Abstract

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis bacteria is still a health problem in the world, including Indonesia. One way to diagnose this disease is by microscopic examination in the form of Acid Resistant Basil (BTA) staining by the Ziehl-Neelsen method using sputum specimens. This check in Indonesia is very often used because the material price is quite affordable and easy to get. Sodium hypochlorite is a disinfectant solution that can be beneficial as a sputum thinner. The purpose of this study was to analyze the degree of positivity of BTA coloring results with the use of NaOCl solution of 0.25%, 0.5% and 1%. The Research Design used in this descriptive descriptive research is Observational. The subject of this study was a suspected sputum sample of TB at Aisyiyah Klaten RSU on February 10, 2025 to February 24, 2025 with a total of 10 samples. From the results of the study obtained degrees of positivity of microscopic TB diagnostics between without use of NaOCl solutions compared with the use of NaOCL 0.25%, 0.5% and 1%. Conclusion: The use of Sodium Hypochlorite (NaOCl) in sputum samples has been shown to increase the degree of positivity of microscopic diagnostic tuberculosis. The number of positive samples increased from 6 samples in the group without NaOCl to 6 postive samples and 2 scanty samples in the group with NaOCl 1%. In addition, there was an increase in positivity score from +1 to +2 in 4 samples and from negative to scanty in 2 samples after the use of NaOCl of 1%. NaOCl concentration 1% is the best concentration capable of increasing the average value of the number of Mycobacterium tuberculosis cells. The average number of BTA cells increased from 809 cells in the group without NaOCl to 2,168 cells in the group with 1% NaOCl. This concentration also converts 2 samples that were initially negative into scanty. In addition, the addition of this bleach solution can also improve the clarity of the visibility of the microscope.

Abstract

Penyakit infeksi merupakan masalah kesehatan yang utama dibeberapa negara berkembang. Salah satu bakteri penyebab infeksi pada manusia adalah bakteri Staphylococcus aureus. Tanaman yang banyak dimanfaatkan sebagai bahan obat tradisional dan sebagai antibakteri adalah daun belimbing wuluh. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanolik daun belimbing wuluh (Averrhoa bilimbi) terhadap Staphylococcus aureus isolat laboratorium dan isolat pus pasien dan mengetahui perbedaan daya hambat antara kedua bakteri tersebut. Daun belimbing wuluh  diekstraksi dengan cara maserasi dengan menggunakan pelarut etanol 96%. Maserat digunakan untuk uji aktivitas antibakteri terhadap Staphylococcus aureus isolat laboratorium dan isolat pus pasien Rumah Sakit menggunakan metode difusi. Ekstrak daun belimbing wuluh dibuat dengan berbagai konsentrasi yaitu 25%, 50%, 75% dan 100%. Hasil penelitian menunjukan zona hambat tertingggi pada uji difusi yaitu pada konsentrasi 100% sebesar 23 mm pada isolate laboratorium dan 25,33 mm pada isolat pus pasien. Namun analisis hasil statistik konsentrasi ekstrak daun belimbing wuluh yang paling efektif dalam menghambat Staphylococcus aureus isolat Laboratorium dan isolat pus pasien Rumah Sakit adalah pada konsentrasi 25%. Ekstrak etanolik daun belimbing wuluh mempunyai aktifitas yang sama terhadap bakteri Staphylococcus aureus  isolat Laboratorium dan isolat pus pasien Rumah Sakit.

Abstract

Abstrak Penularan HIV dari ibu ke anak (transmisi vertikal) masih menjadi tantangan utama dalam pengendalian HIV/AIDS di Indonesia. Berdasarkan data terbaru tahun 2024, mayoritas kasus HIV pada anak disebabkan oleh infeksi yang ditularkan selama kehamilan, persalinan, atau menyusui, dengan estimasi lebih dari 90% berasal dari jalur ini. Penelitian ini menggunakan pendekatan observasional terhadap data penjaringan pemeriksaan HIV serta tinjauan literatur untuk menganalisis faktor risiko penularan vertikal dan efektivitas media edukasi. Hasil menunjukkan bahwa kurangnya pemahaman ibu hamil dan tidak tersedianya intervensi tepat waktu meningkatkan risiko penularan. Studi juga mengungkap bahwa media leaflet dan audiovisual sama-sama efektif dalam meningkatkan pengetahuan ibu hamil tentang pencegahan HIV, meskipun media audiovisual dinilai lebih menarik dan mudah diakses. Penerapan program Pencegahan Penularan HIV dari Ibu ke Anak (PPIA) melalui strategi 4 pilar terbukti mampu menekan risiko penularan hingga di bawah 5% jika dijalankan secara konsisten. Kesuksesan program ini sangat bergantung pada kolaborasi lintas sektor, edukasi berkelanjutan, serta dukungan sistem skrining HIV yang kuat selama masa kehamilan dan pascapersalinan. Abstract Mother-to-child transmission (MTCT) of HIV remains a major challenge in the control of HIV/AIDS in Indonesia. According to the latest data from 2024, the majority of HIV cases in children are caused by infections transmitted during pregnancy, childbirth, or breastfeeding, with over 90% estimated to result from this transmission route. This study employs an observational approach to data from HIV screening programs and a literature review to analyze the risk factors for vertical transmission and the effectiveness of educational media. The findings indicate that a lack of awareness among pregnant women and the absence of timely interventions increase the risk of transmission. The study also reveals that both leaflet and audiovisual media are effective in enhancing pregnant women's knowledge about HIV prevention, although audiovisual media is considered more engaging and accessible. The implementation of the Prevention of Mother-to-Child Transmission (PMTCT) program through a four-pronged strategy has been proven to reduce the risk of transmission to below 5% when applied consistently. The success of this program largely depends on cross-sector collaboration, continuous education, and strong support from HIV screening systems during pregnancy and the postpartum period.

Abstract

Abstract Gout Arthritis is a joint disease caused by high levels of uric acid in the blood exceeding normal limits, causing accumulation in the joints and other organs of the body, this accumulation makes the joints sore, painful, and inflamed. Gout arthritis sufferers control joint damage with non-pharmacological therapy including patient education, diet management, and aiming to reduce foods rich in purine as a natural gout control. Dietary compliance related to the treatment given, reducing the intake of foods high in purine helps control the production of uric acid in the body. Cases of gout arthritis need great support from the family related to nutrition, psychological conditions. Family support is an important factor in realizing compliance with medical programs. The purpose of this study was to analyze family support with low-purine diet compliance in Gout Arthritis sufferers in the Helvetia Medan Health Center work area in 2024. The research design was descriptive correlation with a cross-sectional approach. Purposive sampling technique, the sample of this study amounted to 48 respondents, the research instrument used a family support questionnaire sheet with low-purine diet compliance. Statistical tests using the Spearman Rho test. The results of the study showed that there was a relationship between family support and compliance with a low purine diet in gout arthritis patients at the Helvetia Health Center, Medan.

Abstract

The Microbiology Laboratory at the Department of Medical Laboratory Technology, Poltekkes Kemenkes Aceh, often uses Nutrient Agar (NA) media to count the total number of bacteria in various bacteriology practicums. This media, after being made and cooled to harden, is stored in a refrigerator at 2-8°C for 1-2 weeks. Before use, the media must be reheated at 100°C to thaw it. This process is often done repeatedly to save labor and facilities in the laboratory. However, repeated heating of NA media is thought to lower the pH of the media and reduce the number of bacterial colonies that grow, potentially affecting the accuracy of microbiological test results. Therefore, this study was conducted to determine the extent to which repeated heating of NA media affects the pH and growth of bacterial colonies. Experimental method with a completely randomized design. Data were collected through repeating the heating of NA media 4 times, and the pH and number of colonies growing were measured after certain time intervals until heating was done 5 times. The results showed that repeated heating of NA media affected pH and bacterial growth. The more often the media is heated, the lower the pH of the media, and the fewer bacterial colonies grow. Repeated heating of Nutrient Agar (NA) media has an effect on bacterial TPC or the amount of Escherichia coli bacterial growth.

Abstract

ABSTRACT Acute Respiratory Infections (ARI) are a major cause of morbidity among children under five, with a high prevalence in Indonesia, particularly in the 1–4 year age group. Maternal knowledge plays a critical role in preventing ARI. This study aimed to determine the relationship between maternal knowledge and the incidence of ARI among toddlers at UPT Puskesmas Medan Helvetia in 2024. The study employed a cross-sectional design with accidental sampling, involving 20 respondents. A questionnaire was used as the research instrument. Chi-Square test results showed a significant relationship between maternal knowledge and ARI incidence in toddlers (p-value = 0.025). Abstract

Tithonia diversifolia, commonly known as Kipahit, is a wild plant with promising potential as a natural antibacterial agent due to its effectiveness in suppressing bacterial multiplication. This study aimed to evaluate the antibacterial inhibitory power of ethanol extracts from Kipahit leaves against Salmonella typhi and to determine the most effective concentration for inhibiting its growth. To isolate the active constituents from Kipahit leaves, a maceration technique was employed, with 96% ethanol acting as the organic solvent. The disc diffusion method (Kirby-Bauer) was employed to determine antibacterial potency. The presence of a clear halo around the disc demonstrated the compound's capacity to hinder bacterial growth. SPSS 27 was used for data analysis, which involved normality testing (p > 0.05), Kruskal-Wallis testing, and Mann-Whitney U testing to determine significant variations between the treatment group. The highest antibacterial activity was observed at an extract concentration of 70% (categorized as sensitive), while the 10% extract still demonstrated an inhibitory effect classified as intermediate. Statistically, both 40% and 70% concentrations showed significant differences when compared to the negative control. A noticeable difference also emerged when these concentrations were pitted against the 1% chloramphenicol positive control. A clear positive correlation also existed between the concentration of the extract and the diameter of the inhibition zone that developed. Abstract

Acinetobacter baumannii (Carbapenem Resistant Acinetobacter baumannii CRAB) is a bacterium that is resistant to almost all antibiotics and is difficult to eliminate from the environment. Some CRABs can carry genes that allow bacteria to make carbapenemase enzymes that destroy carbapenem antibiotics. These carbapenemase genes can move between bacteria, increasing the possibility of spreading resistance to various drugs. Commonly detected genes are the oxacillinase group such as OXA-23, ΟΧΑ-24/40, ΟΧΑ-51 and OXA-58. A small portion of CP-CRAB has a mobile gene that encodes carbapenemase which is often found in other gram-negative bacteria, such as Enterobacterales. This study aims to detect whether the OXA-51 and OXA-58 genes are detected in Acinetobacter baumannii isolates, which are genes encoding molecular indicators of resistance to the carbapenem group. The study design was an observational analytical study with a cross-sectional approach. The target population was all Acinetobacter baumannii cultures. From the research results, the sensitivity test showed that all samples were resistant to meropenem and imipenem antibiotics, while the DNA isolation process that had been carried out needed to be optimized to obtain the right isolation procedure in order to obtain clear DNA band visualization results and visualization of the OXA-51 gene showed optimal results, while detection of the OXA-58 gene still needed optimization to be more sensitive and specific. The output of the research for publication has cannot been done yet because the research will be continued to the next stage.

Abstract

Contamination of pathogenic bacteria in animal-based food products is a major cause of foodborne illnesses. This study aims to identify the presence of Gram- negative rods and Gram-positive cocci on fingernail clippings of poultry and beef traders at Pasar Gede Hardjonagoro, Surakarta. This descriptive observational study involved 20 samples, consisting of 10 poultry traders and 10 beef traders. Laboratory analysis included Gram staining, selective media isolation, and biochemical tests. The results showed that 18 out of 20 samples contained Gram- negative rods, while 2 samples contained Gram-positive cocci. The presence of these bacteria is strongly associated with poor personal hygiene practices, such as not washing hands with soap, not trimming nails regularly, and not using gloves while handling raw meat. These findings highlight the need for improved hygiene education and better sanitation measures in traditional markets to prevent the spread of foodborne pathogens

Abstract

Triglycerides are a type of blood fat that plays an important role in energy metabolism, but excessive levels can increase the risk of cardiovascular disease. The examination of triglyceride levels generally requires fasting, but the optimal duration of fasting is still debatable. This study aims to analyze the differences in triglyceride levels in non-fasting conditions, 8-hour fasting, and 12-hour fasting. This study is an analytical study with a cross sectional design involving 39 students of the Bachelor of Applied Medical Laboratory Technology Study Program at Al-Irsyad University Cilacap as samples. Blood was taken in three conditions: non-fasting, 8-hour fasting, and 12-hour fasting, then examined using the GPO-PAP colorimetric enzymatic method. The analysis showed that the average triglyceride level in the non-fasting condition was 104.59 mg/dL, 8-hour fasting was 91.90 mg/dL, and 12-hour fasting was 87.08 mg/dL. The results of the Paired Sample T-Test test showed that there was a significant difference between non-fasting conditions and 8-hour fasting (p = 0.009), as well as between non-fasting and 12-hour fasting (p < 0.001). However, there was no significant difference between 8-hour fasting and 12-hour fasting (p = 0.172). Based on these findings, it can be concluded that fasting for 8 hours is enough to significantly reduce triglyceride levels, so it can be considered as the minimum feasible duration for triglyceride.
