Abstract

Soil-Transmitted Helminth (STH) infection is an infection caused by Ascaris lumbricoides worms, Ancylostoma duodenale worms, Necator americanus worms, and Trichuris trichiura worms. Transmission of infection occurs through soil media containing feces contaminated with STH worm eggs. STH worm infection can be identified by qualitative and semi-quantitative stool examination. Stool qualitative examination consists of the direct slide method (native), the flotation method, and the sedimentation method. This study aims to see the effectiveness of 33% ZnSO4  solution and 33% sucrose solution in examining STH worm eggs in the centrifugation flotation method. This study began by collecting feces samples; then the samples were identified by using the flotation centrifugation method. The solutions used in this method were 33% ZnSO4 solution and 33% sucrose solution. The data obtained were analyzed by using the Mann-Whitney test. There were 35 stool samples identified. The examination results obtained 13 positive samples of Ascaris lumbricoides worm eggs in 33% ZnSO4  solution and 1 positive sample in 33% sucrose solution, and the results of the Mann Whitney test obtained Asymp. Sig <0.05. Therefore, there is a significant difference between the 33% ZnSO4 solution and the 33% sucrose solution. Abstract

Dengue Hemorrhagic Fever (DHF) is a disease caused by the Dengue Virus, which is carried by vectors. One of which is the Aedes albopictus. DHF case data in Indonesia in 2016 was reported to reach 204,171 cases (IR 78.85/100,000 population) with a mortality rate of 1,598 people (CFR 0,78%). The most common way to control DHF is chemically using synthetic insecticides. Improper use of chemicals can have negative impacts, such as the death of non-target organisms, environmental pollution and vector resistance. Because of this impact, efforts are needed to use natural larvicides derived from natural ingredients to kill mosquito larvae that are safe for the environment, one of which is larvicides from butterfly pea flower extract. Based on this description, this study aims to determine the potential of butterfly pea flower extract (C. ternatea L.) on the death of A. albopictus larvae. This research is a pure experimental research. The control group in this study consisted of positive controls using 1% Abate and negative controls using distilled water. The experimental group was the administration of butterfly pea flower extract (C. ternatea L.) to A. albopictus larvae with varying concentrations of 30% and 40%. From the results of research on the potential of butterfly pea extract as an alternative larvicidal of A. albopictus larvae, it can be concluded that the butterfly pea extract has a larvicidal effect on A. albopictus larvae. The concentration of butterfly pea extract has the potential to kill A. albopictus larvae with an LC50 value obtained at a concentration of 39.815%.

Abstract

Corona Virus Infection Disease (COVID)-19 is a disease caused by a new coronavirus C derivative. The SARS-CoV-2 genome has six main open reading frames (ORFs): ORF 1a and 1b, envelope protein/E genes, membrane protein/M genes, spike protein/S gene, and nucleocapsid protein/N genes.  Realtime RT-PCR is a DNA amplification technique in which amplification products can be analyzed at each cycle using fluorogenic probes.  The RT-PCR method is used for amplification, isolation, or identification of sequences from RNA cells or tissues. The extraction of nucleic acids in the form of DNA and RNA is the initial process for biomolecular studies. The principle in the extraction of genetic material in the form of DNA and RNA is to break down cells and genetic material in the cell from other cellular components in the form of fats, proteins, carbohydrates, and other substances. The purpose of this study is to find out the picture of the value of CT Value qRT-PCR SARS-CoV-2 using two extraction methods, namely manual and automatic. This study was descriptive using accidental sampling techniques of positive patients confirmed with COVID-19. Swab samples are carried out using manual and automatic extraction methods, then qRT-PCR examination is carried out. The results of descriptive statistical tests obtained the Mean Gene E value of manual extraction 19.1290 and automatic extraction 18.8187, as well as the Mean Gene value of ORF1ab manual extraction 19.1290 and automatic extraction 19.5177.

Abstract

Tobacco consumption in Indonesia is a health epidemic that has direct or indirect toxicity due to exposure of cigarette smoke which contains harmful substances, especially cadmium metal. Cadmium’s exposure can increase the risk of coronary heart disease due to increased LDL cholesterol. One of the cadmium toxicity treatment by utilizing Moringa oleifera leaves as a natural antioxidant considering the flavonoids, saponins, ascorbic acid, carotenoids, phenolics, interquinones as cadmium chelating agents and β-sitosterol content. Therefore, it was necessary to determine the effectiveness of Moringa leaves extract in lowering blood cadmium and LDL cholesterol levels as indicators of atherosclerosis in white rats exposed to cigarettes smoke. The type of research was an experiment with quantitative analysis, conducted at the nutrition laboratory of the Faculty of Public Health Airlangga University and the clinical chemistry laboratory of Surabaya Health Polytechnic in March-July 2023. The independent variable was the doses of Moringa leaf extracts, while the dependent variables were blood cadmium and LDL cholesterol levels of white rats. The analysis of blood cadmium levels used an atomic absorption spectrophotometer and used precipitation methods by a photometer for LDL cholesterol levels. Based on statistical analysis using one way anova showed that blood cadmium levels had a significant value of < 0.05, followed by post hoc tests it was found that treatment group 2 had significant differences from normal groups. Moringa leaves extract dose of 1500 mg/kgbw has been effective in lowering cadmium and LDL cholesterol levels against the risk of atherosclerosis for 14 days. Abstract

Tuberkulosis adalah penyakit menular langsung yang disebabkan oleh kuman Mycrobacterium tuberculosis. Sitokin pro inflamasi yang berperan dalam proses infeksi MTB salah satu nya adalah interleukin-6 (IL-6), yang berperan penting dalam respons fase akut dan transisi dari peradangan akut ke kronik, peningkatan protein fase akut yang meningkatkan pembentukan rouleaux sehingga akan meningkatkan kecepatan laju endap darah (Pagana, 2013).Jenis penelitian ini adalah penelitian observasional analitik, dengan rancangan cross sectional.Penelitian ini dilakukan di Puskesmas 1, 2, dan 3 Kecamatan Buleleng, Provinsi Bali, pada bulan April 2023 - Mei 2023, dengan jumlah sampel 30 pasien. Distribusi frekuensi hasil pemeriksaan Kadar IL-6 100% meningkat ≥ 4 ng/L, menggunakan metode ELISA. Distribusi frekuensi hasil pemeriksaan nilai LED normal (3,3%) dan 14 pasien memiliki nilai diatas normal (46,7%) pada laki-laki. Pada perempuan terdapat 7 pasien dengan nilai LED normal (23,3%) dan 8 pasien memiliki nilai LED diatas normal (73,3%), menggunakan metode westergreen. Berdasarkan hasil uji kolerasi nonparametrik Spearman didapatkan nilai signifikansi (p) 0,020 dan nilai koefisien korelasi (r) 0,422. Rata-rata hasil pemeriksaan kadar Interleukin-6 pada pasien Tb Paru adalah 27,066, rata-rata hasil pemeriksaan nilai LED adalah 39,066 Terdapat korelasi antara Interleukin-6 dengan nilai Laju Endap Darah pada pasien Tb (p) 0,020. Abstract

Uncontrolled diabetes mellitus will lead to various chronic complications, both microvascular and macrovascular complications. HbA1c examination is effective for long-term monitoring of blood glucose for people with diabetes mellitus. A high HbA1c value is used as a measure of the development of complications of diabetes mellitus. Diabetic nephropathy is the most common complication of diabetes mellitus which can end in kidney failure resulting in damage to the glomeruli. Indications to determine kidney function impairment is creatinine examination. This study aims to determine the correlation between HbA1c levels and creatinine levels in patients with chronic diabetes mellitus at the Clinical Pathology Laboratory of the Bajawa Regional General Hospital. Analytical observational research with an approach cros- sectional using techniques purposive sampling 35 chronic DM patients. Test results who squares shows that the value of p = (0.687) > α (0.05), thus it can be concluded that there is no relationship between HbA1c levels and Creatinine levels in patients with chronic diabetes mellitus. Abstract

Staphylococcus aureus is a bacteria that can cause skin and soft tissue infections such as urinary tract infections, pneumonia, mastitis, and meningitis. Staphylococcus aureus infections are usually treated with antibiotics, but most antibiotics are resistant to the bacteria and have side effects of toxicity to the body. One alternative to finding new antibacterials from natural ingredients is to use turi plants. This study aims to determine the content of secondary metabolites in the water fraction of the bark of turi (Sesbania grandiflora L.) and to determine the antibacterial activity of the water fraction of the bark of turi (Sesbania grandiflora L.) against Staphylococcus aureus. This research is an experimental study, using the bark fraction of the turi plant. The fraction was obtained after the fractionation process from the bark extract of turi (Sesbania grandiflora L.) so that the water fraction was divided into several concentrations, namely 25%, 50%, 75%, and 100%. Antibacterial activity test was carried out by well diffusion method. The results of this study indicate that the water fraction of the bark of turi (Sesbania grandiflora L.) contains secondary metabolites of alkaloids, tannins, saponins, steroids, and phenolics. The antibacterial activity produced was included in the moderate category at a concentration of 25% with an inhibition zone of 9.70 ± 0.14 mm, a strong category at concentration of 50% and 75% with an inhibition zone of 12.37 ± 0.25 mm, and 18.47 ± 0.23 mm, in addition to the category of very strong at a concentration of 100% with an inhibition zone of 23.47 ± 0.09 mm.

Abstract

Connexin-36 (Cx36) merupakan salah satu protein kanal antar sel. Protein ini paling banyak ditemukan pada otak. Gen Cx36 mengekspresikan protein penghubung Cx36 yang membentuk sinaps listrik. Penelitian ini bertujuan untuk memperoleh desain primer terbaik untuk menganalisis ekspresi Cx36 pada otak tikus dengan metode Quantitative Real Time PCR (qRT-PCR). Jenis penelitian ini adalah penelitian true experimental menggunakan mRNA dari 4 otak tikus. Analisis in silico menggunakan tools Primer 3 dan OlygoAnalizerTM by IDTdengan menggunakan database gen pada NCBI. Tiga primer dengan kriteria yang paling optimum kemudian dievaluasi dengan metode qRT-PCR. Berdasarkan hasil penelitian ketiganya mampu mengamplifikasi gen Cx36 dengan menggunakan qRT-PCR. Hasil primer terbaik menunjukkan melt curve tunggal pada primer no.5 gen Cx36 pada kisaran Tm 80-81oC. Hasil perbandingan uji in silico primer Cx36 yang sudah pernah dipublikasi sebelumnya menunjukkan bahwa primer no. 5 juga memiliki nilai optimal pada setiap kriteria. Maka hasil desain primer yang memiliki spesifisitas dan efektifitas yang optimum untuk mengamplifikasi gen Cx36 pada otak tikus Rattus norvegicus ialah primer no.5 (F: 5’-ATTTCCCGCTTCTACATCATCCAAG-3’ dan R: 5’-CACAGCAAACATGAACACCAGAAAG-3’).

Abstract

Pediculosis humanus capitis is a blood-sucking parasite that spends its entire life cycle on the human scalp. Poor personal hygiene can affect pediculosis capitis infection. In school children, the chronic infestation of pediculosis capitis causes anemia and makes children lethargic and sleepy. The aim of the study was to determine the relationship between personal hygiene and anemia status with the incidence of pediculus humanus capitis in first grade students at madrasah tsanawiyah at the "Assaasunnajah" Islamic boarding school Ateuk lung ie, Wanna Jaya District, Aceh Besar Province. The research design used is descriptive analytic cross sectional approach. The sampling technique of this research is total sampling. The instruments in this study were observation and measurement. Based on the data analysis that has been done, with a total of 30 respondents, poor personal hygiene was obtained by 7 respondents with a percentage of 23.3%. For the incidence of pediculus humanus capitis, 7 respondents stated that a pediculus was found with a percentage of 23.3%. While Anemia Status based on Hb results, 2 respondents were declared Anemia with a percentage of 7%. The conclusion of this study based on the results of the chi square test showed that there was no relationship between personal hygiene and the incidence of pediculus humanus capitis because the test results obtained a p-value of 0.708 > confidence probability of 0.05 and there was a relationship between anemia and the incidence of pediculus humanus capitis in class 1 students of madrasah tsanawiyah at the "Assaasunnajah" Islamic boarding school Ateuk lung ie, Want Jaya District, Aceh Besar Province because the test results obtained a p-value of 0.008 < 0.05 confidence probability. Abstract

Increasing population growth, industrialization and the use of motorized vehicles are the factors causing the increase in air pollution. One of the particles contained in the exhaust gas emissions produced is lead. This study was aimed at the operators of the Public Fuel Filling Station in the Industrial Area of West Bandung District who are vulnerable to lead exposure from various pollutant sources. The purpose of this study was to describe the highest and lowest levels of lead accumulated in the hair of gas station operators. The method used is descriptive method. The research sample was taken from respondents who have worked as gas station operators for >1 year. The sample used was 19 gas station operators. Data was collected by filling out a questionnaire followed by laboratory examination. The results showed that 5 people had lead levels exceeding the normal threshold and 14 others had normal lead levels with an average of 13.52 g/g. The highest lead content in hair was 110.75 g/g and the lowest level was -3.50 g/g. With a threshold value of lead levels in hair that is <12 mg/g. The head of the gas station is advised to apply regulations on the use of appropriate PPE for gas station operators to minimize lead exposure. For further research, it is recommended to monitor lead (Pb) levels with different specimens.   Keywords      : Lead, Hair, Gas Station, Industry, Motor Vehicle
