Abstract

Our previous study showed that one of risk factor may lead treatment failure on tuberculosis was non adherence. The current research was carried out to fi nd the role of pharmacist in increasing on medication adherence of tuberculosis outpatients. A non-randomized controlled study was conducted at one of government hospital in Jakarta during April 2015 to July 2015 using MMAS-8. Total sample were 108 patients that divided each group of 54 patients into experimental group who had education and control group without education. Mean of age were 36.93 (SD=15.02) of experimental group and 38.12 (SD=15.11) of control group. More than 50% respondents were male with education level of senior high school. Control group had higher adherence in pre education than post education (P<0.05). Meanwhile, medication adherence in experimental group of pre education was signifi cantly diff erent with post education, 1.94±1.65 vs 0.54±0.64 (P<0.05). There was no diff erence signifi cant of adherence between experimental group and control group in pre education. Furthermore after patients education had been applied experimental group’s adherence had higher than control group (P<0.05). The fi nding of the study may lead that impact of patient education by pharmacist result in better medication adherence.

Abstract

Hiperkolesterolemia dan hiperglikemia mampu memicu terjadinya radikal bebas yang berlebih. Untuk mencegah terjadinya radikal bebas, diperlukan pemberian antioksidan. Kombinasi ekstrak jahe gajah (Zingiber officinalle Rosc.) yang merupakan tanaman yang memiliki kandungan antioksidan dengan zink diharapkan mampu meningkatkan kemampuan antioksidan untuk melawan radikal bebas yang menyebabkan kerusakan pada organ tubuh. Tujuan penelitian ini adalah mengetahui aktivitas kombinasi ekstrak jahe gajah dan zink berdasarkan pengukuran kadar malonyldialdehide (MDA), aktivitas superoksida dismutase (SOD) dan katalase pada mencit hiperkolesterolemia dan hiperglikemia dengan penginduksi streptozotosin. Mencit jantan DDY sebanyak 48 ekor dibagi menjadi 8 kelompok, masing-masing 6 ekor. Kelompok I (normal), kelompok II (negatif), kelompok III (atorvastatin), kelompok IV (ekstrak jahe 37,5mg/kgbb), keblompok V (zink 20mg/kgbb), kelompok VI (ekstrak jahe 37,5mg/kgbb dan zink 20mg/kgbb), kelompok VII (ekstrak jahe 75mg/kgbb dan zink 20mg/kgbb) dan kelompok VIII (ekstrak jahe 150mg/kgbb dan zink 20mg/kgbb). Pengukuran MDA, SOD dan katalase pada hati dan darah mencit menggunakan spektrofotometer UV-Vis. Data yang diperoleh diuji dengan anova satu arah, lalu dilanjutkan dengan uji Tukey. Hasil penelitian menunjukkan bahwa kombinasi pada kelompok VII mampu menurunkan kadar MDA, meningkatkan kadar SOD dan katalase pada mencit jauh lebih baik dibandingkan yang lainnya. Aktivitas antioksidan kelompok VII ini sebanding dengan kontrol positif.

Abstract

Ion-pair chromatography is applicable for separating mixtures of acids and bases, either in ionic or nonionic form, such as mixture of ascorbic acid, thiamine hydrochloride, riboilavin, nicotinamide, pyridoxine hydrochloride, folic acid, and cyanocobalamine, and menadion sodium bisulphite. Separation using ion-pair chromatography is highly influenced by the type and concentration of the counter ion and the selected pH of mobile phase. Eight Water soluble vitamins Were separated in a single analysis Withtotal analysis time never exceeded 12 minutes by using a reversed-phase column (CS) with a mixture of methanol and solution of 5 mM sodium hexanesulphonate at pH 3.50 (49: 151) as mobile phase, at a flow rate of l ml per minute, and ultraviolet detector at 275 nm. Satisfactory separation was shown by the resolution values of more than 1.5, which were between 3.8-10.6.

Abstract

8-Hydroxy 9,12-octadecadienoic acid (8-HODE) is an oxidized aliphatic fatty acid belongs to the group of oxylipins, which shows bioactivity. It was reported previously that Curvularia lunata BioMCC FE-00283, an endophytic fungi from Cibotium barometz be able to produce 8-HODE by submerged fermentation. Glucose, monosodium glutamate, corn oil are media components which showed significant effect toward HODE production by C. lunata BioMCC FE-00283. In this study we reported the enhancement of 8-HODE production by adding potato extract to previouse medium. These four medium components, which were potato extract, glucose, monosodium glutamate and corn oil, were optimized for 8-HODE production by Response Surface Metodology (RSM). A full factorial Central Composite Design (CCD) was used to explain the interaction effect between media components. Maximum HODE production predicted by the quadratic model was 16.46 mg/L where media composition consits of 36.50% potato extract, 3.60 g/L glucose, 15.92 g/L monosodium glutamate and 0.96 ml/L corn oil. Experimentally verification using these medium composition production of 8-HODE reached 16.189 ± 0.526 mg/L. This optimization,which was added potato extract as variabel in medium optimization was enhanced 3 times fold of 8-HODE concentration compare to the 3 variabel optimization.

Abstract

Objectives : Determine the antidepressant activity of some fractions of the leaves of Ocimum basilicum L Methods : The experimental methods are using Force Swimming Test and Tail Suspension Test. The experiment was divided into seven groups. Negative control group (CMC 1 %), Positive control group (fluoxetine 0,052 mg/20 g BW of mice), test I group (fraction of n-hexane 50 mg/Kg BW), test II group (fraction of ethyl acetate 50 mg/Kg BW), test III group (fraction of ethanol 50 mg/Kg BW), test IV group (combination of cyproheptadine 0,0208 mg/20 g BW of mice and fluoxetine 0,052 mg/20 g BW of mice) and test V group (combination of cyproheptadine 0,0208 mg/20 g BW of mice and fraction of ethyl acetate 50 mg/Kg BW). Result : The Force Swimming Test and Tail Suspension Test showed the same result that ethyl acetate fraction was given significant difference (p<0,05) in immobilization time reduction compared to negative control and has a better immobility reduction time than other groups. Added on cyproheptadine to fluoxetine and ethyl acetate fraction groups showed an increased in immobilization time compared to fluoxetine and ethyl acetate fraction alone. Conclusion : Ethyl acetate fraction showed better antidepressant activity than another fraction through the role of serotonin.

Abstract

Isolation and structure elucidation of an alkaloid, a bioactive compound from the roots of the Indonesian medicinal plant Kemuning, Murraya exotica (Rutaceae), including the brine shrimp lethality test (BSLT) have been carried out. The bioactivity test using Artemia salina, isolation and purification of the ethyl acetate extract performed by column chromatography gave yellow crystals. Based on ultraviolet, infrared, nuclear magnetic resonance (1H-&13C-NMR) and gas chromatographymass spectra the isolated bioactive compound is an indole alkaloid typical to yuehchukene.

Abstract

Salam leaves (Syzygium polyanthum (Wight) Walp.) contains flavonoids, volatile oil and phenolic acid that can inhibiting platelet aggregation induced by ADP (adenosine diphospate), arachidonic acid, epinephrine, thrombin, collagen and aff ecting the activity enzyme ectonucleotidase so it can be used as anti-platelet drug. The purpose of this study was to investigated 70% ethanol extract of salam leaves have anti-platelet aggregation eff ect and increase of dose can also increase anti-platelet aggregation. This study was performed in 5 groups, each group consisted of 5 mice there were: groups of positive control, normal control, 70% ethanol extract leaves doses (150, 300, 450) mg/ kg, each group is given orally once daily during 8 days and measurements of parameters at day-0 and day-9th. The results of this study was 70% ethanol extract of leaves of salam leaves doses (150,300, 450) mg/kgBW showed the effects prolong bleeding time, coagulation time, enhance the decreased plasma absorbance after the addition of ADP. From the measurement parameter bleeding time, coagulation time and measurement decreased plasma absorbance after the addition of ADP that 70% ethanol extract of salam leaves doses 150, 300 and 450 mg/kgBW have anti-platelet aggregation eff ect and increase of dose was able to increase the eff ect anti-platelet aggregation.

Abstract

Munculnya penyakit-penyakit infeksi serta adanya sifat resistensi mikrob terhadap antibiotik yang telah ada, telah mendorong penelitian untuk menemukan antibiotik baru dari bakteri, fungi dan Actinomycetes. Saat ini, kelas Actinomycetes menjadi fokus para peneliti karena salah satu anggotanya, yaitu Streptomyces dipromosikan sebagai bakteri yang mampu menghasilkan zat antimikrob terbesar. Sebanyak 45,6% dari 8.000 antimikrob dihasilkan oleh Streptomyces. Tujuan penelitian ini untuk mengisolasi dan mengkarakterisasi Streptomyces yang berasosiasi dengan rizosfer rumput belulang (Eleusine indica) yang berpotensi sebagai penghasil zat antifungi. Jenis penelitian ini adalah eksplorasi dengan pemeriksaan laboratorium. Berdasarkan hasil penelitian ditemukan sebanyak 33 isolat dari rizosfer rumput belulang dan 17 isolat di antaranya berpotensi sebagai penghasil zat antifungi. Delapan isolat hanya mampu menghambat pertumbuhan salah satu fungi uji, 7 isolat mampu menghambat 2 macam fungi uji dan satu isolat, yaitu RRR67 mampu menghambat pertumbuhan ketiga fungi uji, yaitu Trichophyton mentagrophytes, Aspergilus fumigatus dan Candida albicans. Berdasarkan hasil pemeriksaan dengan mikroskop elektron (SEM) diketahui morfologi spora dan ornamen permukaan rantai spora isolat RRR67 termasuk anggota genus Streptomyces. Penelitian ini menyimpulkan bahwa 17 isolat dari rizosfer rumput belulang (Eleusine indica) berpotensi menghasilkan zat antifungi yang diperlukan untuk pengobatan penyakit-penyakit infeksi.

Abstract

Custard apple (Annona squamosa) is a favorite fruit in Indonesia. The leave of this plant is used and known for long time as an astringent, antiinflammatory, birth control, and antitumor agents. Previous experiments on toxicity using brine shrimp lethality test (BSLT) method indicated that the chloroform fraction of custard apple leaves was toxic to the shrimp larvae with LC50 value at 1.77µg/ml. This experiment was carried out to investigate further the possible role of custard apple leaves as an anticancer agent using HeLa cell line. Experiment was initiated by extracting the leaves using 70% ethanol by means of maceration. The macerate was then fractionated further using chloroform as solvent. The ethanol fraction and chloroform fraction were used in cytotoxicity assay using (cervix cancer) HeLa cell line. Cytotoxicity assay Was done using direct counting method. The amount of living cells were observed and counted, death percentage Was then determined and probit analysis Was applied to determine the LC50 value. LC50 value for ethanol fraction from the leaves of custard apple (Annona squamosa) Was 7.695 µg/ml While for chloroform fraction was 4.547 µg/ml based on cytotoxicity analysis using HeLa cells. This experiment concluded that theleaves of custard apple (Annona squamosa) has cytotoxicity toward HeLa cell lines which confirmed the previous toxicity experiment using brine shrimp lethality test (BSLT) method. Overall, this experiment indicated that the leaves of custard apple (Annona squamosa) have anticancer activity.

Abstract

Rhodamine B is a colouring agent for paint industry, textile and paper. It causes irritation at respiration systems and has a carcinogenic effect. At high concentration rhodamine B causes liver damages. Nowadays, it has been reported being added into cosmetics and food product such as lipstick, eye shadow, rouge, food paste, and kerupuk. This research was conducted to determine rhodamine B in the products by using HPLC and spectrophotometry UV-Vis. It was done through sample collection, sample preparation, validation of analytical method, and analysis of rhodamine B using HPLC and spectrophotometry UV-Vis. The results of HPLC and spectrophotometry UV-Vis showed that four samples of kerupuk (57,14%) and four samples of food paste (50%) contained rhodamine B. However, rhodamine B was not found in cosmetics. The results showed the urgency of fast methods to identify rhodamine, thus its content in food products is carefully controlled.
