Abstract

Covid-19 (Corona Virus disease 2019) is a new type of disease caused by a virus from the Corona Virus group, namely SARS-CoV-2. Covid-19 may cause respiratory system disorders, ranging from mild symptoms such as flu to lung infections, such as pneumonia. Laboratory diagnoses for Covid-19 disease generally include hematological (complete blood examinations), and molecular or a combination of serology and molecular. PCR examination that can be carried out using the Insulated Isothermal PCR and Reverse Transcription PCR methods. This research is descriptive observational research. Data were collected at Grha Kedoya Hospital, West Jakarta, North Kedoya. The result showed a sensitivity of iiPCR score 82,6%, which is relatively lower than gold-standard RT-PCR. Our research suggested that RT-PCR is still an effective and sensitive method for Covid-19 examination.

Abstract

ABSTRAK Nutrient agar merupakan jenis media umum yang sering digunakan pada laboratorium, harga media ini relatif mahal. Nutrient agar berbentuk padat, terdiri atas campuran ekstrak daging, pepton serta agar. Ikan teri jengki dan ikan layang deles jumlahnya melimpah di Indonesia. Ikan teri jengki (Stolephorus indicus) mengandung protein 51.54%, sementara ikan layang deles (Decapterus Macrosoma) mengandung protein sebesar 57,89%. Escherichia coli digunakan sebab bakteri ini merupakan salah satu jenis mikroorganisme kontrol positif yang direkomendasikan berdasarkan Data Sheet Nutrient Agar OXOID. Penelitian ini bersifat eksperimental dengan tiga perlakuan pada masing-masing bahan yaitu variasi massa 3 gram, 4 gram dan 5 gram yang dilakukan di Laboratorium Bakteriologi Jurusan Teknologi Laboratorium Medis Poltekkes Kemenkes Surabaya. Hasil penelitian menunjukkan jumlah koloni rata-rata Escherichia coli yang tumbuh pada kontrol positif sebanyak 118 x 1013, sementara pada media ikan teri jengki variasi massa 3 gram adalah 72 x 1013 CFU/mL, 4 gram sebanyak 85 x 1013 CFU/mL, 5 gram sebanyak 94 x 1013 CFU/mL. Pada media ikan layang deles variasi 3 gram sebanyak 88 x 1013 CFU/mL, 4 gram sebanyak 96 x 1013 CFU/mL, 5 gram 108 x 1013 CFU/mL. Variasi massa yang paling baik dan mendekati hasil kontrol positif (nutriet agar) pada ikan teri jengki dan ikan layang deles adalah variasi massa 5 gram.Kata kunci: ikan teri jengki, ikan layang deles, Escherichia coli, nutrient agar

Abstract

The corona virus outbreak first occurred at the end of 2019 in Wuhan, China. This virus then spread rapidly throughout the world, including Indonesia. So, countries in the world take steps to lock down. In Indonesia, large-scale social restrictions have been implemented in big cities with a high prevalence of the spread of the virus. Efforts to control the virus include limiting mobility, wearing masks, frequently washing hands or using hand sanitizers. Hand sanitizers are products made from ethanol which are increasingly limited and expensive during the Covid-19 pandemic. The purpose of this study is to make bioethanol with a variation of the fermentation time made from molasses, so as to produce ethanol content above 70%. This research is an experimental research. Production of bioethanol based on molasses, aquades, NPK, urea, and yeast culture of Saccharomycess cerevisae with time variations of 4, 5, and 6 days and distilled at a temperature of 500C. Testing the ethanol content using gas chromatography mass spectrometry. The results of this study, the fourth day of fermentation produces ethanol content of 50.23±0.42% with an average volume of 490 mL; day 5 produced ethanol content of 62.92±0.38% with an average volume of 385 mL; and the 6th day yielded ethanol content of 72.11±0.46% with an average volume of 243 mL. So, the 6th day of fermentation is the best fermentation to produce bioethanol as a raw material for hand sanitizer with ethanol content above 70%.

Abstract

Chronic diabetes mellitus is carried on by a rise in blood glucose levels as a result of decreased insulin sensitivity to manage blood sugar levels. The purpose of this study was to analyze participants with controlled and uncontrolled type 2 diabetes in relation of blood glucose, insulin, Homa-IR, and SCUBE2 protein levels. This study has a cross-sectional design and is descriptive analytic research. The sample for this study was stored biological fluid which was collected by total sampling from the Roemani Muhammadiyah Hospital Semarang Laboratory Installation in September 2022 to October 2022. The T-Independent test and the Mann-Whitney U-test used to the data analysis. The results of the T-Independent test results suggest that there was no significant variance between the insulin levels in DMT2 patients who were in control and those who unable (p=0.820, p>0.05). According to the results of the Mann-Whitney U-test analysis, there was no significant difference in the levels of scube2 protein between controlled and uncontrolled T2DM, and there was a significant difference in the glucose levels and Homa-IR values between controlled and uncontrolled T2DM, including both.but there was a significant effect of differences in glucose levels and Homa-IR values. The conclusion is that there are differences in glucose levels and Homa-IR values in controlled and uncontrolled DMT2 and there is no difference in insulin and scube 2 protein levels in controlled and uncontrolled DMT2.  Keywords: Glucose, Insulin, Homa-IR, Protein Scube2

Abstract

Bacteriuria is the leading indicator of Urinary Tract Infection (UTI), which is detected through urine culture examination. Significant bacteriuria indicates the growth of microorganisms as much as ≥ 100,000 cfu/ml in urine culture. There are drawbacks to this examination that requires a screening test with a short time and lower cost. This study aimed to determine differences in the results of nitrite and leukocyte esterase examination using a urine dipstick with urine culture as a predictor of bacteriuria. The type of research used is analytic with an experimental approach. The sample used in this study was ten samples of urine. Based on the bivariate analysis results, the p-value was 0.541, which means that there was no difference in the results of leukocyte esterase and urine culture. This indicates the presence of leukocytes in the urine can be an indication of bacteriuria. The results for the nitrite examination obtained a p-value of 0.000, which means there is a difference between the results of the nitrite examination and the results of the urine culture. This shows that the presence of nitrites in the urine cannot indicate of bacteriuria. However the data on the sensitivity and specificity of dipstick examination are still low for leukocyte esterase and urine nitrite tests. However, urine dipstick examination to detect bacteriuria and UTI is still an option for faster screening. Keywords       : leukocyte esterase, nitrites, urine culture

Abstract

Konsumsi alkohol dapat menyebabkan perubahan struktur dan fungsi ginjal serta mengganggu kemampuan ginjal untuk mengatur volume, elektrolit dan komposisi cairan dalam tubuh. Pemeriksaan serum kreatinin merupakan salah satu parameter yang penting untuk mengetahui fungsi ginjal. Penelitian ini merupakan penelitian observasional dengan rancangan cross sectional analitik untuk mengetahui perbedaan kadar serum kreatinin pada peminum arak tingkat ringan, sedang dan berat di Kelurahan Renon, Kecamatan Denpasar Selatan, Kota Denpasar. Dari hasil penelitian didapatkan data peminum ringan berjumlah 7 sampel dengan kadar serum kreatinin paling rendah yaitu 0,6 mg/dL dan paling tinggi yaitu 1,4 mg/dL. Peminum sedang berjumlah 12 sampel dengan kadar serum kreatinin paling rendah 0,8 mg/dL dan paling tinggi 1,8 mg/dL. Peminum berat berjumlah 11 sampel dengan kadar serum kreatinin paling rendah 1,0 mg/dL dan paling tinggi yaitu 2,5 mg/dL. Berdasarkan hasil uji One-Way ANOVA untuk ketiga kelompok peminum bernilai signifikansi 0,000. Hasil uji LSD kelompok peminum rendah – berat bernilai p = 0,000 dan kelompok peminum sedang – berat bernilai p = 0,000.

Abstract

Tahapan pra-analitik merupakan faktor yang penting dalam pemeriksaan di laboratorium. Tahapan ini meliputi persiapan untuk pasien, pemberian identitas pada spesimen, penampungan spesimen, perlakuan pada spesimen, pengiriman spesimen, penyiapan dan pengolahan spesimen. Serum yang terbentuk setelah proses sentrifugasi sebaiknya segera digunakan untuk pemeriksaan dan apabila harus dilakukan penundaan maka serum sebaiknya segera dipisahkan dari bekuan darah , dipindahkan pada wadah serum dan disimpan dalam lemari pendingin. Tidak semua laboratorium melakukan penyimpanan serum yang sesuai. Hal tersebut berpeluang terjadinya metabolisme spesimen yang disebabkan sel hidup sehingga berpengaruh pada stabilitas. Tujuan penelitian ini untuk mengetahui pengaruh variasi waktu penundaan pemisahan serum terhadap hasil pemeriksaan glukosa. Jenis penelitian ini adalah eksperimental dengan desain penelitian One Group Pretest-Pottest Design. Penelitian dilakukan pada bulan April 2022 di Laboratorium Kimia Klinik Jurusan Teknologi Laboratorium Medis Poltekkes Kemenkes Surabaya. Digunakan 10 sampel serum dari mahasiswa dengan kadar glukosa normal. Sampel serum diberikan 5 perlakuan penundaan pemisahan yaitu 0, 1 ,2, 3, 4 jam. Kadar glukosa diperiksa dengan metode GOD-PAP pada alat fotometer 5010 vs+. Data yang terkumpul dianalisis dengan Uji Repeated Measure Anova . Rata-rataa hasil pemeriksaan glukosa yang dilakukan segera, penundaan 1 jam, 2 jam, 3 jam dan 4 jam adalah 78,1 mg/dL; 75,2 mg/dL, 71,8 mg/dL; 66,9 mg/dl dan 65,1 mg/dL. Hasil analisis uji statistik menunjukkan bahwa ada pengaruh variasi waktu penundaan pemisahan serum terhadap hasil pemeriksaan glukosa dengan nilai sig 0,000. Kata Kunci : Glukosa, serum, penundaan pemisahan

Abstract

Alcohol is a psychoactive substance that can cause dependence. Alcohol as an intoxicant can affect various structures and processes in the central nervous system and increase the risk of intentional and unintentional injury. One of the direct risks due to alcohol consumption is an accident while driving a vehicle and accidents at work. Rate alcohol It is measured in various body fluids including urine. This study aims to determine the sensitivity of the modified potassium dichromate method for qualitative test of alcohol in urine without filter paper and water bath. This research is a laboratory experimental study, the data obtained were analyzed in a descriptive way, namely seeing the color change of the reaction between the alcohol sample in urine and the Potassium bichromate reagent to green or blue with a modified working method. Tested all showed positive results with the modified Potassium bichromate method. A color change from green to blue is read as a positive alcohol result. The conclusion of this study is that the modified Potassium bichromate method is effective for the qualitative alcohol test process and there is an increase in the sensitivity of the test.

Abstract

COVID-19 is a virus that can spread through direct contact, indirect contact, or close contact with an infected person. This situation makes a person vulnerable to disease. Good knowledge about COVID-19 and healthy living behaviours as an effort to prevent transmission of COVID-19 are essential to implement. The aim is to determine the description of the understanding of the people of Denpasar City related to obesity being more susceptible to COVID-19 infection. This type of research is descriptive by conducting a survey of 400 people in Denpasar City through purposive sampling. This study used a questionnaire as a data collection tool and the data were analyzed univariately. Based on the characteristics of the respondents, it was found that most respondents were in the age range of 21-30 years. For the latest education, most of the respondents are high school / equivalent and university graduates with a percentage of 53.25% and 46% respectively. In terms of occupation, it was found that most of the respondents consisted of students with a percentage of 70.5% and the least were in the TNI / Police / Retired with a percentage of 0.25%. For the characteristics of the place of residence of the respondents, most of them are found in South Denpasar and the least in East Denpasar with a percentage of 32.5% and 16.75% respectively. The majority of respondents knowledge is known that most people in Denpasar understand that people who are obese will be susceptible to COVID-19 infection with good and sufficient percentages of 44% and 53% respectively.  Based on the results of the research and observations, it can be concluded that the people of Denpasar City have a high level of knowledge about the impact of obesity in relation to COVID-19. Abstract

A test of the inhibitory power of randu honey against Eschericia coli ATCC 8739 has been carried out. Honey contains compounds that function as antibacterials. The honey used is monoflora honey, namely randu honey. The concentration of randu honey in the inhibitory power test against Escherichia coli ATCC 8739 is 100% and 50%. The method used is the diffusion method in order to use nutrient media. The inhibitory power of randu honey with a concentration of 100% against Escherichia coli ATCC 8739 is 39.30 mm and the randu honey solution with a concentration of 50% against Escherichia coli ATCC 8739 is 35.80 mm so that randu honey has the potential to be antibacterial.
