Abstract

Investigation on formulation of film coated tablets containing combination of three active substances, i.e. extracts of soursop leaf, mangosteen rind and Ling Zhi mushroom has been carried out, which was expected to have high antioxidant and immunomodulator activities. The research consists of formulation of film coated tablets F1 containing extracts of soursop leaf-mangosteen rind-Ling Zhi mushroom (100:100:125 mg) and F2 (125:75:125 mg) in each 840 mg tablet. Full examination showed that film coated tablet F1 and F2 comply with the 4th Indonesian Pharmacopoeia, except for the dissolution test. Determination of antioxidant activity with DPPH method showed IC50 of F1 tablet was 215.93 μg/mL, IC50 of F2 tablet was 363.86 μg/mL, whereas IC50 of soursop leaf extract 602.64 μg/mL, IC50 of mangosteen rind extract 111.80 μg/mL and IC50 of Ling Zhi mushroom extract 2199.50 μg/mL. Determination of immunomodulatory activity by in vivo carbon clearance method showed a moderate phagocytic index of 1.385 for F1 tablet, 1.469 for F2 tablet, and <1 for Echinacea extract as positive control. Stability test performed based on the α-mangostin content showed that the shelf-life of F1 tablet was 1.865 years, and F2 tablet was 1.842 years. The result showed that film coated tablet F1 and F2 have both synergistic antioxidant and immunomodulatory activities as compared to their respective single extract. The result showed that the antioxidant activity and stability of film coated F1 tablet is better than F2 tablet, but the imunomodulatory activity of F2 tablet is better than F1 tablet.

Abstract

Subchronic toxicity test is performed to evaluate the eff ect and monitor the toxicity eff ects of a compound. The use of combination of ethanol extract of Detam 1 soybean (EEDS) and Jati Belanda (EEJB) as an antiobesity therapy will be consumed for a long time, therefore this test need to be done. The purpose of this research is to know the subchronic toxic eff ect of the test material on Wistar rat hematology. The study was conducted in accordance with the toxicity test stipulated by the Food and Drug Supervisory Agency of Indonesia, on 120 Wistar rats (60 male, 60 females) for 90 days and 120 days (satellite). Rats were divided into 6 groups. Parameters measured were leukocyte, hemoglobin, MCH, MCHC, MCV, hematocrit, RBC, and platelets using the Hematology Analyzer with the principle of colorimetry. The results showed that the eff ect of the test material on all hematologic parameters was not diff erent from the control group (p <0.005), except for the leukocyte, MCH and platelet levels, but the results could be negligible. Conclusion: subcronic toxicity

Abstract

Mangosteen (Garcinia mangostana L.) rinds extract in capsule form has been marketed. a-mangostin, the active compound of G. mangostana had been known has low solubility in water. This study was aimed to measure and compare antioxidant effect of a-mangostin as cosolvent solution to extracts suspentions. 25 rats were divided into 5 groups: negative (I) and positive (II) control, cosolvent solution (III), extract (IV) and extract® (V). Treatment were given for 14 days, free radicals were induced by CCl . MDA were analyzed using thiobarbituric acid method while SOD by using adenochrome assay. Results showed MDA level were: 1.0±0.19; 0.3±0.05; 0.4±0.04; 0.6±0.04 and 0.5±0.00 nmol/mL, whereas SOD activity were: 17±0.0; 123±25.3; 107±19.0; 73±18.9 and 97±27.0 U/mL for group I, II, III, IV and V, respectively. Kruskal Wallis test showed a significant difference in MDA between group III with IV and V (p>0.05), showing cosolvent has higher ability to decrease MDA than the extracts suspension. There were significant differences of SOD between group III and IV (p>0.05), meaning cosolvent has higher ability to increase SOD activity compare to the extracts. Research showed cosolvent has a higher antioxidant effect with potential to inhibit MDA formation by 70% and increase SOD activity by 623%.

Abstract

Radiopharmaceutical 177Lu-(1,4,7,10-tetraazacyclododecane-N,N’,N”,N”’-tetra acetic acid)-trastuzumab (177Lu-DOTA-trastuzumab), based on antihuman epithelial receptor type 2 (HER-2) monoclonal antibody which is expected to be potential for diagnostic and therapeutic agent of breast cancer positive HER-2, had been successfully prepared with radiochemical purity of > 99%. Preclinical studies aimed in providing basic data for clinical trial and particularly in finding out the effectiveness of 177Lu-DOTA-trastuzumab in killing cancer cells which over express HER-2, have been performed. The data included urine and faeces clearance tests, imaging with gamma camera and cytotoxicity test. The results showed that the excretion of radioactivity post injection of 177Lu-DOTA-trastuzumab in normal rats were more rapid through urine as compared to the excretion through faeces. The gamma camera image on normal rat 144 hours post injection of 177Lu-DOTA-trastuzumab showed that there was remaining a trace of radioactivity in hepatic area. The residue of radioactivity (< 5%, quantified by biodistribution test) was found to be relatively lower than reported for 111In-NSL-trastuzumab. However, this residue of radioactivity has to be seriously considered when 177Lu-DOTA-trastuzumab is going to be applied for treatment of cancer. Cytotoxicity test showed that 177Lu-DOTA-trastuzumab was far more effective in killing cancer cells positive HER-2 (SKOV-3 cell lines) than that of trastuzumab.

Abstract

Lactobacillus isolated from tropical fruits can be used as probiotic and potential to prevent diseases in the digestive tract due to bacteriocin content. One of the bacteriocin called plantaricin F, is produced by Lactobacillus plantarum. Plantarisin F has antimicrobial activity against Gram positive and Gram negative bacteria. The aim of the research was to test the antibacterial activities of several Lactobacillus species on 5 bacteria isolates, and analyzed the gene expression of plantaricin F. Seven isolates of ten selected isolates could inhibit the growth of Gram positive and Gram negative bacteria. Lactobacillus TB (NK) produced the largest inhibition area of 2.05 cm on E.coli, and twice stronger than chloramphenicol control. As confirmation, the plantaricin F gene was analyzed using RT-PCR. The analysis result showed that the size of plantaricin F gene was 70 bp. The highest relative expression of plantaricin F gene was shown by the Lactobacillus Mar A2 isolate of 10,676 Au, followed by Lactobacillus TB (NK) of 2.9207 Au, and the lowest was Lactobacillus TB (CK) of 0.5141 Au. Lactobacillus TB (NK) can be used as candidate of probiotic and alternative antibiotic in preventing of gastrointestinal disease.

Abstract

Natrium askorbil fosfat adalah senyawa hidrofilik yang sulit melewati stratum korneum. Salah satu upaya untuk meningkatkan penetrasi obat melalui stratum korneum adalah dengan mengembangkan sistem niosom. Penelitian ini bertujuan untuk mengetahui konsentrasi optimal span 40 dalam meningkatkan efisiensi penjerapan niosom yang diformulasikan dalam sediaan gel dan menentukan kestabilan serta kemampuan penetrasi sediaan secara in-vitro. Konsentrasi span 40 divariasikan ke dalam tiga formula: F1 (100 μmol), F2 (150 μmol) dan F3 (200 μmol). Sediaan gel dibuat dalam dua formula dengan menggunakan viskolam MAC 10 sebagai basis gel: G1 (natrium askorbil fosfat dengan sistem niosom) dan formula G2 (natrium askorbil fosfat tanpa dibuat sistem niosom). Uji stabilitas sediaan gel dilakukan selama 28 hari meliputi organoleptis, pH dan penetapan kadar. Uji penetrasi dilakukan secara in-vitro menggunakan membran kulit ular. Data dianalisis secara statistik menggunakan program SPSS. Hasil penentuan efisiensi penjerapan menunjukkan formula F1 memiliki efisiensi penjerapan optimal sebesar 99,13±0,10%. Formula G1 lebih stabil selama 28 hari penyimpanan dibandingkan formula G2. Penggunaan span 40 sebagai penyusun niosom dapat meningkatkan penetrasi natrium askorbil fosfat dalam sediaan gel dengan jumlah kumulatif persen difusi selama 8 jam sebesar 89,04±0,01%.

Abstract

The barks of Pentaspadon motleyi Hook. f. (Anacardiaceae) were collected from the Poso district in Central Sulawesi, Indonesia, in 1992 which it has been used traditionally as a remedy for malaria and tuberculosis in the local area. It was found that a mixture of (+)-sesamin (1) and (-)-sesamin (2) in a ratio of 100:86 were present together with lupeol in the bark of Pentaspadon motleyi. This was the first evidence of sesamin being isolated in almost equivalent amounts of (+)- and (-)-forms from a single plant species, and indicates that the biosynthetic pathway of sesamin in the plant is not enantioselective or fuzzy.

Abstract

Carrageenan, a substance of seaweeds made from carrageenophyte, consists of kappa, iota and lamda form. Semi-refined carrageenan can be used as stabilizer, emulsifier, and the thickening agent in industries such as pharmaceutical and cosmetic industries. Tooth paste as a cosmetic product requires a thickening agent in the preparation. In this experiment, semi-refined carrageenan: iota (I) and kappa (K) were used in several concentrations; 0.2, 0.4, 0.6 and 0.8 % and in combination: A(I: K=90: 10), B(I:K=70:30), C(I:K=50:50), D(I:K=30:70), E(I:K=10:90). The determination was based upon organoleptic, physical and chemical analysis. The obtained results showed that 0.8% of combination D gave the best results with some results fulfilling the SNI requirements.

Abstract

The soybean curd residue is the waste product of soybean. It has potential for being excipients in pharmaceutical preparations because of its high carbohydrate element. This study aims to evaluate the soybean curd residue as excipients of piroxicam antiinfl ammatory paste. The excipient of soybean curd residue is made by isolating it with 0.1% KOH, then being characterized by physical, chemical, and functional test after becoming starch. It was prepared in piroxicam antiinfl ammatory paste preparations with ratio of soybean curd residue starch and manihot strach are F1 (0:0.25), F2 (0.25:0), F3 (1.25:1.25), F4 (1.5:1), and F5 (1:1.5). The result of physical and freeze-thaw stability test were analyzed descriptively. The data of viscosity, stickiness, and spreading power test were analyzed using one-way ANOVA. Soybean curd residue starch’s physic is slightly coarse, cubic-shapes powder, uneven surface and nonporous, slightly hygroscopic, and brownish-white. It has distinctive odor and has no taste. Chemical test showed that it contained carbohydrate and had pH 4.9. Functional test showed on average 15% it has viscosity 1959 cps, fl ow time 6.7 seconds and still angle 39.2o. Results of physical and stability tests showed that F3 were the best piroxicam antiinfl ammatory paste formula.

Abstract

In 2014, health insurance entirely colected together into Badan Penyelenggara Jaminan Sosial (BPJS) became Jaminan Kesehatan Nasional (JKN). But there is different payment method that is ASKES used Fee for Services (FFS) method and BPJS use INA-CBGs method. Diabetes mellitus is a chronic disease that patients with that diagnosed should use drug continuously. Therefore, research with conducted by time series longitudinal retrospective method that aimed to observe impact of JKN about drug treatment profile among outpatients of type 2 DM in X Hospital Jakarta. Total of samples collected were 84 patients, 31 (36.90%) of male and 53 (63.10%) of female. Most patients (29.76%) were 66-70 years old. There were 38 patients with 10 times of visite, 27 patients with 11 times of visite, 19 patients with 12 times of visit. Based on drug treatment profile, after JKN has launched, decreasing of total of drug use, number of visit increased to 42 visits, kinds of drug appropriate with DPHO decreased until 79%, kinds of drug appropriate with national formularium rised 97%, generic drugs also increased 54% and non generic drug decreased 47%. Based on descriptive analysis, when JKN has launched, there were impact toward drug treatment profile.
