Abstract

Recently, expanded consideration has been paid to the business and potentiality of microalgae. Some microalgae are at present being studied for their capacity to find important metabolites for the drug industry or aquacultural applications. Concerning these biotechnological challenges, there is a consistent exertion accommodated in both finding and taking advantage of new microalgal assets and fostering their putative business results or modern valorizations. The crude methanol extracts of three microalgae, Dunaliella salina, Tetraselmis chuii, and Isochrysis galbana, have been investigated for antibacterial activity using the disk diffusion method against aquatic pathogens of fish, shrimp, and shellfish e.g. Pseudomonas fluorescence and Vibrio harveyi. This research aimed to analyze the antibacterial activity of crude extract of these microalgae against aquatic pathogens. The disk diffusion method was used to investigate the antibacterial activity. The result showed that only T. chuii has the inhibition zone for both tested bacteria P. fluorescence and V. harveyi with the inhibition zone of 3.0 ± 0.6 mm on 100 µg. g–1 of extract concentration and 4.20 ± 1.1 mm against P. fluorescence on 10000 µg. g–1 of extract concentration. While D. salina and I. galbana have inhibition zone only for V. harveyi with the inhibition zone of 4.4 ± 0.6 mm and 3.2 ± 0.7 mm on 10000 µg. g–1 of extract concentration. These three microalgae may have potential use in aquatic pathogens as antimicrobial agents. It would be possible to develop biologically active compounds of microalgae as a functional feed for aquaculture.

Abstract

The management effectiveness evaluation of marine conservation areas (MCAs) in Indonesia is often conducted assuming that they are likely to be evaluated periodically. However, for good and reliable results, it is recommended to perform the evaluability assessment prior to any evaluation to determine whether an MCA can be evaluated or not. This study aims to assess the evaluability of MCAs managed by the Ministry of Marine Affairs and Fisheries (MMAF) by reviewing their management plans and effectiveness evaluation tools. By employing a qualitative approach, this study found that (i) the management plans are not conceived to support an effective management of MCAs in the field or the evaluation of management effectiveness; and (ii) the current guidelines for evaluating MCA management effectiveness do not evaluate the expected achievements of management actions relative to biodiversity conservation goals and objectives. As a result, MCAs under MMAF direct supervision are not evaluable, and hence, the evaluations are pointless from a biodiversity conservation perspective. To address this discrepancy, it is recommended that the management effectiveness evaluation should only be applied to MCAs with an effectiveness-oriented management plan and should employ tools or guidelines that are devised to evaluate the achievement of MCA conservation goals and objectives.

Abstract

Banyuurip is one of the villages in Ujung Pangkah District which has potential natural resource that is mangrove forests. However, the occurrence of abrasion and conversion of mangrove land has impacted mangrove ecosystem. Rehabilitation efforts by replanting mangroves have been carried out, but they have not been effective in overcoming the current problems. The reduction of mangrove area that led the decreased of mangrove’s function can affect the livelihood of the local community, including fishermen. The installation of geosynthetic bags, which are sand-filled bags arranged on the ground, is done as an alternative solution in Banyuurip Village which functions as coastal protection and a sediment trap. The utilization of geosynthetic material is often used to help the coastal problem, the used of geosynthetic material is rarely carried out in mangrove areas. This study aims to determine the sediment characteristics, sedimentation rates, and sedimentation process based on the relationship between current velocity and sedimentation rate. The result shows that generally the type of sediment fraction in this location is silt (79,12% - 80,12%) and the rest is clay. The current installation of geosynthetic bags can result in the land extension from the trapped sediment behind the structure. In addition, the current velocity conditions and the sediment transport process also affect the sedimentation process that occurs. The average sedimentation rates around the geosynthetic bags installation area ranged from 150.72-305.01 mg.cm-2.day-1. This study may provide a basic information for further development in Banyuurip Mangrove Center and other mangrove conservation area.

Abstract

Seagrass is an aquatic biota that is ecologically and economically beneficial. This study aims to determine the community structure of seagrass in Harapan Island, Seribu Islands, Indonesia. The research was conducted using purposive sampling method, that divided into two sites which is considered to represent the ecology’s condition of Harapan Island. The site 1 was located adjacent to the mangroves area, while the site 2 was placed near a reclamation development and ship anchoring. Every site had three subsites as replications, each was 50 meters apart. Water parameters temperature, salinity, turbidity, pH, dissolved oxygen (DO), depth, and organic compound of each site were measured. The results showed that coverage of seagrass in site 2 is higher than site 1 that categorized as frequent-common coverage. The site 2 had the density of 220 individuals seagrass per square meter that dominated by Cymodocea rotundata. The highest diversity index was found in the site 1 at 1.15, with the lowest dominance index being 0.39. The distribution pattern in site 1 is clustered and uniform, while site 2 is uniform. The Importance Value Index in site 1 was 114,8 by Halodule uninervis, while site 2 was 125,5 by C. rotundata. It is thought that the low diversity of seagrass at the site 2 due to it was near a reclamation development and ship anchoring that disturb the abundance and community structure of seagrass in Harapan Island. Habitat destruction and anthropogenic impact of seagrass need to be minimized to support environmental coastal and fisheries sustainability.

Abstract

The coral reef is massive deposits of calcium carbonate produced from coral animals that are symbiotic with zooxanthellae. One of the activities carried out as an effort to improve the sustainability of coral reefs is monitoring the health condition of coral reefs. The coral reef health index value is based on benthic components and fish components. The benthic component consists of variable life coral cover and the level of coral reef resilience determined based on macroalgae, rubble, and life coral. Fish component is the total biomass of target reef fish. The highest coral reef health index value is at value 10 and the lowest value is at value 1. This study aimed to determine the value of the health index on Sangiang Island. This research was conducted on 16-18 February 2019 on Sangiang Island on three dive stations are Legon Bajo, Legon Waru, and Tembuyung. Data collection of benthic components uses the Underwater Photo Transect (UPT) method and data collection of fish components uses the Underwater Visual Census (UVC) method. The results showed that the coral reef health index on Sangiang Island was at values 6 and 5, means the live coral cover on Sangiang Island was included in the high and medium category, and the level of coral reef resilience or recovery potential was included in the high category, while the total biomass of the target reef fish is still in the low category, only a few species of fish were found that belong to the target reef fish.

Abstract

Currently, there are only six species in the genus Seonamhaeicola, i.e., Seonamhaeicola aphaedonensis, S. algicola, S. marinus, S. acroporae, S. maritimus, and S. sediminis. These bacteria have typical yellow or orange color. Among the identified strains, only S. marinus that had been reported to have a yellow polyene flexirubin pigment. However, the presence of carotenoid pigments has not been reported in this genus. Recently, we successfully isolated a new strain, S. algicola strain CC1, bacterium that was found in association with a red seaweed, Halymenia sp., collected from the coast of South Malang, Indonesia. The strain was grown well in the Zobell marine agar 2216E producing yellowish pigments. According to the 16S rRNA sequencing analysis and BLAST search, the strain is closely related to S. algicola strain Gy8, with 99.78% identity. The pigment composition was separated and analyzed by a high-performance liquid chromatography with tandem mass spectrometry detection (HPLC-MS/MS) and the strain was found to produce zeaxanthin as the major component, which appeared at a retention time (tR) of 28.89 min, showing a typical mass spectrum with a molecular ion at m/z 568.5 [M]+ and four product ions at m/z 261.4 [M−307]+, 476.6 [M−92]+, 429.3 [M−139]+, and 536.5 [M− 32]+. Other carotenoids, including zeaxanthin cis isomers, β-cryptoxanthin, β-carotene cis isomer, and β-carotene, are as minor components. The novel and noteworthy finding of this report is the identification of a Seonamhaeicola species that produces carotenoids and can be used as a source of zeaxanthin.

Abstract

NORM (Naturally Occurring Radioactive Material) is a radionuclide element that naturally already exists in the earth. Its concentration can be increased by industrial activities, such as coal-fired power plant (CFPP). Coal-fired power plant activities produce fly ash and bottom ash which will be carried away by the wind and then can enter the CFPP environment, one of which is marine waters and can affect the existence of marine biota. The determination of the radiation dose rate is essential in assessing the risk of radionuclide exposure to the marine environment. This study aims to determine and evaluate the total dose rate of 210Po to marine biota taken from the Karangsari fish market with catchment areas around the waters of CFPP Tanjung Awar – Awar, Tuban, East Java. This research was conducted in April 2021 in the waters of CFPP Tanjung Awar – Awar.  210Po measurement activity was carried out using alpha ray spectrometry at the Marine Radioecology Laboratory of PTKMR-BATAN, then the radiation dose rate was calculated using the ERICA Tool software. The value of the total radiation dose of 210Po on marine biota ranges from 2.70E-1 µGy.hr-1 to 39.70E+0 µGy.hr-1. The radiation dose of 210Po on marine biota measured in the waters of CFPP, has a lower value range than the research result carried out in other countries. Based on the Erica Tools software analysis, the total radiation dose measured on marine organisms in the waters of CFPP Tanjung Awar – Awar, does not give a negatively impact to the marine ecosystems and the sustainability of marine organisms in the study area.

Abstract

Namanereis sp. belongs to class Polychaeta, a group of main benthic community in marine ecosystems. The purpose of this study to determine the potential of bioactive compounds from Namanereis sp. extract as an antibacterial agent and to determine its extract content through Gas Chromatography–Mass Spectrometry (GC-MS) analysis. Samples of Namaneries sp. were collected from Demak Waters, Central of Java. This research used experimental laboratory method. Samples were extracted with non-polar (N-Hexane), semi-polar (Ethyl acetate) and polar (Methanol) solutions. Different concentrations of extraction, i.e. 5, 10, 20, 40, 80, and 100 µg.disk-1. then were tested against pathogenic bacteria Escherichia coli and Staphylococcus aureus. The extract content analysis was obtained using the GC-MS method.  The results showed that Namanereis sp. extract had a potential as an antibacterial agent. The highest inhibition zone was seen in the fraction with ethyl acetate solvent at a concentration of 100%, namely 13±3.05 mm in E. coli and 14±3.04 mm in S. aureus, which then was analysed with GC-MS. Its results found 20 bioactive compounds in the extract. The five most dominant compounds determined by the highest peak were Dihydrojasmone (12.31%), Hexanoic acid (11.49%), Cholesterol (7.66%), Cyclohexanol (7.53%) and Isopropyl Palmitate (7.52%). The research concluded that Namanereis sp. possessed antibacterial potency and contained several bioactive compounds.

Abstract

Soft coral Nepthea sp. grows in the seas of South-East Sulawesi, Indonesia. However, information on the chemical and pharmaceutical aspects of this genus is still limited. Therefore, this research aims to explore the chemical contents and biological activities of Nepthea sp. The sample was collected from the waters of Saponda Island by SCUBA diving. It was extracted by ethyl acetate and fractionated using vacuum liquid chromatography. The chemical content was analyzed by phytochemical screening, LC-MS/MS analysis, Total Phenolics Content and Total Flavonoids Contents. Antioxidant potency was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals and ABTS (2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid). Cytotoxicity property was analyzed by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays. The result showed that the fractionation of Nepthea ethylacetate extracts produced six fractions (A-F). Fractions A and B contain non-polar compounds. Based on LC-MS/MS data, the non-polar compounds in Fraction A and B include achillin, atractylenolide II, buthyl isobuthyl phthalate, rengyolester, 2a-acetoxycostic acid, ocotillol acetate, petasitolone and some unidentified compounds that are C33H58O4, C15H21NO, C21H33NO, and C16H20O4. In general, the antioxidant and cytotoxic properties of all samples are in the weak category, however, when examined for each sample, the antioxidant properties of fraction B is slightly better than fraction A based on the IC50 value of DPPH and ABTS. Cytotoxicity of Fraction A is better than Fraction B against Breast Cancer cell lines MCF-7. The non-polar fraction of Nepthea sp. can be developed as raw material for the discovery of new compounds, antioxidant and anticancer agents, especially breast cancer.

Abstract

Seagrass plays an important role in aquatic resources, such as to support the sustainable management of small-scale fisheries, ensuring the availability of seagrass stocks for generations of local communities to cultivate in a sustainable manner. The purpose of this study is to provide information on the seagrass health status to support sustainable small-scale fisheries in the South Misool Regional Waters Conservation Areas which is located within the Raja Ampat Marine Protected Area of  West Papua. The research was conducted in January 2019 in the Yefgag, Yellu and Harapan Jaya island. A total of ten quadratic transects measuring 1x1 m were laid perpendicularly to the coastline adapted from the seagrass watch method to collect the seagrass data, i.e. the species and the frequency of seagrass found, the dominance and the percentage of seagrass cover. Additional data on fish species were collected by interviewing the local fishermen directly. The relationship between seagrass cover and the number of fish species was analyzed. Th results showed that there were eight species of seagrass found in three observation stations, i.e. Halophila ovalis, Halodule uninervis, Halodule pinifolia, Halophila minor, Syringodium isoetifolium, Cymodocea serrulata, Cymodocea rotundata and Enhalus acoroides. According to the standard criteria for the health status of seagrass beds, the three locations are classified as less rich/less healthy. It because the seagrass coverage was in the range of 30-59%. The relationship between the percentage of seagrass cover and the number of fish species resulted equation of  Y = 15,923x + 0,3174 with R2 = 0,763. It means that the percentage of seagrass cover affects the abundance of fish species by 76,3% with the remaining being influenced by other variables, such as water quality.
