Abstract

Kalanchoe pinnata (Lmk) Pers is a medicinal plant that has many activities, such as antioxidant, anti-infl ammation and immunosuppressant activities. Based on our previous study, aqueous extract of this plant leaves has a repairing eff ect on lupus nephritis model. The expected active compounds are flavonoids. Those compounds are separated in the ethyl acetate fraction. Therefore, this study observed the activity of the ethyl acetate fraction of Kalanchoe pinnata (Lmk) Pers on reducing the level of Anti- Smith autoantibody and the regulation function of CD4+CD25+ T reg in lupus mice. The experimental groups were a negative control group that received placebo, ethyl acetate fraction of Kalanchoe pinnata (Lmk) Pers (EF-KP) group and a positive control group that received cyclophosphamide. The anti-Sm level was measured by using indirect ELISA. The spleen cells were prepared for CD4+CD25+ T reg assay using fl ow cytometry. The result of this experiment was a reduction of anti-Sm antibody level. The decrease was suffi cient to stabilize lupus condition. The fl ow cytometry assay results showed the increase in the relative percentage of CD4+CD25+ T reg. This function can inhibit the reactivity of autoantigens, so it prevents tissue and organ damage from lupus. However, further research is needed to observe the complete mechanism.

Abstract

Jerbung merupakan jenis udang yang tersebar luas pada perairan Indonesia dan merupakan alergen utama penyebab alergi seafood. Sampai saat ini, menghindari semua penyebab dan pencetus alergi merupakan cara terbaik untuk pengobatan penyakit alergi. Menghindari makanan tertentu layaknya ditegakkan berdasarkan uji alergi seperti skin prick test (SPT). Skin prick test merupakan uji alergi yang umum digunakan untuk menentukan alergen penyebab alergi yang dimediasi oleh IgE. Bahan utama SPT adalah ekstrak protein yang disebut dengan reagent. Protein jerbung diekstrak dari dagingnya menggunakan phospate buffer saline (PBS), kemudian dianalisis menggunakan SDS-PAGE dan konsentrasi proteinnya ditentukan dengan uji Bradford. Ekstrak protein diformulasi sebagai reagen SPT dengan mengikuti standardisasi European Pharmacopoeia Monograph on Allergen Products 7 (2010:1063). Sebanyak 24 subyek alergi makanan dan 16 subyek non alergi makanan dicukitkan ekstrak protein udang jerbung pada kulit tangan bagian bawah. Serum subyek dikumpulkan untuk pengujian IgE total, IgE spesifik dan immunobloting secara in vitro. Sensitivitas ekstrak protein udang jerbung pada penelitian ini adalah 95% dengan kesalahan negatif sebesar 5%, spesifitas sebesar 94% dengan kesalahan positif 6%. Hasil immunobloting menunjukkan bahwa 9 di antara 10 serum subyek alergi udang jerbung yang diuji mampu berikatan secara spesifik (90%) dengan protein reagent udang jerbung pada kisaran berat molekul 28-63 kDa.

Abstract

To recover good appearance of skin, dead skin cells should be removed, among others by using exfoliating agent, and to preserve the fresh skin ethyl vitamin C could be used as an antioxidant. Formulation and evaluation of skin exfoliating gel containing ethyl vitamin C in Macrobead® delivery system, using acrylates copolymer and carboxy vinyl polymer as gel bases have been carried out. The gel evaluation included physical evaluation and safety examination. The results showed that ethyl vitamin C could be trapped into Macrobead® system with good absorption inthe ratio of active content and Macrobead® of 3:4. Based on the physical evaluation of the gel formula, it was found that formula with acrylates copolymer base has altered the consistency and homogenity of gels, but not the formula with carboxy vinyl polymer base. Gels using carboxy vinyl polymer showed good physical stability for28 days of storage. The safety evaluation indicated that the preparation was safe to be used for it did not cause irritation on consumers skin. The examination showed that the preparation could exfoliate dead skin cells and thus freshen the users’ skin effectively.

Abstract

Plant of dewa leaves (Gynura pseudochina ( L. ) DC) from family Asteraceae is a plant that has been widely used for some treatments including blood circulation , trauma, bleeding and blood clot. In this research, the isolation and identification of flavonoid compounds in n-butanol fraction of the methanol extract of dewa leaves (Gynura pseudochina ( L. ) DC) has been carried out. The compound in Dewa leaves was isolated using maceration with methanol which was then partitioned by n-hexane solvent, ethyl acetate and n-butanol. The n-butanol phase was then isolated by paper chromatography with BAW (4:1:5) as a mobile phase which resulted in seven bands. Each of the bands was further identified using spectrophotometry UV-vis to show wavelength of maximum absorption that was given from only three bands for flavonoids namely NB-III, NB-V and NB-VI. Moreover, the bands were further eluated with 15% acetic acid as second mobile phase and the spot appeared in the presence of ammonia. The final isolates were identified by UV-vis spectrophotometer with addition of a shift reagent. The resulting spectrum from the isolates NB-III was considered as the one of flavonol compounds with the OH group at theposition of 3.7 and oxygenation at 6 or 8 and o– at OH group in ring B; NB-V isolates, OH groups at positions 3, 5, 7 and o– at OH group in ring B and NB-VI isolates, the OH group at position 5,7 and 8.

Abstract

Cervical cancer ranks second of cancer that affects women in the world and the first order for women in developing countries. Plant Salung (Psychotria viridiflora Reinw. Ex. Blume) have been used for generations to treat various diseases. The purpose of the study to determine the effects of leaf extracts and fractions salung as the anti-proliferation and inducer of apoptosis in HeLa cervical cancer cells. Concentration of extracts and fractions of 640; 320; 160; 80; 40 µg/mL, while cisplatin using a concentration of 200; 100; 50; 25 µg/mL. The results showed IC50 extract value 380.7 µg/mL. n-hexane fraction amounted to 229.3 µg/mL. ethyl acetate fraction of 116.8 µg/mL, and amounted to 562.8 µg/mL fraksi methanol water, so that the ethyl acetate fraction had enough categories active cytotoxic activity seen from the IC50. To further test the researchers only tested the active fraction is the fraction of ethyl acetate. Results ethyl acetate fraction doubling time is 58, which means replicating or proliferating cells on hour to 58 while the cells replicate cisplatin on the hour to 64.5 and the control cell on the hour to 41. Results of flowcytometry showed ethyl acetate fraction induce apoptosis by 72.82% while cisplatin 87.96%. It can be concluded that the extracts and fractions of leaves salung have a cytotoxic effect, acts as an anti-proliferation and inducer of apoptosis in HeLa cervical cancer cell.

Abstract

This research was carried out to assess the antiplasmodic activity of six fractions from the aethylacetate extract of the stem barks of Garcinia parfivolia. The result showed that fractions C, D, and G were IC50 < 10 µg/ml, fraction E: IC50 24,19 µg/ml, and fraction F and H: > 100 µg/ml against a chloroquine resistant strain of P. falciparum FCR-3.

Abstract

Bacillus megaterium DSM 319 is a microbial neutral protease producer, which is usually used for food and non food (especially for dehairing and bating processes) industries. It was shown in this experiment, that the addition of some additive substances such as sodium benzoat, ammonium quarterner, calcium chloride and zeolite tested as a stabilizer of the ability for protease enzyme activities maintain comparing with control the application for the leather process was sign the difference performance. By using varian analysis and Duncan methode analysis at the 4 additif substances above from 0%, 0.5%, 1%, and 2% can be seen that the highest activity of protease reached at 0.862 U with sodium benzoat and the lowest activity at 0.636 U with zeolite.

Abstract

Structure-Based Virtual Screening (SBVS) protocols targeting C-X-C chemokine receptor type 4 (CXCR4) have been constructed by employing PLANTS 1.2 to perform molecular docking simulations and PyPLIF 0.1.1 to identify Protein-Ligand Interaction Fingerprint (PLIF). By using ChemPLP score from PLANTS 1.2 and Tc-PLIF from PyPLIF 0.1.1 to select best pose in the retrospective SBVS showed Enrichment Factor (EF) values of less than the EF value of the reference SBVS (17.5). Nevertheless, the retrospective SBVS campaigns have also resulted in PLIF bitstrings for all poses resulted from the molecular docking simulations.In this article, binary Quantitative Structure-Activity Relationship (QSAR) analysis employing new predictors ensemble PLIF resulted from the retrospective SBVS campaings, instead of using PLIF bitsrings from the best pose only, are presented. The ensemble PLIF as predictors were calculated by taking into account all poses with ChemPLP score lower than a certain ChemPLP score as the predefi ned cutoff , and subsequently for every compound the percentage of “on” interactions was calculated for every PLIF bitstring. The predefi ned cutoff was selected by performing systematic trials to obtain a ChemPLP score as the cutoff with the highest F-score and Matthews correlation coeffi cient (MCC) value.The results showed that the F-score and MCC values could reach 0.58 and 0.61, respectively with EF value of 323.47, which was much better than the EF value of the reference SBVS protocol.

Abstract

Crude powder of papain is very irritative and unstable, thus it should be formulated in emulgel using factorial design 2. The purpose of the study was to assess the effect of gelling agent (2.5% HPMC, 1 % Carbomer 940 ), oil phase ( liquid paraffin 5% to 7.5 % ), emulsifier ( 1.5% -2% Tween 80 - Span 80) and determine the optimum formula. Viscosity, ease of spread, proteolytic activity and antimicrobial activity of papain emulgel were used as the parameters in formula optimization. Each parameter had a contour plot. With the use of superimposed countor plot, the shaded area could be determined as the optimum formula. The results of the factor effect analysis and its interactions were that gelling agent gave significant effect (p<0.05) and dominant in increasing the viscosity of emulgel and the activity of papain proteolytic enzyme. However, it decreased the diameter of the dispersing area and the inhibition zone diameter. The increase in the concentration liquid paraffin concentration had a dominant and significant effect towards the improvement of the emulgel’s dispersive power and the proteolytic activity of papain. The increase in the combination of Tween 80-Span 80 emulgator had a dominant and significant effect towards the improvement of viscosity and the decrease in the dispersive power, the proteolytic activity, and the inhibition zone diameter. From the superimposed contour plots analysis and response optimizer, the optimum formula was achieved by 2.5% of HMPC gelling agent, 5.6% of liquid paraffin, and 2.4% of emulsifier combination.

Abstract

Telah dilakukan optimasi penandaan sel dengan 51Cr(VI) untuk penentuan respon imun setelah vaksinasi antirabies. Optimasi dilakukan karena, pada pengerjaan awal, penandaan yang didasarkan pada prosedur yang telah dipublikasi memberikan hasil penandaan dan cacah per menit per sel (CPM/ sel) yang rendah. Sangat kontras dengan yang pernah dilaporkan, penandaan dalam media lengkap (media + feral bovin serum, FBS) memberikan efisiensi penandaan hanya 3.7 ± 0,7% dan CPM/sel ≤ 0.02 (persyaratan untuk chromium release assay ≥ 0.2 CPM/sel). CPM/sel dari sel bertanda tetap rendah Walaupun diinkubasi selama lima jam. Penandaan kemudian dilakukan dalam media yang tidak mengandung FBS, atau tanpa media di mana 51Cr langsung ditambahkan pada sel, untuk mengamati apakah ada kompetisi antara sel dengan FBS dalam mengikat 51Cr Efisiensi penandaan dalam media OPTI, EMEM 10, PBS, dan air, atau tanpa media memperlihatkan peningkatan yang sangat berarti. Efisiensi penandaan paling tinggi (96,3 ± 3,9 %) dengan CPM/sel (1 ,7 ± 0,07) Serta kehidupan sel yang terjaga (2 80%) diperoleh dengan media OPTI. Eiisiensi penandaan dan CPM/sel dalam media lain relative lebih rendah, yaitu 12-26% dan 0,07-0,13. Sel Vero dan sel fetus ayam, yang juga dapat digunakan untuk penentuan respon imun setelah vaksinasi antirabies, ditandai pula dengan 51Cr dalam media OPTI. Penandaan dalam kondisi ini memberikan hasil yang memuaskan (efisiensi ~ 90%, dan CPM/sel > 1,5). Hasil penandaan di atas mengindikasikan adanya kompetisi antara FBS (albumin) dalam penandaan sel dengan 51Cr Sel yang ditandai dengan 51Cr dalam media OPTI telah berhasil digunakan untuk penentuan respon imun setelah vaksinasi antirabies secara intradermal dan intramuskular.
