Abstract

Chronic diabetes mellitus is carried on by a rise in blood glucose levels as a result of decreased insulin sensitivity to manage blood sugar levels. The purpose of this study was to analyze participants with controlled and uncontrolled type 2 diabetes in relation of blood glucose, insulin, Homa-IR, and SCUBE2 protein levels. This study has a cross-sectional design and is descriptive analytic research. The sample for this study was stored biological fluid which was collected by total sampling from the Roemani Muhammadiyah Hospital Semarang Laboratory Installation in September 2022 to October 2022. The T-Independent test and the Mann-Whitney U-test used to the data analysis. The results of the T-Independent test results suggest that there was no significant variance between the insulin levels in DMT2 patients who were in control and those who unable (p=0.820, p>0.05). According to the results of the Mann-Whitney U-test analysis, there was no significant difference in the levels of scube2 protein between controlled and uncontrolled T2DM, and there was a significant difference in the glucose levels and Homa-IR values between controlled and uncontrolled T2DM, including both.but there was a significant effect of differences in glucose levels and Homa-IR values. The conclusion is that there are differences in glucose levels and Homa-IR values in controlled and uncontrolled DMT2 and there is no difference in insulin and scube 2 protein levels in controlled and uncontrolled DMT2.  Keywords: Glucose, Insulin, Homa-IR, Protein Scube2

Abstract

COVID-19 is a virus that can spread through direct contact, indirect contact, or close contact with an infected person. This situation makes a person vulnerable to disease. Good knowledge about COVID-19 and healthy living behaviours as an effort to prevent transmission of COVID-19 are essential to implement. The aim is to determine the description of the understanding of the people of Denpasar City related to obesity being more susceptible to COVID-19 infection. This type of research is descriptive by conducting a survey of 400 people in Denpasar City through purposive sampling. This study used a questionnaire as a data collection tool and the data were analyzed univariately. Based on the characteristics of the respondents, it was found that most respondents were in the age range of 21-30 years. For the latest education, most of the respondents are high school / equivalent and university graduates with a percentage of 53.25% and 46% respectively. In terms of occupation, it was found that most of the respondents consisted of students with a percentage of 70.5% and the least were in the TNI / Police / Retired with a percentage of 0.25%. For the characteristics of the place of residence of the respondents, most of them are found in South Denpasar and the least in East Denpasar with a percentage of 32.5% and 16.75% respectively. The majority of respondents knowledge is known that most people in Denpasar understand that people who are obese will be susceptible to COVID-19 infection with good and sufficient percentages of 44% and 53% respectively.  Based on the results of the research and observations, it can be concluded that the people of Denpasar City have a high level of knowledge about the impact of obesity in relation to COVID-19. Abstract

Connexin-36 (Cx36) merupakan salah satu protein kanal antar sel. Protein ini paling banyak ditemukan pada otak. Gen Cx36 mengekspresikan protein penghubung Cx36 yang membentuk sinaps listrik. Penelitian ini bertujuan untuk memperoleh desain primer terbaik untuk menganalisis ekspresi Cx36 pada otak tikus dengan metode Quantitative Real Time PCR (qRT-PCR). Jenis penelitian ini adalah penelitian true experimental menggunakan mRNA dari 4 otak tikus. Analisis in silico menggunakan tools Primer 3 dan OlygoAnalizerTM by IDTdengan menggunakan database gen pada NCBI. Tiga primer dengan kriteria yang paling optimum kemudian dievaluasi dengan metode qRT-PCR. Berdasarkan hasil penelitian ketiganya mampu mengamplifikasi gen Cx36 dengan menggunakan qRT-PCR. Hasil primer terbaik menunjukkan melt curve tunggal pada primer no.5 gen Cx36 pada kisaran Tm 80-81oC. Hasil perbandingan uji in silico primer Cx36 yang sudah pernah dipublikasi sebelumnya menunjukkan bahwa primer no. 5 juga memiliki nilai optimal pada setiap kriteria. Maka hasil desain primer yang memiliki spesifisitas dan efektifitas yang optimum untuk mengamplifikasi gen Cx36 pada otak tikus Rattus norvegicus ialah primer no.5 (F: 5’-ATTTCCCGCTTCTACATCATCCAAG-3’ dan R: 5’-CACAGCAAACATGAACACCAGAAAG-3’).

Abstract

The increasing number of Covid-19 cases, especially the presence of a new Delta variant which causes a risk of severe symptoms in pregnant women, the Indonesian government has established a screening program and implementation of the Covid-19 vaccination as stated in circular letter HK.02.01/I/2007/2021. Based on preliminary studies carried out on pregnant women in the city of Surabaya, there are 90% percent of pregnant women who are still hesitant and afraid to vaccinate. One form of effort being made is the development of an application that facilitates screening and accelerates vaccination of pregnant women, namely the VMil application. The purpose of this study was to find out how the Volunteer program through the V-MiL application affects pregnant women in carrying out Covid-19. This type of research is pre-experimental with one group pre-test and post test design. The dependent variable is the respondent's participation in the implementation of the Covid-19 vaccine, while the independent variable is intervention by volunteers using the V-MiL application. The research sample for pregnant women who had not been vaccinated was 40 people using a purposive sampling technique. Data collection with questionnaires and interviews. The results of the study the majority of respondents aged 21-30 years (58%), gestational age 13-27 weeks (78%). After the intervention the respondents had good knowledge 18 (45%), lack of knowledge 22 (55%), negative attitude positive attitude 16 (40%), negative attitude 24 (60%), strong motivation 15 (38%), weak motivation 25 (63%) to vaccinate. Respondents did the Covid-19 vaccine as many as 15 (38%) and 25 (63%) respondents did not do the vaccine. Analysis using the Paired sample T test obtained a value of λ of 0.001 <0.005 which indicated that there was a significant influence on the intervention given by volunteers to the respondents. Keywords                    : Volunteers, Covid-19 Vaccines, Pregnant Women

Abstract

Free radicals are compounds that are very dangerous for the body because they can attack and cause cell damage in the body, therefore antioxidant compounds are needed that can counteract the presence of these free radicals. One of the fruits that contain antioxidant compounds is dates (Phoenix dactylifera L.) which contain flavonoid compounds that act as antioxidants. The purpose of this study was to determine the antioxidant activity at heating temperatures of 400C, 500C, and 650C. The extraction method used in this study is the maceration method with 80% ethanol as solvent. Testing of antioxidant activity using the method of reducing free radicals, namely the ABTS method and using vitamin C as a comparison control to show antioxidant activity. ABTS is a method of determining antioxidant activity obtained from the oxidation of potassium persulfate with diammonium salt. Absorbance measurements were carried out using a UV-Vis spectrophotometer with a wavelength of 520 nm. Data analysis was carried out using linear regression equations to determine the IC50 value in the sample of dates (Phoenix dactylifera L.). The IC50 value indicates the antioxidant activity in a sample, namely the smaller the IC50 value, the higher the antioxidant in a sample. The results of data analysis using the Kruskal Wallis test obtained a significance value of 0.104 which indicates that > 0.05 which means there is no significant difference between each treatment group. From the results of the analysis data, it is known that there is no effect of heating temperature on the antioxidant stability of dates, this is also strengthened by the value of antioxidant activity in the linear regression equation where the value of antioxidant activity in each treatment group has an equally strong antioxidant activity value. based on the Blois classification. Abstract

Bacteriuria is the leading indicator of Urinary Tract Infection (UTI), which is detected through urine culture examination. Significant bacteriuria indicates the growth of microorganisms as much as ≥ 100,000 cfu/ml in urine culture. There are drawbacks to this examination that requires a screening test with a short time and lower cost. This study aimed to determine differences in the results of nitrite and leukocyte esterase examination using a urine dipstick with urine culture as a predictor of bacteriuria. The type of research used is analytic with an experimental approach. The sample used in this study was ten samples of urine. Based on the bivariate analysis results, the p-value was 0.541, which means that there was no difference in the results of leukocyte esterase and urine culture. This indicates the presence of leukocytes in the urine can be an indication of bacteriuria. The results for the nitrite examination obtained a p-value of 0.000, which means there is a difference between the results of the nitrite examination and the results of the urine culture. This shows that the presence of nitrites in the urine cannot indicate of bacteriuria. However the data on the sensitivity and specificity of dipstick examination are still low for leukocyte esterase and urine nitrite tests. However, urine dipstick examination to detect bacteriuria and UTI is still an option for faster screening. Keywords       : leukocyte esterase, nitrites, urine culture

Abstract

A test of the inhibitory power of randu honey against Eschericia coli ATCC 8739 has been carried out. Honey contains compounds that function as antibacterials. The honey used is monoflora honey, namely randu honey. The concentration of randu honey in the inhibitory power test against Escherichia coli ATCC 8739 is 100% and 50%. The method used is the diffusion method in order to use nutrient media. The inhibitory power of randu honey with a concentration of 100% against Escherichia coli ATCC 8739 is 39.30 mm and the randu honey solution with a concentration of 50% against Escherichia coli ATCC 8739 is 35.80 mm so that randu honey has the potential to be antibacterial.

Abstract

Soil-Transmitted Helminth (STH) infection is an infection caused by Ascaris lumbricoides worms, Ancylostoma duodenale worms, Necator americanus worms, and Trichuris trichiura worms. Transmission of infection occurs through soil media containing feces contaminated with STH worm eggs. STH worm infection can be identified by qualitative and semi-quantitative stool examination. Stool qualitative examination consists of the direct slide method (native), the flotation method, and the sedimentation method. This study aims to see the effectiveness of 33% ZnSO4  solution and 33% sucrose solution in examining STH worm eggs in the centrifugation flotation method. This study began by collecting feces samples; then the samples were identified by using the flotation centrifugation method. The solutions used in this method were 33% ZnSO4 solution and 33% sucrose solution. The data obtained were analyzed by using the Mann-Whitney test. There were 35 stool samples identified. The examination results obtained 13 positive samples of Ascaris lumbricoides worm eggs in 33% ZnSO4  solution and 1 positive sample in 33% sucrose solution, and the results of the Mann Whitney test obtained Asymp. Sig <0.05. Therefore, there is a significant difference between the 33% ZnSO4 solution and the 33% sucrose solution. Abstract

Dengue Hemorrhagic Fever (DHF) is a disease caused by the Dengue Virus, which is carried by vectors. One of which is the Aedes albopictus. DHF case data in Indonesia in 2016 was reported to reach 204,171 cases (IR 78.85/100,000 population) with a mortality rate of 1,598 people (CFR 0,78%). The most common way to control DHF is chemically using synthetic insecticides. Improper use of chemicals can have negative impacts, such as the death of non-target organisms, environmental pollution and vector resistance. Because of this impact, efforts are needed to use natural larvicides derived from natural ingredients to kill mosquito larvae that are safe for the environment, one of which is larvicides from butterfly pea flower extract. Based on this description, this study aims to determine the potential of butterfly pea flower extract (C. ternatea L.) on the death of A. albopictus larvae. This research is a pure experimental research. The control group in this study consisted of positive controls using 1% Abate and negative controls using distilled water. The experimental group was the administration of butterfly pea flower extract (C. ternatea L.) to A. albopictus larvae with varying concentrations of 30% and 40%. From the results of research on the potential of butterfly pea extract as an alternative larvicidal of A. albopictus larvae, it can be concluded that the butterfly pea extract has a larvicidal effect on A. albopictus larvae. The concentration of butterfly pea extract has the potential to kill A. albopictus larvae with an LC50 value obtained at a concentration of 39.815%.

Abstract

Corona Virus Infection Disease (COVID)-19 is a disease caused by a new coronavirus C derivative. The SARS-CoV-2 genome has six main open reading frames (ORFs): ORF 1a and 1b, envelope protein/E genes, membrane protein/M genes, spike protein/S gene, and nucleocapsid protein/N genes.  Realtime RT-PCR is a DNA amplification technique in which amplification products can be analyzed at each cycle using fluorogenic probes.  The RT-PCR method is used for amplification, isolation, or identification of sequences from RNA cells or tissues. The extraction of nucleic acids in the form of DNA and RNA is the initial process for biomolecular studies. The principle in the extraction of genetic material in the form of DNA and RNA is to break down cells and genetic material in the cell from other cellular components in the form of fats, proteins, carbohydrates, and other substances. The purpose of this study is to find out the picture of the value of CT Value qRT-PCR SARS-CoV-2 using two extraction methods, namely manual and automatic. This study was descriptive using accidental sampling techniques of positive patients confirmed with COVID-19. Swab samples are carried out using manual and automatic extraction methods, then qRT-PCR examination is carried out. The results of descriptive statistical tests obtained the Mean Gene E value of manual extraction 19.1290 and automatic extraction 18.8187, as well as the Mean Gene value of ORF1ab manual extraction 19.1290 and automatic extraction 19.5177.
