Abstract

Bullet Tuna (Auxis rochei) is classified under the neritic tuna group, which plays an essential role in small-scale fisheries in Karangasem Bali. The increasing catch of bullet tuna indicated its stock probably under threat. Therefore, genetic characterization is often required as the first step before building a conservation program. This study aims to categorize bullet tuna DNA using microsatellite. Of all five loci used, all were high polymorphism-type, with the number of alleles per locus varied between 18-27. Successful PCR (Polymerase Chain Reaction) created relatively high DNA concentration, ranging from 27.050 to 237.05 ng/ul, with a DNA purity level ranging from 2.073 to 2.239. Overall, the stock condition allegedly still in good condition, marked with high genes diversity (Ho=0.367-0.767 and He=0.934-0.966). All loci used can be amplified and well described.

Abstract

The use of basket traps has contributed to the utilization of reef fisheries resource in Karimunjawa Island. The basket trap is a fishing gear that dominant used for capturing grouper, which is a high economic valueble fish consumption. The demand for groupers continues to increase, so fishermen tend to catch all sizes of groupers, including young immature fish. Currently, there are indications that the condition of grouper fisheries has decreased in stock due to fishing pressure. If this is ignored, there may be opportunities to threaten the sustainability status of grouper fish resources. An option to reduce the catch of immature grouper is the use of escape gaps. However, it is not yet known to what extent its effectiveness for fishing activities in Karimunjawa Island. Therefore, this study aims to examine the effectiveness of using the escape gap in traps for reducing the chances of catching immature groupers. The research was conducted using an experimental fishing method. The data collection method was carried out through field observations. Fish survival ratio were calculated using non linier least square method by the solver facility. The results showed that the catch of immature groupers decreased from 66.7% to 45.5%. The use of escape gaps was able to reduce immature grouper with and effectiveness of 69.2%. Grouper can survive after passing through the escape gap with a survival ratio at the eighth day of observation was 77.8%, where the survival ratio of juvenile (86.4%) is higher than adult groupers (69.2%).

Abstract

Ikan Petek mengikuti persamaan pertumbuhan von Bertalanffy Lt = 170(1-exp-0,66(t+0,08528)).

Abstract

The slow growth of the Java barb (Puntius javanicus Bleeker, 1855) fingerlings is due to the feeding of plant-based protein which is usually lack of lysine.  The lack of it, can hinder the feed efficiency and growth. The best way to solve the problem is by adding lysine in the diet. The objectives of the study were to determine optimal needs of lysine to increase feed efficiency, growth, and nutrient contents of Java barb.  The fish samples were fingerlings of Java barb which weighed from 3.26 to 3.78 g-1 fish. The feed comprised 30% isoprotein and 262 Kcal isoenergy supplemented with lysine amino acid.  The dosages of the lysine in the diet were 1.25% (A1), 1.5% (A2), 1.75% (A3), 2.0% (A4), 2.25% (A5), and 2.5% (A6) of dried diets. The parameters observed were relative growth rate (RGR), apparent digestibility coefficients of protein (ADCp), efficiency of feed usage (EFU), feed conversion ratio (FCR), protein efficiency ratio (PER), survival rate (SR), hepatosomalic index (HSI), viscerosomatic index (VSI) and nutrient contents of Java barb. The results of the study disclosed that the enrichment of lysine into the diet increased the feed utilization, the growth and the nutrient contents of Java barb.  The needs of lysine for Java Barb (Puntius javanicus) fingerlings to optimize feed efficiency, growth, and nutrition contents ranged from 1.58% to-1.70% dried diet (5.3 to 5.6 % of protein diet).

Abstract

Liquid organic fertilizer from Sargassum sp. with the treatment of different concentrations of protease enzymes, namely 0%, 1%, 2% and 3% are expected to increase the content of nutrients and growth hormones in plants. The purpose of this study was to determine the effect of differences in protease concentrations on the nutrient content of liquid organic fertilizer. Size reduction of Sargassum sp. and soaking in phosphoric acid solution. Homogenization and heating with the addition of 3% molasses as a source of microbial nutrition and 1% KOH to maximize mineral content. The fermentation process was carried out for 10 days as the exponential phase of Trichoderma sp. which plays a role in nitrogen fixation and produces phytohormones, as well as decomposers of organic matter. The results of ANOVA analysis and Tukey's test showed that the treatment of different concentrations of protease enzymes gave significant differences. The composition of the protease enzyme with a concentration of 3% gave the highest value for C-Organic (1.56%), Nitrogen (0.90), phosphorus (0.88%) and potassium (0.85%), while the yield (65, 83%) and the highest pH value (6.33%) was the protease enzyme with a concentration of 0%. Growth hormones in liquid organic fertilizers are auxin, gibberellins, abscisic acid, zeatin and kinentin. The yield of liquid organic fertilizer hormones with the addition of enzymes, namely 36% auxin hormone, 29% gibberellin, 16% zeatin and 17% kinentin, while for abscisic acid is lower at 99%.
