Abstract

Curcumin is a potential compound to be developed. However, the clinical use of these compounds is limited due to their low stability and bioavailability. Therefore, to increase the pharmacological activity of curcumin, it is necessary to modify the chemical structure of curcumin. Curcumin analog compound (2E,6E)-2-(4-hydroxy-3-methoxy-5-((pyrrolidine)methyl)benzylidene)-6-(4-methoxybenzyliden)cyclohexane-1-on can be synthesized in 3 steps. Phase I, the reaction between p-methoxybenzaldehyde and cyclohexanone took place aldol condensation reaction with NaOH catalyst. Phase II, the Clasein-Schmidt reaction between compound 1 (the result of the synthesis of stage I) and vanillin involved dilute HCl as the catalyst. Phase III, reacting the results of the synthesis of phase II with Mannich pyrrolidine base by adapting the method used by Geschickter and Maeadow. Compound 3 as a result of purification was observed using Thin Layer Chromatography using Ethyl Acetate:Methanol (1:1) as an eluent to obtain Rf 0.46 and a melting range of 130-131 oC. The yield obtained was 51.52% with the characteristics of a reddish brown powder. This compound needs further identification using the Mass Spectrum and NMR.

Abstract

Science and nanotechnology in the pharmaceutical field, among others, are used to increase drug effectiveness and optimal safety. The research was conducted using the review method with the aim of this study to obtain information about the utilization of nanoscience and nanotechnology which can be used as reference material for development in the pharmaceutical field. Nano-drug delivery system technology is carried out by developing materials used both from natural origin, synthetic, and semi-synthetic as well as developing drug delivery systems. Nano-drug delivery system technology or known as smart drugs promises a very bright future through various modifications of drug dosage forms developed from conventional dosage forms to dosage forms with drug delivery systems with controlled drug release. Nano-drugs with nano-delivery systems can be made using carriers in the form of natural polymers, or synthetic ones that can be biodegradable or non-biodegradable. Efficient therapy using nanotechnology is carried out in order to increase the safety level of the drug, as well as the ability of the drug to reach its target with minimal side effects. There are three ways to do this: increasing the solubility and bioavailability of drugs, creating new dosage forms, and using better routes of administration. Nanotechnology has great potential in contributing to the national economy and the nation's competitiveness, although much needs to be done to transform scientific and technological excellence into the economic field.

Abstract

Inflammation is a protective process against tissue injury caused by infection, trauma, or chemicals. The mechanism of inflammation starts from a stimulus that will cause cell damage, these cells will release several phospholipids, one of which is arachidonic acid. Free arachidonic acid will be activated by cyclooxygenase and lipoxygenase enzymes. This enzyme converts arachidonic acid into an unstable form which is further metabolized to leukotrienes, prostaglandins, prostacyclin, and thromboxane. One of the natural medicines used as an inflammatory treatment is jamblang leaf, because there are flavonoids that can inhibit cyclooxygenase and lipoxygenase enzymes. This study aims to determine the anti-inflammatory effect and the best dose of jamblang leaf extract. The method used was 1% carrageenan induction and a digital plethysmometer to measure the volume of inflammation on the soles of the test animals' feet. The test animals used were 25 mice divided into 5 treatment groups, each group consisting of 5 mice. The treatment group consisted of: negative control of 1% Na-CMC, positive control of Diclofenac Sodium 0.182 mg/20 g BW, dose 1 of jamblang leaf extract 10 mg/20 g BW, dose 2: 15 mg/20 g BW and dose 3: 20 mg/kg 20 g BB. The results showed that all doses of jamblang leaf extract had an anti-inflammatory effect and dose 3 was the best dose that reduced the volume of inflammation.
