Abstract

Soil Transmitted Helminths (STH) infection is still a health problem in Indonesia. This infection is caused by intestinal nematodes which in its transmission require soil media. Vegetables can be a medium of transmission of STH. STH transmission occurs when the soil is contaminated with STH worm eggs and the worm eggs stick to vegetables. In addition, STH contamination can occur in traditional markets with poor sanitation such as humid rooms, floors with lots of puddles, muddy water, and improper placement of waste. The aim of the study was to describe the contamination of Soil Transmitted Helminths on Scallion (Allium fistulosum L.). This research is a descriptive research, the examination of worm eggs is done by indirect method with sedimentation technique. The research sample was taken from 25 traders. Leeks contaminated with STH as many as 15 (60%) samples from 25 traders. The type of STH found was hookworm rhabditiform larvae. There is STH contamination in leeks sold at the Bangetayu market, Semarang City. To avoid the risk of being infected with STH, people are expected to consume vegetables that have been cooked.

Abstract

Tuberculosis is an infectious disease caused by Mycobacterium tuberculosis. Until now, tuberculosis is still a public health problem in the world. Tuberculosis is a dangerous infectious disease, the healing process also takes a long time. Acid-fast bacilli examination is an examination to determine the presence or absence of Mycobacterium tuberculosis bacteria. The factors that influence Tuberculosis patients are age, gender, education and occupation. The purpose is to knowing the description of AFB positivity in patients with clinical diagnosis of pulmonary tuberculosis at Balkesmas Semarang in 2020. This research was an observational study with descriptive research criteria through a retrospective approach. Results from 656 patients with clinical diagnosis of tuberculosis who underwent acid-fast bacilli examination, 220 patients (33.5%) were found positive and acid-fast bacilli were negative (64.5%). The results of the examination of acid-fast bacilli were positive with a scanty positivity degree of 3 patients (1.4%), acid-fast bacilli (+1) were 71 patients (32.3%), (+2) were 83 patients (37.7%), (+3) as many as 63 patients (28.6%). Based on the category of age 0-14 years as many as 1 patient (0.5%), 15-34 years as many as 78 patients (35.5%), 35-54 years as many as 96 patients (43.6%), 55-74 years as many as 44 patients (20%), 75 years as many as 1 patient (0.5%). Based on gender, there were 123 patients (56%) male and 97 female patients (44%). Conclusion the results of the examination of acid-fast bacilli were positive with a scanty positivity rate of 3 patients (1.4%), acid-fast bacilli (+1) 71 patients (32.3%), (+2) 83 patients (37.7%), (+3) 63 patients (28.5%). Based on gender with positive acid-fast bacilli, there were 123 male patients (56%) while female 97 patients (44%). Age with the most positive acid-fast bacilli results in the range 0–14 years 1 patient (0.5%), 15–34 years 78 patients (35.5%), 35-54 years 96 patients (43.6%), 55 –74 years.

Abstract

The cutting stage is the network manufacturing stage which needs to be considered the thickness scale. The standard scale that is commonly used is 2-7 µm, while the thickness of 5 µm which is used as a standard for cutting the thickness of the liver tissue shows that the cell is good for cutting liver tissue in mice (Mus musculus). Mice have many advantages, while the liver organ is one of the organs that are often used in research. The aim of this study was to describe the microscopic quality of liver preparations of mice (Mus musculus) by cutting tissue thickness of 2 µm, 5 µm and 8 µm. This type of research includes experimental research with descriptive research criteria. The study design used a purposive sampling study approach. The results of the quality of microscopic images of mice tissue (Mus musculus) at a thickness of 2 µm there were 12% not good, 20% not good and 68% good. The thickness of 5 µm is 100% good, while the thickness of 8 µm is 8%, the preparation is not good,  36% is not good, and 56% is good. The conclusions of this study were microscopic tissue mice (Mus musculus) preparations with good quality 68%, 8 µm 56%, and 100% at a thickness of 5 µm.

Abstract

During the Covid-19 pandemic, people began to maintain their health, by using herbal medicine as an alternative treatment. One type of herbal medicine used is jamu beras kencur because it has properties to increase stamina, increase appetite, and relieve aches and cough disorders. In the Regulation of the Minister of Health of the Republic of Indonesia No. 007 of 2012 carrying herbs such as jamu beras kencur does not require marketing authorization. The quality of the herbal medicine produced has not been confirmed as feasible from microbial contamination, especially in molds, the number of yeast molds in the jamu beras kencur must comply with BPOM standards Number 32 of 2019 namely AKK 103 colonies/mL. This study aimed to determine the effect of the storage time of jamu beras kencur on mold growth with storage treatments of 0 hours, 4 hours, 8 hours, 12 hours, and 24 hours at room temperature. This type of research is a Quasi Experiment with a research design of One Group Pretest-Postest Design. The results of this study showed that the average AKK value was 0 hours 0.2x100 colonies/mL, 4 hours 0.4x100 colonies/mL, 8 hours 0.4x100 colonies/mL. 12 hours 0.8x100 colonies/mL and 24 hours 2.2x100 colonies/mL. These results stated that the jamu beras kencur met the BPOM standard requirements Number 32 of 2019 which was AKK 103 Colonies/mL. The results of the Kruskal Wallis test obtained a sig value of 0.286, it was concluded that there was no effect of storage time for the jamu beras kencur on the growth of molds. The identification results on the jamu beras kencur obtained fungal species, namely Aspergillus flavus, Aspergillus niger, and Penicillium sp. Suggestions for further researchers can examine the effect of storage time on mold growth in other types of herbal medicine.

Abstract

Process of getting older causes a person to experience physical and mental, spiritual, economic and social changes as well as degeneration of organ function. Age is one of the risk factors for high uric acid. Uric acid is the final product or waste product from purine metabolism, that excess amount in the blood will crystallize. At an advanced age, there will be a decrease in the production of several enzymes and hormones in the body that play a role in the process of removing gout. Exercise such as elderly gymnastic regularly can delay physiological changes that usually occur in a decrease in strength and flexibility, as well as an increased susceptibility to injury. The results are presented in descriptive form which supported by tables to describe uric acid levels and its causative factors in elderly gymnastic participants at the elderly posyandu in Village Timang, Sub District Wonokerto, District Wonogiri. Examination of uric acid levels from 73 respondents were 42 respondents (57.5%) with normal uric acid levels and 31 respondents (42.5%) with uric acid levels above normal. An increase in uric acid levels can be caused by several factors such as age, sex, food intake, physical activity and history.

Abstract

Lemongrass leaves are a typical plant of South Kalimantan which contain anti-insecticide, namely secondary metabolites such as alkaloids, saponins, tannins, steroids and triterpenoids. Pediculosis is a disease caused by a tick infection which is still neglected and becomes a health problem. One way to get rid of head lice is to use natural ingredients that contain anti-insecticide substances. This study aims to examine the possibility of using natural ingredients in lime leaves as vegetable insecticides against head lice mortality. The research method used is experimental, with a research design that is a post-test only control group, which measures and compares the mortality variable of Pediculus humanus capitis after being treated with lime leaf juice with concentrations of 10%, 30%, 60%, 100% during 1, 6, 12 and 24 hours and compared them with the negative control group with aquadest and the positive control with pemethrin. The sample of this study was lime leaf extract which was extracted by the method of squeezing using water. From the results of the study obtained the highest percentage of death at 12 and 24 hours with 100% mortality at a concentration of 100%. Based on the results of statistical tests using mortality observation data, the LC99 values were obtained within 1 hour (197.678%), 6 hours (126.055%), 12 hours (89.847%), and 24 hours (60.758%). Based on the results of the analysis of the data obtained, it can be concluded that there is effectiveness of lime leaf juice (Citrus hystrix) on the mortality of head lice (Pediculus humanus capitis) in vitro. Then the juice of lime leaves can be used as an alternative to getting rid of head lice with natural ingredients.

Abstract

TB patients who do not adhere to OAT treatment can lead to resistance and become MDR-TB and add to the burden of TB management globally. TB control programs including microscopic sputum examination are important for diagnosis, evaluation, follow-up treatment, and follow-up. The results of the sputum examination used the IUATLD scale as an interpretation of the results and the reporting was negative, scanty, positive one, positive two, and positive three according to the number of TB germs found. The results of the sputum examination to determine the classification of the patient, the decision to start treatment, and the patient's recovery. At the step of treatment for MDR-TB patients, the results of sputum examination are an indicator to determine the success rate of treatment which is marked by the conversion of BTA. Research objectives is to determine the description of the results of sputum examination in Multidrug-Resistant Tuberculosis (MDR-TB) patients at the Kendal District Health Center. Research methods is observational research with descriptive research criteria through a cross sectional approach. Respondents of the study were multidrug-resistant tuberculosis (MDR-TB) patients from three health centers in Kendal district with the highest case prevalence. The study respondents were 7 MDR-TB patients. All study respondents, totaling 7 MDR-TB patients (100%), were included in the advanced step of treatment and showed negative sputum examination results. All study respondents (100%) showed that the results of microscopic sputum examination were negative, which means that BTA conversion occurred at the advanced treatment step.

Abstract

Clearing results in histological processing can be affected due to tissue density and viscosity of the clearing agent. Xylene is the most commonly used aromatic solvent for clearing agents and deparaffinizing agents in histopathology laboratories, but xylene is one of the most dangerous chemicals found in histology laboratories. Based on the dangers posed by xylene, a replacement material is needed. Several xylene substitutes such as reagents derived from limonene, aliphatic hydrocarbons, vegetable oils and mineral oils have been commercially developed, but the available xylene substitutes are still less effective, more expensive, and still as dangerous as xylene itself. Substitutes that are considered safer are from natural oils, one of which is olive oil which has similarities with xylene, namely in hydrocarbon and phenol compounds. This study used two types of tissue, namely skin and liver from mice (Mus musculus) which were cut into two parts; the first part uses xylene as and the other part uses olive oil as a clearing agent. The assessment of hematoxylin eosin staining results were determined based on the category of assessment of cell structure, such as the cell nucleus, cytoplasm, and color uniformity carried out by three readers and five fields of view for each microscopic slide at 40X magnification of the objective lens. Comparison of observations of the xylene group in skin and liver tissue 100% got a good score on the cell nucleus, cytoplasm, and color uniformity. (p=1,000). The olive oil group had a slight difference in color uniformity in liver tissue when compared to skin, but not statistically different (p=0.773). The comparison of the overall readings of the xylene and olive oil groups on skin and liver tissue also showed no statistically different (p=0.262). So it can be concluded that olive oil can be recommended as a substitute for xylene in the clearing process in histological tissue processing.

Abstract

Endospores are formed by members of several genera of gram-positive bacteria, such as Bacillus and Clostridium. The spores are very resistant structurally; withstand the temperature of boiling water for two hours or more. They contain very little water and show very few chemical reactions. The favorable external environment will cause the spores to break down and the vegetative cells to appear to grow and reproduce. Identification of endospore-forming pathogens is important in food and medical microbiology. However, the spores contain multiple protective layers, which cannot be easily penetrated by simple staining or Gram staining techniques. Therefore, it is necessary to apply heat to help penetrate the color, with various methods of Gram stain, fluorescent spore painting, Schaeffer and Fulton spore painting and Client's spore painting method. The results of the fluorescent painting method have better results but require much more expensive equipment using a fluorescent microscope considering the facilities that must be provided for the Schaeffer and Fulton painting method as an alternative if they do not have these facilities.

Abstract

Blood glucose levels are influenced by factors related to pastients and laboratory factors. There are various ways to treat blood glucose that have increased or decreased. Water consumption helps the process of removing all toxins in the body, including excess sugar. In James’s research (2010), that drinking water causes a breakdown of sugar. This research is to Know the description of fasting blood glucose levels with and without drinking water. Descriptive type research with cross sectional approach. Blood glucose levels of 28 student of TLM was examined by POCT methode and the respondent filled out the questionnare. The results of blood glucose levels are described based on questionnares that have been filled in the form of respondents diagram. From the results of examination 28 respondents, mean blood glucose level in conditions without drinking is 82,03 mg/dl, respondents with a minimum level of 65 mg/dl and a maximum of 97 mg/dl. Mean blood glucose level in conditions with drinking is 78,71 mg/dl, respondents with minimum level of 69 mg/dl and maximum of 104 mg/dl. Respondent’s fasting blood glucose levels in conditions without drinking and drinking decreased by 4%. There was a decrease in fasting blood glucose levels in respondents conditions without drinking and by drinking water, also influenced by physical activity factors, nutrition intake, drugs/vitamin and obesity.
