Abstract

Some genes like 18S rRNA, 26S rRNA, elongation factor 1-alpha (EF1a), and beta-tubulin (TUB) are members of housekeeping genes group that are commonly used as internal control in gene expression study. This study aimed to isolate those four housekeeping genes of tuntun angin (Elaeocarpus floribundus). The research material included fresh leaves of E. floribundus that were picked up from Kajuik Lake in Riau Province and four primer pairs. The procedures consisted of total DNA isolation using Genomic DNA Mini Kit Plant (Geneaid), polymerase chain reaction (PCR), electrophoresis on 1% agarose gel, sequencing, and bioinformatic analysis. This study has been isolated 18S rRNA, 26S rRNA, EF1a, and TUB genes with the size of 422 bp, 922 bp, 856 bp, and 877 bp, respectively. The EF1a and TUB genes has never been reported in Elaeocarpaceae family. Thus, those partial DNA sequences are the first sequences reported from this species and can be used as reference genes in this plant after validation.

Abstract

Orchid is a plant with a high aesthetic value.The existence of orchids directly by the community from their natural habitat, causing the existence of orchid in nature was threatened. The potential of orchid diversity in the Gebugan Nature Reserve area needs exploration to maintain its sustainability. This research would explore the diversity and abundance of orchid species and determined the environmental conditionof Gebugan Nature Reserve. The study was done on 2 stations with altitude difference are 900 m asl and 1045 m asl. Five plots of 10 m x 10 m were systematically constructed at each station with a distance of plots 50 meters. Species of orchids found in the Gebugan Nature Reserve were 12 species, including 11 species of epiphytic orchids and 1 species of terrestrial orchids. The abundance of individual orchid species was relatively higher in places with higher altitudes. Micropera sp was the most abundant orchid species and Corymborkis veratrifolia was the species of orchid with the lowest abundance. It is found that the environmental conditions of Gebugan Nature Reserve are suitable for orchid. The novelty of the research is providing a new data base of orchid in the Gebugan Nature Reserve. The result of the study would be beneficial for developing strategy of genetic conservation of orchird germplasm.

Abstract

Analysis fingerprinting teak is supported by information about teak’s character molecularly. The information is gotten by analyzing 3 primary microsatellite in 46 teak samples that produce total 18 kinds of alel averagely 6 alel per locus. Genetics variety level is shown by Observed Heterozigosity (Ho), Expected Heterozygosity (He), Polimorfi sm Information Content (PIC) and Genetics Differentiation Coeffi cient successively 0.5122; 0.6221; 0.5818; and 0.0629. Kinship connection through dendogram analysis produces coeffi cient resemblance 0.3-1.00 that form 2 groups. However it has not shown inclination of forming group based on origin grows yet. Genetics differentiation analysis indicates different genetics in a population is higher then it is between populations. Analysis result has not found specifi c alel yet but has found 12 specifi c genotipe that is potential for analyzing fi ngerprinting if it relates to important useful genes. Abstract

Research about the relationship of body length and ratio papilla with sex in goby fish (S. macrostetholepis Blkr.) has been done at Animal Structure and Developmental Laboratory, Biologi Department, Faculty of Matematics and Natural Sciences, Andalas University, Padang, which purposed to analyse the relationship of body length and ratio papilla with sex of goby fish (S. macrostetholepis Blkr.). The samples were taken in wild stream area at Batangkuranji river, Padang City. This research used descriptive method and data were analyzed by qualitatively and quantitatively. The results of investigation showed that in several goby fishes (S. macrostetholepis Blkr.) with different sex had the same of body length and the same of ratio papilla. So, there was not relationship between of body length and ratio papilla with sex.  Goby fishes (S. macrostetholepis Blkr.) it belongs to the hermaphrodite protogini, which the androgynous young females, while in adulthood, it would change sex to male. The results of this study are expected to add to the treasures of knowledge and information about reproductive gobies (S. macrostetholepis Blkr.) in the preservation and development of fish farming.

Abstract

Kaili is one of the ethnic region in Central Sulawesi which saves a lot of cultural values and traditions. As a traditional community, their life is very dependent upon natural resources contained in the environment. They still have knowledge, traditional culture, treatment and utilization system against various types of plants. The purpose of the study was to examine the knowledge understanding and utilization of medicinal plants by local community Tompu District of Kaili. Data knowledge and utilization were collected through interview, literature study, exploratory survey methods, PEA (participatory ethnobotanical appraisal), questionnaire and from interviews with the informants. The results from interviews showed that of public knowledge is still based on the traditional concept. Based on the results identifications obtained by (90 species). As many as six species medicinal plants to often used the Tompu community are Euphorbia hirta L. Phyllanthus niruri L. Ageratum L. Blumea conyzoides balsaminifera L. (DC). Kleinhovia hospita L and Tabernaemontana pandacaqui. Abstract

Penelitian bertujunan mengamati serangga-serangga hama penting yang menyerang tanaman tebu di PTPN VII, Cinta Manis Sumatera Selatan, dilakukan dari bulan Februari sampai September 2012. Metode survei langsung ke pertanaman dengan mengikuti jadual early warning system (EWS) PTPN VII. Hasil penelitian menemukan serangga hama penting tanaman tebu ialah penggerek batang bergaris (Chilo saccharipaghus), penggerek batang berkilat (Chilo auricilius), dan penggerek pucuk (Scirpophaga nivella). Gejala serangan penggerek batang dan pucuk tebu ditemukanpada umur 2 bulan, serangan tinggi pada umur 3-5 bulan, hal ini berkaitan dengan cuaca. Pada saat penelitian dilakukan serangan tertinggi terjadi pada bulan Mei sampai Juli, dengan suhu, curah hujan, jumlah hari hujan, dan kelembaban nisbi berturut-turut adalah 26,6OC, 245,5 mm, 17 hari dan 98%. Serangan pada tanaman muda, menyebabkan kematian. Pada tanaman yang sudah membentuk ruas, gejala penggerek batang jelas terlihat dari luar jika daun tebu “diklentek”. Gejala serangan penggerek pucuk terlihat pada helai daun yang berlubang.Intensitas serangan penggerek batang tertinggi pada umur tebu 3 bulan (6,69%), sedangkan intensitas serangan (2,97%) dan populasi penggerek pucuk (44,60 larva) tertinggi pada umur 3,5 bulan. The research aimed to observe the important pest insects attacking sugarcane in PTPN VII, Cinta Manis,South Sumatra. Direct Survey methodto the crop by following the schedule ofearly warning system (EWS) of PTPN VII, conducted from February to September 2012. The results found that important insect pests attacking sugarcane werestriped stem borer (Chilo saccharipaghus), shiny stem borer (Chilo auricilius), and shoot borers (Scirpophaga nivella). Symptoms attack of shotand stemborer of sugarcane were found at 2 months of sugarcane age, high attack at the age of 3-5 months, it is highly related to weather.The highest attack occurred in May and July, with the temperature, precipitation, number of rainy days, and the relative humidity in were 26.6 oC, 245.5 mm, 17 days and 98% respectively. Attack on young plants, causing death. In plants that were already established segments, stem borer symptoms clearly visible from the outside ifsugarcane leaves were “diklentek”. Shoot borers attack symptoms seen in the perforated leaves. The highest intensity attackof stem borer was at 3 months of sugarcane age (6.69%), while the attack intensity (2.97%) and the highestpopulation of shoot borer(44.60 larvae) were at3.5 months of sugarcane age.

Abstract

A survey has been conducted in grade XI natural science of senior high school, at odd semester, academic year 2016/2017 in Samarinda, Indonesia. The focus of this research are to determine the students’ ethnicities, and the responses of biology teachers about students' critical thinking skills, and Problem-Based Learning. Sampling technique was total sampling. The research instrument was a questionnaire. Data was analysed by techniques used descriptive. The results of the data analysis show that the compositions of students’ ethnics are: Java (12.7%), Bugis (6.9%), Kutai (13.3%), Banjar (13.3%), and other ethnics (28.5%). Most of the teachers (86.0%) who already understood about the learning approaches, said that they already understood, while the remaining 14% did not understand the patterns of Problem-Based Learning. In addition, 23.3% teachers said that students were less able to explain, express opinions, and make conclusions; 76.7% said that students are sufficiently able to explain, express opinions, and make conclusions. Future research related to biology learning in multiethnic students needs to be conducted to get more information. The design of learning tools needs to consider the ethnic  of the students in strengthening critical thinking skills. Similar research needs to be done to obtain more information.The findings of this research are the students’ ethnicities of the XI class of Natural Sciences, as well as the responses of high school biology teachers about students' critical thinking skills in Biology.

Abstract

Gandarusa (Justicia gendarussa Burm.f.) is an Indonesian medicinal plant that has many benefits as drug and male contracetive. For industrial needs, Gandarusa must be  available in large quantity. Hairy root culture is one of methode to produce phytochemistry compound. The objective of the study was to examine the effect of various density of Agrobacterium rhizogenes strain LB510 on hairy roots induction of gandarusa (Justicia gendarussa Burm.f.) leaf plant. Leaf explants were inoculated in MS liquid medium with various density of OD600 = 0.1; 0.2; 0.3; 0.4; and 0.5. Explants were co-cultivated for 2 days on MS solid medium without any hormone then sub-cultured on MS solid medium containing antibiotic cefotaxim 300 ppm, in dark condition. The data were analyzed descriptively and statistically. The results showed that various density of Agrobacterium rhizogenes strain LB510 was affected the lenght of hairy roots induction of J. gendarussa Burm.f., but these was not effected toward lenght formation time and number of hairy root. The treatment of OD600 0.2 was the best treatment for hairy root induction on Justicia gendarussa Burm. f. This data could be used for optimized the quality of methode of hairy root induction.

Abstract

One of soybean problem in Indonesia is leaf damage caused by the Spodoptera litura. The aim of the study was to identify the resistance of soybean genotypes againts army worm based on the preference index and morphophysiological characters. 150 soybean genotypes were grown under field condition from February to May 2018 in Malang (East Java, Indonesia). The preference test was conducted in the laboratory using G100H as resistant reference plant to armyworm. The result showed that the preference index was ranged from 0.09-1.82. A total of 105 genotypes showed a low preference index (resistant), 39 genotypes showed a high preference index (susceptible), and a genotype has similar preference index with the reference plant (has equal resistance with G100H). Based on the preference index and morphophysiological characters, 150 soybean genotypes can be grouped into eight clusters. A genotype from cluster VIII (Anjasmoro/Rajabasa-41) has characteristics of low leaf water content, dense leaf trichomes, and the preference index of 0.14. Those characteristics could be the attributes of the soybean genotypes that is resistant to S. litura. The Anjasmoro/Rajabasa-41 was recommended to be cultivated in Indonesian, or may be used as a gene source in the breeding program for S. litura resistance

Abstract

The high rate of urbanization in Semarang have caused the land conversion from forest into various human need spaces. The land conversion is not only affect the life of humans but also the animals, including ants. The objectives of this study were to examine the dominance of tramp ants in the settlement area of Semarang City. The method used was the bait trap method using chicken intestine as a bait. Samples of ants were collected from four habitats, i.e., waste disposal area, traditional markets, house area, and urban parks. This study found four dominant ants, i.e., Anoplolepis gracilipes, Paratrechina longicornis, Solenopsis geminata and Monomorium pharaonis. Two of them were categorized as tramp ants (Anoplolepis gracillipes and  Paratrechina longicornis). The highest diversity of ants was found in house  (H'= 2.72).  The waste area tends to be an ideal habitat for particular species, which was proved by the low value of Simpson's dominance index (iD = 0.06) and a high value of evenness index (E=0 0.89). This study provides new information about tramp ants population as pest in urban area. The findings would be beneficial for controlling the population of tramp ants, which are disturbing humans in the settlement area.
