagi kehidupan masyarakat di sekitar pulau. Abstract

Microalgae are a group of phytoplankton that live in waters and in damp places; Microscopic in size and can only be seen with a microscope. The stages of microalgae growth are started with the lag phase, the exponential phase, the growth rate decreasing phase, the stationary phase, and the death phase. Porphyridium cruentum is a single-celled microalgae, belonging to the class Rhodophyceae, free-living or in colonies bound in mucilago. The purpose of this study was to observe the growth of the marine microalgae Porphyridium cruentum in culture media treated with lead acetate at different concentrations, namely 30 ppm, 50 ppm, 80 ppm, and control. Observation of cell growth was carried out by counting the number of cells every day, at the same time until Porphyridum cruentum entered the death phase. The cell density of Porphyridium cruentum marine microalgae showed a good growth pattern where until day 11 the average number of microalgae cells was 9.8 x104. Furthermore, the culture media was treated with lead acetate in different concentrations. Cell density with lead acetate treatment can affect the growth of Porphyridium cruentum. Keywords : Microalgae; Porphyridium cruentum; Lead Acetate Abstrak Mikroalga adalah kelompok fitoplankton yang hidup di perairan dan di tempat yang lembab; berukuran mikroskopis dan hanya bisa dilihat menggunakan mikroskop. Tahapan pertumbuhan mikroalga adalah dimulai dari fase lag, fase eksponensial, fase penurunan laju pertumbuhan, fase stasioner dan fase kematian. Porphyridium cruentum merupakan mikroalga bersel tunggal, termasuk kelas Rhodophyceae, hidup bebas atau berkoloni yang terikat dalam mucilago. Tujuan dari penelitian ini adalah mengamati pertumbuhan dari mikroalga laut Porphyridium cruentum pada media kultur dengan perlakuan timbal asetat pada konsentrasi berbeda yaitu 30 ppm, 50 ppm, 80 ppm dan kontrol. Pengamatan pertumbuhan sel dilakukan dengan menghitung jumlah sel setiap hari, pada waktu yang sama sampai Porphyridum cruentum memasuki fase kematian. Kepadatan sel mikroalga laut Porphyridium cruentum menunjukkan pola pertumbuhan yang baik dimana sampai hari ke 11 jumlah rata-rata sel mikroalga 9,8 x104. Selanjutnya media kultur diberi perlakuan timbal asetat dengan konsentrasi berbeda. Kepadatan sel dengan perlakuan timbal asetat dapat mempengaruhi pertumbuhan Porphyridium cruentum. Abstract

This study aims to identify and determine the phylogenetic position of two bacterial isolates (F0031 and F0033), isolated from culture media of Rotifer Brachionus plicatilis sp. complex. Genomic DNA of the bacteria was extracted using Qiaprep Miniprep Kit and the 16S rRNA gene was amplified using primer pairs 8F (5'-AGAGTTTGATCCTGGCTCAG-3') and 1492R (5'-ACCTTGTTACGACTT-3'). The sequences were analyzed using Geneious Prime ver. 2020 and bacterial identification was performed using the Basic Local Alignment Tool (BLAST) method.  Phylogenetic analysis was conducted using MEGA X, where the phylogenetic tree was constructed using the Maximum Likelihood method, and evaluated using the Bootstrap method.  Results of this study showed that both bacterial isolates (F0031 and F0033) belong to different genera, namely Bacillus (F0031) and Mesobacillus (F0033). Bacterial isolate F0031 had the closest similarity and was located at the same phylogenetic branch as B. cereus, while isolate F0033 was with M. jeotgali. Abstrak Penelitian ini bertujuan untuk mengidentifikasi dan menentukan posisi filogeni dua isolat bakteri (F0031 dan F0033) yang diisolasi dari media kultur rotifer Brachionus plicatili sp. complex.  DNA genom dari isolat bakteri diekstrak menggunakan Kit Qiaprep Miniprep dan gen 16S rRNA diamplifikasi menggunakan pasangan primer 8F (5'-AGAGTTTGATCCTGGCTCAG-3') dan 1492R (5'-ACCTTGTTACGACTT-3').  Hasil sekuens dianalisis menggunakan Sequence scanner ver. 2.0 dan analisis sekuens dilakukan menggunakan Geneious Prime ver. 2020.  Identifikasi isolat bakteri dilakukan dengan metode BLAST (Basic Local Alignment Tools) dan analisis filogenetik dilakukan dengan menggunakan MEGA X, dimana konstruksi filogeni menggunakan metode Maximum Likelihood yang dievaluasi dengan metode Bootstrap. Hasil penelitian menunjukan bahwa kedua isolat bakteri (F0031 dan F0033) merupakan isolat bakteri yang berasal dari genus berbeda, yaitu genus Bacillus (F0031) dan genus Mesobacillus (F0033).  Isolat bakteri F0031 memiliki kesamaan terdekat dan berada pada posisi percabagan filogenetik yang sama dengan B. cereus sedangkan Isolat F0033 dengan M. jeotgali Kata kunci: Filogeni, molekuler, bakteri, bacillus, mesobaccilus, rotifer, pakan

Abstract

Penelitian ini dilaksanakan di Pulau Kumo Kabupaten Halmahera Utara. Tujuan penelitian ini untuk menganalisis kepadatan, indeks keanekaragaman dan dominasi dari spesies makro alga yang ditemukan di lokasi penelitian, mendeskripsikan morfologi makro alga dari masing-masing spesies yang ditemukan di lokasi penelitian, menganalisis kepadatan, indeks keanekaragaman dan dominasi dari spesies makro alga yang ditemukan di lokasi penelitian. Metode penelitian ini menggunakan metode survei. Perbedaan setiap spesias disebabkan karena beberapa faktor lingkungan yaitu faktor fisika, faktor kimia dan faktor biologi. Kepadatan spesies pada Stasiun 3 lebih kecil dibandingkan dari Stasiun I dan 2. Indeks-indeks ekologi seperti keanekaragaman dan dominasi menunjukan bahwa keanekaragaman dan keadaan komunitas sedang pada ketiga stasiun penelitian, sedangkan untuk dominasi pada ketiga stasiun penelitian menunjukan dominasi rendah. Abstract

This study aims to determine the meristic and morphometric characteristics of the flying catfish in Manado Bay. It is hoped that it will be useful in the optimal management process based on characteristics, and become reference material for policymakers, practitioners, and the public so that they are more concerned about the preservation of flying catfish, and serve as a basis for managing flying fish resources. Data for flying fish are taken from the Fish Auction Place (TPI), fishing boat landing sites, and several markets in Manado City. Measurement of morphometric and meristic data was carried out directly at the sampling location, including observations of fish fins and scales. Morphometric data analysis is presented in the total length frequency distribution graph. The relationship between two certain morphometric characters is determined by the formula: regression Y = a + b X and the correlation value is r to see the closeness of the relationship between the two characters. The relationship between the two calculated characters is presented in graphical form. The morphometric value of the blue swallow is obtained, namely the Y axis which is the same as FL with a value of 5.297, and the X axis which is the same as TL with a value of 0.887. Diharapkan bermanfaat dalam proses pengelolaan secara optimal berdasarkan karakteristik, dan menjadi bahan referensi bagi pengambil kebijakan, praktisi, dan masyarakat agar lebih peduli dalam pelestarian ikan layang, serta dijadikan sebagai dasar dalam pengelolaan sumber daya  ikan layang. Data ikan layang diambil dari Tempat Pelelangan Ikan (TPI), tempat pendaratan perahu nelayan, dan beberapa pasar di Kota Manado. Pengukuran data morfometrik dan meristik dilakukan secara langsung di lokasi pengambilan sampel, termasuk pengamatan sirip dan sisik ikan. Analisis data morfometrik disajikan dalam grafik sebaran frekuensi panjang total. Hubungan antara dua karakter morfomterik tertentu ditentukan dengan rumus : regresi Y = a + b X dan nilai korelasinya yakni r untuk melihat keeratan hubungan kedua karakter. Hubungan kedua karakter yang dihitung akan disajikan dalam bentuk grafik.  morfometrik ikan layang biru diperoleh nilai yaitu sumbu Y yang sama dengan FL dengan nilai 5.297, dan sumbu X yang sama dengan TL dengan nilai 0.887.   Kata kunci: morfometrik, meristik, ikan layang, Teluk Manado

Abstract

Abstract The Government of Indonesia has targeted 32.5 million hectares of marine conservation areas or 10% of the total area of ​​Indonesian waters in 2030. The location of the Water Tourism Park Conservation Area of North Minahasa Regency has been reserved by the North Sulawesi Provincial Government through Governor Decree Number 407 of 2018 covering an area of ​​25,838.91 hectares. The purpose of this research is to formulate a strategy for the sustainable development of the Water Tourism Park Conservation Area in North Minahasa Regency. This research was conducted for 6 months starting from June - December 2022. The method used in this research is a descriptive research method with survey techniques, namely by collecting data through interviews with relevant stakeholders as respondents. The results of this study obtained 10 (ten) priority strategies for the sustainable development of Water Tourism Park Conservation Area in North Minahasa Regency which will be recommended to the local government, in this case the Marine and Fisheries Office of North Sulawesi Province and related stakeholders as suggestions and materials that can be proposed in the context of making public policy in the field of marine and fisheries. Lokasi Kawasan Konservasi Taman Wisata Perairan Kabupaten Minahasa Utara telah dicadangkan oleh Pemerintah Provinsi Sulawesi Utara melalui Surat Keputusan Gubernur Nomor 407 Tahun 2018 seluas 25.838,91 hektar. Tujuan penelitian ini untuk merumuskan strategi pengembangan berkelanjutan Kawasan Konservasi Taman Wisata Perairan di Kabupaten Minahasa Utara. Penelitian ini dilakukan selama 6 bulan dimulai dari Juni - Desember 2022. Metode yang digunakan dalam penelitian ini adalah metode penelitian deskriptif dengan teknik survey, yaitu dengan cara pengumpulan data melalui wawancara dengan para stakeholders terkait sebagai responden. Hasil dari penelitian ini didapatkan 10 (sepuluh) strategi prioritas untuk pengembangan berkelanjutan Kawasan Konservasi Taman Wisata Perairan di Kabupaten Minahasa Utara yang akan direkomendasikan kepada pemerintah daerah dalam hal ini Dinas Kelautan dan Perikanan Daerah Provinsi Sulawesi Utara beserta stakeholders terkait sebagai saran dan bahan yang dapat dipertimbangkan dalam rangka pembuatan kebijakan publik pada bidang kelautan dan perikanan. Abstract

The existence of seagrass beds in a shallow water environment has an important ecological role for the organisms that depend on this ecosystem. This research was conducted with the aim of knowing the types of seagrass and the condition of the seagrass beds in the waters near Mokupa Village, where the beach is between Mokupa Resort and Lotus Resort Manado. The method used is a quadrant transect (vertically to the shoreline), the coordinates point 1°24'53" N 124°42'22" E is transect 1, 1°24'54" N 124°42'22" E is transect 2,  and 1°24'56.7" N 124°42'23.5" E is transect 3. In this study, 5 species of seagrass were found, namely: Syringodium isoetifolium, Cymodocea rotundata, Thalassia hemprichii, Enhalus acoroides, and Halophila ovalis. Cymodocea rotundata had the highest average seagrass cover per species, 15.349%, and Enhalus acoroides had the lowest average seagrass cover, 0.053%. The condition of the seagrass beds at the study site was included in the unhealthy category with a seagrass cover of 31.432%. Keywords: Mokupa Waters, Seagrass conditions, Cover, Seagrass bed Abstrak Keberadaan padang lamun di lingkungan perairan dangkal memiliki peranan ekologis yang penting bagi organisme yang bergantung dalam ekosistem ini. Penelitian ini dilakukan dengan tujuan untuk mengetahui jenis-jenis lamun dan kondisi padang lamun di perairan sekitar Desa Mokupa, dimana pantai berada di antara Mokupa Resort dan Lotus Resort Manado. Penelitian ini mengikuti Pedoman Status Padang Lamun KEPMEN LH 200/2004. Metode yang digunakan yaitu transek kuadran (tegak lurus garis pantai), dengan titik koordinat Transek 1 1°24'53" N 124°42'22" E, Transek 2 1°24'54" N 124°42'22" E, Transek 3 1°24'56.7" N 124°42'23.5" E. Pada penelitian ini ditemukan 5 jenis lamun yaitu: Syringodium isoetifolium, Cymodocea rotundata, Thalassia hemprichii, Enhalus acoroides dan Halophila ovalis. Cymodocea rotundata memiliki rata-rata penutupan lamun per jenis tertinggi yaitu 15,349%, dan Enhalus acoroides memiliki rata-rata penutupan lamun terendah yaitu 0,053%. Kondisi padang lamun di lokasi penelitian termasuk dalam kategori kurang sehat dengan penutupan lamun sebesar 31,432%. Kata kunci: Perairan Mokupa, Kondisi lamun, Penutupan, Padang lamun
