Abstract

One of the natural ingredients that is utilized as a medicinal ingredient is the binahoang plant. Binahoang has roots, leaves, tubers, stems, flowers that contain active compounds namely flavonoids, alkaloids, torpenoids, and saponins. The purpose of this research is to determine whether binahoang leaf extract can inhibit the growth of Staphylococcus aureus bacterial biofilm. The research method used is True Experimental Design. This research design uses Posttest Only Control Deisgn. Where there are three groups, namely the treatment group, positive control group and negative control group. The data obtained will be tested statistically using the JASP v.18 analysis of variance method and followed by the Paired Sample T-test parametric test. The results of this research obtained that binahoang leaf extract is able to inhibit biofilm against Staphylococcus aureus bacteria with an average value in the biofilm formation inhibition activity test in 3 treatments namely 0.372; 0.441; 0.314 and positive control 0.610. So it can be stated that the results of bacterial growth in the treatment have a smaller value than the positive control of tetracycline 0.03 mg/ml. The conclusion of this research is that binahoang (A. cordifolia) leaf extract can inhibit biofilm growth against Staphylococcus aureus bacteria.

Abstract

Typhoid fever is a systemic disease caused by Salmonella typhi bacteria with LPS-09 antigens that can stimulate an immune response in the form of IgM antibodies, so it can be detected through Tubex TF and leukocyte counts as predictors to reduce morbidity and mortality. This study aims to determine the relationship between fever duration and leukocyte count based on Tubex TF examination results in diagnosing typhoid fever. The type of research used was associative analytical observational with secondary data from 70 typhoid fever patients at Pasar Rebo Hospital from March 2023 to February 2024. The results showed that most patients were female (41 or 58.6%) and the largest age group was children (6-11 years) with 20 patients (28.6%). Based on the Fisher’s Exact test, there was a significant association between fever duration and Tubex TF result (p=0.000), while there was no association between leucocyte count and Tubex TF result (p=0.645). In conclusion, fever duration can be used as an indicator in the diagnosis of typhoid fever with Tubex TF, while leucocyte count cannot always be used as a specific marker. These results are expected to improve individual health information of typhoid fever patients through early detection and intervention.

Abstract

Vegetables are an important food source because they contain vitamins and minerals. However, vegetables that are contaminated with intestinal nematode eggs and then consumed raw or not washed and cooked properly can become a medium for transmitting intestinal nematode eggs and cause worms. Worms are included in the category of neglected tropical diseases (Neglected Tropical Diseases) where this disease receives little attention and treatment, even though it can cause serious health problems, especially in children, it can cause growth disorders. The purpose of this study was to determine the presence of intestinal nematode eggs in various types of vegetables, namely basil (Ocimum sanctum), kale (Ipomoea aquatica), spinach (Amaranthus tricolor) and lettuce (Lactuca sativa) sold in traditional markets in Palangka Raya as an effort to ensure food safety and control parasitic diseases. The study used a descriptive survey method by conducting microscopic examinations in the laboratory, examination of worm eggs was carried out using the flotation method using 0.9% NaCl. The total number of vegetable samples was 20 samples. The results showed that vegetable samples were contaminated with intestinal nematode eggs with a percentage of positive vegetable samples contaminated with intestinal nematode eggs of 8 samples (40%) and negative samples of 12 (60%). The species of intestinal nematodes that contaminate are Ascaris lumbricoides, Trichuris trichiura and Enterobius vermicularis. The results of this study can add information to always maintain food hygiene, especially vegetables that will be consumed

Abstract

Malaria is a disease caused by the Plasmodium parasite and transmitted through the bite of an infected female Anopheles mosquito. Accurate diagnosis is essential for effective malaria management, especially in endemic areas such as East Kalimantan. This study aims to evaluate the sensitivity and specificity levels of the Rapid Diagnostic Test (RDT) in detecting malaria, correlating the results with thin blood smear microscopy, which serves as the gold standard at Dr. Hardjanto Hospital in Balikpapan. A cross-sectional approach was used, involving 30 samples from patients suspected of having malaria. The analysis showed that RDT had a sensitivity of 86.67% and a specificity of 93.33%, with a positive predictive value (PPV) of 92.85% and a negative predictive value (NPV) of 87.56%. The conclusion of this study is that the specificity of the Rapid Diagnostic Test (RDT) met the minimum criteria set by the World Health Organization (WHO), which is greater than 90%, with a result of 93.33%. However, the sensitivity level was still below the WHO's recommended standard of ≥95%, as it was found to be 86.67% in this study

Abstract

Kendari City is one of the cities with the highest cases of Dengue Hemorrhagic Fever (DHF) in Southeast Sulawesi Province. DHF is caused by infection of the Aedes Sp. mosquito whose infection spreads rapidly. Monitoring and control of vector density is carried out to see high cases of DHF. The installation of ovitraps is a way to describe the density of eggs in the area so that prevention can be carried out more effectively. In addition to the installation of ovitraps, chemical larvicide, namely temephos (abate) is used as a control of DHF vectors by sprinkling abate powder into water reservoirs. The most common vector control currently carried out is chemical control, because it is considered more effective and efficient, but if used continuously it can cause resistance in the vector. The purpose of this study was to determine the distribution and abundance of Aedes Sp. mosquito eggs in ovitraps and the susceptibility of larvae to temephos (abate). The research method used was observational by observing ovitraps, identifying larvae and calculating the percentage of larvae that successfully hatched from the ovitrap results. The results of the study showed that the number of eggs trapped in the ovitrap outside the house was 191 eggs (56.3%0 and inside the house was 148 (43.6%) Aedes Sp. The results of the Aedes Sp. larvae susceptibility test after 24 hours of exposure showed that 100% of Aedes Sp. larvae died and were classified as vulnerable. From the results of the study, it can be concluded that the abundance of Aedes sp. eggs outside the house is higher than inside the house, and the results of the susceptibility of Aedes sp. larvae are included in the vulnerable category

Abstract

Antibiotic Susceptibility Testing (AST) is a fundamental element in the management of bacterial infections and in determining effective antimicrobial therapy for patients. AST methods continue to evolve; however, not all hospitals are equipped with automated testing facilities. Hence, conventional manual AST remains the primary option. This study explored the factors that may influence the results of AST by introducing variations to the reference standard procedures outlined in the CLSI guidelines. AST was conducted using the Kirby-Bauer disc diffusion method on Staphylococcus aureus and Escherichia coli bacteria. The independent variables in this study were the type of media, media thickness, incubation temperature, and incubation time. The dependent variable was the diameter of the inhibition zone around the antibiotic discs. The inhibition zone diameters were compared to the CLSI references. Statistical analysis was performed to determine whether there were significant differences in sensitivity test results across the variations for each tested variable. Based on the research results, it can be concluded that variations in the type of media (MHA, NA, and PCA), media thickness (2, 4, and 6 mm), incubation temperature (25, 35, and 45°C), and incubation time (18, 24, and 48 hours) affect the results of the AST. With these findings, it is recommended that Medical Laboratory Technology personnel continue to follow the standard procedures according to CLSI guidelines to ensure that the results and their interpretations are accountable.
