Abstract

Avian orthoreovirus (ARV) is widespread and found in almost every commercial poultry farm and other poultry species. Virus isolation from local isolates is necessary to obtain vaccine seeds that are homologous to viruses in the field.  The R&D Laboratory of PT Vaksindo Satwa Nusantara successfully isolated ARV from field cases. Inoculation of SPF Embryonated Chicken Eggs (ECE) is the protocol used to evaluate virus titer, especially to calculate the antigen titer of vaccine prototype candidates. The gross pathology of chicken embryos infected with ARV to evaluate virus growth has not been widely reported. This study aims to observe the gross pathology of chicken embryos infected with ARV to measure the virus titer using infectivity test such as the embryo infectious dose 50 (EID50). The virus suspension was diluted from 10-1 to 10-6 then dilutions of 10-4 to 10-6 were inoculated each into five ECE. Incubated ten days and observed for embryonic death. The observations found the embryonic death on day seven. Pathological changes in the embryos are stunted, thin feather, oedema, and less allantois fluid compared to control of uninfected embryo. The results of the embryos necropsy showed cardiac necrosis, greenish liver, and gelatinous exudate in the abdominal cavity. Keywords - Avian Orthoreovirus, Embryonated Chicken Eggs, Embryo Pathology, EID50.

Abstract

This research aims to determine the most optimal type of filter paper in making strip tests from crude anthocyanins of purple sweet potato to detect borax. The experimental design consisted of paper type optimization by testing magenta color intensity, sensitivity tests of strip tests using borax standard solution by testing cyan color intensity, and application of strip tests on food. Optimization of paper type was tested quantitatively with the treatment of ordinary filter paper and whatman paper number 1, 40, 41, and 42. The sensitivity was tested quantitatively with four concentrations of borax. The results of the application of borax test strip tests on food products were analyzed qualitatively. The results of the research on paper type optimization showed that the color intensity on 5 types of paper was significantly different, with the highest color intensity value owned by ordinary filter paper. Subsequent sensitivity tests on ordinary filter paper at four borax concentrations showed results with a detection limit of 1 ppm. The application test of purple sweet potato strip test, using ordinary filter paper, demonstrated its effectiveness as a borax detection tool in food products, with a sensitivity limit of up to 0.5%. Abstract

PT. Indah Kiat Pulp and Paper Perawang is a company that prioritizes paper production. There is a crucial unit known as the lime kiln within the company. Issues occurring within this unit often disrupt the overall production process. Therefore, conducting reliability analysis on the instrumentation of this unit is highly important. This research utilizes the Reliability Centered Maintenance (RCM) method. The objective of this study is to determine which devices within the lime kiln are most vulnerable to problems by calculating the Risk Priority Number (RPN) for each component, evaluating the reliability level of these devices, and providing optimal maintenance schedule recommendations. The research findings indicate the sequence of RPN values for lime kiln instrumentation from highest to lowest as follows: temperature sensor 336, on-off valve 288, flow transmitter 252, control valve 245, proximity sensor 240, pressure gauge 216, sensor indication of motorized damper 210. Furthermore, the reliability values of each instrumentation did not meet the threshold set by the Indonesian Industry Standard (SII), which is 0.7. Therefore, it can be concluded that maintenance action is required for these instrumentation devices. Maintenance schedule recommendations for temperature sensor are 252 days, on-off valve 294 days, flow transmitter 293 days, control valve 251 days, proximity sensor 293 days, pressure gauge 352 days, and sensor indication of motorized damper 353 days. Keywords – Keandalan, Instrumentasi, Maintenance, Lime kiln, RCM.

Abstract

The use of electric motors in industry often experiences damage caused by several things, including high starting current surges, overload or full load, loss of one phase in the electric motor, and other causes of damage to the 3-phase motor. The main problem that occurred in this research was damage caused by high starting currents, where in the case study many 3-phase motors were used to drive factory machines. The use of 3-phase motors to drive factory machines is often damaged by unstable currents in the machines. This is because the time and workers for monitoring are very minimal and the materials for repairing machines are inadequate. In this way, the solution for repairing a 3-phase motor is to replace and rewind the damaged motor so that it can be used again. This research aims to find a solution on how to reduce high starting currents in 3-phase motors by utilizing the influence of replacing damaged induction motor components as well as repair methods used in starting when rewinding 3-phase motors. The research results from the groove distance with a wire size of 0.90 mm showed that the groove distance of 9, 9, 8 was 29.3, and at a distance of 10, 10, 10 it had a current of 31 A with no-load testing and recommended groove distances of 10, 10, 10. Next, testing wire sizes of 0.75 mm and 0.90 mm with distances of 10, 10, 10, and 9, 9, 8 without load obtained 26.1 A and 27.4 A while 0.90 mm was 30, 7 A and 28, 1 A. The load was 43.5 A and 43.9 A, while 0.90 mm was 51.3 A and 49.3 A. From the test results, the largest current decrease was at a distance of 10, 10, 10 with a wire size of 0.75 mm.
