Abstract

Chalcones are compounds derived from nature, it is show the anticancer activity. The aim of thisstudy is to determine the binding free energy (affi nity) and interaction of chalcone derivatives as inhibitors for A549 lung cancer cells using 4HFZ protein as receptors. This study used 10 chalcone derivatives which were selected with three lowest IC50 values, fi ve medium IC50 values and two with higher IC50 values (i.e. more than 100 μg/mL) and 4HFZ protein was used as receptor. The structure of the chalcone derivative is transformed in 3D structure, then the docking process of chalcone compounds with amino acids on the MDM2 receptor (4HFZ) is carried out. The simulation results shown that data in the form of binding free energy value (kcal / mol) shown the stability of ligand interaction of chalcone compounds on amino acids on MDM2 receptors. The docking results shown that, compound 2 is able to bind strongly with MDM2 receptor (4HFZ) and it is also stable because it has a low binding-free energy value of -7.2 kcal / mol, with a RMSD value of 0,000. Construction of some hydrogen bonds is the same as active receptor (gefi tinib), they are Leu54 on the NH2.

Abstract

Obesity or overweight is defi ned as the accumulation of abnormal or excessive fat in adipose tissue whichcan damage health.Several plants have been used empirically as antiobesity.This study aims to obtain candidate antiobesity compounds that act as pancreatic lipase enzyme inhibitors, fi nd some amino acids that involved on the active site of the receptors and modelling the interactions of these active compounds at antiobesity receptors.The study start with internal validation of the receptors used (4DOQ), then carried out the docking process for the native ligand, comparison compound (Orlistat) and each test compound with the same receptor. The softwares used are PLANTS, YASARA, ChemSketch, and Pymol. The results of the 4DOQ receptor validation have an RMSD value of 1.7097 Ǻ. The results of the docking score for orlistat (-65.3258), and 4 active compounds namely chlorogenic acid (-69,3009), 5-O-caff eoylquinat acid (-70.6911), 4,5-O-dicaff eoilkuinic acid (-85 , 3375), 5-Op-coumaroilkuinic acid (-67,1886). Amino acids that play a role in the affi nity on the active side of the ligand bond with the pancreatic lipase receptor are THR21 (Threonine), ASN84 (Asparagine), GLY19 (Glycine), GLN156 (Glutamine), ALA85 (Alanine), ILE63 (Isoleucine), ARG62 (Arginine) . The conclusion of this study there are 4 chemical compounds in green coff ee beans that are active as antiobesity with the mechanism of action of the pancreatic lipase enzyme inhibitor, namely chlorogenic acid, 5-O-caff eoylquinic acid, 4,5-O-diphoeilquinic acid, 5-op-coumaroilquinic acid.

Abstract

Cocor bebek leaves contain fl avanoids which have anti-infl ammatory power by inhibitingprostaglandin synthesis by blocking the work of cyclooxygenase and inhibiting leukotriene by blockinglipookigenase. Infl ammation is a local protective response caused by damage to tissue caused by physicaltrauma, damaging chemicals, or microbiological substances. This study aims to determine the eff ect ofcocor bebek leaf extract as an anti-infl ammatory in leg edema of white mice (Rattus novergicus) with2% carrageenan induction. In this test using the in vivo activity test method for white rats (Rattusnovergicus) which has been induced by carrageenan. The volume of the rat’s feet is measured usinga pletismometer. Rats were divided into fi ve groups, the fi rst group as negative control were givendiclofenac sodium gel, groups two, three, four were given ethanol extract of leaves of cocor bebek 200,400 and 600 mg respectively, group fi ve as negative controls then calculated volume of feet. The resultsobtained, extract of 400 mg better anti-infl ammatory eff ects.

Abstract

Continuation of research on Macaranga plant has been conducted. In this study, the totalphenol content (TPC) of Macaranga hispida Blume Mull. Arg leaves extracts were investigated. Gallicacid (1) and methyl gallate (2) have been isolated from the ethyl acetate fraction which highest phenoliccontent (8.41 mg GAE/g). Serial techniques of chromatographic and spectroscopic methods, namelyextraction, fractionation, purifi cation, FTIR, LCMS, and FT-NMR were used to isolate and identify theisolates. Cytotoxicity of compound 1 and 2 against P-388 cell line showed that compound 2 has higheranticancer activity compared to compound 1 with IC50 value of 75.53 μg/mL.

Abstract

Curcumin has antioxidant, antiinfl ammatory, antitumor, apoptotic-inducing, andantiangiogenesis eff ects. In order to study the pharmacokinetics of curcumin, a method for analysisof curcumin in plasma levels is required. The aim of this study was developing of a simple HighPerformance Liquid Chromatography (HPLC) for analysis curcumin in human plasma in-vitro. TheHPLC system was using isocratic technique in column reversed phase of C18 (Reliant® RP-18ecolumn (4.6x250 mm; 5 μm)) and mobile phase of acetonitrile–acetic acid–aquabidest (60:1:39) at fl owrate of 1.0 mL/min. Irbesartan was used as an internal standard. Detector was performed at a wavelengthof 428 nm for curcumin and 270 nm for irbesartan. Linearity test shown linear results with a correlationcoeffi cient (r) of 0.9970. LLOQ value was 0.0196 μg/mL with a diff erentiation value of 10.48-18.09%.The accuracy and precision of this method met requirement with a diff erentiation value and relativestandard deviation of between -5.51-3.04 % and 1.04-1.86 %, respectively. Recovery of this method was94.74-103.12%. This method provides good selectivity for the analysis curcurmin in human plasma.The developed of HPLC was a simple and valid method for analysis curcumin in human plasma.

Abstract

HIV/AIDS has become a global issue in many countries. The implementation of ARV / HAARTprograms is always changing, due to clinical reasons, population factors, and aff ordability. Pharmacoeconomic studies can be one of the considerations in the policy process for procurement and selection of therapy regimens. The aims of this study were to determine the eff ectiveness of treatment based on CD4 increase and cost eff ectiveness analysis of HAART A and HAART B treatment regimens. This study used a cross sectional observation method. Retrospective data collection and non-experimental observation where conducted followes by a quantitative descriptive analysis according to the patient's perspective to obtain costeffectiveness data. Data were statistically analyzed to obtain the cost-eff ectiveness of treatment. The results showed that the cost-eff ectiveness analysis (CEA) of two regimens found that HAART A was more dominant than HAART B with a signifi cant diff erence p<0.05. While ACER regimen HAART A (16,227) was greater than the value ACER therapy B (28,330.7).

Abstract

Allergies are health problems that can affect a person’s quality of life. It is currently unknown whether the use of anti-allergic herbs can improve the quality of life for allergic patients. The purpose of this study was to determine the effect of giving anti-allergic herbs on the quality of life of patients with allergic rhinitis at the Tawangmangu Hortus Medicus Clinic. The design of this research was a pre-post treatment quasi experimental. A total of 55 subjects were given anti-allergic herbal decoction consisting of sembung, javanese chilies, ginger, and nut grass for 4 weeks and drunk 3 times a day. Before the study and on day 28th, the subjects were examined for quality of life using the Short Form-36. The results were analyzed using the Wilcoxon test. The results showed that there was a significant increase in quality of life between before and after drinking anti-allergic herbal medicine (p<0.05), especially for the dimensions of physical function, energy and general health. The conclusion is that anti-allergic herbs can improve the quality of life for allergic rhinitis patients.

Abstract

Diarrhea is a condition in which a person defecates with a soft or liquid consistency, which can evenbe water. Diarrhea can be treated with drugs derived from plants, one of which is white pomegranate skin. Theskin of white pomegranate (Punica granatum L.) is thought to have anti-diarrheal activity because it has fl avonoidsand tannins. This study aims to determine the activity and optimal dose of pomegranate skin infusion.Mice were grouped into 5 groups: negative control (Na CMC 0.5%), positive control (Loperamid HCl), andpomegranate peel infusion test group with a dose of 8 mg / 20 g mice BW, 16 mg / 20 g weight mice and 32 mg./ 20 gr BB mouse. Mice induced diarrhea with Oleum ricini, 30 minutes later the mice were given a test preparation.The onset of diarrhea, stool consistency, diarrhea frequency, duration of diarrhea and stool weight weremeasured every 30 minutes for 6 hours. The results showed that the fi rst dose (8 mg / 20 gr BW of mice) wasthe optimal dose for the initial parameters of diarrhea and fecal weight, which means that there was a signifi cantdiff erence with negative control and positive control (<0.05). The parameters of stool consistency, frequency ofdiarrhea and duration of occurrence of diarrhea showed no signifi cant diff erence between groups of 8 mg / 20 gmice BW, 16 mg / 20 g mice BW and 32 mg / 20 g mice BW, negative control and positive control (p> 0, 05).

Abstract

Andaliman fruit (Zanthoxylum acanthopodium DC.) contain flavonoids that has antioxidantactivity. This study was conducted to obtain the effervescent granules andaliman fruit extract which has the highest antioxidant activity by DPPH free radical scavenging method. Andaliman fruit was extracted in stages using n-hexane, ethyl acetate and 70% ethanol, then dried using freeze drying method. Furthermore, the dry extract of ethanol is formulated using variation acid sources, namely citric acid and tartaric acid. The results showed that the formulas had moisture rate of 2.30 – 3.95%; fl ow rate of 2.53-4.12 g/sec; the particle size distribution with the fi nes <10%; dissolved time < 2 minutes; and a pH value of 5.48-5.65. The results of the antioxidant activity of formula I-III were 74.0; 84.9 and 91.9 ppm respectively. The conclusion of the research showed that the formula I with a combination of citric acid and tartaric acid is a formula that meets the physical quality requirements and has a strong antioxidant activity with IC50 value of 74.0 ppm.

Abstract

Michelia champaca L. is known to have effi cacy as an antimicrobial, antioxidant, and anticancer.In general, endophytic fungi that live in plant tissues is capable of producing secondary metabolites as theirhost plant. The objective of this research is to examine the antimicrobial potential of endophytic mold isolates C.1.1 and C.3.3 from yellow cempaka branches (Michelia champaca L.). Isolates obtained from previous research, fi rst was rejuvenated on PDA. Fermentation was carried out on each isolate for 12 days. The fermented supernatant was then extracted partially with n-hexane, ethyl acetate, and n-butanol solvent. The extract was then concentrated and tested for antimicrobial activity using disk diff usion and microdilution methods. N-butanol extract of endophytic fungi isolates of yellow champaca branches was able to inhibit the microbial growth with the largest inhibition area in isolate C.1.1 in Staphylococcus aureus ATCC 6538 (12.26 mm), Escherichia coli ATCC 8739 (11.40 mm), Candida albicans ATCC 10231 (10.50 mm), and Metichillin-Resistant Staphylococcus aureus (11.30 mm) with Minimum Inhibitory Concentration and Minimum Concentration of 12,5% . It was concluded that both endophytic fungi isolates of yellow champaca branches had the potential to inhibit the growth of pathogenic microbes.
