Abstract

Tuberculosis (TB) is still a public health problem and is the 13th leading cause of death in the world. TB is an infectious disease caused by the bacterium Mycobacterium tuberculosis. Molecular Rapid Test (TCM) is one method that can be used to diagnose TB. This study aims to find out the description of the results of pulmonary TB examination using the Molecular Rapid Test Method (TCM) at the 7 Ulu Public Health Center, Palembang City in 2022. The results of the TCM examination obtained a total of 2635 patients with suspected pulmonary TB, of the number of patients negative results were more dominant compared to Rif Sen, Rif Res, and Rif Indet results. In patients with pulmonary TB based on male sex, the total was 272 patients and female sex as many as 187 patients. In this study, the male sex was more dominantly affected by pulmonary TB than the female sex. Meanwhile, in patients with pulmonary TB, the total productive age was 395 patients and 65 patients were not productive. In this study, productive age was more dominantly affected by pulmonary TB than non-productive age. The conclusion in this study is that male sex and productive age are more at risk of developing TB. Gender and age influence in this study. It is hoped that the community will prevent pulmonary TB by practicing a clean and healthy lifestyle, carrying out the BCG vaccination, and opening windows in the morning so that the house gets sunlight

Abstract

Bacteriological examination to confirm the presence of Mycobacterium tuberculosis or Acid-Fast Bacilli (AFB) in pulmonary tuberculosis refers to the smear examination of sputum samples. Direct microscopic examination of AFB in sputum has limitations, as the sample often contains a large amount of tissue and mucus, which reduces the likelihood of collecting a specimen containing Mycobacterium tuberculosis bacilli. Therefore, a decontamination process is performed by adding NaOH solution to liquefy the sputum, thus increasing the effectiveness of microscopic examination. Decontamination is carried out by adding NaOH solutions at different concentrations, namely 2% and 4%. This study aims to compare the results of AFB microscopic examination using the conventional method and the decontamination method with 2% and 4% NaOH concentrations. A total of 20 samples were used, with mucoid and purulent sputum consistency. Decontamination was performed with a sample-to-NaOH ratio of 1:1. This research is an experimental analytic study with a cross-sectional approach. The average number of positive AFB counts using the conventional (control) method was 19, while the average AFB count in samples decontaminated with 2% NaOH was 24, and the average count in samples decontaminated with 4% NaOH was 31. Statistical analysis using One-Way ANOVA yielded an F-value of 12.718 with a significance value (sig.) of <.001 (< 0.05), indicating a significant difference in the AFB counts per 100 fields of view between the samples treated with 2% and 4% NaOH decontamination. Based on the findings of this study, it can be concluded that the more effective NaOH concentration for sputum sample decontamination in microscopic AFB examination is 4%.

Abstract

Diabetes is a disease characterized by increased blood sugar levels, caused by pancreatic damage or insulin resistance or both. Various serious complications can occur of patient diabetes mellitus  one of which is diabetic ulcers caused by peripheral neuropathy. Diabetes Mellitus (DM) cases at Kraton Hospital, Pekalongan Regency in 2023 is 232 cases. This study aims to sensitivity of bacterial findings in isolates of acute ulcers and chronic ulcers in Diabetes Mellitus patients at Kraton Hospital, to prescribed antibiotics. The type of method used was analytical descriptive research, namely to determine and test differences in the sensitivity of bacterial findings in acute ulcer and chronic ulcer isolates in Diabetes Mellitus patients at Kraton Hospital, to prescribed antibiotics. There are differences in the results of sensitivity tests for acute ulcer samples and chronic ulcer samples. There is a difference in the sensitivity test results of acute ulcer samples and chronic ulcer samples, namely in sample 1 (acute) Staphylococcus aureus is sensitive to Ampicilin while Escherichia coli is sensitive to all antibiotics tested (Ampicilin, Meropenem, Metronidazole, and Ciprofloxacin), in sample 2 ( acute) Escherichia coli and Pseudomonas aeruginosa were sensitive to Meropenem, while in sample 3 (chronic) Proteus mirabilis was sensitive to Ampicilin.

Abstract

Klebsiella pneumoniae is a pathogenicbacteria that causes infectious diseases in humans. Klebsiella pneumoniae is an opportunistic bacterium that always shows higher resistance to antibiotics. The development of herbal medicines as an alternative to antibiotics is an effort to overcome bacterial resistance to antibiotics. There are substances in randu honey that have bactericidal and bacteriostatic properties, such as antibiotics. Therefore, the aim of the research is to find out how effective the ethanol extract of randu honey is in stopping the development of Klebsiella pneumoniae bacteria. This research used diffusion techniques to test various concentrations of randu honey extract. The data collected shows the diameter of the zone that inhibits the use of honey extract to prevent the growth of the test bacteria. To analyze the data, a one-way ANOVA test was used using the SPSS 29.0 program. The results showed that inhibition started with a 60% concentration, showing an inhibitory zone diameter of 14.25 ± 2.06 mm, an 80% concentration with an inhibitory zone diameter of 16.25 ± 1.89 mm, and a 100% concentration with an inhibitory zone diameter of 18.25 ± 1.71 mm. The greater the concentration level of the extract, the greater the diameter of the inhibition zone formed. It can be concluded that the minimal concentration of extract that can be used to inhibit bacterial growth is 60%.

Abstract

Candida albicans is a fungus that has dimorphic abilities that can grow in the form of yeast or hyphae. Candida albicans fungus can be found in various parts of the human body, including the urinary tract. In individuals with Diabetes Mellitus, high blood glucose levels can increase the risk of fungal infections, because glucose in the urine provides an environment that supports fungal growth. Candida albicans fungus can be detected molecularly which aims to identify DNA in the fungus and the initial molecular stage carried out is DNA isolation. The purpose of this study was to determine the presence or absence of Candida albicans fungus in urine isolate samples of women with diabetes mellitus at the Pajang Health Center, Surakarta City. The research method used was descriptive using a cross-sectional approach. The samples used were urine samples of women with diabetes mellitus at the Pajang Health Center, Surakarta City. Based on the results of the study, data analysis and discussion in this study, it can be concluded that Candida albicans fungus can be detected in the ITS1 region at 219 bp and ITS2 at 338 bp using the Polymerase Chain Reaction (PCR) method.

Abstract

Tofu is a daily staple food that is often consumed by Indonesian people because it is affordable and contains high protein. Poisoning with preservatives (borax) can occur through food or snacks, one of which is tofu. This research aims to analyze the borax content in white tofu food at the Wage Banjarharjo Brebes Market. This research method is laboratory observation. The sample used was white tofu which was bought and sold at the Banjarharjo wage market, which was taken randomly from 14 traders. The method used for the qualitative test of borax is using the flame test and the quantitative test is the acid-base titration test. Analysis of research results was carried out descriptively. Based on the results of qualitative research using the flame test, it shows that tofu contained positive borax in samples from traders B, H and L. Meanwhile, tofu that did not contain borax or was negative was A, C, D, E, F, G, I, J, K, M, and N. In addition, the results of research using a quantitative method using the acid base titration method showed that the volume of borax with levels in trader B was 1.1% (w/w), in trader H 2.9% (w/w) and in the sample trader L 2.5 % (w/w).

Abstract

Leukocytes and lymphocytes play a role in the immune response to tuberculosis (TB), but their specific relationship with TB severity remains unclear. This study aims to analyze the association between leukocyte and lymphocyte counts with the IUATLD scale as an indicator of TB severity stratification. This study employed statistical analysis using SPSS version 26. Spearman’s correlation test was used to assess the relationship between leukocyte and lymphocyte counts with the IUATLD scale, while ordinal logistic regression was applied to evaluate the effect of each variable on TB severity stratification. The analysis showed that leukocyte count had a significant but weak correlation with the IUATLD scale (ρ = 0.263, p = 0.002). Conversely, lymphocyte count did not significantly correlate (ρ = 0.092, p = 0.285). Ordinal logistic regression confirmed that leukocyte count had a significant effect on the IUATLD scale (p = 0.018, Estimate = 0.000), whereas lymphocyte count had no statistically significant impact (p = 0.906, Estimate = -1.964E-5). Leukocytes can be used as an additional hematological indicator in TB severity stratification, although their effect is relatively small. In contrast, lymphocytes do not significantly correlate with TB severity, likely due to immune suppression mechanisms induced by Mycobacterium tuberculosis. This study underscores the need for further exploration of other hematological biomarkers to enhance the accuracy of TB severity stratification.

Abstract

The source of microorganism contamination in wet cake snacks can occur in food raw materials, the behavior of sellers and buyers when selling the wet cakes in the market. Cases of bacterial contamination in food often occur and this can cause serious diseases. Based on the above, the problem and purpose of this study is how microbial contamination in wet cake culinary snacks sold in traditional markets in Manado City. Samples of wet cake culinary snacks were taken randomly from six traditional markets in Manado City, then examined in the bacteriology laboratory of the Medical Laboratory Technology Department of the Manado Ministry of Health Polytechnic. The parameters tested were the Total Plate Count (ALT) value and the isolation and identification of Escherichia coli, Staphylococcus aureus and Salmonella sp. bacteria. Based on the results of the study, it can be concluded that bacterial contamination in culinary snacks of wet cakes in traditional markets in Manado City is Escherichia coli contamination of 15 (9.5%) samples, Staphylococcus aureus contamination of 77 (48.7%) samples, Salmonella Sp contamination of 2 (1.3%) samples and contamination identified 2 types of bacteria, namely E. coli and S. aureus of 6 (4.8%) samples. The Total Plate Count (TPC) value of the samples is 76 (48.1%) samples do not meet the requirements and 82 (51.9%) samples meet the requirements.

Abstract

Background: Pregnancy is one of the factors that contribute to increased blood glucose levels. Although Gestational Diabetes Mellitus (GDM) in pregnant women is temporary, some studies show that more than 50% of pregnant women with GDM are at risk of developing type 2 diabetes, preeclampsia, eclampsia, and complications during labor which can result in overweight babies and can also be born prematurely and can even result in physical disabilities in babies. Objective: This study was to determine the picture of blood glucose levels in pregnant women at the Sukarami Health Center, Palembang City in 2025. Research Sample: totaled 30 samples. Type of Research: descriptive, and the sampling technique uses accidental sampling based on a researcher's consideration. Method: examination using POCT. Results: of 30 samples of pregnant women with elevated blood glucose levels as many as 2 samples and with normal blood glucose levels as many as 28 samples. Suggestion: to the community, especially pregnant women, it is recommended to increase knowledge information, especially about blood glucose levels in pregnant women.

Abstract

The incidence of anemia among pregnant women in Puskesmas Wuryantoro in 2022 was 31 cases or 13.08%, then in 2023 there were still 22 cases of anemic pregnant women or 9.65%, where the coverage target for anemic pregnant women should be less than 6.60%. Fe tablets given to pregnant women aim to build iron reserves and red blood cell synthesis. Vitamin C as a combination of Fe tablets can increase the bioavailability of iron, so that iron can maximally increase the hemoglobin content in the body. This study focuses on the effect of the use of iron tablets supported by the administration of Vitamin C to increase the amount of hemoglobin in pregnant women who visit the Wuryantoro Health Center. This research is analytic comparative through pre-experimental one-group pretest-posttest design method. The subjects studied were 25 pregnant women who had never received iron tablets combined with Vitamin C at the Wuryantoro Health Center in February 2025 taken by purposive sampling. The research time was February to March 2025. Examination of hemoglobin levels using Hematology Analyzer. Data analysis with paired sample t-test (related) at a confidence level of 95% data processing with the Statistical Product and Service Solutions (SPSS) program version 27. The research analysis gave a significance value (p) of 0.000 <0.05. The conclusion of this study is that there is a significant difference in hemoglobin levels of pregnant women before and after being given Fe tablets with Vitamin C at the Wuryantoro Health Center, Wonogiri.
