Abstract

The bacteria Shigella dysentriae and Salmonella enteritidis are gram-negative bacteria in the family Enterobactericeae that cause gastroenteritis. Sabrang onion (Eleutherine americana) is a family Iridaceae, containing flavonoids, alkaloids and saponins. The aims of this study was to determine the antibacterial ablitiy and contact time of ethanol extracts sabrang onion on Shigella dysentriae and Salmonella enteritidis. Research methods of Laboratory experimental. The antibacterial ability of sabrang onions was measured by the agar diffusion modification method and contact time was measured based on the minimum kill concentration values ​​of Shigella dysentriae and Salmonella enteritidis. Research data analyzed with One way ANOVA. The results of ethanol extract of Sabrang onion have antibacterial ability against Shigella dysentriae and Salmonella enteritidis, there is a correlation between the concentration of Sabrang onion extract on inhibition zone diameter. The contact time for Shigella dysentriae showed ethanol extract of Sabrang on minimal kill concentration with an 8 hour contact time had a bactericidal effect with the result of a decrease in the number of bacteria 1000 times on the initial inoculum. The bactericidal effect on Salmonella enteritidis shows ethanol extract of Sabrang onions at a minimum kill concentration with contact time of 5 and 10 minutes. Ethanol extract of Sabrang onion has antibacterial ability against Shigella dysentriae and Salmonella enteritidis. There are differences in the antibacterial effects of ethanol extracts of Sabrang onions against Shigella dysentriae and Salmonella enteritidis at the same contact time with different minimum kill concentration.

Abstract

Mulberry Leaf (Morus alba L.) contains oxyresveratrol has the potential as a skin lightener. The activity of mulberry leaf callus from tissue culture is unknown as an inhibitor of the tyrosinase enzyme. The purpose of this study was to determine the activity callus extracts of mulberry leaf which obtained from tissue culture with the addition of growth regulator NAA 1.0 ppm and BAP 2.5 ppm and to formulate callus extract in Nanostructured Lipid Carrier and its characteristics. Callus was extracted by maceration-sonication. NLC is made by solvent evaporation method, NLC characterization including particle size, polydispersity index, zeta potential and particle morphology. The best NLC was made into a gel and evaluated organoleptic, viscosity, flow properties, pH, and tyrosinase enzyme inhibitory on extracts and NLC gels. The characterization of NLC includes particle size of 189.8 nm-632.8 nm; polydispersity index, 0.387-0.582. The morphology of NLC were spherical and zeta potential of -7.37 mV. NLC gel was semi-solid, greenish yellow, homogeneous, viscosity 530000 cPs, plastic thixotropic flow properties, pH 5.26, and inhibitory activity of tyrosinase enzymes (IC50) of callus extract and NLC gel has 72.51 µg/ml and 79.69 µg/ml, respectively. It can be concluded that callus extract of Mulberry leaf can be prepared into Nanostructured Lipid Carriers, NLC gels are physically and chemically stable and have the potential of lightening activity.

Abstract

Garlic (Allium sativum L) contains allicin which has the property of reducing blood sugar levels. In previous studies, methanol extract of garlic can reduce blood sugar levels in mice at a dose of 400 mg/kg BW. Penetration of allicin can be increased by formulating extracts in the phytosome system. In this study, it will be observed the effect of making a phytosome system on the diffusion rate of allicin. Garlic extract phytosome was made using a thin layer hydration method which was then evaluated for its physicochemical properties which included testing of entrapment efficiency, density, particle size, index polydispersity, and zeta potential. Determination of the value of the diffusion rate of allicin in the extract and in the phytosome system was carried out using a modified diffusion cell for 420 minutes. The phytosome system formed has an entrapment efficiency value of 64.8789% with a density of 1.0051 g/mL, the particle size of 270 nm, the zeta potential of -32.55 mV and polydispersion index of 0.571. Diffusion rate testing indicates the diffusion rate of the phytosome system can increase the diffusion rate of allicin when compared to the diffusion rate of allicin in garlic extract.

Abstract

Mulberry leaves (Morus alba L.) contain quercetin, a flavonoid compound with antibacterial activity against Staphylococcus aureus. For convenience, dosage accuracy, and hygiene, a gel formulation with a mixture of carbopol and propylene glycol is required. This study aimed to determine the right concentration of carbopol and propylene glycol to obtain a gel with good physical properties and antibacterial activity on Staphylococcus aureus. Mulberry leaf extract was obtained by maceration method. A factorial design method was used to optimize carbopol and propylene glycol concentrations. The parameters of gel physical properties were the viscosity, spreadability, adhesion, and pH test. The optimal area was determined by creating a contour plot of the parameters test result. Validation of the optimum formula was by comparing the prediction result to the actual research result. The data were analyzed using SPSS with a 95% confidence level. The results showed that the concentration of carbopol and propylene glycol was 1.04% and 7.85%, 1832.42 ± 62.48 cP viscosity, 5.96 ± 0.08 cm spreadability, 64.63 ± 0.97 seconds of adhesion, and pH 5.26 ± 0.03. The antibacterial test showed an inhibitory power with 28.67 ± 0.76 mm. From the teat result, it can be concluded that the gel had antibacterial activity and good physical properties.

Abstract

Bacterial resistance to antibiotics is a serious concern. The search for new antibacterial compoundsfrom medicinal plants can result in overexploitation of natural ingredients. Therefore, it is necessary to search for new alternative sources. This study was aimed to fi nd new antibacterial agents derived from soil fungi isolated from swampy soil in the Pasir Putih Beach area of Situbondo Regency in inhibiting Pseudomonas aeruginosa (P. aeruginosa). Isolation was carried out using Potato Dextrose Agar (PDA) dissolved in sea water. The isolation results showed that 5 isolates fungi. The fungi IS-STB-III-4 and IS-STB-III-5 have the potential to inhibit bacterial using antagonist tests. Potential soil fungi were fermented for 14 days and extracted with ethyl acetate as a solvent. The extraction results were dissolved in 10% DMSO with a concentration series of 1000 μg /mL, 2000 μg/mL, 4000 μg/mL, 6000 μg/mL, 8000 μg/mL which will be used for antibacterial activity test using the disc diff usion method. The extract activity test showed results that were directly proportional to the extract concentration. The analysis and profi ling of the extract using High Performance Liquid Chromatography (HPLC) instrument showed that the extract contained alkaloids which were then isolated and identifi ed by HPLC.

Abstract

The use of cooking oil repeatedly is one of liver damage trigger which usually be marked byenhancement of SGPT and SGOT level. The objective of this study is to know the eff ect of P. speciosa peelethanolic extract in used cooking oil exposed Wistar rats (R. norvegicus). The indicator are SGPT and SGOTconcentration. Parkia speciosa peel was extracted then use to antioxidant source. Rattus norvegicus was divided into 4 group. Group 1 with no treatment, group 2 with 1 ml used cooking oil 118 mek/kg and 100 mg/l extract, group 3 with 1 ml used cooking oil 118 mek/kg and 200 mg/l extract, and group 4 as positive control with 1 ml used cooking oil 118 mek/kg. Each group serum sample was analyzed for SGPT and SGOT. The result showed that 200 mg extract treatment is signifi cantly bring lower SGPT and SGOT concentration compared to positive control. The higher dose of ethanolic extract of P. speciosa peel is highly prevent liver damage which indicated by the lower SGPT and SGOT concentration in serum. The ethanolic extract of P. speciosa peel has potential to ward off free radicals which caused by used cooking oil exposed. That fact was be marked by SGPT and SGOT decreasing persentage.

Abstract

Wualae (Etlingera elatior (Jack) R. M. Smith) is plant that traditionally used as traditional medicine in Southeast Sulawesi. It contains fl avonoids, tannins, and terpenoids that act as anti-infl ammatory. The studyaims to investigate the anti-infl ammatory activity of ethanolic extract of wualae by stabilizing human red bloodcell (HRBC) membrane. Hemolysis and stability due to oxidative induction and hypotonic solutes are used asmeasures of anti-infl ammatory activity. The positive controls used in this study were diclofenac sodium withvarious concentrations of 250 ppm; 500 ppm; 750 ppm; 1000 ppm; 1250 ppm; and 1500 ppm. Sample wasalso having similar identical concentration. According to the results of stability and hemolysis percentage ofE. elatior fruit ethanol extract to HRBC were 57,75% and 42,25% (250 ppm), 66,71% and 33,29% (500 ppm),74,10% and 25,90% (750 ppm), 75,72% and 24,28% (1000 ppm), 79,87% and 20,13% (1250 ppm), and 84,89%and 15,11% (1500 ppm). Compared with diclofenac sodium stability and hemolysis percentage were 62,61%and 37,39% (250 ppm), 66,71% and 33,29% (500 ppm), 71,02% and 28,98% (750 ppm), 72,10% and 27,90%(1000 ppm), 73,94% and 26,06% (1250 ppm), 76,63% and 23,37% (1500 ppm). These data suggest that the ethanolic extract of wualae is potentially have activity as anti-infl ammatory.

Abstract

Curcumin has low solubility in water and has low stability so that a drug delivery system is needed that can overcome these problems. One delivery system that can be used to overcome these problems is transethosome. Transethosome is composed of several component, one of them is surfactant. Surfactant is a component that can help improve the stability of the transethosome system so that in this study research was conducted on the eff ect of the use of a combination of surfactants (tween 60 and span 60) on the physical stability of transethosome curcumin. In this study used a combination of tween 60 and span 60 with a ratio of 1: 2, 1: 1, 2: 1 at a concentration of 5%. Transethosome formed then characterized include organoleptic, viscosity, particle size and particle size distribution, zeta potential, adsorption effi ciency and pH. Transethosome characteristics were observed for 8 weeks to see the physical stability of the transethosome. The results at week 0 indicate that the transethosome formed a suspension with orange color and smelled of phosphatidylcholine with the highest viscosity in formula 3, has a particle size of 1-1000 nm, polydispersity 0-0.571, has a negative zeta potential value, has an absorption effi ciency above 80 % and pH 8.5-10.0. Physical stability test results show the use of a combination of tween 60 and span 60 with rasio 3:1 (F3) can improve the physical stability of transethosome curcumin.

Abstract

Research on the Estrogenic Mechanism Study of the Combination of Basil Herb (Ocimumamericanum) and Fennel Fruit (Foeniculum vulgare) against the background of research results whichshowed that the combination of 70% ethanol extract of basil herbs 0.35g / 200g BB and fennel fruit0.0725g / 200g BB had effi cacy estrogenic because it greatly aff ects the duration of the estrus phase,vascularization and weight of the ovaries and uterus. The aim and target to be achieved from thisresearch is to know the working mechanism of the combination of estrogenic basil and fennel fruitherbs. The parameters observed were histopathological examination of the ovary and uterus. Histopathictest results in the ovary and uterus show the eff ect of treatment on the development of follicles in theovary and uterine wall thickening. So it can be concluded that the mechanism of estrogenic combinationof basil and fennel fruit extracts occurs through the mechanism of genomics, where phytoestrogencompounds in both plants pass through cell membranes and interact with receptors so that they canproduce estrogen-like eff ects.

Abstract

Administration of antibiotics in the private hospital "X" adopted a concept called RonaldIrwanto Antimicrobial Steweardship Program (RASPRO). The suggestion of an empirical antibioticcombination in type III stratifi cation community pneumonia patients was meropenem-levofl oxacinor meropenem-amikasin. The aim of the study was to determine the eff ect of empirical antibioticcombination meropenem-levofl oxacin with meropenem-amikacin to RASPRO type III stratifi cationcommunity pneumonia patients towards the LOS and decreased leukocytes. The test sample calculatedusing diff erence between two population proportions formula and analysed using Chi-square method.Diabetes mellitus, immobilisation and geriatrics as confounding variables were controlled by logisticregretion multivariate analysis. The results showed that meropenem-levofl oxacin had a tendency 1.81times to experience LOS < 5 days and 0.92 times to experience decreased leukocytes ≥ 10% comparedto meropenem-amikacin, but both were not signifi cant (p 0.161 and p 0.835). The result control ofconfounding variables were found that geriatrics as a meaningful confounding variable eff ect of LOSand no confounding variables were considered to aff ect the decreased leukocytes. In conclusion, thereis no eff ect of empirical antibiotic combination meropenem-levofl oxacin with meropenem-amikacintowards the LOS and decreased leukocytes to RASPRO type III stratifi cation community pneumoniapatients using statistics after controlling the confounding variables.
