Abstract

Enteropathogenic Escherichia coli (EPEC) is a Gram-negative pathogenic bacterium that causes diarrheal disease, especially in infants and children. Aptamers are short chain oligonucleotides that have high affinity, specificity, and selectivity to their targets, which have potential to be developed as a method for diagnosing pathogens. In this study, aptamer was isolated through the Systematic Evolution of Ligands by Exponential Enrichment (SELEX) method using whole cells bacteria (Bacterial-SELEX) for recognizing pathogenic E. coli EPEC K1.1 which was isolated from children with diarrhea in Indonesia. Ten rounds of bacterial-SELEX procedure were conducted with modification conditions by using Top10, DH5a E. coli cells, Listeria monocytogenes, and Lactobacillus plantarum S34 as counter-selections. The selection process was started with a pool of ssDNA random library consisting of a random base with 40-nucleotides long flanked with fixed primers sequence for aptamer amplification purpose. Short single-stranded DNA amplification was done by symmetric and asymmetric PCR. The highly enriched oligonucleotide pools (pooled 8, 9, and 10) were cloned and the resulting ssDNA aptamers were identified by Sanger DNA sequencing. Finally, twelve aptamers with unique sequences and various secondary structures including G-quadruplex sequence motif within aptamers were obtained as candidates specific aptamer for detection and capturing of EPEC K1.1.

Abstract

This study aims to describe the molecular characteristics of DNA barcodes in chocolate hind grouper (Cephalopholis boenak) in Indonesian waters with new sequence records from Madura Island waters. Partial sequences of the cytochrome c oxidase 1 (CO1) gene were successfully obtained from two samples of C. boenak in Madura Island waters. In contrast, the other sequences were obtained from the BOLD system database, covering the Islands of Bali, Lombok, and Ambon waters. The overall molecular analysis involved 10 C. boenak sequences with a length of 625 bp. The results indicated that the population of C. boenak in Indonesia waters has high genetic diversity, as evidenced by the value of haplotype diversity (Hd) = 0.956 and nucleotide sequence diversity (Pi) = 0.01746, in which the population is distributed into eight haplotypes. Results of phylogenetic tree reconstruction of neighbor-joining and maximum-likelihood indicate similar topology. The branching of NJ and ML phylogenetic trees of C. boenak in Indonesia waters is grouped into two geographical clades. Clade 1 with the subpopulations of the waters from Madura, Bali, and Lombok Islands. Clade 2 with the subpopulations of the waters from Ambon Island. The results of the median-joining network reconstruction depicted a similar topology with both phylogenetic trees.

Abstract

This research presents the unusual staining characteristic of the teleosts red blood cells–the common bluestripe snapper Lutjanus kasmira, caught at Toc Tan (Allison) Reef, Truong Sa Archipelago, Vietnam, due to side effects of the delayed staining procedure. In addition to the quantitative data measured, the results of microscopic image research showed that, on slides performed with the delayed Romanovsky staining procedure using Giemsa dye, the cell "nucleus" was not dye-stained as seen in the erythrocytes of much other fish. The AFM and SEM images of blood slides show that the structure of the nuclear zone and the nuclear membrane was destroyed. Although methanol is used as one of the common fixatives in biology, the delayed staining still causes artifactual side effects, disrupts the structure of the red blood cell nucleus and can cause confusion when studying cytogenetics.

Abstract

Tobacco armyworm (Spodoptera litura Fab.) is one of the major insect pests of crops in Indonesia. The management of this pest still depends on the use of chemical insecticides. The use of bio-insecticides, such as Bacillus thuringiensis (Bt.), are known to be alternatives, but it easily degraded by sunlight. This research aimed to study the effects of UV B on Bt. pathogenicity and to explore plant-based additives as UV B protectants for Bt. against armyworm. Thirteen plant extracts were screened based on their UV spectra absorbencies using UV spectrophotometry. The extracts, namely cloves, Jicama, Celebes pepper, turmeric, and Moringa, then used for the formulations of Bt. and exposed under UV B lights for 0, 72, and 144 h. After exposure to UV B, Bt. formulations were tested for bioassay against one- day-old 1st and one day old 2nd larval instars of tobacco armyworm. The result indicated that at 72 and 144 h of UV B exposures, the Bt. added formulations were signifcantly different compared to Bt. alone. At 72 h exposure against the 2nd larval instar, the larval mortality of tobacco armyworm on the second day of observation on Bt. formulated with Celebes pepper and turmeric was 97.3 and 80%, respectively, whereas, at 144 h exposure, the mortality was 96 and 89.3%, respectively. This study concluded that Celebes pepper and turmeric extracts were the potential to be used as the Bt. protectants against UV B.

Abstract

The mesogenic and velogenic strains of the avian Newcastle Disease Viruses (NDV) have been investigated for use in cancer virotherapy. However, despite its promising results, these strains have the potential to cause outbreaks leading to morbidity and mortality in large numbers of avians that can devastate the poultry industry. Therefore, there is a growing interest towards the use of lentogenic strains such as NDV LaSota vaccine strain for virotherapy. This study investigated the oncolytic potential of the NDV LaSota strain on colorectal cancer (HT29) and cervical cancer (HeLa) cell lines. The NDV LaSota stain was propagated in pathogen free embryonated chicken eggs and quantified using a hemagglutination assay. The oncolytic activity was evaluated by comparison of HT29 and HeLa cell lines to the non-cancerous human embryonic kidney (HEK293) cell line using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) viability assay. The highest NDV concentration of 16 HAU was found to reduce the viability of HT29, HeLa, and HEK293 cell lines to 42±3% (p = 0.0001), 49±12% (p = 0.0009), and 77±5% (p = 0.007), respectively, indicating its potential selective cytotoxicity, as supported by the disturbed cell morphology. The NDV LaSota strain exhibits a potential selective oncolytic activity towards cancer cell lines, thus may serve as an interesting candidate for virotherapy against colorectal and cervical cancer.

Abstract

The oyster mushroom (Pleurotus ostreatus) is the most cultivated mushroom which its growth performance and nutritional composition depend on substrate types. Thus, this study investigated the effect of coconut coir on minerals and vitamins B contents in the oyster mushroom cultivation medium. The composition of coconut coir and Sengon’s sawdust as growth medium were variated at ratios of 4:0 (F1), 3:1 (F2), 2:2 (F3), 1:3 (F4), 0:4 (F5) (w/w). Furthermore, the mineral and vitamin B contents of the harvested oyster mushrooms were analyzed by ICPMS and LCMS, respectively. The highest minerals contents of potassium 26,909 mg/kg, 1,136 mg/kg of phosphorus, 313 mg/kg of magnesium, 4,346 mg/kg of calcium, 15.4 mg/kg for zinc, 2.07 mg/kg of copper, 0.623 mg/kg of Selenium were identified at F1, whereas the highest manganese was at F2 (15.3 mg/kg). Potassium was the highest mineral content in oyster mushrooms in all growth media, and low toxic minerals content of cadmium was detected between 0.015-0.058 mg/kg in all variant substrates. Lead (0.525 mg/kg) and mercury (0.012 mg/kg) was only detected at F1, while arsenic was detected at F4 (0.002 mg/kg) and F5 (0.029 mg/kg). LCMS analysis showed that vitamin B1 (Thiamine), B3 (niacin), B6 (pyridoxine) and B12 (cyanocobalamin) were detected in all growth media, while vitamin B5 (pantothenic acid) was only detected at F5. These results indicated that the P. ostreatus cultivation on coconut coir become valuable nutritional resources to alleviate malnutrition and help dispose of coconut coir in an environment-friendly manner.

Abstract

The exosome is a nano-sized vesicular particle commonly secreted from eukaryotic cells to extracellular space. Exosome functions in cellular communication, nutrients or bioactive compounds delivery, and cellular immunity. It is believed that plant-derived exosome-like particle (PDEN) potential to deliver nano-bioactive compounds to the human body. As Indonesia is one of the herbal centers of the world, we are encouraged to develop natural richness for human welfare. The study aimed to verify various methods to isolate exosomes from the fresh ginger rhizome and quina cells and examine the ginger PDENs' capacity to internalize in vitro human cells. The PDENs isolation was carried out by centrifugation and precipitation. The particles were observed through a transmission electron microscope, dynamic light scattering, and particle size analysis. Differential ultracentrifugation combined with PEG6000 provided a better size range of particles. Nanoparticles tracking analysis performed the concentration of particles within a specific size range. Ginger and quina PDENs size ranged from 40-650 nm, and the concentration was 3.6 × 107 and 2.8 × 106, respectively. The PKH67-labelled ginger PDENs were taken up by human dermal fibroblast and human Wharton's jelly mesenchymal stem cells after 30 minutes and 21 hours. DPPH assays revealed that ginger PDENs have antioxidant activity, indicating the presence of some nano-molecules in those particles. This study would be the baseline for developing ginger PDENs as a functional food with nano-ingredients, or as an effective vehicle of nano-drug, for human health.

Abstract

This study aimed to obtain a functional lipase (LipRM) from Ralstonia pickettii BK6 through a co-expression involving its foldase. The ORF of the LipRM was 999 bp while the gene encoding of lipase-specific foldase (LifRM) was 1,030 bp. LipRM and LifRM genes were cloned into a plasmid and were successfully co-expressed in Escherichia coli strains to produce functional LipRM. Enzyme activity from partially purified enzymes showed quite surprising results, LipRM activity in E. coli BL21 (DE3) was 25.84 U/ml, while the other strains (DH5α, HB101, S17-1λpir) were 628.98 U/ml, 761 U/ml, and 1206.46 U/ml, respectively. The highest relative activity of LipRM was found at 50-55°C and pH 7-8 with pNP-laurate (C12) as the preferred substrate specificity. LipRM activity was enhanced sharply in the presence of 30% organic solvents (methanol and ethanol) but decreased by more than 50% in the presence of detergents. This study was the first to report heterologous expression of Ralstonia pickettii lipase employing its native foldase resulting in functional lipase from subfamily I.2.

Abstract

Fungi from South Sumatra (Indonesia) were identified morphologically and molecularly, and their pathogenicity to egg, larvae, and adult Aedes aegypti was evaluated. The fungal isolates used for bioassay were 11 isolates from this study and 4 isolates from the laboratory collection. Fifteen isolates of five fungal species (Metarhizium anisopliae, Penicillium citrinum, Talaromyces diversus, Beauveria bassiana, and Purpureocillium lilacinum) from South Sumatra, Indonesia, were pathogenic to the egg, larvae, and adult of Ae. aegypti. Egg mortality caused by M. anisopliae isolate MSwTp3 was the highest (38.31%). A novel finding of this study was that the eggs exposed to the fungus not only killed the eggs but could continue to kill the emerging larvae, pupae, and adults. The five fungal species induced larval mortality between 52.22−94.44% and adult mortality between 50.00−92.22%. Fungal strains belonging to M. anisopliae, P. citrinum, T. diversus, and B. bassiana from South Sumatra seem to possess remarkable ovicidal, larvicidal and adulticidal activity against Ae. aegypti. M. anisopliae, P. citrinum, T. diversus, and B. bassiana had the potential as entomopathogens to be developed into ovicides, larvicides, and adulticides for controlling Ae. aegypti.

Abstract

Squamocin is one of the annonaceous acetogenins produced by the Annonaceae family and displays potent anti-cancer activity against cancer cell lines. This study aimed to investigate the growth inhibition activity of squamocin coupled with nanodiamond on rats (Rattus norvegicus)-induced breast cancer. Twenty-five female R. norvegicus were divided into five groups (n = 5), including normal control (without any treatment), negative control, group treated with nanodiamond only (ND), group treated with squamocin only (SQ), and the group treated with squamocin coupled with nanodiamond (NDSQ). All of the animal models were induced for breast cancer, except for the normal control group. Breast cancer induction was performed using two doses of N-nitroso-N-methylurea (NMU) injection (50 and 30 mg/kg body weight) intraperitoneally and waited for 22 weeks until the tumor was detected to formed. Nanodiamond coupled with squamocin were administered by intraperitoneal injection (1.5 mg/kg body weight) for 5 weeks, one injection per 3 days. This study showed that the treatment with squamocin coupled with nanodiamond (NDSQ) significantly reduced the proliferation (Ki-67) and induced apoptosis (Caspase-3) of breast cancer cells, corresponding to the reduction of the thickness of the mammary ductal epithelium (p<0.001) and the lower level of CA-153 in serum. In addition, the treatment significantly reduced the malondioldehyde (MDA) and PI3KCA and increased the p53 level significantly. Altogether, in this study, we are the first to report the anti-cancer activity of squamocin in rat-induced breast cancer and the potency of nanodiamond as a carrier of squamocin to increase its anti-cancer activity.
