Abstract

This research aims to investigate the effect of protein extraction time using 90% ethanol on the protein concentrate product's physical, chemical, and organoleptic characteristics. The findings of this study could promote the use of protein concentrate developed from SAA. The study was conducted from April to June 2024, utilizing SAA collected from rice fields in the PALI (Penukal Abab Lematang Ilir) Regency, South Sumatra Province, Indonesia. A completely randomized design was employed, using different extraction times of proteins from SAA with 90% ethanol. Four extraction time levels were tested: 0 hours (P0 or control), 24 hours (P1), 28 hours (P2), and 32 hours (P3), each with three repetitions. Data were analyzed using ANOVA, followed by a post-hoc test employing Duncan's multiple range test, utilizing SPSS version 20.0. Results have shown that the optimal protein extraction time was P2 (28 hours), yielding a protein concentration of 51.6% on a dry basis. Additionally, protein profile analysis conducted via SDS-PAGE revealed the presence of histone proteins and an antibacterial protein derived from snail mucus in SAA. Treatment P2 exhibited five distinct protein bands with molecular weights of 17 kDa, 20 kDa, 25 kDa, 35 kDa, and over 48 kDa respectively. On the other hand, treatments P1 and P3 displayed only three protein bands that are 20 kDa, 35 kDa, and over 48 kDa. Results from the organoleptic analysis indicated that protein isolates with suitable attributes in terms of color and aroma were obtained from treatment P1 (24-hour extraction). This study has potential applications in the food industry.

Abstract

This study aimed to determine the influence of spiritual Mindfulness on increasing adolescent self-confidence. Evaluating the benefits of spiritual therapies such as mindfulness for the mental health of adolescents is highly needed. Adolescence is difficult to adjust to and is especially susceptible to declining self-confidence. Low self-confidence causes most adolescent crises in the current school system. Spiritual Mindfulness is an easy-to-use strategy that can help adolescents become aware of their problems without passing judgment, which reduces costs and saves time. The study design was a pre-experiment with a pre and post-test with the control group. In purposive sampling, the intervention and control students chose twenty-six respondents from senior high school students in Kudus. The research instrument used in this study was the Student Self-Confidence Questionnaire. The researcher conducted six 30-minute sessions of spiritual mindfulness therapy over two days. Research results showed the difference in self-confidence scores before and after the intervention in the intervention group was found statistically significant with a p-value of 0.013 (p < 0.05) and in the control group with a p-value of 0.000 (p < 0.05). The effect of spiritual mindfulness therapy on self-confidence was not statistically significant, with a p-value of 0.110 (p> 0.05). So, it can be concluded that Spiritual mindfulness therapy has no significant influence on self-confidence. The way spiritual mindfulness therapy is given to teenagers needs to be altered to see more noticeable improvements in self-confidence.

Abstract

This study aimed to determine the phytochemicals and antimicrobial activities of Psidium guajava leaves. Psidium guajava (guava) belonging to the Myrtaceae family is an evergreen tree cultivated for its precious fruits and high biological properties. This research work was done to screen the phytochemical present and determine the antimicrobial properties of Psidium guajava leaf fractions. Ethanol solvent was used to extract the sample (using the cold maceration method), and the crude extract was further fractionated using n-hexane, acetone, and methanol solvents respectively. The screening of the phytochemicals (to test for alkaloids, tannins, steroids, flavonoids, polyphenols, terpenoids, and saponins) was done using the standard procedure, and evaluation of the antimicrobial activity of the plant was done using agar well diffusion method. The results showed that Psidium guajava leaf extracts contain all the phytochemicals tested, with an n-Hexane fraction containing Alkaloids, flavonoids, tannins polyphenols, and steroids. The acetone fraction reveals Alkaloids, tannins, terpenoids, polyphenols, and steroids, while the methanol fraction contains Alkaloids, flavonoids, terpenoids, tannins, and saponins. The results also showed Psidium guajava's promising antimicrobial properties, with the n-hexane fraction showing the greatest antimicrobial activity. This research has confirmed the widespread traditional medicinal applications, which is most prevalent among people living in northern Nigeria.
