Abstract

Urinary tract infections (UTI) was an inflammatory response of the urethra when attacked by microorganisms. This disease can happen to people at all ages beginning with asymptomatic to moderately symptomatic infections. Thus far, antibiotic treat-ment was the best for curing the UTI although the chances for being resistance to the antibiotics were also high. Resistance to gram negative bacteri,a in particular, often occur with the antibiotics treatment. Our study at the Faculty of Medicine, University of Indonesia (UI), Bacterial isolation from patient urine sample was performed in the microbiology laboratory of UI. A total of 50 urine samples were collected from X patients volunteered in our study for bacterial isolation, however, only 23 bacterial isolates were successfully obtained. Study was carried out to moni-tor the susceptibility of bacterial isolate towards several types of antibiotics (ofloksazim, amoxyicillin, fosfomisin and sefepim) using Cakram disffusion method. Study re-sults demonstrated that susceptibility of oflokazim to both gram positive and nega-tive bacteria was very low while amoxycillin showed desecding trend of efficacy towards all types of bacteria. Fosfomisin and sefepim, on the other hand, demon-strated strong susceptibility to both gram positive and negative bacteria found in the isolates. Furthermore, it is very interesting to observe ofloksazim was resistant to gram postivie and negative bacteria. Collectively, these research findings strongly illustrated the susceptibility patten and resistance scale of baterial isolates towards various antibiotic tested in the study.

Abstract

Since policy sector pharmaceutical affairs 1993 by government about deregulations sector community pharmacy, competition business community pharmacy in Jakarta was very strict. For win something this competitions, Kimia Farma community pharmacy as the biggest community pharmacy network in Indonesia have propagan-dized vision be the foremost business networking community pharmacy in Indonesia and mission first-rate services for customers with customer satisfaction. The objec-tive of this research is detect satisfaction level of service quality of Kimia Farma community pharmacy Jakarta, in particular Kimia Farma community pharmacy -1 Kemayoran, Kimia Farma community pharmacy-48 Matraman, Kimia Farma com-munity pharmacy-147 Duren Sawit. Beside This research also objective for knowed vision, mission of Kimia Farma community pharmacy was reached. This research using survey method with quisionaire Single Cross Sectional Study approaches. Research using Servqual (Service Quality) model was discovered by Parasuraman, Zeithaml and Berry use five dimensions of service quality, that was tangible, reli-ability, responsiveness, assurance, and emphaty. Satisfaction level measured with Gap analysis that was difference expectation before customer receiving the service and perception after that; and Cartesian Diagram analysis, that was mapping atribut service quality on Cartesian Diagram. Analysis difference on customer satisfaction within community pharmacy used analysis varian (Anova). Result: dimension that highest satisfaction level was emphaty (gap -0.37 or satisfactions levels 91.88 %); attribute that highest satisfaction level was well designed interior/exterior building (gap -0.14 or satisfaction levels 96.4 %). Mapping attribute on Cartesian Diagram majority on quadrat B, means service quality Kimia Farma community pharmacy was sufficient. Based on hypothesis test, was not difference significant satisfactions level Kimia Farma community pharmacy was research (alpha > 0.05). Conclusions: generally satisfactions level Kimia Farma community pharmacy that researched nearly satisfy, with satisfaction levels within community pharmacy was not significant difference.

Abstract

The administration of antibiotic at RSUD dr. H. Abdul Moeloek Lampung is based on sensitivity test result and empirical therapy on diabetic ulcer. Therefore, the objective of the research is to analyze the effectivity of various antibiotics adminis-tration based on the sensitivity test result on clinical outcome reached by consider-ing influenced factors. On the other hand, total evaluation of the resistances of empirical antibiotic used by considering germ and sensitivity test feature was done. The research was conducted at dr. H. Abdul Moeloek Regional Hospital from April - July 2008 by applying cross sectional design retrospectively. Data was taken by the diabetic ulcer patients medical record which were treated from January 1st 2005 to Mei 30, 2009. Requirement of patient inclution are patient with diabetic ulcer infection, receiving antibiotic recipe, antibiotic given is based on sensitivity test result, patient was cured until getting the doctor approval to go home. The research which involved 98 patient inclution showed that clinical outcome of diabetic ulcer patient who was administered antibiotic based on sensitivity test result was influ-enced by chronic level of patient and age.

Abstract

Japanese encephalitis virus (JEV) is a neuropathogenic virus commonly caused cen-tral nervous diseases such as meningitis and severe encephalitis. Although vaccine has been developed, no specific and effective drug is available so far. We previously carried out a screening of inhibitor of JEV RNA helicase, an enzyme that essential for virus replication, from Actinomycetes and found that Streptomyces chartreusis produce the inhibitor of JEV RNA helicase. In this study, an extracellular protein which has inhibition activity on ATPase activity of JEV RNA helicase was purified from supernatant of Streptomyces chartreusis culture by ammonium sulfate pre-cipitation and size exclusion chromatography. SDS-PAGE analysis showed a single band with aproximate molecular mass of 11,4 kDa, suggesting that the inhibitor was successfully purified into a single protein.

Abstract

One of pharmacy’s duty is being the place of pharmacist to serve patient profesionally. The fact, it was many report in mass media that pharmacist ‘s performance is not good enough. Many pharmacy regulation already issued, this review report and discuss pharmacy regulations related to pharmacy. Regulation that have issued by goverment about pharmacy ie: St. No.419 in 1949, Goverment regulations (PP) No.25 in 1980, Rule of Minister of Health (Permenkes) No.26 in 1981, Kepmenkes No. 278, 279 and 280 in 1981, Permenkes No.240 in 1990, Kepmenkes No.347 in 1990, Permenkes No.922 in 1993, Act (UU) No.5 in 1997, UU No.22 in 1997, PP No.72 in 1998, UU No.8 in 1999, Kepmenkes No.1332 in 2002, UU No.29 in 2004, Kepmenkes No.1027 in 2004, and Permenkes No.384 in 2007. Pharmacy regulations are enouh and relevance to support pharmacist practice, commitment to adhere the regulations and enforcement its implementation still required.

Abstract

Lipid metabolism disorder is one of common condition among HIV (Human Immunodeficiency Virus)/AIDS (Acquired Immune Deficiency Syndrome) patients who used first line antiretroviral efavirenz and nevirapine. This study was conducted to compare LDL (Low Density Lipoprotein) cholesterol between two group HIV/AIDS patients with efavirenz and nevirapine. The prospective study was performed in 2 months. HIV/AIDS patients who used first line antiretroviral at least for 6 months, aged ≥18 years old, not pregnant, without Mycobacterium ovium complex opportunistic infection and antihyperlipidemic, corticosteroid, retinoid in last 3 months were included. The 70 patients who enrolled in this study were divided into two groups. The independent T test was carried out to analyze the different of LDL cholesterol between two group. The group of patients using efavirenz showed 142.31 ±41,461 mg/dL and the group of nevirapine 126.83 ±40,55 mg/dL LDL cholesterol. This study showed that patient with efavirenze had higher LDL than nevirapine, but not statistically different.

Abstract

Silver nanoparticles have been studied for its application in wound healing therapy, while aqueous extract of Plantago major L has been studied also for its antibacterial and wound healing activity. This study aimed to obtain silver nanoparticles (AgNPs) through a green synthesis pathway using aqueous extract of Plantago major L. The initial study was performed to optimize the three concentration points of Plantago major L. aqueous extract, namely 0.125%, 0.25% and 0.5% and three synthetic temperature points (60 °C, 70 °C, and 80 °C). The formation of AgNPs were confirmed using a UV-Vis spectrophotometer which shows a peak at a wavelength of 445 nm. The obtained silver nanoparticles were then characterized using Dynamic Light Scattering (DLS Nano), Scanning Electron Microscope (SEM) dan X-Ray Diffraction (XRD). The study showed that the extract concentration of 0.25% with an optimum temperature of 70 °C was the optimal combination to produce the best AgNPs, based on the organoleptic parameters (as the color of silver was produced), the highest yield with weight of 9.13 mg and particle size of 129.20 nm with Polydispersity Index (PDI) of 0.25. The morphology of AgNPs showed the spherical shape and the crystallization of AgNPs showed a diffractogram pattern similar to the silver standard using X-Ray Diffraction (XRD). Antibacterial activity of AgNPs was tested against three bacterial strains, namely Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa and was measured with diffusion method using paper disc. The antibacterial activity testing showed that AgNPS has better antibacterial activity than silver nitrate and is equivalent to its comparative antibiotics, gentamicin sulfate for gram-negative bacteria and Chloramphenicol for gram-positive bacteria.

Abstract

Nicotine, the active compound in cigarettes, was considered as the risk factor for type 2 diabetes mellitus (T2DM). In the human body, nicotine would be metabolized by the enzyme cytochrome P450 2A6 (CYP2A6). CYP2A6 was known to be highly polymorphic. The active form of this gene was CYP2A6 *1, while the CYP2A6 *4 and CYP2A *9 alleles were the inactive alleles. The presence of this inactive allele caused the decreased activity of CYP2A6 so that it would affect the level of nicotine in the blood and would eventually cause an increased blood sugar levels. This study aimed to determine the effect of CYP2A6 polymorphism on glycohemoglobine levels among Javanese smokers. The blood sugar levels were measured by hemoglobin A1c (HbA1c). In this study, 33 active smokers involved in the study were identified as slow metabolizers, by which 63.9% of all test participants had CYP2A6 *1/*4 genotype and as many as 6.1% of the test participants had the CYP2A6 *1/*4/*9. The HbA1c levels among the participants have been analyzed, 28 participants were in normal range (4.83-5.56%); 4 participants were identified in prediabetes condition (5.70% - 5.97%) and 1 participant was in diabetes with HbA1c level was 7.16%. This condition indicates that the presence of CYP2A6 *4 and *9 alleles will affect HbA1c levels which can eventually lead to T2DM disease.

Abstract

Nanofibers dressing exhibit several advantageous characteristics for accelerating wound healing, such as its similar structure to the extracellular matrix (ECM), high surface area/volume ratio, high porosity and high loading capacity of drug. The nanofibers dressing which were prepared by the electrospinning technique using combination of synthetic and natural polymer excipients capable of fulfilling the ideal wound dressing criteria. This study aimed to develop nanofibers dressing prepared from polyvinyl alcohol (PVA), Arabic gum (GA) and honey by electrospinning method. This study focused on the effect of electrospinning parameters and the morphology of electrospun nanofibers of the blended solutions made from PVA (9% w/v) – GA (1% w/v) and honey (at varied concentrations of 0;1;3% w/v) with or without Triton X-100 (0.05% v/v). The effect of varied process parameters such as voltage and flow rate in electrospinning was also investigated. The blended solutions with the various concentration of honey at 0;1;3% w/v and Triton X-100 were named as FAt, FBt, and FCt, respectively, while the blended solutions without Triton X-100, were named as FA, FB, and FC. The optimum electrospining parameter were 18 KV and 5 µl/minute for FAt, FBt, and FCt; and 20 KV and 10 µl/minute for FA, FB, and FC. Electrospun nanofibers of FAt, FBt, FCt showed smoother and more uniform fibers in comparison to the nanofibers FA, FB, and FC. The average nanofibers diameter of FAt, FBt, FCt were 244±45; 266±45; 283±57 nm, respectively, while the average nanofibers diameter of FA, FB, FC was 406±140, 457±168, 594±204 nm, respectively. Higher concentration of honey increased the diameters of nanofibers. The average nanofibers diameter of FAt, FBt, and FCt were within nanoscale in range of the ECM (50-500 nm), which were suitable for accelerating wound healing. Therefore, this study indicated that PVA-GA-honey nanofibers dressing is suitable to be further developed as carrier for growth factors

Abstract

Optimization of composition of Lactobacillus bulgaricus and Streptococcus thermophilus in yoghurt fortified by lakum fruit (Cayratia trifolia (L.) Domin) extract as an antibacterial against Escherichia coli was investigated for the first time in this research. Yoghurt was made by fermenting milk with several compositions of L. bulgaricus and S. thermophilus as starter cultures and also adding lakum fruit extract as an antibacterial against E. coli. The fermentation process was depended on the bacterial culture used. The aim of the research was to determine the optimal composition of L. Bulgaricus and S. thermophilus bacteria in lakum fruit yoghurt as an antibacterial against E. coli. The research was done by making variation of composition of L. bulgaricus and S. thermophilus bacteria culture in lakum fruit yoghurt. Lakum fruit yoghurt was analysed with acidity stability test, pH, anthocyanin level and inhibitory activity against E. coli. The data analysis was done with MANOVA (Multivariate Analysis of Variance). The results showed that lakum fruit yoghurt with composition of L. bulgaricus and S. thermophilus 2:1 (v/v) had a good stability with increased acidity dan decreased pH. The value of anthocyanin in lakum fruit yogurt decreased and plain yoghurt (without lakum fruit extract) with composition of L. bulgaricus and S. thermophilus 2:1 (v/v) gave the highest inhibitory value against E. coli.
